    Notes: To help the achievement of integrated product and process development, there is a need for tools that can assist designers in creating manufacturable parts with less design routines and tryouts. This paper presents a systematic approach to developing automated manufacturability assessment tools by identifying the functional and informational requirements and proposing an assessment model. The work presented in this paper includes: (1) identification of characteristics and tasks of design for the die-casting process; (2) determination of functional and informational requirements for automatic manufacturability assessment; (3) formalization and modularization of assessment knowledge; and (4) modeling of product definition data to support the assessment. Object-oriented techniques are employed to model the assessment knowledge and manage the complicated and diverse types of product definition data by taking advantage of data abstraction, modularity, inherent concurrence, and the concept of encapsulation and extendibility.

    Notes: Abstract Hamiltonian systems with chaotic regions are particularly slippery to treat numerically. Numerical treatments can introduce nonphysical features. Simple examples illustrate some of the pitfalls. Integer, or discrete, arithmetic is a favorite “workaround.” While it does not cure chaos, it clarifies the interaction of computational methods with the underlying mathematical structure. Be forewarned: I won't give any prescription that is guaranteed to give a good and reliable method to handle chaotic problems numerically. Instead, I'll stress a few of the concerns and describe one or two pitfalls.

    Notes: Abstract A series of perfectly alternating copolymers containing oligomeric dimethylsilylene and cis or trans stilbene units has been prepared by the condensation polymerization of silyl ditriflates with the corresponding stilbene dianion. As expected, the spectral characteristics and the polymer processability were very different for the isomeric polymers. Evidence of σ–π interaction was obtained from absorption measurements and the excited state of the Si4 catenates was characteristic of a polar charge transfer state. The polymers are all photoactive and irradiation in the long-wavelength absorption band leads to cis–trans isomerization followed by complete disappearance of the long-wavelength (λ〉300 nm) absorption. The latter was not observed for the monomeric model compounds.

    Notes: Abstract  A full-length and several partial cDNAs encoding IGK light chains from the marsupial South American opossum, Monodelphis domestica, were isolated and characterized. Using these clones as a starting point, the expressed IGKV repertoire was sampled by anchored polymerase chain reaction using an IGKC-specific primer. Based on nucleotide sequences of twenty unique, expressed IGKV-J combinations, there are at least four IGKV families and two J segments. Southern blot analysis revealed each IGK-V family contains multiple gene segments totaling at least thirty-five IGKV in the opossum genome. No evidence for particular, recurrent IGKV-J combinations in the opossum IGK repertoire was seen, rather the V-J combinations appeared random and diverse. Each of the four IGKV families appear more closely related to V segments from placental mammals than to each other, suggesting the duplication of the IGKV families prior to the separation of marsupials and placental mammals more than one-hundred-million years ago. Overall, the complexity of opossum light chain V segments appears greater than that found in the heavy chain, and light chains are likely to contribute significantly to Ig diversity in this species.With this report, the homologues encoding all three classes of eutherian Ig chains, IGH, IGL, and IGK, have been described in a non-placental mammal.

    Notes: Abstract The Solar Energetic Particle Ionic Charge Analyzer (SEPICA) is the main instrument on the Advanced Composition Explorer (ACE) to determine the ionic charge states of solar and interplanetary energetic particles in the energy range from ≈0.2 MeV nucl−1 to ≈5 MeV charge−1. The charge state of energetic ions contains key information to unravel source temperatures, acceleration, fractionation and transport processes for these particle populations. SEPICA will have the ability to resolve individual charge states and have a substantially larger geometric factor than its predecessor ULEZEQ on ISEE-1 and -3, on which SEPICA is based. To achieve these two requirements at the same time, SEPICA is composed of one high-charge resolution sensor section and two low- charge resolution, but large geometric factor sections. The charge resolution is achieved by the focusing of the incoming ions, through a multi-slit mechanical collimator, deflection in an electrostatic analyzer with a voltage up to 30 kV, and measurement of the impact position in the detector system. To determine the nuclear charge (element) and energy of the incoming ions, the combination of thin-window flow-through proportional counters with isobutane as counter gas and ion-implanted solid state detectors provide for 3 independent ΔE (energy loss) versus E (residual energy) telescopes. The multi-wire proportional counter simultaneously determines the energy loss ΔE and the impact position of the ions. Suppression of background from penetrating cosmic radiation is provided by an anti-coincidence system with a CsI scintillator and Si-photodiodes. The data are compressed and formatted in a data processing unit (S3DPU) that also handles the commanding and various automatted functions of the instrument. The S3DPU is shared with the Solar Wind Ion Charge Spectrometer (SWICS) and the Solar Wind Ion Mass Spectrometer (SWIMS) and thus provides the same services for three of the ACE instruments. It has evolved out of a long family of data processing units for particle spectrometers.

    Notes: Abstract. Difference constraints systems consisting of inequalities of the form x i - x j $ \leq $ b i,j occur in many applications, most notably those involving temporal reasoning. Often, it is necessary to maintain a solution to such a system as constraints are added, modified, and deleted. Existing algorithms handle modifications by solving the resulting system anew each time, which is inefficient. The best known algorithm to determine if a system of difference constraints is feasible (i.e., if it has a solution) and to compute a solution runs in Θ (mn) time, where n is the number of variables and m is the number of constraints. This paper presents a new efficient incremental algorithm for maintaining a solution to a system of difference constraints. As constraints are added, modified, or deleted, the algorithm determines if the new system is feasible and updates its solution. When the system becomes infeasible, the algorithm continues to process changes until it becomes feasible again, at which point a feasible solution will be produced. The algorithm processes the addition of a constraint in time O(m + n log n) and the removal of a constraint in constant time when the original system is feasible. More precisely, additions are processed in time O( || Δ || + |Δ| log|Δ| ) , where |Δ| is the number of variables whose values are changed to compute the new feasible solution, and || Δ || is the number of constraints involving the variables whose values are changed. When the original system is infeasible, the algorithm processes any change in O(m + n log n) amortized time. The new algorithm can also be used to check for the existence of negative cycles in dynamic graphs.

    Notes: Abstract A recA mutant was constructed of a soil isolate of Burkholderia cepacia, strain ATCC 17616. Prior to mutagenesis, the recA gene was cloned from this strain by its ability to complement the methyl methanesulfonate sensitivity of an Escherichia coli recA mutant. Sequence analysis of the strain showed high sequence similarity (94% nucleic acid and 99% amino acid identity) with the recA gene previously cloned from a clinical isolate of B. cepacia, strain JN25. The subcloned recA gene from B. cepacia ATCC 17616 restored UV resistance and recombination proficiency to recA mutants of E. coli and Pseudomonas aeruginosa, as well as restoring the ability of D3 prophages to be induced to lytic growth from a RecA− strain of P. aeruginosa. The recA mutant of B. cepacia ATCC 17616 was constructed by λ-mediated Tn5 mutagenesis of the cloned recA gene in E. coli, followed by replacement of the Tn5-interrupted gene for the wild-type allele in the chromosome of B. cepacia by marker exchange. The RecA− phenotype of the mutant was demonstrated by the loss of UV resistance as compared to the parental strain. Southern hybridization analysis of chromosomal DNA from the mutant indicated the presence of Tn5 in the recA gene, and the location of the Tn5 insertion in the recA allele was identified by nucleotide sequence analysis. A test using the recA mutant to see if acquired resistance to d-serine toxicity in B. cepacia might be a result of RecA-mediated activities proved negative; nevertheless, RecA activity potentially contributes to the overall genomic plasticity of B. cepacia and a recA mutant will be useful in bioengineering of this species.

    Notes: Abstract We fused the Pseudomonas aeruginosa recA promoter to a promoterless Vibrio fisherilux operon. This recA–lux fusion (pMOE15) was introduced into wild-type P. aeruginosa strain FRD1 and recA expression was monitored by measuring 490-nm light production. The RM4440 strain responded to increasing doses of ultraviolet radiation by an increase in its bioluminescence. RM4440 has the potential to be useful as a biosensor for the presence of DNA-damaging agents in the environment.

    Notes: Abstract A unique decomposition reaction of the novel squalene synthase inhibitors called zaragozic acids has been studied. Under very mild conditions, e.g. by merely exposing their solutions to air and visible light at ambient temperature, these compounds, characterized by the 2,8-dioxabicyclo[3.2.1]octane-4,6,7-trihydroxy-3,4,5-tricarboxylic acid core, rapidly decompose. As relatively stable intermediates in the cascade of decomposition, the biologically active 2,8-dioxabicyclo[3.2.1]octane-6,7-dihydroxy-4-keto-5-caroxylic acid (or 3,4-decarboxy-4-dehydro) derivatives of these compounds have been isolated in ca. 20% yield. Derivatization on the highly reactive 4-carbonyl group yields stable derivatives, several of which are potent inhibitors of squalene synthase. Further decomposition results in the elimination of C3 and C4 atoms and the carboxylic acid on C5, the oxidation of C5 to carboxylic acid and the liberation of the oxo group on C1. Specific results obtained with zaragozic acid A, a key representative of the family of these potent cholesterol-lowering agents, are presented in this study.
