    Beschreibung: The study of soil N cycling processes has been, is, and will be at the centre of attention in soil science research. The importance of N as a nutrient for all biota; the ever-increasing rates of its anthropogenic input in terrestrial (agro)ecosystems; its resultant losses to the environment; and the complexity of the biological, physical, and chemical factors that regulate N cycling processes all contribute to the necessity of further understanding, measuring, and altering the soil N cycle. Here, we review important insights with respect to the soil N cycle that have been made over the last decade, and present a personal view on the key challenges of future research. We identify three key challenges with respect to basic N cycling processes producing gaseous emissions: 1. quantifying the importance of nitrifier denitrification and its main controlling factors; 2. characterizing the greenhouse gas mitigation potential and microbiological basis for N2O consumption; 3. characterizing hotspots and hot moments of denitrification Furthermore, we identified a key challenge with respect to modelling: 1. disentangling gross N transformation rates using advanced 15N / 18O tracing models Finally, we propose four key challenges related to how ecological interactions control N cycling processes: 1. linking functional diversity of soil fauna to N cycling processes beyond mineralization; 2. determining the functional relationship between root traits and soil N cycling; 3. characterizing the control that different types of mycorrhizal symbioses exert on N cycling; 4. quantifying the contribution of non-symbiotic pathways to total N fixation fluxes in natural systems We postulate that addressing these challenges will constitute a comprehensive research agenda with respect to the N cycle for the next decade. Such an agenda would help us to meet future challenges on food and energy security, biodiversity conservation, water and air quality, and climate stability.

    Beschreibung: In the present work, an advanced tsunami generation, propagation and coastal inundation 2-DH model (i.e. 2-D Horizontal model) based on the higher-order Boussinesq equations – developed by the authors – is applied to simulate representative earthquake-induced tsunami scenarios in the Eastern Mediterranean. Two areas of interest were selected after evaluating tsunamigenic zones and possible sources in the region: one at the southwest of the island of Crete in Greece and one at the east of the island of Sicily in Italy. Model results are presented in the form of extreme water elevation maps, sequences of snapshots of water elevation during the propagation of the tsunamis, and inundation maps of the studied low-lying coastal areas. This work marks one of the first successful applications of a fully nonlinear model for the 2-DH simulation of tsunami-induced coastal inundation; acquired results are indicative of the model's capabilities, as well of how areas in the Eastern Mediterranean would be affected by eventual larger events.

    Beschreibung: Breaking wave energy in the surf zone is modelled through the incorporation of the time dependent energy balance equation in a non linear dispersive wave propagation model. The energy equations solved simultaneously with the momentum and continuity equation. Turbulence effects and the non uniform horizontal velocity distribution due to breaking is introduced in both the energy and momentum equations. The dissipation term is a function of the velocity defect derived from a turbulent analysis. The resulting system predicts both wave characteristics (surface elevation and velocity) and the energy distribution inside surf zone. The model is validated against experimental data and analytical expressions.

    Beschreibung: Continuously monitoring cell cultures is essential for both controlling critical parameters and improving understanding of key processes. An ideal technique in this context is surface plasmon resonance (SPR) spectroscopy, which essentially exploits changes in the angle of incident light that occur when molecules bind to a surface. It provides the ability to monitor real-time changes in small concentrations of various molecules, with no need for additional labels or sample preparation. Here we present an SPR-based immunoassay for monitoring concentrations of human serum albumin (HSA), and compare its sensitivity when used in conjunction with a Biacore platform and the cheaper, smaller liSPR system. In conjunction with either system, the immunoassay can detect HSA (a hepatocyte viability marker) at concentrations typically present in three-dimensional hepatocyte cultures mimicking the liver used to evaluate effects of drug candidates before exposure to humans or animals. Furthermore, in conjunction with the liSPR system, it is sufficiently sensitive to measure the much lower HSA levels present in skin–hepatocyte co-cultures.

    Beschreibung: Die Neu-Untersuchung vom „Brörup-Hotel-Moor" und anderer interglazialer Fundstätten aus Jütland haben gezeigt, daß der eigentlichen letzten Interglazialzeit eine kalte Solifluktionsperiode folgte. Ein langes, temperiertes Interstadial mit Birken- und zuletzt auch Fichten- und Föhrenwäldern ging der Hauptphase der letzten Vereisung voraus. Das Interstadial wird dem Göttweiger Interstadial und die vorhergehende kalte Periode dem Altwürm gleichgesetzt. Während der Solifluktionsperiode fanden Umlagerungen von interglazialem Material statt.

    Beschreibung: A high-resolution parallel ocean model is set up to examine how the sill depth of the Atlantic connection affects circulation and water characteristics in the Mediterranean Basin. An analysis of the model performance, comparing model results with observations of the present-day Mediterranean, demonstrates its ability to reproduce observed water characteristics and circulation (including deep water formation). A series of experiments with different sill depths in the Atlantic–Mediterranean connection is used to assess the sensitivity of Mediterranean circulation and water characteristics to sill depth. Basin-averaged water salinity and, to a lesser degree, temperature rise when the sill depth is shallower and exchange with the Atlantic is lower. Lateral and interbasinal differences in the Mediterranean are, however, largely unchanged. The strength of the upper overturning cell in the western basin is proportional to the magnitude of the exchange with the Atlantic, and hence to sill depth. Overturning in the eastern basin and deep water formation in both basins, on the contrary, are little affected by the sill depth. The model results are used to interpret the sedimentary record of the Late Miocene preceding and during the Messinian Salinity Crisis. In the western basin, a correlation exists between sill depth and rate of refreshment of deep water. On the other hand, because sill depth has little effect on the overturning and deep water formation in the eastern basin, the model results do not support the notion that restriction of the Atlantic–Mediterranean connection may cause lower oxygenation of deep water in the eastern basin. However, this discrepancy may be due to simplifications in the surface forcing and the use of a bathymetry different from that in the Late Miocene. We also tentatively conclude that blocked outflow, as found in experiments with a sill depth ≤10 m, is a plausible scenario for the second stage of the Messinian Salinity Crisis during which halite was rapidly accumulated in the Mediterranean. With the model setup and experiments, a basis has been established for future work on the sensitivity of Mediterranean circulation to changes in (palaeo-)bathymetry and external forcings.

    Beschreibung: Interaction of biogenic volatile organic compounds (VOC) with anthropogenic VOC affects the physicochemical properties of secondary organic aerosol (SOA). We investigated cloud droplet activation (CCN activity), droplet growth kinetics, and hygroscopicity of mixed anthropogenic and biogenic SOA (ABSOA) compared to pure biogenic SOA (BSOA) and pure anthropogenic SOA (ASOA). Selected monoterpenes and aromatics were used as representative precursors of BSOA and ASOA, respectively. We found that BSOA, ASOA, and ABSOA had similar CCN activity despite the higher oxygen to carbon ratio (O/C) of ASOA compared to BSOA and ABSOA. For individual reaction systems, CCN activity increased with the degree of oxidation. Yet, when considering all different types of SOA together, the hygroscopicity parameter, κCCN, did not correlate with O/C. Droplet growth kinetics of BSOA, ASOA, and ABSOA was comparable to that of (NH4)2SO4, which indicates that there was no delay in the water uptake for these SOA in supersaturated conditions. In contrast to CCN activity, the hygroscopicity parameter from hygroscopic tandem differential mobility analyzer (HTDMA) measurement, κHTDMA, of ASOA was distinctively higher (0.09–0.10) than that of BSOA (0.03–0.06), which was attributed to the higher degree of oxidation of ASOA. The ASOA components in mixed ABSOA enhanced aerosol hygroscopicity. Changing the ASOA fraction by adding BVOC to ASOA or vice versa AVOC to BSOA changed the hygroscopicity of aerosol, in line with the change in the degree of oxidation of aerosol. However, the hygroscopicity of ABSOA cannot be described by a simple linear combination of pure BSOA and ASOA systems. This indicates that additional processes, possibly oligomerization, affected the hygroscopicity. Closure analysis of CCN and HTDMA data showed κHTDMA was lower than κCCN by 30–70 %. Better closure was achieved for ASOA compared to BSOA. This discrepancy can be attributed to several reasons. ASOA seemed to have higher solubility in subsaturated conditions and/or higher surface tension at the activation point than that of BSOA.

    Beschreibung: We measured a large suite of gas and particle phase multi-functional organic compounds with a Filter Inlet for Gases and AEROsols (FIGAERO) coupled to a high-resolution time-of-flight chemical ionization mass spectrometer (HR-ToF-CIMS) developed at the University of Washington. The instrument was deployed on environmental simulation chambers to study monoterpene oxidation as a secondary organic aerosol (SOA) source. We focus here on results from experiments utilizing an ionization method most selective towards acids (acetate negative ion proton transfer), but our conclusions are based on more general physical and chemical properties of the SOA. Hundreds of compounds were observed in both gas and particle phases, the latter being detected upon temperature programmed thermal desorption of collected particles. Particulate organic compounds detected by the FIGAERO HR-ToF-CIMS are highly correlated with, and explain at least 25–50% of, the organic aerosol mass measured by an Aerodyne Aerosol Mass Spectrometer (AMS). Reproducible multi-modal structures in the thermograms for individual compounds of a given elemental composition reveal a significant SOA mass contribution from large molecular weight organics and/or oligomers (i.e. multi-phase accretion reaction products). Approximately 50% of the HR-ToF-CIMS particle phase mass is associated with compounds having effective vapor pressures 4 or more orders of magnitude lower than commonly measured monoterpene oxidation products. The relative importance of these accretion-type and other extremely low volatility products appears to vary with photochemical conditions. We present a desorption temperature based framework for apportionment of thermogram signals into volatility bins. The volatility-based apportionment greatly improves agreement between measured and modeled gas–particle partitioning for select major and minor components of the SOA, consistent with thermal decomposition during desorption causing the conversion of lower volatility components into the detected higher volatility compounds.

    Beschreibung: Impacts of soil moisture on de novo monoterpene (MT) emissions from Holm oak, European beech, Scots pine, and Norway spruce were studied in laboratory experiments. The volumetric water content of the soil, Θ, was used as the reference quantity to parameterize the dependency of MT emissions on soil moisture and to characterize the severity of the drought. When Θ dropped from 0.4 m3 × m−3 to ~0.2 m3 × m−3 slight increases of de novo MT emissions were observed but with further progressing drought the emissions decreased to almost zero. In most cases the increases of MT emissions observed under conditions of mild drought were explainable by increases of leaf temperature due to lowered transpirational cooling. When Θ fell below certain thresholds, MT emissions decreased simultaneously with Θ and the relationship between Θ and MT emissions was approximately linear. The thresholds of Θ (0.044–0.19 m3 × m−3) were determined, as well as other parameters required to describe the soil moisture dependence of de novo MT emissions for application in the Model of Emissions of Gases and Aerosols from Nature, MEGAN. A factorial approach was found appropriate to describe the impacts of Θ, temperature, and light. Temperature and Θ influenced the emissions largely independently from each other, and, in a similar manner, light intensity and Θ acted independently on de novo MT emissions. The use of Θ as the reference quantity in a factorial approach was tenable in predicting constitutive de novo MT emissions when Θ changed on a time scale of days. Empirical parameterization with Θ as a reference was only unsuccessful when soil moisture changed rapidly

    Beschreibung: The three-dimensional quantification of small-scale processes in the upper troposphere and lower stratosphere is one of the challenges of current atmospheric research and requires the development of new measurement strategies. This work presents the first results from the newly developed Gimballed Limb Observer for Radiance Imaging of the Atmosphere (GLORIA) obtained during the ESSenCe (ESa Sounder Campaign) and TACTS/ESMVal (TACTS: Transport and composition in the upper troposphere/lowermost stratosphere, ESMVal: Earth System Model Validation) aircraft campaigns. The focus of this work is on the so-called dynamics-mode data characterized by a medium-spectral and a very-high-spatial resolution. The retrieval strategy for the derivation of two- and three-dimensional constituent fields in the upper troposphere and lower stratosphere is presented. Uncertainties of the main retrieval targets (temperature, O3, HNO3, and CFC-12) and their spatial resolution are discussed. During ESSenCe, high-resolution two-dimensional cross-sections have been obtained. Comparisons to collocated remote-sensing and in situ data indicate a good agreement between the data sets. During TACTS/ESMVal, a tomographic flight pattern to sense an intrusion of stratospheric air deep into the troposphere was performed. It was possible to reconstruct this filament at an unprecedented spatial resolution of better than 500 m vertically and 20 × 20 km horizontally.
