    Notes: Abstract. A four-step procedure was used to identify an aquatic macroinvertebrate with which to monitor organic contaminant burdens in trophically simple lake food webs of Alberta, Canada. We identified leeches (Oligochaeta: Hirudinea) as the potential indicator assemblage (Step 1), and then documented their abundance and distribution in 16 lakes to identify a species-level bioindicator (Step 2). The latter two steps involved identifying the number and level of organic contaminants within the indicator species (Step 3) and a comparison with other taxa within the food web (Step 4).  The majority of lakes within the watershed contained seven to nine leech species with individuals of Nephelopsis obscura being one of the most abundant and widely distributed species that is sufficiently large to allow for rapid collection of sufficient biomass for organic contaminant analyses. Concentrations of organic contaminants in leeches from 22 sites and in sediments (two lakes) within the watershed were low, and in the majority of cases less than analytical detection levels (i.e., 0.05 μg/kg wet weight). Nevertheless, leech tissues contained trace amounts of a fungicide (i.e., hexachlorobenzene) and four pesticides including: pp′-DDE, pp′-DDD, representing intermediate and final breakdown products of pp′-DDT, a-HCH (i.e., hexachlorocyclohexane) and g-HCH (Lindane). Concentrations of these contaminants did not differ significantly between leeches collected from lakes within a national park compared to areas outside the park that may have been more vulnerable to loadings of contaminants from industrial and agricultural sources.  Lastly, we compared organic contaminant concentrations in N. obscura with Gammarus lacustris and Brook's stickleback, Culaea inconstans (Gasterosteidae). Organic contaminant concentrations in N. obscura were similar or higher than those present in G. lacustris and C. inconstans. Eight organic contaminants were found in N. obscura, whereas, C. inconstans and G. lacustris contained only three and four, respectively. The fact that N. obscura typically contained higher concentrations and greater numbers of detectable compounds suggest that it is a suitable indicator of organic contamination. While current levels of organic contaminants in the Beaver Hills watershed are low, despite the intense agriculture and industrial activities, they should be monitored to act as an early warning indicator of ecosystem degradation.

    Notes: Abstract The glyoxylate cycle, catalysed by two unique enzymes: isocitrate lyase (ICL; EC 4.1.3.1) and malate synthase (MS; EC 4.1.3.2), is necessary for the net conversion of acetate into glucose. This metabolic pathway operates in microorganisms, higher plants and nematodes. Two bacterial genes, encoding ICL and MS, were modified in order to introduce them into the mouse germ line. The ovine metallothionein-Ia (MT-Ia) promoter-aceB gene-ovine growth hormone (GH) gene (3′ GH sequence) construct was fused to the ovine MT-Ia promoter-aceA gene-ovine GH gene (3′ GH sequence). Therefore, in this single DNA sequence, bothaceA andaceB are under independent MT-Ia promoter control and can be induced by zinc. Transgenic mice were generated by pronuclear microinjection of theaceB-aceA gene construct. We now report the establishment of four mouse lines carying these two transgenes. Studies on the progeny of these lines indicate that one line (No. 91) is expressing both genes at the mRNA and enzyme levels in the liver and intestine, whereas another line (No. 66) has a much lower expression. Both enzyme activities were detected in the liver and intestine at levels up to 25% of those measured in fully derepressedEscherichia coli cells.

We prove a number of cognate results and extensions. For instance, we show that Beurling's result remains true if the intervals inI are not just one-dimensional, but if they in fact constitute polar rectangles centred on the negative real axis and having some fixed constant angular width. In doing this, we obtain a new proof of Beurling's result. We also discuss a conjecture of Matheson and Pruss [25] and some other open problems.

    Notes: The recent surge of discoveries concerning the structural organization of nucleosomes, together with genetic evidence of highly specialized roles for the histones in gene regulation, have brought a renewed need for a detailed understanding of nucleosomal anatomy. Here we review recent structural advances leading to a new level of understanding of the nucleosome and chromatin fibre structure. We discuss the problems and challenges for existing models of chromatin structure and, in particular, consider how linker histones may bind within the nucleosome, together with the implications of their association for the structure of the chromatin fibre.

    Description: Histone-DNA contacts within a nucleosome influence the function of trans-acting factors and the molecular machines required to activate the transcription process. The internal architecture of a positioned nucleosome has now been probed with the use of photoactivatable cross-linking reagents to determine the placement of histones along the DNA molecule. A model for the nucleosome is proposed in which the winged-helix domain of the linker histone is asymmetrically located inside the gyres of DNA that also wrap around the core histones.
