    Description / Table of Contents: Summary The necessary chemical preparation and accumulation preliminary to the detection of minute quantities of thallium in toxicological analysis are discussed together with their quantitative determination by chemical and spectral analytical methods. The importance of the application of emission spectral analysis for the unequivocal detection of metallic poisons is shown by means of four cases taken from practice, and it is also pointed out that this procedure yields quantitative results even in cases where purely chemical methods no longer suffice. It has been found that examination of the muscular tissue is of the highest importance to the detection of thallium poisonings, since thallium is stored there for long periods. In this way it was possible, for the first time, to detect thallium after the victim had survived small amounts of the poison, and also to prove the presence of this element in the remains of a corpse that had lain buried for 7 years after the fatal dose had been administered. In both cases, the amount of poison was established quantitatively.
    Abstract: Résumé On expose le traitement chimique requis et l'enrichissement pour la recherche de très petites quantités de thallium, en analyse toxicologique et leur détermination quantitative par voie chimique et spectrographique. L'importance de la spectrographie d'émission pour la recherche sûre des poisons métalliques est démontrée dans 4 cas pratiques et l'on montre qu'avec de tels procédés de recherche, on peut encore obtenir des résultats quantitatifs dans des cas où normalement les méthodes chimiques pures ne peuvent plus accéder. Pour la recherche des empoisonnements par le thallium, on a montré que l'examen du tissu musculaire est de la plus grande importance car le métal y est mis en réserve pendant longtemps. De cette manière, on a pu mettre en évidence, pour la première fois, un empoisonnement avec survie par de petites quantités de thallium, ainsi qu'un empoisonnement fatal, en se servant des restes d'un cadavre enterré depuis 7 ans, et, dans les deux cas, déterminer la quantité de poison.
    Notes: Zusammenfassung Es wird die erforderliche chemische Aufbereitung und Anreicherung zum Nachweis kleinster Thalliummengen in der toxikologischen Analyse und deren qualitative und quantitative Erfassung auf chemischem und spektralanalytischem Wege dargelegt. Die Wichtigkeit der Anwendung der Emissionsspektralanalyse für den eindeutigen Nachweis von Metallgiften wird an Hand von 4 Fällen der Praxis aufgezeigt und darauf hingewiesen, daß mit diesem Untersuchungsverfahren noch quantitative Ergebnisse in solchen Fällen erzielt werden können, wo naturgemäß rein chemische Methoden nicht mehr ausreichen. Es hat sich gezeigt, daß für den Nachweis von Thalliumvergiftungen die Untersuchung der Muskelgewebe von größter Wichtigkeit ist, da in diesen Thallium für lange Zeit gespeichert wird. Wir konnten auf diese Weise erstmalig eine überlebte Vergiftung mit kleinen Thalliummengen bzw. eine tödliche Vergiftung an den Resten einer vor 7 Jahren beerdigten Leiche nachweisen und in beiden Fällen die Giftmenge quantitativ erfassen.

    Description / Table of Contents: Summary A description is given of a method for the polarographic determination of k-strophantine and strophantidine, which at −2.3 volts give reduction stages in alcohol solutions of quaternary ammonium salts. Under given conditions, the detection limit for k-strophantine is 0.4 mg/ml, for strophantidine 0.2 mg/ml.
    Abstract: Résumé On décrit une méthode pour le dosage polarographique de la strophantine-K et de la strophantidine qui donnent, en solution alcoolique de leurs sels d'ammonium quaternaires, des paliers de réduction au voisinage de −2,3 V. Dans les conditions données, la limite pour la recherche de la strophantine-K est de 0,4 mg/ml et celle pour la strophantidine est de 0,2 mg/ml.
    Notes: Zusammenfassung Es wird eine Methode zur polarographischen Bestimmung des k-Strophanthins und des Strophanthidins beschrieben, die in alkoholischen Lösungen von quartären Ammoniumsalzen Reduktionsstufen bei−2,3 V bezogen auf die n-KE. geben. Unter gegebenen Bedingungen beträgt die Nachweisgrenze für k-Strophanthin 0,4 mg/ml, für Strophanthidin 0,2 mg/ml.

    Notes: Abstract d-Ribulose 1,5-diphosphate carboxylase has been purified from autotrophically grown cells of the facultative chemolithotrophic hydrogen bacteriumAlcaligenes eutrophus. The enzyme was homogeneous by the criteria of polyacrylamide gel electrophoresis. The molecular weight of the enzyme was 505000 determined by gel filtration and sucrose density gradient centrifugation, and a sedimentation coefficient of 18.2 S was obtained. It was demonstrated by sodium dodecyl sulphate-polyacrylamide gel electrophoresis that the enzyme consists of two types of subunits of molecular weight 52000 and 13000. Electron microscopy on the intact and the partially dissociated enzyme lead to the construction of a model for the quaternary structure of the enzyme which is composed of 8 large and 8 small subunits. The most probable symmetry of the enzyme molecule is 4:2:2. Michaelis constant (K m ) values for ribulose 1,5-diphosphate, Mg2-, and CO2 were 0.59 mM, 0.33 mM, and 0.066 mM measured under air. Oxygen was a competitive inhibitor with respect to CO2 suggesting that the enzyme also exhibits an oxygenase activity. The oxygenolytic cleavage of ribulose 1,5-diphosphate was shown and a 1:1 stoichiometry between oxygen consumption and 3-phosphoglycerate formation observed.

    Notes: Abstract A new genus of marine methanogenic bacteria and two species within this genus are described.Methanogenium is the proposed genus andMethanogenium cariaci the type species. Cells of the type species are Gram-negative, peritrichously flagellated, irregular cocci with a periodic wall surface pattern. Colonies formed by these bacteria are yellow, circular and umbonate with entire edges. The DNA base composition is 52 mol% guanine plus cytosine. Formate or hydrogen and carbon dioxide serve as substrates for growth. Cells ofMethanogenium marisnigri are of similar shape but smaller diameter thanM. cariaci. The colonies ofM. marisnigri are convex, and the DNA base composition is 61 mol % G+C. Formate or hydrogen and carbon dioxide are growth substrates. Sodium chloride is required for growth of both methanogens.

    Notes: Abstract The coryneform hydrogen bacterium strain GZ 29, assigned to Corynebacterium autotrophicum fixed molecular nitrogen under autotrophic (H2, CO2) as well as under heterotrophic (sucrose) conditions. Physiological parameters of nitrogen fixation were measured under heterotrophic conditions. The optimal dissolved oxygen concentration for cells grown in a fermenter with N2 was rather low (0.14 mg O2/l) compared with cells grown in the presence of NH 4 + (4.45 mg O2/l). C. autotrophicum GZ 29 had a doubling time of 3.7 h at 30°C with N2 as N-source and sucrose as carbon source and at optimal pO2. Acetylene reduction reached values of 12 nmoles of ethylene produced/minxmg protein. Although the oxygen concentration in the growing culture was kept constant, the optimal dissolved oxygen tension for the acetylene reduction assay shifted to higher pO2-values. The overall efficiency of nitrogen fixation amounted to 22 mg N fixed/g sucrose consumed; it reached a maximal value of 65 mg N fixed/g sucrose consumed at the beginning of the exponential growth phase. Intact cells reduced acetylene even under anaerobic test conditions; further anaerobic metabolic activity could not be ascertained so far.

    Notes: Abstract The cell morphology, the arrangement and fine structure of flagella and the piliation of the following Gram-negative aerobic hydrogen bacteria have been studied: Alcaligenes eutrophus, Alcaligenes paradoxus, Alcaligenes ruhlandii, Pseudomonas flava, Pseudomonas pseudoflava, Pseudomonas palleronii, Pseudomonas facilis, Aquaspirillum autotrophicum, Paracoccus denitrificans, Corynebacterium autotrophicum, and strains MA 2 and SA 35. The identity of the bacteria was examined by their substrate spectra and type of flagellation. Three types of flagellar fine structure were differentiated. The presence of pili was noted in strains of Alcaligenes paradoxus, Pseudomonas flava, P. pseudoflava, P. palleronii, and P. facilis.

    Notes: Abstract The alcohol-acetaldehyde dehydrogenase complex of Clostridium kluyveri has been separated from contaminating β-hydroxybutyryl-CoA dehydrogenase by repeated precipitation with manganese and ammonium sulfate. Mn++ was required for maximum alcohol dehydrogenase activity. The molecular weight of the enzyme complex was 194,000 as determined by sucrose density gradient centrifugation. The enzyme complex has been shown to contain two types of subunits with molecular weights of 55,000±2,600 and 42,000±1,200, respectively which are arranged in “H”-shaped particles. In solutions with an ionic strength above 25 mM the enzyme complex precipitated in the form of lumps as has been shown with specific ferritin-conjugated antibodies. These lumps are assumed to be aggregated polygonal bodies present in C. kluyveri.
