Description / Table of Contents: Abstract Marine sediment location spread over a wide range of environments contain evidence of anoxic intervals dated early Late Cretaceous. In the north and south deep Atlantic deposition of anoxic layers followed a widespread sedimentary hiatus of Middle Cenomanian age. In the eastern-central parts of the Atlantic (Angola and Cape Verde basins) rythmic deposition of anoxic layers commenced in the Latermost Cenomanian and lasted as late as the Santonian. In the rest of the deep north Atlantic on the contrary anoxia duration was probably limited to the Cenomanian-Turonian boundary or thereabout. Late Cretaceous anoxic layer is not yet recorded south of the Walvis-Rio Grande Rises in the South Atlantic. In a number of locations in marginal and epicontinental seas in America, Africa and Europe previously related to the Tethys a few anoxic layers dated Cenomanian-Turonian are intercalated within thick oxic series. Coniacian to Santonian anoxic layers are limited to small areas in epeiric and marginal domains in the southern Alps, North Africa, Israel, North America (Santonian in the Western Interior) and (?) Australia all with no clear link with any ocean-wide event of early Senonian age. Rythmical alternation of oxic/anoxic layers helps to document the emplacement of sediment and disposition of anaerobic water masses. Development of anoxia at about the Cenomanian-Turonian boundary induced abnormal concentration of trace metals and a partial renewal of the marine fauna. To understand the relationship between a number of factors such as sea-level changes, climates, terrestrial influences, biogenic fertility, sedimentation rates, etc., is essential to explain of the very peculiar anoxic global event of early Late Cretaceous age.

Notes: Abstract The edible parts of several species of fish and other marine organisms inhabiting the Calcasieu River/Lake, Louisiana were analyzed for Cd, Cr, Cu, Pb, Hg, Ag, Zn, and As. Concentrations of all metals measured in both intra and interspecies showed no significant variation with sample location. Differences in elemental concentrations were related to organism mobility and not to site-specific variations in metal loadings. Different species of finfish contained only trace amounts of Cd (0.02-0.08), Ag (〈0.01–0.3), Pb (〈0.2–0.5), and As (〈0.1–0.3) mg kg−1. Concentrations of Cu and Cr were highly variable, in contrast to Zn, which was present in relatively constant amounts across all species (28 ± 7 mg kg−1). Sessile organisms such as oysters had the highest concentrations of heavy metals, including Cd. Periphyton and Zooplankton were the only groups that showed differences in metal concentrations with sampling location.

Notes: Summary Linkage data of the four gene markers Pi, Gm, GLO, and HLA exclude Gm and Pi gene assignment to the short arm of chromosome 6. The findings, however, do not disprove the following gene order: HLA-GLO-centromere-Gm-Pi.

Notes: Abstract The lymphoma cell line BJAB.B95.8.6 was gamma-irradiated to induce mutations of major histocompatibility complex (MHC) encoded genes. Cloned “wild-type” cells were phenotyped HLA-A1, A2, B 13, 1335, Bw4, Bw6, Cw4, DR5, DRw52, DQwl, DQw3, DPw2, DPw4, GLO1*1, PGM3*2-1, and ME1*0 and possessed two apparently normal chromosome 6s prior to mutagenesis. Loss mutants were selected 5 days after 3 Gy gamma-irradiation employing three complement-fixing monoclonal antibodies specific for HLA-A2 (TÜ101) and Bw4 (TÜ48, TÜ109). Fifteen independently arising mutants were isolated and cloned. Typing with monospecific alloantisera and cell-mediated lympholysis revealed the presence of HLA-A1, 835, Bw6, Cw4, DR5. DRw52, DQw3, and DPw4 specificities on all mutant clones. HLA-A2, B13, and Bw4 were absent. Mutants differed in their expression of class 11 antigens. One group retained DQw1 and DPw2, another was DQw1−, DPw2+, and a third was DQw1−, DPw2−. Karyotyping of the “wild-type” line and selected mutant clones showed that the loss of HLA specificities correlated with deletions which map the HLA-A and -B loci directly to the distal part of the 6p2l.33 region and the class II genes to the region 6p21.33 (proximal) to 6p21.31 (distal) on the short arm of chromosome 6.

Notes: Abstract Gene cloning and sequencing of theHLA-B locus split antigens B38 (B16.1) and B39 (B16.2) allowed localization of their subtypic as well as their public specificities HLA-Bw4 or-Bw6 to the α-helical region of the α 1 domain flanked by the amino acid positions 74–83. Comparison of their amino acid sequences with those of otherHLA-B-locus alleles established HLA-Bw6 to be distinguished by Ser at residue 77 and Asn at residue 80. In contrast, HLA-Bw4 is characterized by at least seven different patterns of amino acid exchanges at positions 77 and 80–83. Reactivity patterns of Bw4-or Bw6-specific monoclonal antibodies reveal two alloantigenic epitopes contributing to the HLA-Bw4 or-Bw6 specificity residing next to the region of highest diversity of the α 1 domain.

Notes: Summary Interspersed simple repetitive DNA is a convenient genetic marker for analysis of restriction fragment length polymorphisms (RFLPs) because of the numbers and the frequencies of its alleles. Oligonucleotide probes specific for variations of the GA C T A simple repeats have been designed and hybridized to a panel of human DNAs digested with various restriction enzymes. Numerous RFLPs were demonstrated in AluI and MboI digested DNA with “pure” GATA oligonucleotides as probes. The optimal length of the probe for RFLP analysis was 20 bases taking into account fragment lengths (1.5-7 kilobases = kb), signal to background ratio, and number of clearly evaluable RFLPs. By using different restriction enzymes individual-specific hybridization patterns (“DNA fingerprints”) can be established. Hypervariable simple repeat fragments are stably inherited in a Mendelian fashion. Advantages of this method are discussed.

Notes: Summary A novel procedure is presented to estimate the ratio of male to female mutation rates for Duchenne muscular dystrophy (DMD). X-specific restriction fragment length polymorphisms are used to establish DNA haplotypes in three-generation DMD families. From the proportion of DMD patients who have inherited their maternal grandfather's X chromosome, the ratio of mutation rates can be calculated. In contrast to classical methods, the proposed procedure is not restricted to sporadic or familiar cases nor is any information on the carrier status of female relatives required.

Notes: Abstract Potassium hexacyanoferrate(II) trihydrate, K4Fe(CN)6·3H2O, was heated under controlled conditions of mass and rate in a derivatograph in the presence of oxygen. The heating was stopped at different temperatures and Mössbauer spectra and X-ray diffractograms were taken on the quenched material at room temperature. The reaction pathway was studied in this way and the advantages and drawbacks of each of the techniques are described. At different stages of the thermal process we were able to show the presence of K4Fe(CN)6,α-Fe2O3, Fe3O4, Fe3C, Fe, FeO, KFeO2,Β-FeOOH, KOCN, K2CO3 and KCN.

Notes: Conclusions Although more laborious at first sight than commercial ELISA procedures, our assay system can compete with them even in terms of turn-around time: with simple microtitre equipment assay time is practically uninfluenced by the sample number. Costs per single assay are reduced to less than 0.15 DM. More important, an assay design is presented consisting of a “universal” solid phase/detection-system combination that merely requires (commercial) monoclonal mouse and unconjugated polyclonal antibodies to be completed to a specific test system. Competitive assays requiring a monoclonal antibody and conjugated marker analyte may be attractive as well.
