    Description: For the first time white spot disease (WSD) was reported in shrimp farms of khoozestan province, in southwest of IRAN in 2002. Then in 2005 the neighbor province, boushehr, was contaminated. In 2008 WSD outbreak reported in sistan-bloochestan province in southeast of Iran. In 2015 all of southern shrimp farms of country except Hormozgan, the middle southern province, which has remained free of WSD, are being contaminated. White Spot disease suspended shrimp culture in thousands hectares of shrimp farms. Considering that white spot disease has not been observed in Hormozgan province yet, the question is; to what extent environmental and management factors participated in preventing WSD outbreak or cause WSD outbreak. In this study (20102012), the effects of environmental factors and management, stressors that decrease immune system function of shrimp are discussed. In addition, the role of pathogen as the main factor of outbreak is discussed. The goal of this study is to define environmental parameters and management practices associates with outbreak of white spot disease in affected provinces and discover reasons of being Hormozgan province free of this disease. In this study the role of the local environmental factors and management practice stressors in susceptibility to WSD was determine. Both the effects of environmental factors in water of ponds including total ammonia, nitrogen, dissolved oxygen, pH, salinity, transparency, and temperature and management issues related to biosecurity are studied. There were overlaps on physical and chemical parameter values obtained in clear areas with contaminated areas .Results of the data analysis suggest that lack of association with WSD incidence was 7 times greater than WSD incidence despite of disease outbreak in sistan-bloochestan province, so other sources of white spot disease virus incidence was suspected in affected areas. Histopathological examinations and polymerase chain reaction (PCR) tests during project performance did not reveal white spot disease virus evidences in post larvae examined from khoozestan province stocked in farms but disease outbreak was happened in that farms , so we suspected to management practice include feed , pond preparation and carrier of disease. Recorded values of temperature and salinity in some months during inspection in Hormozgan province specified stressful condition that may lead to WSD outbreak, however the disease did not appear. Therefore the hypothesis that the water physical and chemical conditions are reasons to prevent disease outbreak in Hormozgan province is being rejected. The policy of Hormozgan’s fishery authorities, to replaced Fenneropenaeus indicus with specific pathogen free Litopenaeus vannamei, that is more resistant to some of diseases, before incidence of WSD in farms and to before being endemic in the Hormozgan province, made an advantage compare to affected southern provinces that introduced Litopenaeus vannamei after WSD prevalence to their farms. However it does not guarantee to maintain current trend of being Hormozgan province farms free of white spot disease. Therefore establishing the principals of biosecurity are strongly emphasized. Strategies taken by the proficient authorities in preparation of SPF shrimp broodstock can be the most important factor in preventing WSD. Regarding biosecurity principals purchased feed must be free of shrimp head powder. Construction the new shrimp farms should be as far as it could be away from contaminated areas.

    Notes: Using an adaptive strategy, Chinese hamster ovary (CHO) cell lines were developed that are capable of robust growth in serum-free suspension culture. These preadapted derivatives of the commonly used strain of CHO cells (CHO DUKX), termed PA-DUKX, were used for the introduction and stable expression of several heterologous human genes. A significant advantage of recombinant PA-DUKX cells was their ability to readily resume growth in serum-free suspension culture after transfection and amplification of heterologous genes. Expression of recombinant human proteins in PA-DUKX cells was quantitatively similar to that of lineages generated using conventional CHO DUKX cells. In addition, recombinant human proteins expressed by transfected PA-DUKX lineages were shown to be biochemically and structurally similar to those expressed in CHO DUKX cells, PA-DUKX host cell technology provides an opportunity for reducing the time and resources required to develop large-scale, suspension culture-based manufacturing processes employing serum-free medium. © 1996 John Wiley & Sons, Inc.

    Description: Upland stream systems have been extensively investigated in Europe, North America and Australasia and many of the central ideas concerning their function are based on these systems. One central paradigm, the river continuum concept is ultimately derived from those North American streams whose catchments remain forested with native vegetation. Streams of the tropics may or may not fit the model. They have been little studied. The Amani Nature Reserve in the East Usambara Mountains of north-eastern Tanzania offers an opportunity to bring these naturally forested systems to the attention of the ecological community. This article describes a comparison made between two lengths of the River Dodwe in this area. The work was carried out by a group of postgraduate students from eighteen European and African countries with advice from five staff members, as part of a course organised by the Tropical Biology Association. Rigorous efforts were made to standardise techniques, in a situation where equipment and laboratory facilities were very basic, through a management structure and deliberate allocation of work to specialists in each area.The article offers a summary of invertebrate communities found in the stream and its biomass. Crabs seem to be the key organism in both sections of the streams.

    Description: This study aimed to investigate the effects of 5- parameter dissolved oxygen, PH, nitrite, nitrate and temperature of the water on Streptococcosis incidence in two groups of fry and grower fish. Research was performed at west Mazandaran province -Tonkabon region in 12 rainbow trout selected farms. Research conducted in 3 farms with well source water, 8 farms with water source of the river of Dohezar and one farm with water source of the river of Azarood, during 12 consecutive months. From 1390.04.01 to 1391.04.01, once time each month, and in each time 10 fish randomly sampled, inlet water were sampled simultaneously Of 1350 sampled fish 607 fish with an average weight 22.04 gr, average length 12.59 cm were in fry category and 743 fish with an average weight 156.25 gr, average length 23.32 cm in were grower category. Streptococcosis observed only in grower category Of 72 fish with clinical signs of the disease, 14 numbers were positive Streptococcosis (19.44%) and 58 numbers were negative Streptococcosis (80.55%). Three fish from grower category has not any clinical signs of disease and seemed to be healthy but were positive Streptococcosis in examinations (0.22% of total fish sampled. Fish with clinical signs of the disease but negative Streptococcosis were of at least 55.1 gr and at most weight 417 gr respectively. The results showed that 47.07% cases of Streptococcosis happened at 15.6°C water temperature, 35.29% at 16.98°C and 17.64% cases happened at 18.04°C so that 100% of Streptococcosis cases was observed at the average temperature of 16.99°C. In addition, the survey results show that despite relatively high levels of nitrite in source water of farms from wells, nitrite does not effect on the disease incidence. It seems that an optimal level of dissolved oxygen in water is effective in reducing the effectiveness of nitrite in this disease. According to equation coefficients logit model is as follows: According to Logit model, it seems that for every degree change in temperature and pH of water, morbidity change will diminish 0.37 % and 1.74 % respectively. 80.56% of fish sampled that had the clinical symptoms, was ranging from an average weight of 5.1 gr (fry) and 417 gr (grower fish), that despite having clinical signs of the disease were negative Streptococcosis. Isolation of Staphylococcus bacteria as well as Gram-negative bacilli from fish with clinical symptoms similar to the symptoms of Streptococcosis, may indicate the involvement of other pathogens in fish clinical signs.

    Description: This project has been started as monitoring study since 2001 that in first phase has lasted for two years and because of its importance, it was decided that this study should be continued for two other years. In the early 1980s, an alien ctenophore Mnemiopsis leidyi already known as a gelatinous zooplankton was transported (likely via ballast waters) to the Black Sea from its native waters of the western Atlantic. The possibility of Mnemiopsis leidyi introduction into other sensitive, neighboring ecosystems, notably the Caspian Sea, had been mentioned during the GESAMP meeting in 1994. And, as expected, this ctenophore was reported to be present in the Caspian Sea by November 1999. In this report, spatial and temporal distribution of M. leidyi had been investigated in southern Caspian Sea (Iranian waters) in 6 transects consisted of Lisar, Anzali, Sefidroud, Nowshar, Babolsar and Amirabad at 24 stations during 2003 and 2004. As a whole, 818 samples had been analyzed included 236, 302, 136 and 144 belong to ctenophore, phytoplankton, zooplankton and benthos. Meantime 811 physicochemical parameters had been measured. Water temperature varied between 7.8 to 29.3 co. Transparencies also fluctuated from 0.20 to 10.5 m. Salinity in different regions was from 7.80 to 13.90 ppt and the average was 12.09 ppt. pH was between 7.2- 8.6. Demand oxygen (O2) has been measured very high in water surface (12.89 µg.l) and it decreased at minimum level in deep down to 4 µg.l. Nutrients measurements such as NO_2, NO_3 and NH_4 were 0.10-15.17 , 3.80- 133.40 and 0.36- 86.40 µg.l, respectively. Total nitrate and phosphate were measured between 337.00- 1635.00 and 14.90-103.40 µg.l, respectively. Comparison of M. leidyi distribution in the southern Caspian Sea shown that maximum abundance recorded in warm summer months (e.g. August) with 11627 ind.m^2 and its biomass maximum was in autumn months (September and October) about 446.11 gr.m^2 . Mean abundance and biomass of M. leidyi were 2504 ind.m^2 and 142.42 gr.m^2, respectively. Maximum size of the ctenophore recorded as 70 mm while the less than 10 mm length frequency consisted 92.2 per cent of total population. Spatial distribution of M. leidyi in different regions shown that Sefidroud not only had the maximum Mnemiopsis abundance but also consisted higher biomass in the southern Caspian Sea in 2003-4. At present study, 142 phytoplankton species were identified which belongs to Chrysophyta (67, 48), Chlorophyta (23, 16%), Cyanophyta (22, 15%), Pyrhophyta (21, 15%) and Euglenophyta (9, 6%). Mean phytoplankton abundance and biomass were 11137032 ind. m^3 and 40.26 mg. m^3, respectively. Chrysophyta (diatoms) consisted 42 and 47% of whole phytoplankton abundance and biomass. In the other hand, this group included 50 % of phytoplankton population. In zooplankton study, there were identified 21 species in which 10 species were only meroplankton (zoobenthic) and 11 species was holoplankton. These species belongs to Copepoda (4, 37%), Rotatoria (4, 36%), Cilliophora (2, 18%) and Cladocera (1, 9%). Monthly zooplankton abundance and biomass data shown that its population completely effected by Copepoda. Among copepoda, Acartia clausi was the main species duri ng 2003-4. As a whole, maximum abundance recorded in spring (March) and autumn (September) about 11497 and 11321 ind. m^3 while its maximum biomass occurred in summer (August) with 48.95 mg. m^3. Zooplankton minimum abundance and biomass were in wintertime about 3894 ind. m^3 and 10.16 mg. m^3. Benthic fauna in recent study consisted 27 species included Gammaride (8, 29%), Cumacea (7, 26%), Polychaeta (4, 15%), Bivalvia (3, 11%) and Oligochaeta, Balanidae and Xantidae each one (4%) species. Mean benthic abundance and biomass were 1215 ind. m^ 2 and 20.09 g. m^2, respectively. Polychaeta and Oligochaeta were established maximum frequency about 49 and 34% of total fauna while Bivalvia consisted 82.8% biomass overall.

    Description: In 1999 Mnemiopsis leidyi was introduced to the Caspian Sea from the Black Sea with ballast waters from the ships. The comprehensive study on probability of controlling Caspian Sea invasive Ctenophora planned after a remarkable of decreeing in Kilka fish catches stocks and fisheris community problems. This study focus on reproduction experiments of Beroe ovata as the best candidate for control of Mnemiopsis population size in the Caspian Sea that was performed in Turkey and Iran during 2002-2003. At 2002, 87 specimens of B. ovata, 10-50 mm transferred to Caspian sea ecology research center from Marmareh sea where acclimated with Caspian sea water gradually. At 2003, experiments were performed near to Black sea (at Sinop) with freshly collected Beroe ovata, 40-65 mm size in three salinity level treatment, the Black sea water 18 , Mixed water 15 and Caspian water 12 . 130 individuals of Beroe ovata were brought from Sinop (Turkey) to Iran during 2003. A number of Beroe specimens were sent to Guilan province for reproduction studies and another part were sent to Mazandaran province for both reproduction and mesocosem studies. For control we had 1 Beroe, length 30 mm in the Black Sea water that was alive during of study in Iran. The Jars were examined each day for ova and larvae and they were collected and put into glass container of Caspian water for hatching and developing survey, some of them were left without any handling for larvae developing. Also in another experiment the eggs collected from jars were placed in the same three treatments for studying of growth and survival. The results were unsuccessful on propagation experiments at 2002 since the spawning and hatching rates were very low (20 ova) and, none of the larvae developed into adults in Caspian Sea water. The spawning was more in Marmareh sea water with 138 ova where only 7 larvae was hatched. Results showed that Beroe specimens is able to survive and reproduce in Caspian water but was not as well as Black Sea also the Beroe larvae growth rate is low in the Caspian Sea water. Maximum fecundity of Beroe individual was 2212 and 235 ovae in Caspian Sea water in site Sinop and Iran respectively. Results showed 34-100% eggs in Caspian Sea water were destroyed and did not develop. In Iran we obtained only one larvae with 5 mm length, other larvae were at different stages of development but most of them were 1.2 2 mm. The results of mesocosm survey showed most of ova and larvae have been obtained from the tanks where individuals B. ovata were with Mnemiopsis. Fecundity of Beroe in the control with Black Sea water were between 17 to 1879 with average of 828 ± 112 ova. The poor results of B. ovata reproduction obtained in this survey in Caspian Sea may be due to transportation and acclimation stress and low salinity of Caspian Sea water.

    Notes: Deactivation data and fluorescence intensity changes were used to probe functional and structural stability of cutinase in reverse micelles. A fast deactivation of cutinase in anionic (AOT) reverse micelles occurs due to a reversible denaturation process. The deactivation and denaturation of cutinase is slower in small cationic (CTAB/1-hexanol) reverse micelles and does not occur when the size of the cationic reverse micellar water-pool is larger than cutinase. In both systems, activity loss and denaturation are coupled processes showing the same trend with time. Denaturation is probably caused by the interaction between the enzyme and the surfactant interface of the reversed micelle. When the size of the empty reversed micelle water-pool is smaller than cutinase (at W0 5, with W0 being the water:surfactant concentration ratio) a three-state model describes denaturation and deactivation with an intermediate conformational state existing on the path from native to denaturated cutinase. This intermediate was clearly detected by an increase in activity and shows only minor conformational changes relative to the native state. At W0 20, the size of the empty water-pool was larger than cutinase and the data was well described by a two-state model for both anionic and cationic reverse micelles. For AOT reverse micelles at W0 20, the intermediate state became a transient state and the deactivation and denaturation were described by a two-state model in which only native and denaturated cutinase were present. For CTAB/1-hexanol reverse micelles at W0 20, the native cutinase was in equilibrium with an intermediate state, which did not suffer denaturation. 1-Hexanol showed a stabilizing effect on cutinase in reverse micelles, contributing to the higher stabilities observed in the cationic CTAB/1-hexanol reverse micelles. © 1998 John Wiley & Sons, Inc.     Notes: The secretion rate of activated protein C (APC) by BHK cells was increased 35-fold by increasing the cDNA copy number per cell from 50 to 240. In this range, the relation between APC secretion and cDNA copy number was not linear and the rate of APC secretion per cDNA copy increased sevenfold. This apparent cooperative effect of multiple cDNA copies could be related to their integration in tandem. For cDNA copy numbers higher than 240, the APC secreation rate per cDNA and per cell decreased dramatically. The γ-carboxylation of glutamic acid residues, a posttranslational modification required for APC biological activity, was also investigated. The proportion of APC that was fully γ-carboxylated decreased as the secretion rate of APC increased. © 1995 John Wiley & Sons, Inc.

    Notes: Using a pure culture of Pseudomonas fluorescens as a model system nitrite inhibition of denitrification was studies. A mineral media with acetate and nitrate as sole electron donor and acceptor, respectively, was used. Results obtained in continuous stirred-tank reactors (CSTR) operated at pH values between 6.6 and 7.8 showed that growth inhibition depended only on the nitrite undissociated fraction concentration (nitrous acid). A mathematical model to describe this dependence is put forward. The maximum nitrous acid concentration compatible with cell growth and denitrification activity was found to be 66 μg N/L. Denitrification activity was partially associated with growth, as described by the Luedeking-Piret equation. However, when the freshly inoculated reactor was operated discontinuosly, nitrite accumulation caused growth uncoupling from denitrification activity. The authors suggest that these results can be interpreted considering that (a) nitrous acid acts as a proton uncoupler; and (b) cultures continuoulsy exposed to nitrous acid prevent the uncoupling effect but not the growth inhibition. Examination of the growth dependence on nitrite concentration at pH 7.0 showed that adapted cultures (grown on CSTR) are less sensitive to nitrous acid inhibition than the ones cultivated in batch. © 1995 John Wiley & Sons, Inc.

    Notes: Antisense technology is potentially a powerful means by which to selectively control gene expression. We have used antisense oligonucleotides to modulate the response of the hepatoma cell line, HepG2, to the inflammatory cytokine, IL-6, by inhibiting the expression of its multifunctional signal transducer, gp130. HepG2 cells respond to IL-6 by upregulating acute phase proteins, such as haptoglobin, by five- to tenfold. Gp130 is central to this response, as the upregulation of haptoglobin is almost completely blocked by the addition of high concentrations (∼100 μg/ml) of a monoclonal antibody to gp 130. Antisense oligodeoxynucleotides complementary to the mRNA encoding gp 130 inhibited the upregulation of haptoglobin by IL-6-stimulated HepG2 cells by about 50%. However, a nonsense sequence also inhibited haptoglobin secretion by about 20%. To improve the specificity and efficiency of action, we targeted the antisense oligonucleotides to HepG2 cells using a conjugate of asialoglycoprotein-poly-L-lysine. The targeted antisense reduced the binding of IL-6 to HepG2 cells, virtually eliminating high affinity binding. In addition, it inhibited haptoglobin upregulation by over 70%. Furthermore, the dose of targeted antisense required for biological effect was reduced by about an order of magnitude as compared with unconjugated antisense. These results demonstrate the potential of antisense oligonucleotides as a means to control the acute phase response as well as the need for a greater understanding of the mechanism and dynamics of antisense molecules as they are developed toward therapeutic application. © 1997 John Wiley & Sons, Inc.
