    Notes: To determine whether detection of fecal contamination on cantaloupes is possible using fluorescence imaging, hyperspectral images of cantaloupes artificially contaminated with a range of diluted bovine feces were acquired from 425 to 774 nm in responses to ultraviolet-A (320 to 400 nm) excitation. Evaluation of images at emission peak wavelengths indicated that 675 nm exhibited the greatest contrast between feces contaminated and untreated surface areas. Two-band ratios compared with the single-band images enhanced the contrast between the feces contaminated spots and untreated cantaloupe surfaces. The 595/655-nm, 655/520-nm, and 555/655-nm ratio images provided relatively high detection rates ranging from 79% to 96% across all feces dilutions. However, both single band and ratio methods showed a number of false positives caused by such features as scarred tissues on cantaloupes. Principal component analysis (PCA) was performed using the entire hyperspectral images data; 2nd and 5th principal component (PC) image exhibited differential responses between feces spots and false positives. The combined use of the 2 PC images demonstrated the detection of feces spots (for example, minimum level of 16-μg/mL dry fecal matter) with minimal false positives. Based on the PC weighing coefficients, the dominant wavelengths were 465, 487, 531, 607, 643, and 688 nm. This research demonstrated the potential of multispectral-based fluorescence imaging for online applications for detection of fecal contamination on cantaloupes.

    Notes: To verify the compatibility behaviour of the almond cultivar ‘Francolí’ and to clarify its S genotype a combination of pollination tests, stylar ribonuclease and allele specific PCR analysis was used. ‘Francolí’ was released from IRTA's breeding programme in 1994, having been putatively raised from the cross ‘Cristomorto’ (S1S2) × ‘Gabaix’ (S10S25). This cultivar was also reported to be self-incompatible but revealing only one S band in the zymograms after S-RNases analysis. ‘Francolí’ sets nuts after test crossing with two S1S25 cultivars, having a different genotype from that earlier reported. ‘Francolí’ was also observed to be self-compatible after selfing flowers in the field and in the laboratory. ‘Francolí’ was re-assigned the S1Sf genotype after test crossing, stylar ribonuclease and PCR data analysis. After microsatellite analysis, the self-compatible ‘Tuono’ (S1Sf) cultivar is suggested as the male parent of ‘Francolí’ instead of the earlier reported ‘Gabaix’.

    Notes: Three transgenic sugarcane populations produced by Agrobacterium tumefaciens infection were analysed for the presence of genomic variability. Plants of the original cultivar, plants regenerated without transformation, as well as transformed and untransformed calli were used as control treatments. Amplified fragment length polymorphism (AFLP) of DNA extracted from leaves or calli assessed genomic profiling. The average DNA polymorphism within each population was determined by calculating the polymorphism index, while the extent of genomic dissimilarity among individual plants within transgenic populations was verified in unweighted pair group method using arithmetic averages dendrograms. The results showed that the production of transgenic sugarcane plants by A. tumefaciens infection is accompanied by limited but detectable genomic changes and that, on average, these occur at the same rate in plant populations carrying different transgenes. Main factors contributing to somaclonal variation in transgenic sugarcane plants have been verified by pre-existing DNA polymorphism into the donor genotype and in vitro culture steps during the transformation procedure. The relevant practical conclusion from this finding is that the AFLP analysis may be effectively used to identify individual transgenic plants with the least genomic deviation from the parental ones. The selected genotype would be conserved as cultivated sugarcane is asexually propagated.

    Notes: Penaeid shrimps are among the most studied crustaceans, mainly regarding their immune system, and several proteins involved in the defense against pathogens have been described. Haemocytes are very dynamic cells responsible of recognition, phagocytosis, degranulation and nodule formation, but still little is known about their gene expression. Using differential display, we found modification in haemocyte gene expression after inoculation with an abiotic particle (DEAE-Sephadex) or potential pathogenic bacteria (Vibrio alginolyticus). We also noticed that some of the newly expressed genes are exclusive from a specific treatment. Here we report that haemocytes from Litopenaeus vannamei are capable of recognizing and distinguishing different foreign materials, and respond specifically to each treatment, indicating some specificity in shrimp immune response.

    Notes: Haemocytes from penaeid shrimp (Farfantepenaeus californiensis, Litopenaeus vannamei and L. stylirostris) were separated using a discontinuous Percoll gradient. Shrimp haemocytes were spontaneously adhered to glass, allowing slide preparations for staining and microscopic differential counting. Like other crustaceans, shrimp has three main populations differing in presence and size of cytoplasmic granules and each population seems to be biochemical or functionally compromised. Prophenoloxidase (proPO )activity was mainly located in large granules haemocytes (75%) while the small granules cells participate with 25%, but seem to be responsible for encapsulation. Haemocyte discrimination ability was tested using Sephadex™ (Seph™), DEAE-Seph™ and CM-Seph™. Only DEAE-Seph™ was encapsulated by shrimp haemocytes and provoked the release of proPO activating system, indicating the role of particle charge in the activation of shrimp immune response.

    Notes: A sudden overaccumulation of methylglyoxal (MG) induces, in Saccharomyces cerevisiae, the expression of MG-protective genes, including GPD1, GLO1 and GRE3. The response is partially dependent on the transcriptional factors Msn2p/Msn4p, but unrelated with the general stress response mechanism. Here, we show that the high-osmolarity glycerol (HOG)-pathway controls the genetic response to MG and determines the yeast growth capacity upon MG exposure. Strains lacking the MAPK Hog1p, the upstream component Ssk1p or the HOG-dependent nuclear factor Msn1p, showed a reduction in the mRNA accumulation of MG-responsive genes after MG addition. Moreover, hyperactivation of Hog1p by deletion of protein phosphatase PTP2 enhanced the response, while blocking the pathway by deletion of the MAPKK PBS2 had a negative effect. In addition, the activity of Hog1p affected the basal level of GPD1 mRNA under non-inducing conditions. These effects had a great influence on MG resistance, as hog1Δ and other HOG-pathway mutants with impaired MG-specific expression displayed MG sensitivity, whereas those with enhanced expression exhibited MG resistance as compared with the wild-type. However, MG does not trigger the overphosphorylation of Hog1p or its nuclear import in the parental strain. Moreover, dual phosphorylation of Hog1p appears to be dispensable in the triggering of the transcriptional response, although a phosphorylable form of Hog1p is fundamental for the transcriptional activity. Overall, our results suggest that the basal activity of the HOG-pathway serves to amplify the expression of MG-responsive genes under non-inducing and inducing conditions, ensuring cell protection against this toxic glycolytic by-product.

    Notes: : A novel method to assess the ripening state of Persea Americana Mill avocados was developed. The method was validated and its results compared with standard ones such as the penetrometer method. Also, comparison were made between the proposed method and the ultrasonic system reported by other authors. The correlation between the proposed method and both the standard and former ultrasonic method is quite good. The measurement system overcomes the problem of alignment between the emitter and receiver found in the resent execution development for the same purposes. Extension of proposed systems could allow the measurement with a single ultrasonic source and possibility the development of an online evaluation system for all the fruits.

    Notes: Wounding lettuce (Lactuca sativa L., var. Longifolia) leaves induced an eight-fold increase in the activity of phenylalanine ammonia-lyase (PAL; EC 4.3.1.5), and the subsequent accumulation of phenolic compounds and tissue browning at 10°C. PAL is a key enzyme in the synthesis of phenolic compounds. A PAL cDNA was previously isolated by reverse-transcription PCR using total RNA from wounded lettuce leaves. RNA gel blots showed that maximum accumulation of both PAL mRNA and PAL enzyme activity occurred 24 h after wounding. A 2-min heat shock at 45°C administered within 5 min of wounding delayed the wound-induced increase in PAL activity, but did not delay the increase in wound-induced PAL mRNA. Changes in the content of PAL protein were also followed by immunoblot using anti-PAL antibody raised against the bacterially expressed protein from the cDNA. Immunoblots showed that the level of PAL protein in wounded lettuce tissue was significantly reduced by the heat shock treatment. These data suggest that heat shock reduces the rise in wound-induced PAL enzyme activity by reducing the translation of wound-induced PAL mRNA, or by increasing the turnover of the induced PAL protein.
