    Notizen: Abstract The thermal decomposition of Zn[NFA]2⋅5H2O (NFA=C16H18FN3O3, norfloxacin) and its kinetics were studied under non-isothermal conditions in air by TG-DTG and DTA methods. The intermediate and residue for each decomposition were identified from the TG curve. The non-isothermal kinetic data were analyzed by means of the Achar method and the Madhusudanan-Krishnan-Ninan (MKN) method. The possible reaction mechanisms were investigated by comparing the kinetic parameters. The kinetic equation for the second stage can be expressed as dα/dt=Aexp(−E/RT)(1−α).

    Notizen: Abstract Studies presented here show that cellular NAD, which we hypothesize to be the relevant biomarker of niacin status, is significantly lower in humans than in the commonly studied animal models of carcinogenesis. We show that nicotinamide and the resulting cellular NAD concentration modulate expression of the tumor suppressor protein, p53, in human breast, skin, and lung cells. Studies to determine the optimal NAD concentrations for responding to DNA damage in breast epithelial cells reveal that DNA damage appears to stimulate NAD biosynthesis and that recovery from DNA damage occurs several hours earlier in the presence of higher NAD or in cells undergoing active NAD biosynthesis. Finally, analyses of normal human skin tissue from individuals diagnosed with actinic keratoses or squamous cell carcinomas show that NAD content of the skin is inversely correlated with the malignant phenotype. Since NAD is important in modulating ADP-ribose polymer metabolism, cyclic ADP-ribose synthesis, and stress response proteins, such as p53, following DNA damage, understanding how NAD metabolism is regulated in the human has important implications in developing both prevention and treatment strategies in carcinogenesis.

    Notizen: Abstract Acetone transformation into methyl isobutyl ketone (MIBK) was studied using a fixed-bed dynamic reactor at 160°C, 1 atm pressure and acetone/H2 molar ratio=3. The reaction was carried out over Pt/HMFI bifunctional catalysts, with 0.30 wt% of platinum which was supported over three aluminosilicates (Si/Al ratio=40, 95 and 160) and a borosilicate (Si/B ratio=44) with similar dispersion. The results show that catalytic properties depend greatly on density and strength of the acidic sites of the catalysts. Moreover, the limiting step of the MIBK synthesis reaction (aldolization of two acetone molecules) is carried out over the acidic sites of the aluminosilicates, but not over those of the borosilicate, which considerably affects reaction selectivity.

    Notizen: Abstract Potentiometric and electrochemical impedance spectroscopic investigations of calcium-selective membranes containing poly(vinylchloride), dioctylphenylphosphonate, calcium (bis[4-(1,1,3,3-tetramethylbutyl)phenyl] phosphate) and different amounts of the lipophilic anionic additive tridodecylmethylammonium chloride were carried out. The addition of the lipophilic additive changes the properties of calcium-selective electrodes, e.g. slope and calcium selectivity. The selectivity for calcium in presence of H+, Na+, K+, NH4 +, Mg2+, Ba2+, Sr2+ and (C2H5)4N+ was measured by three different methods, namely separate solution method, fixed interference method and matched potential method. Membranes with different concentration ratios between the calcium-exchanger and tridodecylmethylammonium chloride were investigated within half a year. The tendency of changing from cationic into anionic response for membranes containing nearly equivalent concentrations of cation- and anion-exchanger was shown. This inversion of the electrode response depends not only upon the concentration ratio of both ion-exchangers but also upon the total concentration of calcium-exchanger. Electrochemical impedance spectroscopy was used for monitoring the development of membrane resistances during a soaking period of one month. Based on these results dielectric constants for the calcium-selective membranes depending on the membrane composition were determined. Furthermore, the dependence of the membrane resistance on the membrane thickness and the concentration of tridodecylmethylammonium chloride was evaluated.

    Notizen: Abstract Severe, malignant osteopetrosis is a disease characterized by osteoclasts that fail to resorb bone. Serious defects in the ability of white blood cells to eradicate infectious agents confound the clinical course. Defective superoxide generation by neutrophils, monocytes, and lymphocytes contributes to this inability to fight infection. To elucidate the mechanisms resulting in the defective superoxide generation observed in osteopetrotic leukocytes, gene expression, translocation, and phosphorylation of the major components that form the fimctional NADPH oxidase complex were studied in transformed B-lymphocytes. The expression of the p47 subunit of NADPH oxidase was reduced in B-lymphocytes collected from osteopetrotic patients compared to those from controls. Phosphorylation and translocation of p47 to the cell membrane after PMA stimulation was similar in B-lymphocytes from both patients and normal controls. However, total amount of p47 phosphorylation and translocation was reduced in patient samples. This was further supported by the experiment using p47 antisense oligonucleotide. The other major components of the oxidase (p91, p22, p67) were found to be present at normal levels. Thus, the reduction in p47 expression results in reduced ability to assemble a ftmctional NADPH oxidase complex at the membrane of lymphocytes from osteopetrotic patients. This defect translates into reduced superoxide generation and an increased propensity for infection.

    Notizen: Abstract We have recently described the isolation and characterization of bovine cDNA encoding poly(ADP-ribose) glycohydrolase (PARG). We describe here the preparation and characterization of antibodies to PARG. These antibodies have been used to demonstrate the presence of multiple forms of PARG in tissue and cell extracts from bovine, rat, mouse, and insects. Our results indicate that multiple forms of PARG previously reported could result from a single gene. Analysis of PARG in cells in which poly(ADP-ribose) polymerase (PARP) has been genetically inactivated indicates that the cellular content of PARG is regulated independently of PARP.

    Notizen: Abstract The aim of this work was to obtain a freeze-dried kit for direct99mTc-labeling of human polyclonal IgG. The labeling procedure was carried out by Schwarz's method. The best yields of99mTc-IgG were obtained by using sodium pyrophosphate decahydrate as a weak chelating agent. Performed tests showed the stability of the radiopharmaceutical up to 24 hours. Plasma clearance in rats was fitted to a biexponential curve withT 1/2α=(0.1 ±0.9) h andT 1/2β=(10±3) h. The organs with higher uptake of radiopharmaceutical were lung, kidneys and blood. In a rabbit model the abscess target/background ratio was 3–6 according to time of the scintigraphic images. Thirty patients with musculoskeletal infection were studied. Twenty-one lesions were detected and confirmed by culture/biopsy.

    Notizen: Abstract The effect is studied of the layer formation conditions on the molecular arrangement of copper tetra- tert-butyl phthalocyanine (CuPctBu4) and copper tetra- tert-butyl tetrabenzotriazaporphin (CuThptBu4) at the air–water interface. The decrease in initial surface concentration of these compounds is shown to affect the molecular orientation, as indicated by the increase in the area per molecule. The data are interpreted in terms of formation of CuPctBu4 and CuThptBu4 monolayers with a ‘face-on’ molecular arrangement when the initial surface concentration is sufficiently small (N less than 1.6 × 10−7 mole/m2 for CuPctBu4 and N less than 4 × 10−7 mole/m2 for CuThptBu4) and changes in the molecular orientation to ‘edge-on’ when the N values are higher. It is emphasized that the ‘edge-on’ orientation on the water surface is not only a molecular but a collective property of the azaporphyrine supramolecular assembly.
