    Notes: Abstract. Bone morphogenetic proteins (BMPs) are novel growth and differentiation factors that act on mesenchymal stem cells to initiate new bone formation in vivo and promote the growth and differentiation of cells in the osteoblastic lineage. In the present study, we examined the effects of recombinant human osteogenic protein-1 (also known as BMP-7) on the expression of related members of the BMP family using SaOS-2 and U2-OS, two human osteosarcoma cell strains. Evaluation of BMP-2, -4, and -6 mRNA expression indicates that OP-1 stimulated the mRNA levels of BMP-6 in both SaOS-2 cells (threefold) and U2-OS cells (fivefold) after 24 hours of treatment, while decreasing the mRNA levels of BMP-4 in SaOS-2 cells (80%) and BMP-2 and BMP-4 in U2-OS cells by 50% and 72%, respectively. BMP-2 mRNA expression, as examined by Northern blot analysis, was below detectable limits in SaOS-2 cultures. These results demonstrate that OP-1 modulates the mRNA expression of related members of the BMP family, suggesting a possible mode of action of OP-1 on the growth and differentiation of cells in the osteoblastic lineage in vitro.

    Notes: Abstract  It has been reported that in the chick embryonic retina, N-cadherin first appears at the very early stages and is subsequently substituted by R-cadherin at the middle to late stages of development. To examine the role of R-cadherin in the morphogenesis of chick retinal pigment epithelium (RPE), the distribution of this adhesion molecule was studied by immunofluorescence cytochemistry and immunoelectron microscopy from embryonic day (E) 6 to hatching. R-cadherin immunoreactivity was detected at E6, and was strongest at E12–13. During these stages, R-cadherin was expressed uniformly on the lateral plasma membranes of RPE cells in contact with each other. Thereafter, R-cadherin immunoreactivity was markedly decreased, with intense immunoreactivity restricted to zonulae adherentes in latero-apical regions at E16. R-cadherin immunoreactivity was no longer detectable in the newly hatched chick RPE, even though morphologically well developed zonulae adherentes were present in latero-apical regions. No immunoreactivity was detected on the apical side facing the neural retina or on the basal side facing the basal lamina at any stage of development. These findings indicate that R-cadherin plays an important role as a major cadherin subtype in the morphogenesis of chick embryo RPE, and is involved initially in non-specific cell-cell adhesions, and subsequently in the formation and maintenance of developing zonulae adherentes.

    Notes: Abstract  CD1 and TL were once thought to be genetic homologues because of their thymus-specific expression. We investigated their equivalents in the rat to clarify whether their structure and pattern of expression are conserved in rodents. Two rat class Ib genes, containing 3′ sequences very similar to mouse TL, were identified and designated RT1.P. Neither of them, however, can encode ordinary class I molecules due to the accumulation of harmful mutations in the 5′ regions that are unique to RT1.P, while the 3′TL-like regions still retain protein-coding capacity. Comparison of the structural organization of three types of TL family genes, which include mouse T3/T18-encoding TL antigens, mouse T1/T16, and rat RT1.P1/P2 pseudogenes, revealed the presence of a clear demarcation between the type-specific and TL-specific sequences at intron 3. This finding suggests that recombination plays an important role in creating the TL family genes in rodents. Characteristic features of TL, such as a low level of polymorphism and linkage to the major histocompatibility complex, were also observed in the rat. On the other hand, rat CD1 molecules were expressed at a high level on the surface of thymocytes. Absence of authentic TL antigens and thymic expression of CD1d molecules in the rat suggest the plasticity and conservation of class Ib genes in rodent evolution. Functions of TL may be substituted with CD1 or other class Ib molecules expressed by rat thymus.

    Notes: Abstract Objectives: The present study was conducted to compare pharmacokinetic behaviors of nicardipine enantiomers given in different doses with different formulations of racemic nicardipine in healthy volunteers. Methods: One or two 20-mg racemic nicardipine tablets, and a 40-mg sustained-release capsule of nicardipine were administered to eight healthy volunteers in a cross-over fashion and pharmacokinetic parameters were evaluated. Enantiomer concentrations were determined by GC-MS combined with chiral stationary phase HPLC. Results and conclusions: Serum concentration of (+)-nicardipine was approximately 2–3 times higher than that of (−)-nicardipine in 20- and 40-mg doses of conventional formulations and a non-linear increase in bioavailability with dose was demonstrated. The value for AUC of (+)-nicardipine was approximately 2.3–2.8 times greater than that of the (−)-nicardipine (P 〈 0.05) when 20 and 40 mg racemic nicardipine were administered in a conventional preparation. Relative bioavailability of the sustained-release preparation vs the conventional preparation was 28% and 44% for (+)- and (−)-nicardipine, respectively, for the 40-mg dose.

    Notes: Abstract By means of gas chromatography with electroantennographic detection (GC-EAD), gas chromatography–mass spectrometry (GC-MS), and a series of bioassays, (Z)-11-tetradecenyl acetate (Z11–14:OAc) and (E)-11-tetradecenyl acetate (E11–14:OAc) at a ratio of 100:3 were identified as the female sex pheromone of the adzuki bean borer,Ostrinia scapulalis. The average amounts ofZ11–14: OAc andE11–14:OAc in a single sex pheromone gland were 6.6 ± 2.4 ng and 0.2 ± 0.1 ng, respectively. In a wind-tunnel bioassay, the binary blend ofZ11- andE11–14:OAc elicited almost the same male behavioral responses as did virgin females and sex pheromone gland extract. In field trapping experiments, rubber septa impregnated with the binary blend (50 μg/septum) attracted more males than virgin females. The sex pheromone ofO. scapulalis thus turned out to be similar to that of theZ-type European corn borer,O. nubilalis, in both components and their ratio.

    Notes: Abstract Host-plant chemicals stimulating oviposition by a Leguminosae-feeding pierid butterflyColias erate poliographyswere isolated and identified from one of its primary host plants, white clover (Trifolium repens). Females readily deposited eggs in response to methanolic extracts of the plant, and subsequent partition of the extracts with organic solvents revealed that chemical constituents critical for host recognition reside in the water-soluble fraction. Further fractionation of the hydrosoluble fraction by column chromatography led to the separation of an active fraction and two cyanoglucosides, linamarin and lotaustralin. Conspicuous oviposition response was evoked by unidentified polar compound(s), while these cyanoglucosides exerted no stimulatory activity by themselves. However, ovipositing females preferred samples containing either of the two cyanoglucosides. In dual-choice bioassays, significantly more eggs were laid on samples admixed with the cyanoglucosides, suggesting that the cyanoglucosides serve as synergistic oviposition stimulants and could play an important role in host selection.

    Notes: Abstract A giant danaid butterfly, Idea leuconoe, specializes on apocynaceous plants such as Parsonsia laevigata, which has been reported to contain pyrrolizidine alkaloids. Females of I. leuconoe deposited eggs in response to methanolic extract of P. laevigata, and subsequent bioassay-guided fractionation of the extract revealed that phytochemicals crucial for host recognition by ovipositing females are Parsonsia-specific macrocyclic pyrrolizidine alkaloids including parsonsianine, parsonsianidine, and 17-methylparsonsianidine. Parsonine, another P. laevigata pyrrolizidine component with a keto-dihydropyrrolizine moiety that is closely related in structure to male pheromones of the butterfly, and several nonhost pyrrolizidine alkaloids were entirely inactive. We interpret these data as strong evidence for an ancestral association through herbivory between danaid butterflies and pyrrolizidine alkaloids.

    Notes: Abstract We investigated the enhancement of cytokine expression by heat treatment in transiently transfected glioma cells. The cells were transfected with plasmid bearing the interferon (IFN)-β gene under control of the MMTV promoter, which is inducible by glucocorticoid (dexamethasone). Then magnetite particles (10 nm diameter) as intracellular heating material were incorporated to the cells as the form of magnetoliposome. After 5 hours of incorporation, alternative magnetic field (384Oe, 118kHz) was applied for intracellular heating. IFN-β secreted in the medium was assayed and its concentration was compared to the extracellular heating induced expression, both in the presence and absence of dexamethasone. Higher IFN-β concentration was detected in intracellular heating even at lower temperature, 39 °C, than 43 °C in extracellular heating. The IFN-β expression level reached in the presence of dexamethasone was about three times higher than in the absence of inducer. In intracellular heating of 60 min, the surviving cell number reduced until 20%.

    Notes: Abstract The Virtual Society (VS) project is a long term research initiativethat is investigating the evolution of the futureelectronic society. Our vision for this electronic societyis a shared 3D virtual world where users, from homes and offices, canexplore, interact and work. Our first implementation of aninfrastructure to support our investigation is known asCommunityPlace and has been developed to support large-scale shared 3D spaces onthe Internet using the Virtual Reality Modeling Language (VRML). Obviously, such an ambitious project cutsacross many different domains. In this paper we outline thegoals of the Virtual Society project, discuss the architecture andimplementation of CommunityPlace with particular emphasis on Internetrelated technologies such as VRML and present our views on the role ofVRML and the Internet to support large-scale shared 3D spaces.

    Notes: Abstract Reciprocal crosses were made to introduce the red flower color of Delphinium cardinale Hook and D. nudicaule Torr. & A. Gray into the dwarf species, D. grandiflorum L., and an ovule culture was applied to keep hybrid embryos from aborting. Interspecific hybrid plants of D. cardinale × D. grandiflorum and D. grandiflorum × D. nudicaule were successfully obtained, and their hybridity was confirmed by isozyme analyses. Morphological studies indicated that these hybrids were almost intermediate between the female and male parents in leaf shape and flower color. Dwarf plants with red flowers were not obtained. All hybrids showed low pollen fertility and failed to yield viable seeds by self- or backcross-pollination using fertile pollen grains of their parents. The applicability of the ovule culture technique for genetic improvement of the genus Delphinium through interspecific hybridization is discussed.
