    Notes: Abstract Microwave surface resistance measurement constitutes one of the most advantageous procedures for the investigation and characterization of the new superconducting oxides. Keeping in mind that the bulk ceramic materials usually have cylindrical symmetry, a new automatic measuring system, based on a resonant cavity, has been developed with special emphasis on superconducting pellets. The resolution capability of our system is pointed out by measuring the surface resistance of YBCO and BSCCO samples.

    Notes: Abstract The time resolved emission spectrum of the blue band of Ti:sapphire laser crystal has been investigated as a function of temperature (range 10–290 K) and UV (266 nm) laser excitation intensity. Two blue emission bands, centred at 420 nm and 460 nm, have been detected. The 420 nm band is attributed to Ti4+ centres whereas the 460 nm one is proposed to be due to Ti3+ ions. The evolution of the emission spectrum vs the UV excitation intensity has shown that the concentration of Ti4+ centres is increased under UV irradiation at the cost of the centres responsible for the 460 nm band.

    Notes: Abstract Anl-amino-acid oxidase (EC 1.4.3.1) that catalyzes the oxidative deamination of twelvel-amino acids has been purified 21-fold and with 14% yield to electrophoretic homogeneity fromChlamydomonas reinhardtii cells by ammonium-sulfate fractionation, gel filtration through Sephacryl and Superose, anion-exchange chromatography and preparative electrophoresis in polyacrylamide gels. The native enzyme is a protein of 470 kDa and consists of eight identical or similarsized subunits of 60 kDa each. Optimum pH and temperature were 8.2 and 55° C, respectively, with a Q10 (45–55° C) of 1.7 and an activation energy of 45 kJ · mol−1. Its absorption spectrum showed, in the visible region, maxima at 360 and 444 nm, characteristic of a flavoprotein with a calculated flavin content of 7.7 mol FAD per mol of native enzyme. ApparentK m values of the twelvel-amino acids which can act as substrates ofl-amino-acid oxidase ranged between 31 μM for phenylalanine and 176 μM for methionine. The effect of several specific group reagents, chelating agents and bivalent cations on enzyme activity has also been studied.

    Notes: Abstract Petrocoptis glaucifolia, a paleoendemic member of the Caryophyllaceae from the North of Spain, was found to contain at least five proteins that inhibit protein synthesis in a rabbit reticulocyte lysate. One of them, for which the name petroglaucin is proposed, was purified to apparent electrophoretic homogeneity by chromatography through S-Sepharose Fast Flow, Sephadex G-75 and CM-Sepharose Fast Flow. The apparent Mr of the preparation was 27500. This protein does not contain appreciable glycan chains and displays 45.8% of NH2-terminal amino-acid sequence homology with some ribosome-inactivating proteins from Saponaria officinalis, another member of the Caryophyllaceae. Petroglaucin shows the following functional properties: (i) it strongly inhibits the rabbit-reticulocyte-lysate system and Vicia sativa cell-free extracts, both coded by endogenous messengers, and also inhibits poly(U)-directed polyphenylalanine synthesis by Vicia sativa cell-free extracts and purified rat-liver ribosomes; (ii) it shows much less inhibitory capacity in wheat-germ, Cucumis sativus and rat-liver cell-free systems coded by endogenous messengers; (iii) the inhibitory effects on purified rat-liver ribosomes were irreversible; (vi) it promotes the release of adenine from purified rat-liver ribosomes. The total activity of this translational inhibitor has been found to increase up to 11-fold during its purification, indicating that some regulatory factor that normally blocks the translational inhibitory activity of the ribosome-inactivating protein in crude extracts of the plant is removed during purification.

    Notes: Abstract Lamellar crystals of nylons 5, 9 and 13 inα-structure were grown from solution and systematically studied by electron microscopy and X-rays. Electron diffraction showed that they all consist of a twinned structure with hydrogen bonds running parallel to the long dimension of the crystals. Polyethylene decoration of such crystals revealed folding sectorization only in the case of nylon 9. Diffraction data from films both uniaxially and biaxially oriented were used to propose a unifying model for theα-structure of odd nylons based on the antiparallel model given by Tadokoro. In this model successive sheets are progressively sheared along the chain axis by approximately twice the interatomic height. Such a model is able to account well for most of the structural observations we have made on solution grown crystals.

    Notes: Abstract The precise role of protein kinase C in insulin action in skeletal muscle is not well defined. Based on the fact that inhibitors of protein kinase C block some insulin effects, it has been concluded that some of the biological actions of insulin are mediated via protein kinase C. In this study, we present evidence that inhibitors of protein kinase C such as staurosporine, H-7 or polymyxin B cannot be used to ascertain the role of protein kinase C in skeletal muscle. This is based on the following experimental evidences: a) staurosporine, H-7 and polymyxin B markedly block in muscle the effect of insulin on System A transport activity; however, this effect of insulin is not mimicked in muscle by TPA-induced stimulation of protein kinase C, b) H-7 and polymyxin B block insulin action on System A transport activity in an additive manner to the inhibitory effect of phorbol esters, c) staurosporine, H-7 and polymyxin B block the effect of insulin on lactate production, a process that is activated by insulin and TPA in an additive fashion, and d) staurosporine completely blocks the tyrosine kinase activity of insulin receptors partially purified from rat skeletal muscle.
