    Notes: Changes induced by NaCl in the sizes of both cells and vacuolar compartments of tomato seedling roots were determined using morphometric and stereological analysis. Parameters evaluated showed that cellular volume was smaller in roots grown under saline conditions than in control roots. This reduction was greater when NaCl concentration was increased. Similar behaviour can be observed in the vacuolar compartment where high concentrations of NaCl produce a reduction in vacuolar volume although the number of vacuoles per cell does not change under saline conditions.

    Notes: The triallate tolerance of wheat (Triticum aestivum L.) cultivars Marius and Mexicali was determined under growth-chamber conditions. Mexicali was found to tolerate triallate at levels about fourfold higher than Marius. The susceptible wheat cultivar was protected against triallate by seed treatment with MON 5501. The herbicide concentration in the aerial part of the plants was determined by gas chromatography. Triallate levels were similar in both cultivars treated with 0.5 mg 1−1 of triallate. The safener did not affect the triallate content per plant in the susceptible wheat cultivar treated with 1 mg 1−1 of herbicide. Augmentation de la tolerance du blé au triallate avec le phytoprotecteur MON 5501La tolerance au triallate des cultivars de ble Marius et Mexicali a été déterminée en chambre climatique. Mexicali s'est avéré 4 fois plus tolérant que Marius. Le cultivar de blé sensible a été protege du triallate par un traitement des grains avec MON 5501. La concentration en herbicide dans la partie aérienne des plantes a été determined par chromatographie gazeuse. Les taux de triallate ont été equivalents chez les 2 cultivars traites avec 0,5 mg l−1 de triallate. Le phytoprotecteur n'a pas affecté lateneur en triallate par plante chez le cultivar sensible traite avec 1 mg 1−1 d'herbicide. Verbesserung der Toleranz von Weizen gegenuber Triallat mil dem Antidot MON 5501Die Triallat-Toleranz der Weizensorten ‘Marius’ und ‘Mexicali’ wurde im Phytotron bestimmt. ‘Mexicali’ war etwa 4mal toleranter als ‘Marius’. Die empfindliche Weizensorte wurde durch Beizung mit MON 5501 gegen Triallat geschützt. Die Herbizidkonzentration in den oberirdischen Pflanzenteilen wurde gaschromatographisch bestimmt Der Wirkstoff-Gehalt war in beiden mit 0,5 mg 1−1 Triallat behandelten Sorten ahnlich hoch. Das Antidot hatte keinen Einfluβ auf den Triallat-Gehalt der empfindlichen Weizensorte, die mit 1 mg 1−1 behandelt worden war.

    Notes: Twenty-four near-isogenic barley lines, with a cv.‘Pallas’ background, carrying different mildew resistance genes were subjected in 1987, 1989 and 1990 to natural infection by the pathogen at several different and contrasting Spanish sites in order to study its virulence. The virulence genes proved to be geographically grouped into three regions: western (Valladolid), southern (Sevilla) and northern and northeastern (Navarra, Lleida and Girona). The mildew population of Lleida was more variable when compared with Navarra and Valladolid. Overall, the most effective resistance genes were: Ml-a13 + Ml-(Ru3), mlo and Ml-(1402).

Multispecies stock assessment based upon Schaefer's theory was applied to a coastal pelagic fishery (Spanish mackerel, Scomberomorus maculatus (Mitchill); king mackerel, S. cavalla (Cuvier) and the blue runner, Caranx fusus (Geoffroy-St Hilaire)), from the west central Gulf of Mexico. Linear and non-linear systems of equations were estimated by using a multiple stepwise regression technique. The values of interaction parameters show a clear competition between mackerels, and technological interdependences between the blue runner and mackerels, The maximum yield estimation was from 4000 to 5000 tonnes, obtained with 23 and 34 beach seines respectively, depending on the applied model. Two stages were observed from the statistical records; in the first the Spanish mackerel is the most important species while in the second the abundance of this species declines and the others remain at the same level. Significant interactions were found from the first data group.

    Notes: In order to develop plasmids adequate for non-integrative genetic transformation of Candida albicans, a DNA fragment of 15.3 kb was cloned from this organism on the basis of its capacity to convert the integrative Saccharomyces cerevisiae vector Ylp5 into a non-integrative one. Southern hybridization analysis, carried out with a labelled DNA probe of 3.6 kb derived from the cloned fragment, showed that it consisted of C. albicans DNA, the hybridization pattern indicating that the corresponding sequences were homologous to several chromosomal regions. The size of the C. albicans DNA promoting autonomous replication In S. cerevisiae was substantially reduced by subcloning. A 5.1 kb subfragment, defined by BamHI and SalI restriction sites, retained autonomous replication sequences (ARS) functional in the heterologous S. cerevisiae system and in C. albicans, when inserted in plasmid constructions that carried a S. cerevisiae trichodermin-resistance gene (tcm1) as selection marker. C. albicans transformants were both of the integrative and the non-integrative type and the plasmids recovered from the latter very often carried a reorganized ARS, indicating that recombination of the inserted ARS DNA had occurred in the homologous host. Successive reorganizations of the ARS insert in C. albicans eventually led to a more stable and much smaller fragment of 687 bp that was subsequently recovered unchanged from transformants. Sequence analysis of the 687 bp fragment revealed four 11-base blocks, rich in A+T, that carried the essential consensus sequence considered relevant for yeast ARS elements in addition to other features also described as characteristic of yeast replication origins.

    Notes: Pea ovaries are induced to enter a fruit development pathway involving physiological and morphological changes by pollination or application of plant growth regulators. In the absence of these stimuli, overies stop growing and enter an alternative pathway of senesecence that leads to their degeneration. We have used two dimensional polyacrylamide gel electrophoresis in search of molecular changes underlying fruit development and ovary senescence at the level of total accumulated proteins, newly synthesized proteins, and translatable, RNA populations. We have found changes in gene expression during the processes of ovary formation and ovary senescence. Stimuli that induce fruit set do not appreciably alter the overall patterns of synthesized proteins or translatable RNAs, indicating that fruit development is apparently a natural continuation of ovary formation. However, ovary senescence is an alternative pathway that involves the presence of new RNA messengers and proteins as well as the disappearance of others. These changes were detected earlier than any morphological or structural changes could be observed in the ovary.

    Notes: Susceptibility of alfalfa (Medicago saliva L. cv. Aragón) nodules and leaves to water stress has been investigated. Nodule acetylene reduction activity (ARA), leaf CO2 exchange rate (CER) as well as soluble protein, proline and total soluble sugar (TSS) contents were determined during drought. Water status was estimated as water potential (Ψw) and Relative water content (RWC) of the respective tissues. Maximum rates of ARA required higher Ψw than CER. Nodules had lower RWC for a given Ψw than leaves. Water stress reduced soluble protein content in both tissues; however, the decline in soluble protein content was detected at greater Ψw in nodules than in leaves. Proline and TSS increased in leaves and nodules, and again the threshold Ψw triggering such accumulation was higher in nodule tissues. Oior results suggest that alfalfa nodules are more susceptible to water shortage than leaves. Effects of accumulated TSS and proline upon leaf and nodule physiology are discussed in relation to protein stability (proline), pH control (proline) and osmotic adjustment (proiine and TSS). The TSS accumulation induced by water stress suggests that substrate shortage would not be the primary effect of drought on nodule activity.

    Notes: The effect of drought upon phosphoenolpyruvate carboxylase (PEPC; EC 4.1.1.31), malate ddiydrogenase (MDH; EC 1.1.1.37), alcohol dehydrogenase (ADH; EC 1.1.1.1) and β-hydroxybulyrate dehydrogenase (β-OH-BDH; EC 1.1.1.30) enzyme activities as well as the leghemoglobin (Lb), malate and ethanol contents of alfalfa nodules (Medicago sativa L. cv. Aragon) were examined. Both the ieghemoglobin (Lb) content and the Lb/soluble protein ratio were significantly reduced at a nodule water potential (Ψnod) of—1.3 MPa. At lower Ψnod, Lb content decreased further, but the ratio remained unchanged. Slight stress (—1.3 MPa) drastically affected acetylene reduction activity (ARA; 60% reduction) whereas in vitro PEPC activity was main-tained at relatively constant values. As stress progressed (—2.0 MPa), a simultaneous reduction in both activities was observed. Severe stress (Ψnod lower than —2.0 MPa) stimulated in vitro PEPC. Bacteroid β-J-OH-BDH activity was stimulated by slight (—1.3 MPa) and moderate (—2.0 MPa) drought. MDH activity rose in slightly stressed nodules (Ψnod—1.3 MPa). Greater water deficits sharply decreased MDH activity to values significantly lower than those found in control nodules. Nodule malate content followed the same pattern as MDH. The plant fraction of the nodule showed constitutive ADH activity and contained ethanol. ADH was stimulated at slight (— 1.3 MPa) and moderate drought levels (—2.0 MPa). Ethanol content showed similar behavior to ADH activity. Inhibition of ARA, reduction of Lb content and stimulation of the fermentative metabolism induced by water stress suggest some reduction ira O2 availability within the nodule.
