    Description: Yeast artificial chromosomes (YACs) were obtained from a 550-kilobase region that contains three probes previously mapped as very close to the locus of the fragile X syndrome. These YACs spanned the fragile site in Xq27.3 as shown by fluorescent in situ hybridization. An internal 200-kilobase segment contained four chromosomal breakpoints generated by induction of fragile X expression. A single CpG island was identified in the cloned region between markers DXS463 and DXS465 that appears methylated in mentally retarded fragile X males, but not in nonexpressing male carriers of the mutation nor in normal males.     Description: High-resolution radio images of PKS1830 - 211 are obtained to study the possibility that the double structure is a gravitationally lensed object. The VLBI observations, taken from interferometric radiotelescope networks, reveal an elliptical ring that connects two bright spots of similar composition. Because the lens and the lensed object are closely aligned, and because of the structure of the two spots, the source is concluded to be a radio Einstein ring. The source is found to be close to the galactic plane, and the lens and the lensed object are extragalactic. The source is also found to be unusually bright, suggesting that it is aligned with a bright background source or amplified by some mechanism related to a source that is not so bright.

    Notes: Thin sputtered films of the high-Tc material YBa2Cu3O7-Δ were investigated using 180° back-scattering geometry with Fourier transform (FT) Raman spectroscopy. It is established that the main bands in the spectra of a YBa2Cu3O7-Δ film on strontium titanate substrate exhibit similarity with those obtained for oriented untwinned crystallites of the superconductor having their c-axis perpendicular to the surface of the substrate. No degradation or excessive heating of the film was observed with 1.064 μm excitation for low laser powers. The film on a magnesium oxide substrate gives unique evidence of the isolated strong, featureless, almost flat continuum, producing a broad maximum near the region of the antiphase axial bending O(2)—Cu(2)—O(3) motions of planar oxygens at very low excitation energy. This appears to be the first application of FT-Raman spectroscopy using near-infrared excitation (1.064 μm) to the field of high-temperature superconducting films and their detailed characterization.

    Description: The human immunodeficiency virus-1 (HIV-1) trans-activator Tat is an attractive target for the development of antiviral drugs because inhibition of Tat would arrest the virus at an early stage. The drug Ro 5-3335 [7-chloro-5-(2-pyrryl)-3H-1,4-benzodiazepine-2(H)-one], inhibited gene expression by HIV-1 at the level of transcriptional trans-activation by Tat. The compound did not inhibit the basal activity of the promoter. Both Tat and its target sequence TAR were required for the observed inhibitory activity. Ro 5-3335 reduced the amount of cell-associated viral RNA and antigen in acutely, as well as in chronically infected cells in vitro (median inhibition concentration 0.1 to 1 micromolar). Effective inhibition of viral replication was also observed 24 hours after cells were transfected with infectious recombinant HIV-1 DNA.     Description: Severe combined immunodeficient (SCID) mice reconstituted with human peripheral blood leukocytes (hu-PBL-SCID mice) have inducible human immune function and may be useful as a small animal model for acquired immunodeficiency syndrome (AIDS) research. Hu-PBL-SCID mice infected with human immunodeficiency virus-1 (HIV-1) contained virus that was recoverable by culture from the peritoneal cavity, spleen, peripheral blood, and lymph nodes for up to 16 weeks after infection; viral sequences were also detected by in situ hybridization and by amplification with the polymerase chain reaction (PCR). Mice could be infected with multiple strains of HIV-1, including LAV-1/Bru, IIIB, MN, SF2, and SF13. HIV-1 infection affected the concentration of human immunoglobulin and the number of CD4+ T cells in the mice.     Description: Three spatially distant surface loops were found to mediate the interaction of the coagulation protein factor X with the leukocyte integrin Mac-1. This interacting region, which by computational modeling defines a three-dimensional macromotif in the catalytic domain, was also recognized by glycoprotein C (gC), a factor X receptor expressed on herpes simplex virus (HSV)-infected endothelial cells. Peptidyl mimicry of each loop inhibited factor X binding to Mac-1 and gC, blocked monocyte generation of thrombin, and prevented monocyte adhesion to HSV-infected endothelium.     Description: Photometry and colorimetry of 951 Gaspra were obtained on nine nights during the 1990 opposition. A composite lightcurve constructed using data from eight of those nights yielded a synodic rotational period of 7.04346 +/- 0.00006 hours, a mean absolute V magnitude of 11.8026 +/- 0.0025, and a slope parameter of 0.285 +/- 0.005. The apparent discrepancy can be easily resolved by realizing that their determination is based primarily on data obtained after opposition. Different phase functions pre- and post-opposition are a natural consequence of a changing aspect during an opposition. If the sub-Earth latitude on Gaspra is at a less equatorial aspect after opposition than it was before opposition, then we would expect to see a shallower phase function (corresponding to a larger numerical value of the slope parameter). Adding weight to this hypothesis is the last observation of the opposition, made in May after Gaspra had passed post opposition quadrature, which is displaced toward brighter absolute magnitudes relative to the rest of our data, indicating an even more poleward sub-Earth latitude than earlier in the opposition. Because the orbits of Earth and Gaspra are nearly coplanar, a substantial change in sub-Earth latitude during the opposition would not have been possible unless the obliquity of the asteroid's rotational axis is not small.

    Description: The observation that the K-Ar, Pb-Pb, and Rb-Sr ages of a significant number of lunar highland rocks were reset in the interval of 4.1-3.8 Ga ago led to the concept of a cataclysmic bombardment of the moon during this period. An important consideration for understanding the early bombardment history of the solar system, including the moon, is whether evidence also exists in meteorites for resetting of radiometric ages by cataclysmic bombardment of their parent bodies. A comparison of Ar-39-Ar-40 ages of achondritic meteorites with ages of lunar highland rocks should help elucidate the nature of the early bombardment of the solar system. We are participating in various consortia studies of primarily Antarctic eucrites and howardites for which we measured Ar-39-Ar-40 ages of various clasts and matrix samples. The results of these studies are presented.
