    Notes: The process performance and metabolic rates of samples of activated sludge dosed with vitamin supplements have been compared. After initial screening, four vitamins and two metals as single supplements and in pairs, were dosed continuously into the mixed liquor of an activated sludge simulation. Toxicity, oxygen demand removal, respiration rates and suspended solids were measured to monitor the effect on process efficiency. It was confirmed experimentally that an industrial wastewater stream did not contain a sufficient supply of micronutrients for efficient biological treatment. This was concluded from the observation that control sludge batches (receiving no supplements) averaged chemical oxygen demand removal efficiency of 58%. Dosing micronutrients into the mixed liquor produced removal efficiencies of up to 69%. Some of the supplements increased the respiration rate of the sludge while some decreased it, indicating a range of stimulatory and inhibitory effects. Complex interactions between micronutrients that were dosed simultaneously were evident. Several positive effects led to the conclusion that micronutrients have the potential to optimise process performance of activated sludge plants treating industrial wastewater. The addition of phosphorus/niacin and molybdenum/lactoflavin removed wastewater components that were toxic to nitrifiers as indicated through toxicity testing, thus protecting downstream nitrification/denitrification treatment processes.     Notes: Abstract Vegetation pattern on landscapes is the manifestation of physical gradients, biotic response to these gradients, and disturbances. Here we focus on the physical template as it governs the distribution of mixed-conifer forests in California's Sierra Nevada. We extended a forest simulation model to examine montane environmental gradients, emphasizing factors affecting the water balance in these summer-dry landscapes. The model simulates the soil moisture regime in terms of the interaction of water supply and demand: supply depends on precipitation and water storage, while evapotranspirational demand varies with solar radiation and temperature. The forest cover itself can affect the water balance via canopy interception and evapotranspiration. We simulated Sierran forests as slope facets, defined as gridded stands of homogeneous topographic exposure, and verified simulated gradient response against sample quadrats distributed across Sequoia National Park. We then performed a modified sensitivity analysis of abiotic factors governing the physical gradient. Importantly, the model's sensitivity to temperature, precipitation, and soil depth varies considerably over the physical template, particularly relative to elevation. The physical drivers of the water balance have characteristic spatial scales that differ by orders of magnitude. Across large spatial extents, temperature and precipitation as defined by elevation primarily govern the location of the mixed conifer zone. If the analysis is constrained to elevations within the mixed-conifer zone, local topography comes into play as it influences drainage. Soil depth varies considerably at all measured scales, and is especially dominant at fine (within-stand) scales. Physical site variables can influence soil moisture deficit either by affecting water supply or water demand; these effects have qualitatively different implications for forest response. These results have clear implications about purely inferential approaches to gradient analysis, and bear strongly on our ability to use correlative approaches in assessing the potential responses of montane forests to anthropogenic climatic change.

    Notes: Abstract The acute toxicity of a technical formulation of pentachlorophenol (PCP) and pure pentachlorophenol to three age classes ofDaphnia magna, and adultD. pulex andD. galeata mendotae was determined by static toxicity tests. The influence of a number of factors on toxicity of PCP was also examined. The 48-hr LC50 estimates for adult daphnids of the three species exposed to pure PCP were 1.78, 4.59 and 0.51 mg/L, respectively, while those for the technical formulation were 2.57, 3.66 and 0.33 mg/L, respectively. There was little difference in toxicity between the technical and pure PCP; however, toxicity of both forms of PCP was influenced by duration of exposure, age (and/or size) and species of test organism and pH of the test solution. Pentachlorophenol caused a toxic response over a very narrow range of concentrations, with the greatest response occurring immediately between 0 and 24 hr. Pure PCP was equally toxic to all age classes ofD. magna but susceptibility to technical PCP decreased with maturation.D. g. mendotae was ten times more sensitive thanD. pulex to PCP. Pure PCP was significantly more toxic toD. magna at pH 5.5 than 7.0 with mean 48-hr LC50 values of 0.082 and 1.78 mg PCP/L, respectively. At 12°C, the toxicity of both forms of PCP toD. g. mendotae andD. pulex did not differ significantly from that at 20°C; however, technical PCP was significantly more toxic toD. magna at 12°C for an exposure duration of 48 hr. There was no effect of test container size (100, 250, 600 and 1,000 mL) on the toxicity of PCP toD. magna at 20°C with the lower pH of 5.5, suggesting that adsorption to glassware was not a factor in availability of PCP to test organisms. Beaker size had no effect on the toxicity of PCP toD. pulex at 20°C with test solutions having a pH of 7.0-8.0.

    Notes: Abstract Chronic toxicity test procedures (static, with renewal) were used to determine the chronic toxicity of sublethal concentrations of a technical formulation of pentachlorophenol (PCP) and pure pentachlorophenol toDaphnia magna. Test organisms 48+-12 h old were exposed for their entire lifespan (i.e., until death) to 0.01, 0.05, 0.1 and 0.5 mg technical PCP/L and 0.01, 0.087 and 0.1 mg pure PCP/L. Criteria used to assess chronic toxicity were mean time to appearance of the primiparous instar in the brood chamber, mean number of days to release of the first brood, mean number of broods produced per female, mean brood size per female, mean number of reproductive days, mean number of young produced per reproductive day per female and survivorship. Pentachlorophenol differentially affected maturation and reproduction but not survivorship or longevity. Mean number of broods produced per daphnid, length of the reproductive period, longevity and survivorship were insensitive criteria relative to mean time to appearance of the primiparous instar, time to release of first brood, brood size, and number of young produced per daphnid per reproductive day. Generally, there was little difference in toxicity of the three concentrations of pure PCP, for they significantly reduced mean brood size and rate of reproduction of young and significantly but differentially affected maturation. Technical PCP, at the highest concentration of 0.5 mg/L, significantly reduced mean brood size and the rate of production of young, and significantly delayed both time to appearance of the primiparous instar and release of the first brood. When differences in toxicity occurred, generally, pure PCP was more toxic than comparable concentrations of technical PCP. Although enhanced maturation was observed there was no compensatory reproduction. Similar conclusions regarding maturation and survivorship would have been derived from this study had it been terminated after the standard 21 d; however, the conclusions would have been different for reproduction. Only the highest concentration of technical PCP reduced brood size, the rate of production of young and total number of young produced per daphnid in 21 d. Only pure PCP at 0.05 mg/L caused daphnids to produce significantly fewer broods and, although the mean brood size was significantly larger than those in the controls, the mean number of young produced in 21 d was significantly reduced. These results differ substantially from those based on the entire life-cycle study and one of the most obvious differences is the much lower rates of young production in the entire life cycle study (2.41–3.03 young per daphnia per reproductive day) opposed to the first 21 days of the study (5.13–7.5 young per daphnia per reproductive day).

    Notes: Abstract Red-tailed hawks were exposed to sublethal levels of lead acetate for periods of 3 or 11 weeks. Alterations in the heme biosynthetic pathway were demonstrated after the first week of exposure to 0.82 mg lead per kilogram body weight per day. Activity of erythrocyte porphobilinogen synthase (aminolevulinic acid dehydratase) was depressed significantly and did not return to normal levels until 5 weeks after the termination of lead treatments. A rapid and relatively brief increase in erythrocyte free protoporphyrin and a slower but more prolonged increase in its zinc complex were also demonstrated with exposure to this dose of lead for 3 weeks. Less substantial decreases in hematocrit and hemoglobin levels occurred but only in the longer experiment with exposure to higher lead levels. Short term, low level lead exposure did not effect immune function significantly in the hawks, as measured by antibody titers to foreign red blood cells or by the mitogenic stimulation of T-lymphocytes. Increased lead exposure produced a significant decrease in the mitogenic response but had no effect on antibody titers.

    Notes: Abstract  Sinkhole collapse is one of the main limitations on the development of karst areas, especially where bedrock is covered by unconsolidated material. Studies of sinkhole formation have shown that sinkholes are likely to develop in cutter (enlarged joint) zones as a result of subterranean erosion by flowing groundwater. Because of the irregular distribution of pinnacles and cutters on the bedrock surface, uncertainties arise when "hit-or-miss" borehole drilling is used to locate potential collapse sites. A high-resolution geophysical technique capable of depicting the details of the bedrock surface is essential for guiding the drilling program. Dipole-dipole electrical resistivity tomography (ERT) was used to map the bedrock surface at a site in southern Indiana where limestone is covered by about 9 m of clayey soils. Forty-nine transects were conducted over an area of approximately 42,037 m2. The electrode spacing was 3 m. The length of the transects varied from 81 to 249 m. The tomographs were interpreted with the aid of soil borings. The repeatability of ERT was evaluated by comparing the rock surface elevations interpreted from pairs of transects where they crossed each other. The average difference was 2.4 m, with a maximum of 10 m. The discrepancy between interpreted bedrock-surface elevations for a transect intersection may be caused by variations in the subsurface geology normal to the transect. Averaging the elevation data interpreted from different transects improved the ERT results. A bedrock surface map was generated using only the averaged elevation data at the transect junctions. The accuracy of the map was further evaluated using data from four exploratory boreholes. The average difference between interpreted and actual bedrock surface-elevations was less than 0.4 m. The map shows two large troughs in the limestone surface: one coinciding with an existing sinkhole basin, while the other is in alignment with a small topographic valley. Because sinkholes were observed at the same elevation interval in similar valleys in the vicinity, the delineated trough may have implications for future land use at the site.

    Notes: Abstract  We analyzed a functional homologue of the swallow gene from Drosophila pseudoobscura. The swallow gene of D. melanogaster plays an essential role in localizing bicoid mRNA in oocytes, and swallow mutant embryos show anterior pattern defects that result from the lack of localization of the bicoid morphogen. The pseudoobscura homologue rescues the function of swallow mutants when introduced into the genome of D. melanogaster, and its expression is similar to that of the melanogaster gene. The predicted pseudoobscura and melanogaster proteins are 49% identical and 69% conserved. The coiled-coil domain previously identified in the melanogaster swallow protein is strongly conserved in the pseudoobscura homologue, but the weak similarity of the melanogaster swallow protein to the RNP class of RNA-binding proteins is not conserved in the pseudoobscura homologue. These and other observations suggest a structural role for swallow in localizing bicoid mRNA, perhaps as part of the egg cytoskeleton.

    Notes: Summary We conducted a controlled crossing experiment to examine the effects of maternal and paternal parentage, the size of the pollen load, and prior fruit production on the proportion of flowers that set fruit, seed number per fruit and seed weight in a natural population of Campanula americana. Effects due to the maternal parent were large for all measures of fruit and seed production, while the paternal parent had a significant effect only upon mean seed weight. As the number of prior fruits on the maternal plant increased the probability that a flower would produce a mature fruit, the number of seeds per fruit, and total seed weight per fruit all decreased. We found no effect of the size of the pollen loads used in this study on fruit or seed production. These results are consistent with those of other studies that suggest in natural plant populations maternal effects, especially environmental maternal effects, can have an overwhelming effect on fruit and seed production and on seed characteristics.

    Notes: Summary Phaseolus coccineus typically has six linearly arranged ovules per ovary. The three ovules near the stylar end of the fruit (positions one, two, and three) are more likely to produce mature seeds, to produce heavier seeds, and to produce more vigorous progeny than the ovules in positions near the peduncular/basal end of the fruit (ovule positions four, five, and six). We conducted a series of field experiments designed to supplement our understanding of the mechanisms determining these position effects. We found that approximately 98% of the ovules in 752 fruits were fertilized — about 0.6% of the stylar ovules were not fertilized, whereas 3.2% of the basal ovules were unfertilized. Moreover, we found that only about 49% of the ovules in these 752 fruits produced mature seeds. Over 60% of the stylar ovules produced mature seeds, whereas only 37% of the basal ovules produced mature seeds. Consequently, the proportion of fertilized ovules cannot explain the differences in seed maturation among the ovule positions. We found that after 6.5 h most of the fertilized ovules were located in the stylar ovule positions, and that there were no fertilized ovules in ovule positions five and six, indicating that the stylar ovules are fertilized first. When only the fastest growing pollen tubes were permitted to enter the ovary (due to exision of the style), only the ovules at the stylar end were fertilized, indicating that the ovule positions that are fertilized first are indeed fertilized by the fastest growing pollen tubes.

    Notes: Summary This study examines the assumption of the pollen competition hypothesis that genetic differences among microgametophytes lead to differences in pollen performance and result in non-random fertilization. In addition, we examined the assumption that pollen performance is genetically correlated with sporophyte vigor due to an overlap in gene expression between the two stages of the life cycle. The results from a pollen mixture experiment in which two cultivars of common zucchini were used show that the ability to sire seeds is nonrandom with respect to the cultivar of the pollen donor plant. The proportion of the progeny sired by the two cultivars is not independent of the region of the fruit where the seeds are produced. The progeny sired by the yellow cultivar outperformed the progeny sired by the green cultivar in a greenhouse study. In addition, the progeny sired by the yellow cultivar from the stylar region of the fruit germinated faster and had more leaf area than the progeny sired by the same cultivar from the peduncular end of the fruit. Thus, the most vigorous progeny are obtained from the stylar region of the fruit where the ovules are fertilized by the most vigorous microgametophytes.
