    Notes: A new approach to gross error detection provides unbiased estimates and 100(1-α)% simultaneous confidence intervals of process variables when biased process measurements and process leaks exist. Presented in this article are estimation equations for process variables, as well as equations that help identify biased measurements and process leaks. These equations include the power function for a global test, and two types of α-level component tests and their power functions. Important strengths and weaknesses of this approach are compared to those of the serial compensation strategy, in particular, by varying the significance level (α), the variance-covariance matrix (Σ), the size of measurement bias (δ), the number of biased variables, and the sample size (N). Accuracy of δ estimation and performance in detecting the presence of process leaks (γ) are also evaluated and compared. The proposed approach has unique features that can provide a basis for improving the reconciliation of variables in process operations.

    Notes: A new plasmid construct has been used in conjunction with selective recycle to successfully maintain otherwise unstable plasmid-bearing E. coli cells in a continuous bioreactor and to produce significant amounts of the plasmid-encoded protein β-lactamase. The plasmid is constructed so that pilin expression, which leads to bacterial flocculation, is under control of the tac operon. The plasmid-bearing cells are induced to flocculate in the separator, whereas cell growth and product synthesis occur in the main fermentation vessel without the inhibiting effects of pilin production. Selective recycle allows for the maintenance of the plasmid-bearing cells by separating flocculent, plasmid-bearing cells from nonflocculent, segregant cells in an inclined settler, and recycling only the plasmid-bearing cells to the reactor. As a result, product expression levels are maintained that are more than ten times the level achieved without selective recycle. All experimental data agree well with theoretical predictions.

    Notes: Recombinant bacterial cells express various levels of model product proteins if the genes of interest are regulated by controllable promoters. The level of gene expression influences the growth-rate differential between plasmid-bearing and plasmid-free cells, and thereby affects the culture dynamics of a plasmid-containing cell population. An expression system has been designed in which host Escherichia coli cells contain the pil operon controlled by a tac promoter; these cells are transformed with plasmids that contain the repressor gene, lacl, for the tac promoter, in combination with an expression system for a model protein, chloramphenicol acetyl tranferase (CAT). Experimental and theoretical results show that plasmid-bearing cells can be maintained as dominant in continuous cultures without selective pressure when 12% or less of the cells' total protein is the model product protein, CAT. This is because the segment cells produce pili greatly in excess of normal wild-type levels, and thus have more of a metabolic burden than do the plasmid-bearng cells that overproduce CAT. However, when the level of the plasmid-directed CAT expression is increased above 12% of the cells' total protein, the growth rate of the plasmid-bearing cells decreases to a value lower than that of the segregant cells. Therefore, plasmid-containing cells lose their selective advantage at this expression level, and cannot be maintained as the dominant cell type in a continuous culture unless antibiotic or other positive selection methods are used. By controlling the growth rate differential of this bacterial host/plasmid system, a variety of interesting competitive culture dynamics is investigated. All experimental measurements for continuous cultures are in very good agreement with theory using kinetic parameters determined from independent batch experiments. © 1992 John Wiley & Sons, Inc.

    Notes: In order to prevent enzymatic degradation of β-casomorphin-5 (1) and morphiceptin, reduced peptide bonds were incorporated at the 2-3 and 3-4 bonds, respectively. The analogues were synthesized by a combination of solid phase methodology and reductive alkylation of resin-bound peptide amines with Boc-amino acid aldehydes (Boc: tert-butyloxycarbonyl) in the presence of NaBH3CN. During reversed phase high pressure liquid chromatography purification, peak shape distortions could be observed. Epimerization was excluded, based on gas chromatography/mass spectroscopy analysis, which indicated acceptable levels of racemization (〈3%) in the crude product. Instead, the phenomena could be attributed to slow cis/ trans isomerizations originating from the Xxx-Pro bonds in the sequence. The presence of different conformational isomers was also established by 1H-nmr spectroscopy in DMSO-d6. All analogues showed high stability in blood plasma, enhanced binding affinity for the μ receptor, and very low binding to the δ receptor. While the Phe3Ψ(CH2-N) Pro4 analogues (3) and (5) displayed agonist activity, the Pro2Ψ(CH2-NH) Phe3 modified analogue (2) showed antagonist activity comparable to D-Phe-Cys-Tyr-D -Trp-Arg-Thr-Pen-Thr-NH2.

Die erhaltenen Xerogele wurden anschließend mit Wasser bis zum Erreichen der Gleichgewichtsquellung behandelt. Um die drei Vernetzungsmittel vergleichen zu können, wurde unter Berücksichtigung von Molmasse und Funktionalität der Xerogele die theoretische Konzentration an Vernetzungsstellen zwischen 0,032 und 0,644 mol/l variiert. Die relative Hydrophobie der drei Vernetzungsmittel hat einen ausschlaggebenden Einfluß sowohl auf die Gleichgewichtsquellung als auch auf den Elastizitätsmodul der gequollenen Gele. Der Gehalt an nicht-einfrierbarem Wasser in mit 1,1,1-Trimethylol-propan-trimethacrylat vernetzten Gelen wurde anhand von DSC-Untersuchungen analysiert. Das Verhältnis von nicht-einfrierbarem zum Gesamtgehalt an Wasser in den Gelen nimmt mit steigender Vernetzungsdichte zu.
    Notes: By means of γ-irradiation 2-hydroxyethyl methacrylate has been copolymerized in bulk with two different tetrafunctional monomers, ethylene glycol dimethacrylate and 1,3-bis(acryloxymethyl) tetramethyl disiloxane and with a hexafunctional monomer, 1,1,1-trimethylolpropane trimethacrylate. The resultant xerogels were subsequently swollen to equilibrium in water. In order to allow a comparison among the three crosslinkers, their molar masses and funtionalities were taken into account, the theoretical crosslink concentrations being varied from 0.032 to 0.644 mol dm-3. The relative hydrophobicity of the three crosslinkers was found to have a significant effect on both the equilibrium swelling and elastic modulus of the swollen gels. Non-freezing water in several gels crosslinked with 1,1,1-trimethylolpropane trimethacrylate was analysed by differential scanning calorimetry (DSC). The proportion of non-freezing water in the gel relative to the overall water content was found to increase with increasing crosslink density.

    Notes: An ultrasensitive method capable of detection and quantification of β-phenylethylamine in 1 ml of human plasma has been developed using gas chromatography/electron capture negative ion mass spectrometry. Phenylethylamine and tetra-deutero phenylethylamine internal standard in plasma were acetylated, extracted into organic solvent and then further acylated with pentafluorobenzoyl chloride. The N-acetyl-N-pentafluorobenzoyl-phenylethylamines were detected by high-resolution single ion monitoring of the molecular ions. Normal plasma levels were found to be 41.5 ± 10.7 pg ml-1, in accordance with results of a previous high-performance liquid chromatographic method.

    Notes: High-performance adhesive bonds require clean, microscopically rough adherend surfaces that are compatible with the adhesive or primer. Such surface allow wetting by the polymer and a complete filling of the pores of ‘nooks and crannies’. The resulting mechanical bonds supplement or supplant the chemical bonds present. For durability in aggressive environments, aluminum bonds that are exposed to high humidity also require hydration-resistant oxide surfaces. Such hydration resistance is inherent in some surfaces, e.g. phosphoric acid-anodized surfaces, and can be high temperatures, such bonds require oxide-free surfaces when exposed to temperatures above 300°C. These surfaces can be formed by plasma spraying a metallic powder coating onto the titanium alloy. Although preparation of high-quality surfaces and bonds is achieved readily in the laboratory, occasional failures are observed in the manufacturing environment owing to, for example, contamination, pH or temperature variations. These processing problems can prevent wetting of the surface or can destroy the microrough morphology. A basic understanding of the mechanisms involved in the production of appropriate surfaces and their resulting adhesive bonds and in their degradation is critical to the evaluation of the failures and their remedy.
