    Notes: Summary Mechanically skinned fibre preparations from the extensor digitorum longus muscle of the rat were used to test whether a rise in myoplasmic [NH4 +] in the range 2–10 mm interferes with the mechanism of excitation-contraction coupling in fast-twitch mammalian muscle. Under our conditions (pH 7.10, Mg2+ 1 mm, temperature 23° C), [NH4 +] up to 10 mm had little effect on the Ca+-activated force and on the peak of the t-system depolarization-induced force response. However, the duration of the depolarization-induced force response was decreased significantly at [NH4 +] ≥2 mm. From these data we conclude that the intracellular accumulation of NH4 + is not likely to play a major role in fatigue. Nevertheless, the build up of NH4 + during fatigue, may have a significant inhibitory effect on the force output by decreasing the duration of the t-system depolarization-induced activation of the contractile apparatus.

    Notes: Abstract Sediments of Lake 382, Experimental Lakes Area, Canada, were sampled at six sites using a 5-cm Wildco KB core sampler (KB), a similar device incorporating a ball check valve (BC), and a 0.2 m by 1.2 m flat-faced aluminum freeze core sampler (FC). Cores were sectioned at 1-cm intervals to a depth of 15 cm. Contaminant (210Pb and 137Cs) concentrations (Bq g-1) were measured by gamma spectroscopy, and inventories (Bq cm-2) were calculated following standard methods. Sediments collected using FC, BC and KB had similar contaminant concentrations, however, cores collected by FC and BC had lower estimated inventories than KB cores. Differences between estimates appear to be caused by differences in the water content (WC) of core material. Laboratory studies confirm that FC sediments have higher WC than tube-cored sediment. We hypothesize that ice crystal formation increases the WC of freeze cores, resulting in lower contaminant inventories. Loss of surficial sediment caused by a ‘bow wave’ may have a similar effect on BC samples. We conclude that KB core gear is appropriate for sampling sediments to measure contaminant concentrations and inventories in recently deposited sediments.

    Notes: Abstract Bioassays using rapid-cycling plants allow measurement of multiple endpoints and assessment of impacts on both growth and reproduction. Selections of Brassica rapa develop rapidly in a broad range of soils and are very consistent in production of flower and seed. Their sensitivity to variation in growth conditions was investigated to define the variables that most affect performance. Yield differences between soils were substantial, indicating the need for careful selection and use of control treatments. The sensitivity to contaminants was investigated with applications of mercury (Hg) and zinc (Zn) to three soils. In a sand soil, bloom initiation was slowed by 〈10 mg Hg kg−1 soil and 〈50 mg Zn kg−1 soil. In contrast, lettuce emergence and earthworm survival were less sensitive to these metals in this soil. Survival of Daphnia magna and the Microtox® assay in soil extracts were more sensitive to Hg than bloom initiation, but less sensitive to Zn. A similar relationship among the bioassays was observed for two finer-textured soils, although for these, effects were usually apparent only at soil metal concentrations 〉200 mg kg−1. Enzyme assays were included for comparison, but were not sensitive to Hg contamination. Rapid-cycling B. rapa selections are suitable for routine bioassays, and are representative of several widely distributed and utilized species.

    Notes: Abstract Soil persistence and lateral movement of 2,4-D (2,4-dichlorophenoxy acetic acid) and picloram (4-amino-3,5,6-trichloropicolinic acid) were examined following their application as a stem-foliage spray for brush control on two power line rights-of-way. Ditches to collect runoff water were located 3, 10, 20, and 30 m downslope from the treated areas. Runoff water and soil samples were collected after 0.14, 0.43, 0.57, 1, 2, 4, 7, 8, 11, 15, 16, and 48 weeks and were analyzed for picloram and 2,4-D residues. Only 3 of 85 soil samples downslope from the target areas contained residues of 2,4-D, and only 1 of 85 down slope samples contained a detectable residue of picloram. Of 56 runoff water samples, only 11 contained 2,4-D residues and only 1 contained residues of picloram. The greatest distances downslope at which residues were detected in runoff water were 20 and 10 m for 2,4-D and picloram, respectively. No residues of either herbicide were recovered in soil or water at 15 weeks or 48 weeks after spraying. Despite normal rainfall frequency and amounts in the first several weeks after spraying in mid-June, significant runoff of either herbicide was not evident at either study site.

    Notes: Summary In a survey of 528 unrelated haemophilia A patients, six partial deletions of the factor VIII (FVIII) gene were detected by Southern blotting. These deletions were further mapped by a combination of Southern blotting and polymerase chain reaction amplification and found to vary in length between 4.7kb and 57kb. The frequency of detectable FVIII gene deletions (about 1%) is thus considerably lower than previously reported. Statistical analysis of currently available data did not provide any evidence for a deletion “hotspot”. Four of the six deletion patients reported here possessed inhibitors. Taken together with previous data, deletion of the FVIII gene was found to be associated with an approximately fivefold higher risk of developing inhibitors compared with other severe haemophiliacs without gene deletions.

    Description / Table of Contents: Zusammenfassung Mehrere Formationen im unteren Teil der Appin Gruppe der Dalradians sind durch karbonatische und kalk-silikatische Lithologien gekennzeichnet. Amphibole treten in diesen metasedimentären Einheiten in verschiedener Ausbildung auf, als feinkörnige, disseminierte Kristalle, als radialstrahlige, die Gesteine durchwachsende Aggregate und in dunkelgrünen, zu mehr als 90% aus Amphibolen bestehenden Lagen. Elektronenstrahl-Mikrosondenuntersuchungen belegen einfache mineralchemische Zusammensetzungen, die Tremolite, tremolitische Hornblende und Magnesio-Hornblende umfaßt. Die einzige bedeutende Substitution ist die zwischen Tremolit und Alumino-Tschermakit [MgSi ⇌ AlviAliv] und zwischen Tremolit und Ferri-Tschermakit [MgSi ⇌ FeviAliv]. Im Gegensatz dazu zeigen Amphibole aus den nahegelegenen Metavulkaniten und intrusiven Metadoleriten vier Substitutionstrends von Tremolit zu Ferro-Akttinolith, Edenit, Alumino-Tschermakit und Ferri-Tschermakit. Diese unterschiedlichen Trends könnten auch anderswo bei der Unterscheidung zwischen Para- und Orthoamphiboliten behilflich sein. Tremolitische Amphibole machen normalerweise einen Großteil der kalk-silikatischen Gesteine aus, in denen ansonsten nur Quarz ± Phologpit auftritt. Karbonate fehlen bemerkenswerterweise. Dies weist auf Mg-reiche sedimentäre Protolithe, wie dolomitische Mergel mit möglicherweise etwas Magnesit, hin. Sedimentologische Überlegungen lassen die Ablagerung in abgeschnürten Küstenlagunen tropischer Breiten, die Teil eines weiträumigen stabilen Schelfes waren, vermuten.
    Notes: Summary Several formations within the lower part of the Dalradian Appin Group are characterised by calcareous and calc-silicate lithologies. Amphibole occurs in these metasedimentary units in a variety of habits, from fine-grained disseminated crystals, through radiating aggregates permeating the whole rock, to dark green layers consisting of over 90% amphibole. Electron microprobe analyses of these amphiboles exhibit simple compositional trends from tremolite, through tremolitic hornblende to magnesio-hornblende. The only significant substitutions are from tremolite towards alumino-tschermakite [MgSi ⇌ AlviAliv] and from tremolite towards ferri-tschermakite [MgSi ⇌ FeviAliv]. This is in marked contrast to amphiboles from nearby suites of metavolcanic rocks and intrusive metadolerites which exhibit four coupled substitutions, leading from tremolite towards the ferro-actinolite, edenite, alumino-tschermakite and ferritschermakite end-members. These distinctive compositional trends may aid the distinction between para- and ortho-amphibolites elsewhere. Tremolitic amphibole commonly constitutes a high proportion of the calc-silicate rocks, the only other phases present being quartz ± phlogopite. Carbonate minerals are notably absent. This suggests that the sedimentary protolith was high in Mg, such as a dolomitic marl, possibly containing some magnesite. Sedimentological considerations suggest deposition in tropical latitudes in restricted coastal lagoons forming part of an extensive stable shelf bordering a mature landscape.

    Notes: Abstract Atomic emission spectroscopy (AES) is used to monitor volatilization during waste vitrification with thermal plasmas. Model baste specimens consist of am alumino-silicate clay spiked with 10 wt. % Fe, Ni, or Cr. Specimens are vitrified in processing atmospheres with 0, 8.5, and 17 vol. % oxygen/argon concentrations. Particulate generated from condensation of the volatilized materials is collected following each specimen run and analyzed by energy dispersive X-ray analysis to determine composition. Spectra are collected from the ultraviolet range 240–380 nm in where the presence of metal vapor is readily detected. Correlation between line emission intensities and volatilization rates allows the establishment of provisional detection limits for the volatilization of Ni, Cr, Si, and Fe, of 1 × 103 g/s, 4 × 104 g/s, 2 × 104 g/s, and 5 × 105 g/s, respectively. The results of this investigation support the concept of using AES as an in-situ process monitor for feedback to optimize plasma processing of hazardous metal-containing waste.

    Notes: Abstract   We examined the effects of pollen selection for rapid pollen-tube growth on progeny vigor. First, we crossed a wild gourd (Cucurbita texana) to a cultivated zucchini (Cucurbita pepo cv `Black Beauty') to produce an F1 and then an F2 generation. Half of the F1 seeds were produced by depositing small loads of C. texana pollen onto the stigmas of C. pepo. These small pollen loads were insufficient to produce a full complement of seeds and, consequently, both the fast- and the slow-growing pollen tubes were permitted to achieve fertilization. An F2 generation was then produced by depositing small loads of F1 pollen onto stigmas of F1 plants. The F2 seeds resulting from two generations of small pollen loads are termed 'the non-selected line' because there was little or no selection for pollen-tube growth rate on these plants. The other half of the F1 and F2 seeds were produced by depositing large pollen loads (〉10000 pollen grains) onto stigmas and then allowing only the first 1% or so of the pollen tubes that entered the ovary to fertilize the ovules. We did this by excising the styles at the ovary at 12–15 h after pollination. The resulting F2 seeds are termed `the selected line' because they were produced by two generations of selection for only the fastest growing pollen tubes. Small pollen loads from the F2 plants, both the selected and the non-selected lines, were then deposited onto stigmas of different C. pepo flowers, and the vigor of the resulting seeds was compared under greenhouse and field conditions. The results showed that the seeds fertilized by pollen from the selected line had greater vegetative vigor as seedlings and greater flower and fruit production as mature plants than the seeds fertilized by pollen from the non-selected line. This study demonstrates that selection for fast pollen-tube growth (selection on the microgametophyte) leads to a correlated increase in sporophyte (progeny) vigor.
