    Notes: Abstract Previous studies from our laboratory have demonstrated the presence of complex alterations in the activities of antioxidant enzymes in various tissues of rats with streptozotocin (STZ)-induced diabetes. In the present investigation, it is shown that rats made diabetic with alloxan (ALX), an agent differing from STZ both chemically and in its mechanism of diabetogenesis, show virtually identical tissue antioxidant enzyme changes which, as is the case with STZ, are preventable by insulin treatment. The finding that the patterns of antioxidant enzyme alterations in chemically-induced diabetes are independent of the diabetogenic agent used and the presence of similar abnormalities in tissues of spontaneously diabetic (BB) Wistar rats (particularly when diabetic control is less than optimal) suggest that the changes observed are a characteristic feature of the uncontrolled diabetic state and that these may be responsible for (or predispose to) the development of secondary complications in clinical diabetes. Comparative studies involving red cells of diabetic rats and human diabetics revealed a number of common changes, namely an increase in glutathione reductase activity, a decreased susceptibility to oxidative glutathione depletion (which was related to the presence of hyperglycemia) and an increased production of malondialdehyde (an indirect index of lipid peroxidation) in response to in vitro challenge with hydrogen peroxide. In the diabetic patients, the extent of this increase in susceptibility of red cell lipids to oxidation paralleled the severity of diabetic complications. Our results suggest that increased (or uncontrolled) oxidative activity may play an important role in the pathogenesis of complications associated with the chronic diabetic state.

    Notes: Abstract Ethane production was evaluated as a method for assessing freeze damage to loblolly pine (Pinus taeda L.) seedlings by comparing it to the widely used electrolyte leakage method. Paired measurements, first ethane production and then electrolyte leakage, were conducted on the pooled needle samples at temperatures between 0° and −12°C. Ethane production rates increased in a linear fashion with decreasing temperatures between 0° and −12°C for both Virginia Coastal Plain (R2=0.80) and Marion County, Florida (R2=0.87) seed sources. The Florida seedlings were consistently 2° to 4°C higher than the Virginia seedlings at a given ethane level. Electrolyte leakage expressed as Index of Injury initially increased with decreasing temperatures, but then leveled off at or decreased below −8°C. The log-log linear regression of ethane production against Index of Injury indicated good correspondence for both seed sources (Virginia − R2=0.81; Florida − R2=0.91). Ethane production appears more rapid and to require less sampling than does electrolyte leakage while producing comparable results to the electrolyte leakage method.

    Notes: Abstract Most organisms are constantly exposed to molecular oxygen, and this has become a requirement of life for many of them. Oxygen is not totally innocuous, however, and it has long been known to be toxic to many organisms, including humans. The deleterious effects of oxygen are thought to result from its metabolic reduction to highly reactive and toxic species, including superoxide anion radical and hydroxyl radical. Peroxidation of lipids is a major consequence of exposure to these species and the cell possesses various enzymes, including superoxide dismutase and catalase, as well as cellular antioxidants which are able to scavenge oxygen free radicals and repair peroxidized lipids. These aspects of oxygen toxicity are reviewed, as well as the involvement of oxygen free radicals in the toxicity of the herbicide paraquat.
