Description: The search for new congeners of the leading anticancer drug doxorubicin has led to an analog that is approximately 1000 times more potent, noncardiotoxic at therapeutic dose levels, and non-cross-resistant with doxorubicin. The new anthracycline, 3'-deamino-3'-(3-cyano-4-morpholinyl)doxorubicin (MRA-CN), is produced by incorporation of the 3' amino group of doxorubicin in a new cyanomorpholinyl ring. The marked increase in potency was observed against human ovarian and breast carcinomas in vitro; it was not accompanied by an increase in cardiotoxicity in fetal mouse heart cultures. Doxorubicin and MRA-CN both produced typical cardiac ultrastructural and biochemical changes, but at equimolar concentrations. In addition, MRA-CN was not cross-resistant with doxorubicin in a variant of the human sarcoma cell line MES-SA selected for resistance to doxorubicin. Thus antitumor efficacy was dissociated from both cardiotoxicity and cross-resistance by this modification of anthracycline structure.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Sikic, B I -- Ehsan, M N -- Harker, W G -- Friend, N F -- Brown, B W -- Newman, R A -- Hacker, M P -- Acton, E M -- CA 24543/CA/NCI NIH HHS/ -- CA 32250/CA/NCI NIH HHS/ -- CA 33303/CA/NCI NIH HHS/ -- New York, N.Y. -- Science. 1985 Jun 28;228(4707):1544-6.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/4012308" target="_blank"〉PubMed〈/a〉

Description: To test the hypothesis that interfamily variability in Huntington's Disease (HD) is due to mutation at different loci, linkage analysis was undertaken in two large HD kindreds that differed in ethnicity, age-at-onset, and neurologic and psychiatric features. Both families showed linkage of the HD locus to the G8 probe. Several recombinants were documented in each family, and the best estimate of the recombination fraction for the two families was 6 percent with a 95 percent confidence interval of 0 to 12 percent. Although the data support the existence of a single HD locus, use of the G8 probe for presymptomatic testing in these kindreds would have resulted in a 12 percent error rate in genotype assignment at the HD locus.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Folstein, S E -- Phillips, J A 3rd -- Meyers, D A -- Chase, G A -- Abbott, M H -- Franz, M L -- Waber, P G -- Kazazian, H H Jr -- Conneally, P M -- Hobbs, W -- AM 13983/AM/NIADDK NIH HHS/ -- NS 16367/NS/NINDS NIH HHS/ -- NS 16375/NS/NINDS NIH HHS/ -- New York, N.Y. -- Science. 1985 Aug 23;229(4715):776-9.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/2992086" target="_blank"〉PubMed〈/a〉

Description: A new approach to in situ observations of trace reactive species in the stratosphere is described. A balloon-borne system, floating 40 kilometers above the earth's surface, successfully lowered and then retracted a cluster of instruments a distance of 12 kilometers on a filament of Kevlar. This instrument cluster is capable of detecting gas-phase free radicals at the part-per-trillion level. The suspended instrument array has excellent stability and has been used to measured atomic oxygen concentrations in the stratosphere.

Description: A number of prominent Q-branches of the nu-7 band of C2H6 have been identified near 3000/cm in aircraft and ground-based infrared solar absorption spectra. The aircraft spectra provide the column amount above 12 km at various altitudes. The column amount is strongly correlated with tropopause height and can be described by a constant mixing ratio of 0.46 ppbv in the upper troposphere and a mixing ratio scale height of 3.9 km above the tropopause. The ground-based spectra yield a column of 9.0 x 10 to the 15th molecules/sq cm above 2.1 km; combining these results implies a tropospheric mixing ratio of approximately 0.63 ppbv.

Description: The ozone fluxes and flux convergences within the region between 30 and 0.4 mbar for the Northern Hemisphere are investigated for the period January 1 to March 31, 1979, using Nimbus 7 solar backscatter ultraviolet ozone profiles and the meteorological data derived from NOAA satellites. The results indicate that large contributions to the ozone transport and the transport convergence are due to mean vertical advection and horizontal eddy processes. The contribution from the vertical eddies is insignificant. The transport of ozone into the polar latitudes during winter and early spring is mainly accomplished by the large-scale horizontal eddies. The results also show that more ozone is transported into the polar region by the large-scale eddies on disturbed days than on quiet days.

Description: Measurements of the nighttime atmospheric sodium layer have been performed since 1977, using a dye lidar at Fukuoka (33.4 deg N, 130.2 deg E.). The Kyushu lidar uses a flashlamp pumped dye laser tuned to the sodium D sub 2 line (589.0 nm) as a transmitter. The dye laser used in the Kyushu lidar system has been described elsewhere. Sporadic enhancements of the total column abundance of the layer during the Perseids meteor shower were observed on the nights of August 12 and 13, 1978, 1979, 1981, and 1983. Degrading weather conditions prevented observation on the nights during 1980 and 1983. A contour plot of the sodium layer for the period of 8 hours from 21:00 to 5:00 JST on the night of August 12 to 13 1983, is shown. The predominant feature visible is the increase in the height of peak density from 21:00 to 2:00 JST, and the slight decrease in the height of peak density from 2:00 to 5:00 JST. The average abundance had a maximum of about 7 x 10 to the 13th power/sq m on August 13. A plot is presented of the nightly average abundance for available data. The sodium layer abundance increased on the night of August 12 to 13, 1983, during the peak of the Perseids meteor shower, and the following night, August 13 to 14, almost went back to the monthly mean.

Description: Stratospheric chlorine monoxide concentrations were measured in situ using a new and improved observational method in which the detection package is lowered on a tether from a balloon-borne platform floating near 40 km altitude and then reeled back up. Two reel down and reel up sequences covering 38 to 31 km produced (ClO) profiles which agree with each other to + or - 12 percent. The measured (ClO) profile falls within the envelope of nine previous fast parachute drop results, but exhibits a different shape, peaking near 32 km as opposed to approximately 39 km for the average of the previous profiles. These results are lower than but consistent with the profiles derived from remote sensing methods, and indicate the need for simultaneous measurements of relevant species.

Description: The present study has the objective to document more fully a somewhat surprising heating pattern for 10 of the bow shock crossings investigated by Russell et al. (1982). A description of the detailed evolution of the actual distribution functions across the subcritical shocks is also provided. It is pointed out that the shocks examined are all very near the theoretical one-fluid critical Mach number. None of the shocks has a significant magnetic foot or overshoot. It is argued that the observed heating is not due to reflected ions but rather to physical processes characteristic of subcritical shocks.

Description: Global monthly mean charts for both hemispheres are given for four mid-season months, and for the pressure levels 30, 10, 1, and 0.1 mbar for temperature and 0.4 mbar for ozone. Charts of total ozone are provided separately. This set of charts shows clearly the very close coupling between the temperature and ozone distributions and demonstrates the influence of the large-scale planetary waves which give rise to very large longitudinal variations. The regular and interannual variability of temperature and ozone are discussed.
