    Description: Ovarian cancer is the leading cause of death among gynecological cancers, but is poorly amenable to preoperative diagnosis. In this study, we investigate the feasibility of “optical biopsy,” using high-optical-resolution photoacoustic microscopy (OR-PAM) to quantify the microvasculature of ovarian and fallopian tube tissue. The technique is demonstrated using excised human ovary and fallopian tube specimens imaged immediately after surgery. Quantitative parameters are derived using Amira software. The parameters include three-dimensional vascular segment count, total volume and length, which are associated with tumor angiogenesis. Qualitative results of OR-PAM demonstrate that malignant ovarian tissue has larger and more tortuous blood vessels as well as smaller vessels of different sizes, while benign and normal ovarian tissue has smaller vessels of uniform size. Quantitative analysis shows that malignant ovaries have greater tumor vessel volume, length and number of segments, as compared with benign and normal ovaries. The vascular pattern of benign fallopian tube is different than that of benign ovarian tissue. Our initial results demonstrate the potential of OR-PAM as an imaging tool for fast assessment of ovarian tissue and fallopian tube and could avoid unnecessary surgery if the risk of the examined ovary is extremely low.

    Notes: Abstract The production of photons ine + e −→γ+hadrons is investigated at three centre of mass energies around 14, 22 and 34 GeV. On average, photons carry 25% of the total available energy, with a multiplicity similar to the charged multiplicity. The inclusive photon spectra are found to scale with the centre of mass energy as a function of the Feynman variablex. π0 and η mesons are reconstructed from their decay photons. The slopes of the spectra are similar to that for charged pions and approximate scaling is observed for π0 production. The mean π0 and η multiplicities are given. The observed photon yield can be fully accounted for by hadron decays and initial state radiation. However, up to one extra photon per event from other sources cannot be excluded.

    Notes: Abstract  The fibrogenic and cytotoxic potential of coal mine dust is independent of the amount of quartz and other inorganic parameters. Results of coal petrographical and organic geochemical investigations of coals and coal mine dust from the Ruhr and Ibbenbüren Region of Germany demonstrate variations of organic dust amount possibly influencing these noxious properties. Coal mine dust of high rank coals is characterized by a pronounced fibrogenic risk. This risk, independent of the quantity of quartz, is probably based on shape variations of different coal macerals. With increasing coalification of the corresponding seam, the vitrinite is enriched in its dust; however, lower concentrations have been determined for inertinite. Vitrinite shows constant shapes and sizes independent of the rank of coal. Inertinite particles with elongated to fibrous shapes tend to larger sizes with increasing coalification. Strikingly, coal mine dust from miners' lungs with high degrees of coal mine workers' pneumoconiosis (CWP) is enriched in inertinite. In contrast, high cytotoxicities in cell tests are known for coal mine dust from low coalified coals. High concentrations of phenolic compounds can be extracted by dichloromethane from low coalified coal mine dust. These compounds, which are characterized by a high water solubility and therefore high bioavailability, explain the high cytotoxicities of coal mine dust. Contamination of dust by diesel emissions in the coal mine can act as additionally supporting parameters for extended cytotoxicities.

    Notes: Abstract  The representation of hydrological land surface processes has, so far, been treated inadequately in global models of the atmospheric general circulation (GCMs). In particular the lateral waterflows from the continents into the ocean have been described unsatisfactorily. The aim of this study is to develop a model for the lateral waterflow on the global scale which describes the translation and retention of the lateral discharge as a function of the spatially distributed land surface characteristics that are globally available. Here, global scale refers to the resolution of 0.5° and smaller, corresponding to a typical GCM gridbox area of about 2500 km2. Discharge models need a number of specific input variables that are usually not available from measurements such as runoff and drainage. Therefore, these variables have to be derived from observed data such as precipitation and surface air temperature. For the model development in this study, a new simplified land surface scheme was applied to compute these variables. The discharge from a catchment of approximately the size of a 0.5° gridbox was simulated using several types of models. The intercomparison of the simulated lateral flows of the different models shows that as a minimum a separation between flow processes such as overland flow, baseflow and riverflow is required to yield good discharge simulations. As both the retention and translation of a flow process need to be simulated, a two-parameter model is required for the representation of overland flow and riverflow. For the baseflow, a one-parameter model is sufficient. The resulting model structure is called the hydrological discharge model or HD model. A first parametrization approach was defined using the gridbox characteristics of topography gradient and gridbox length. The skill of the discharge simulation depends not only on the formulations of the model, but also on the precise definition of the boundaries of the model catchments. The sizes and the positioning of the model catchments on the globe were defined by using a river direction file as well as a modified topography dataset. In a first application, the HD model successfully simulated river discharge using runoff and drainage from a five year atmospheric GCM integration (ECHAM4-T42) as input. The annual cycles of the monthly means of the simulated discharge of several large rivers were compared to the observed discharges provided by the Global Runoff Data Centre. The HD model achieves a considerable improvement of the simulated discharge compared to the model which is currently operational at MPI. The improved volume of the discharge is directly related to the definition of more realistic model catchments and the improved timing of the flow is mainly due to the newly introduced separation of the flow processes.

    Notes: Summary The present study describes the intracellular distribution of Tamm-Horsfall protein (THP) in rat kidney. The localization was determined by immunoelectron microscopy using the protein A-gold technique. Various fixation and embedding protocols were evaluated for this purpose. Brief perfusion fixation (3 min) with 1% glutaraldehyde and embedding in a highly hydrophilic glycol methacrylate-polyester mixture were most appropriate for antigen-antibody recognition and structural preservation. The overall tissue distribution of THP was evaluated by indirect immunofluorescence microscopy; reaction was strong along the entire thick ascending limb of the loop of Henle (TAL) with enhanced fluorescence in the apical cytoplasm. On the electron microscopic level immunogold labelling was concentrated over numerous membrane-bound vesicles which form a compartment in the apical cytoplasm. The Golgi region was consistently labelled, whereas the plasma membranes revealed only sporadic labelling at the luminal side, and basolateral membranes were mostly unlabelled. Quantitative evaluation of the gold labelling, which was separately done for the inner stripe, outer stripe and cortical TAL, consistently showed the highest particle density in the apical cytoplasm. Middle and basal levels in the TAL cells were only moderately labelled. The results are discussed with respect to the current opinion which describes THP as a membrane glycoprotein. We speculate that the accumulation of THP in the apical vesicular compartment of TAL cells indicates a storage site of the protein, possibly prior to extrusion via exocytosis of the vesicle contents.

    Notes: Abstract Accumulation of exogenously supplied osmoprotective compounds was analyzed in the cyanobacterium Synechocystis sp. PCC6803, which synthesizes glucosylglycerol as the principal osmoprotective compound. Glucosylglycerol and trehalose were accumulated to high levels and protected cells of a mutant unable to synthesize glucosylglycerol against the deleterious effects of salt stress. In the wild-type, uptake of trehalose repressed the synthesis of glucosylglycerol and caused metabolic conversion of originally accumulated glucosylglycerol. Trehalose cannot be synthesized by Synechocystis and was not or only insignificantly metabolized. Sucrose, which can be synthesized in low quantities by Synechocystis, was also taken up, as indicated by its disappearance from the medium. Sucrose was not accumulated to high levels, probably due to a sucrose-degrading activity found in cells adapted to both low- and high-salt conditions. Despite its low intracellular concentration, sucrose showed a weak osmoprotective effect in salt-shocked cells of a mutant unable to synthesize glucosylglycerol.

    Notes: Summary Green-white variegated plants of the pm line of Oenothera hookeri were reported in the literature to be due to the action of a nuclear recessive ‘plastome mutator’ (pm). The plastid DNAs of green and white leaves have been studied by restriction analysis and Southern hybridization. Remarkable differences in restriction pattern have been found between them. The green plastids show the typical pattern of plastome I; the white plastids give the pattern of plastome III. The conclusion from our experiments is: variegated ‘pm’ plants contain two genetically different types of plastids. Their variegation is not due to nuclear gene-induced plastome mutations, but is the result of hybrid bleaching of the type III plastids, which were combined with type I plastids following — as we suppose-inadvertent pollination.

    Notes: Abstract The genome of Drosophila bifasciata harbours two distinct subfamilies of P-homologous sequences, designated M-type and O-type elements based on similarities to P element sequences from other species. Both subfamilies have some general features in common: they are of similar length (M-type: 2935 bp, O-type: 2986 bp), are flanked by direct repeats of 8 by (the presumptive target sequence), contain terminal inverted repeats, and have a coding region consisting of four exons. The splice sites are at homologous positions and the exons have the coding capacity for proteins of 753 amino acids (M-type) and 757 amino acids (O-type). It seems likely that both types of element represent functional transposons. The nucleotide divergence of the two P element subfamilies is high (31%). The main structural difference is observed in the terminal inverted repeats. Whereas the termini of M-type elements consist of 31 by inverted repeats, the inverted repeats of the O-type elements are interrupted by non-complementary stretches of DNA, 12 by at the 5′ end and 14 by at the 3′ end. This peculiarity is shared by all members of the O-type subfamily. Comparison with other P element sequences indicates incongruities between the phylogenies of the species and the P transposons. M-type and O-type elements apparently have no common origin in the D. bifasciata lineage. The M-type sequence seems to be most closely related to the P element from Scaptomyza pallida and thus could be considered as a more recent invader of the D. bifasciata gene pool. The origin of the O-type elements cannot be unequivocally deduced from the present data. The sequence comparison also provides new insights into conserved domains with possible implications for the function of P transposons.

    Notes: Abstract A search has been made for the chargino, the supersymmetric partner of theW ± (or the partner of the charged Higgs bosons), ine + e − annihilation at PETRA using the JADE detector. The chargino decay modes investigated are $$l\tilde v, l\bar v\tilde \gamma , q_1 \bar q_2 \tilde \gamma$$ and $$q_1 \bar q_2 \tilde g$$ (with subsequent gluino decay $$\tilde g \to q\bar q\tilde \gamma$$ ). No evidence for such events has been observed. The 95% C.L. lower limit for the chargino mass is typically 22.5 GeV, depending only slightly on the masses of other supersymmetric particles. If the chargino is stable, the lower mass limit is 21.1 GeV (95% C.L.).

    Notes: Abstract A salt shock of 684mm NaCl reduced RNA and DNA synthesis to about 30% of the control level inSynechocystis. DNA synthesis recovered to the initial level within 4 h, while for recovery of RNA synthesis about 8 h were necessary. In cells completely adapted to different salt concentrations (from 171 to 1026mm NaCl), a continuous decrease in the RNA content with increasing salt concentrations up to 684mm NaCl was found, whereas the lowest DNA content was measured around 342mm NaCl, i.e., the salinity at which maximal growth occurred. With the uracil and thymidien incorporation technique, maxima in DNA and RNA synthesis were detected in control cells. Comparing these rates with nucleic acid synthesis rates calculated from the contents of DNA and RNA and the growth rates indicated that adaptation to 1026mm NaCl seemed to lead to an increased RNA turnover inSynechocystis. Analysis of protein synthesis with35S-methionine labeling showed alterations in salt-adapated cells ofSynechocystis. At least three proteins (20.5, 25.8, and 35.8 kDa) were synthesized with highest rates at salinities leading to maximal growth, the synthesis of nine proteins (12.5, 16.9, 19.2, 22.2, 24.7, 28.5, 30.5, 50.3, and 63.5 kDa) increased and that of several other proteins decreased with increasing salinity; but only three proteins (12.5, 22.2, and 30.5 kDa) accumulated under these conditions. The adaptation ofSynechocystis to enhanced salt concentrations led also to increased contents of glucosylglycerol, glycogen, and significant amounts of K+ as well as Na+ ions.
