    Notes: An anion exchange column of DE-52 has been used to purify Eimeria sporozoites from a post-excystation mixture of oocysts, oocyst shells, sporocysts, sporocyst shells, and sporozoites. The mean recovery from several experiments was 94% and virtually all non-sporozoite material was removed. Infectivity studies in vitro with sporozoites showed that they were viable after purification and were at least as infectious as the unpurified sporozoites; furthermore, oocysts in the crude preparation could be recovered from the DE-52 cellulose by resuspending them in a 20% (w/v) sodium chloride solution.

    Notes: Abstract Seven dives in the submersible ALVIN and four deep-towed (ANGUS) camera lowerings have been made at the eastern ridge-transform intersection of the Oceanographer Transform with the axis of the Mid-Atlantic Ridge. These data constrain our understanding of the processes that create and shape the distinctive morphology that is characteristic of slowly-slipping ridge-transform-ridge plate boundaries. Although the geological relationships observed in the rift valley floor in the study area are similar to those reported for the FAMOUS area, we observe a distinct change in the character of the rift valley floor with increasing proximity to the transform. Over a distance of approximately ten kilometers the volcanic constructional terrain becomes increasingly more disrupted by faulting and degraded by mass wasting. Moreover, proximal to the transform boundary, faults with orientations oblique to the trend of the rift valley are recognized. The morphology of the eastern rift valley wall is characterized by inward-facing scarps that are ridge-axis parallel, but the western rift valley wall, adjacent to the active transform zone, is characterized by a complex fault pattern defined by faults exhibiting a wide range of orientations. However, even for transform parallel faults no evidence for strike-slip displacement is observed throughout the study area and evidence for normal (dip-slip) displacement is ubiquitous. Basalts, semi-consolidated sediments (chalks, debris slide deposits) and serpentinized ultramafic rocks are recovered from localities within or proximal to the rift valley. The axis of accretion-principal transform displacement zone intersection is not clearly established, but appears to be located along the E-W trending, southern flank of the deep nodal basin that defines the intersection of the transform valley with the rift floor.

    Notes: Abstract During the Fall of 1979, a manned submersible program, utilizing DSRV ALVIN, was carried out at the intersection of the East Pacific Rise (EPR) with the Tamayo Transform boundary. A total of seven dives were completed in the vicinity of the EPR/Tamayo intersection depression and documented the geologic relationships that characterize the juxtaposition of these types of plate boundaries. The young volcanic terrain of the EPR axis can be traced into and across the Tamayo Transform valley but becomes buried by sedimentary talus that is being shed from sediment scarps along the unstable sediment slope that defines the north side of the intersection depression. Within 4 km of the transform boundary, the dominant trend (000°) of the fissures and faults that disrupt the rise-generated volcanics is markedly oblique to the regional direction of sea floor spreading (120°). Since no evidence was found to suggest that these structures accommodate significant amounts of strike-slip displacement, they are taken to reflect a distortion of the EPR extensional tectonic regime by a transform generated shear couple. The floor of the Tamayo Transform valley in this area is inundated by mass-wasted sediment, and the principal transform displacement zone is characterized at the surface by a narrow (〈1.5 km) interval of fault scarps in sediment that trends parallel with the transform valley. Extrapolated to the west, this zone links with zones of transform deformation investigated during earlier submersible studies (CYAMEX and Pastouret, 1981). Evidence of low-level hydrothermal discharge was seen at one locality on the EPR axis and at another 8 km west of the axis at the edge of the zone of transform deformation.

    Notes: Summary A comparative study has been carried out on the micro-localization of catalase in mouse tissues subsequent to treatment with a representative range of hypolipidemic drugs. A commonality of effect was shown by clofibrate (ethyl-α-p-chlorophenoxyisobutyrate), Wy-14,643 (4-chloro-6-[2,3 xylidino)-2-pyrimidinylthio] acetic acid), RMI-15,414 (5-tetradecyloxy-2-furancarboxylic acid) and aspirin (acetyl salicylic acid), in that treatments with each of these drugs was associated with the release of peroxisomal catalase into the cytoplasmic compartment of liver and kidney. It was also noticeable that this increased cytosolic activity was characterized by the presence of an ‘aged’ form of the enzyme with different mobility and activity characteristics to that of the peroxisomal enzyme. Possible molecular bases for these effects and their relationship to peroxisomal biogenesis are discussed.

    Notes: Summary Recent investigations into the role of peroxisomes in mammalian lipid metabolism have employed double isotope methodologies to examine the influence of peroxisomal agents on lipid turnover in the liver and extra hepatic tissues of the living animal. The action of these agents, all of which caused extensive changes in the flux of lipid metabolism in the treated animals, may best be viewed in relation to their effects on the common pathway of fatty acid oxidation in peroxisomes. Clofibrate, for example, acts through induction of peroxisomal oxidases and catalase; glycolate and ethanol through activation of this pathway; and aminotriazole and allylisopropylacetamide through inhibition of the catalase step in the sequence. The data from these studies provide support for the concept of an important contributory and regulatory role of peroxisomes in relation to the overall balance of lipid metabolism, and emphasize that these organelles play a significant role in the oxidation of common fatty acids, as well as a potential for the elimination of fatty acids that are poorly oxidized by mitochondria. Additionally, the data raise intriguing questions on the extension of peroxisomal influence to include phospholipid metabolism and the substantial degree of inter-tissue communication which is involved in the balance of lipid metabolism in the whole animal.

    Notes: Summary Oxidation of NADH by rat erythrocyte plasma membrane was stimulated by about 50-fold on addition of decavanadate, but not other forms of vanadate like orthovanadate, metavanadate aad vanadyl sulphate. The vanadate-stimulated activity was observed only in phosphate buffer while other buffers like Tris, acetate, borate and Hepes were ineffective. Oxygen was consumed during the oxidation of NADH and the products were found to be NAD+ and hydrogen peroxide. The reaction had a stoichiometry of one mole of oxygen consumption and one mole of H2O2 production for every mole of NADH that was oxidized. Superoxide dismutase and manganous inhibited the activity indicating the involvement of superoxide anions. Electron spin resonance in the presence of a spin trap, 5, 5′-dimethyl pyrroline N-oxide, indicated the presence of superoxide radicals. Electron spin resonance studies also showed the appearance of VIV species by reduction of VV of decavanadate indicating thereby participation of vanadate in the redox reaction. Under the conditions of the assay, vanadate did not stimulate lipid peroxidation in erythrocyte membranes. Extracts from lipid-free preparations of the erythrocyte membrane showed full activity. This ruled out the possibility of oxygen uptake through lipid peroxidation. The vanadate-stimulated NADH oxidation activity could be partially solubilized by treating erythrocyte membranes either with Triton X-100 or sodium cholate. Partially purified enzyme obtained by extraction with cholate and fractionation by ammonium sulphate and DEAE-Sephadex was found to be unstable.

    Notes: Abstract Carrot (Daucus carota L.) cells grown in suspension culture oxidized exogeneous NADH. The NADH oxidation was able to stimulate K+ (86Rb+) transport into cells, but it did not affect sucrose transport.N,N'-Dicyclohexyl-carbodiimide, diethylstilbestrol, and oligomycin, which only partially inhibited NADH oxidation, almost completely collapsed the K+ (86Rb+) transport. Vanadate, which is less effective as an ion transport inhibitor, was less effective in inhibiting the NADH-driven transport of K+ (86Rb+).p-Fluormethoxycarbonylcyanide phenylhydrazone inhibits the K+ transport over 90% including that induced by NADH. The results are interpreted as evidence that a plasma membrane redox system in root cells is closely associated with the ATPase which can drive K+ transport. Because of the inhibitor effects, it appears that membrane components common to the redox system and ATPase function in the transport of K+.

    Notes: Abstract Doxorubicin (adriamycin) is cytotoxic to cells, but the biochemical basis for this effect is unknown, although intercalation with DNA has been proposed. This study suggests that the cytotoxicity of this drug may be due to inhibition of the plasma membrane redox system, which is involved in the control of cellular growth. Concentrations between 10−6–10−7 M adriamycin inhibit plasma membrane redox reactions 〉50%. AD32, a form of adriamycin which does not intercalate with DNA, but is cytotoxic, also inhibits the plasma membrane redox system. Thus, the cytotoxic effects of adriamycin, which limit its use as a drug, may be based on the inhibition of a transplasma membrane dehydrogenase involved in a plasma membrane redox system.
