    Description / Table of Contents: Zusammenfassung Der Ecdysteroidtiter weiblicher Puppen von Xyleborus ferrugineus (Fabr.) wurde geschätzt, indem ganze Tiere homogenisiert und radioimmunologisch untersucht wurden. Ein ausgeprägtes Maximum an Ecdysteroiden wurde bei 36 Stunden Puppenent-wicklung beobachtet (743 pg/mg Körpergewicht). Der Titer nahm ab auf 299 pg/mg im Pharatstadium und auf 193 pg/mg unmittelbar vor Schlüpfen der Adulten. Qualitative Studien mit HPLC ergaben in frischen Puppen ein Verhältnis von 3:1 Ecdyson zu 20-Hydrooxyecdyson. Pharatstadien enthielten vor allem 20-Hydrooxyecdyson. Das beobachtete einzige Maximum im Titer stimmt überein mit den Resultaten bei andern untersuchten Coleopteren.
    Notes: Abstract Ecdysteroid titers were estimated on the whole body homogenates of Xyleborus ferrugineus (Fabr.) female pupae during development by radioimmunoassay. A distinct peak of ecdysteroids was observed at 36-hr pupal development (743 pg/mg body wt). Titer declined to 299 pg/mg by the pharate adult stage and to 193 pg/mg body wt just before adult emergence. Qualitative studies by HPLC revealed a ratio of 3:1 ecdysone to 20-hydroxyecdysone in the initial pupal stage. Pharate adults had mainly 20-hydroxyecdysone. The observed single peak in ecdysteroid titer agrees with findings in other studied coleopteran species.

    Notes: Abstract Nitrate, nitrite and nitrous oxide were denitrified to N2 gas by washed cells ofRhizobium japonicum CC706 as well as by bacteroids prepared from root nodules ofGlycine max (L.) Merr. (CV. Clark 63). Radiolabelled N2 was produced from either K15NO3 or Na15NO2 by washed cells ofRh. japonicum CC705 grown with either nitrate only (5 mM) or nitrate (5 mM) plus glutamate (10 mM). Nitrogen gas was also produced from N2O. Similar results were obtained with bacteroids ofG. max. The stoichiometry for the utilization of15NO 3 - or15NO 2 - and the produciton of15N2 was 2:1 and for N2O utilization and N2 production it was 1:1. Some of the15N2 gas produced by denitrification of15NO 3 - in bacteroids was recycled via nitrogenase into cell nitrogen.

    Notes: Abstract In seeking to develop an NDE technique for verifying the serviceability of NiTi couplers for plumbing unions, it was found that the reflection coefficient for a normally incident ultrasonic wave correlated with the stress level at the NiTi-tubing interface. Current theory pertaining to interface reflection is not complete but, even at present status, there is semiquantitative accord between theoretical predictions and normalized data for reflection coefficient versus stress level. These results are at fixed frequency and with comparable surface roughness. Variation of either frequency or surface roughness further affects the reflection coefficient. Current studies are being pursued to define this effect.

    Notes: Abstract The development of a successful and accurate instrument for measuring surface-breaking cracks in metals using the ac field technique has raised several interesting theoretical problems. Measurements with the instrument, known as the Crack Microgauge, do not rely on any prior calibration against an artificial flaw such as a saw-cut in a test block, but some users accustomed to such a calibration from other devices have nevertheless wished to use the instrument in that fashion and have in some instances reported erroneous results. In this paper, we develop a simple theory to explain and quantify this phenomenon. We calculate the parasitic voltages induced in the instrument probe due to the finite opening possessed by an artificial flaw, and we use these results to reinterpret the instrument readings. Controlled experimental measurements on artificial flaws of rectangular cross-section made in aluminum and in steel are found to be in good agreement with the theory. It is shown, however, that application of the theory requires additional information about the internal phase shift associated with the instrument. To enhance the accuracy of the theory, the effect of the corners of the artificial flaws was also considered, although it was not very influential in this work.

    Notes: Abstract An outline is given of the existing nondestructive vibration tests for cast irons and their drawbacks in terms of the dimensional accuracy of the components. A proposal is made for a new test based on the change of resonant frequency with vibration amplitude exhibited by cast irons. This proposal is evaluated using material data obtained from dynamic tests of a wide range of cast iron specimens. A test rig is described suitable for testing cylindrical cast bars, and typical results are presented in relation to material strength. There is also a brief description of preliminary testing on cast products.

    Notes: Abstract Mountain pine beetles,Dendroctonus ponderosae Hopkins, and California five-spined ips,Ips paraconfusus Lanier, were reared axenically from surface-sterilized eggs on aseptic pine phloem. After 24 hr in host logs, axenip femaleD. ponderosae and maleI. paraconfusus produced the aggregation pheromones,trans-verbenol (D. ponderosae), and ipsenol and ipsdienol (I. paraconfusus). Emergent, axenically reared maleD. ponderosae contained normal amounts of the pheromoneexo-brevicomin. Axenic femaleD. ponderosae treated with juvenile hormone or exposed to vapors of α-pinene, produced the pheromonetrans-verbenol. By 25–35 days after eclosion, axenic females exposed to α-pinene vapors produced over six times as muchtrans-verbenol as wild females, suggesting that while microorganisms in wild females may producetrans-verbenol, they may also inhibit production of the pheromone or use it as a substrate.

    Notes: Abstract The effect of P nutrition on phosphate uptake and alkaline phosphatase activity was studied in chemostat culture for four rhizobial and three bradyrhizobial species. Phosphate-limited cells took up phosphate 10- to 180-fold faster than phosphate-rich cells. The four fast-growing rhizobial strains contained high levels of alkaline phosphatase activity under P-limited conditions compared to the repressed levels found in P-rich cells; alkaline phosphatase activity could not be detected in three slow-growing rhizobial strains, regardless of their P-status. Glycerol 1-phosphate-uptake in the cowpea Rhizobium NGR234 was derepressed over 50-fold under P-limited conditions, and appeared to be co-regulated with phosphate uptake. The phosphate-uptake system appeared similar in all strains with apparent K m values ranging from 1.6 μM to 6.0 μM phosphate and maximum activities from 17.2 to 126 nmol · min-1 · (mg dry weight of cells)-1. Carbonyl cyanide m-chlorophenyl hydrazone strongly inhibited phosphate uptake in all strains and a number of other metabolic inhibitors also decreased phosphate uptake in the cowpea Rhizobium NGR234. The phosphate uptake system in all strains failed to catalyse exchange of 32P label in preloaded cells or efflux of phosphate. The results suggest a single, repressible, unidirectional and energy-dependent system for the transport of phosphate into rhizobia.

    Notes: Abstract With continuous cultures in a fully defined minimal salts medium steady states were achieved at both limiting and non-limiting concentrations of phosphate in the inflowing medium for Rhizobium trifolii WU95, cowpea Rhizobium NGR234, and Bradyrhizobium CB756. Millimolar growth yields obtained from P-limited cultures varied over 2-fold from 3.2 g dry weight·(mmol P)-1 for WU95 to 5.3 g dry weight·(mmol P)-1 for CB756 and 7.2 g dry weight·(mmol P)-1 for NGR234. For both WU95 and NGR234 growth under P-excess conditions resulted in elevated levels of total biomass P and the storage compound polyphosphate, compared with P-limited cultures. However, P-limited cultures of these two strains still contained significant quantities of polyphosphate. The P-status for CB756 cultures did not affect either total biomass P or polyphosphate levels. Alkaline phosphatase was maximally derepressed in P-limited cultures of WU95 and NGR234. However, in CB756 alkaline phosphatase was not detected at significant levels regardless of its P supply. These data suggest that growth of rhizobia is controlled predominantly by the attainment of a critical internal P level.

    Notes: Abstract Lysozyme/EDTA treatment of four fast-growing rhizobia released repeatable protein profiles after polyacrylamide slab gel electrophoresis. Similar treatment of slow-growing rhizobia failed to release such periplasmic proteins. For the four-fast-growing rhizobia, both P-repressible and P-inducible protein bands occurred. The only P-repressible protein identified was alkaline phosphatase, which showed strain differences in both electrophoretic mobility and activation by Mg2+. The derepression of the P-repressible periplasmic proteins in cowpea Rhizobium NGR234 correlated with derepression of both phosphate and glycerol 1-phosphate uptake.

    Notes: Abstract For the purpose of ultrasonic nondestructive testing of materials, holography in connection with digital reconstruction algorithms has been proposed as a modern tool to extract crack sizes from ultrasonic scattering data. Defining the typical holographic reconstruction algorithm as the application of the scalar Kirchhoff diffraction theory to backward wave propagation, we demonstrate its general incapability of reconstructing equivalent sources, and hence, geometries of scattering bodies. Only the special case of a planar measurement recording surface, that is to say, a hologram plane, and a planar crack with perfectly rigid boundary conditions parallel to the hologram plane and perpendicular to the incident field yields a nearly perfect correlation between crack size and reconstructed image; the reconstruction algorithm is then referred to as the Rayleigh-Sommerfeld formula; it therefore represents the optimal case matched to that special geometrical situation and, hence, may be interpreted as a quasi-matched spatial filter. Using integral equation theory and physical optics, we compute synthetic holographic data for a linear cracklike scatterer for both plane and spherical wave incidence, the latter case simulating a synthetic aperture impulse echo situation, thus illustrating how the Rayleigh-Sommerfeld algorithm or its Fresnel approximation increasingly fail for cracks inclined to the hologram plane and excited nonperpendicularly. Furthermore, we point out how the physical data recording process may additionally influence the reconstruction accuracy, and, finally, guidelines for a careful and serious application of these holographic reconstruction algorithms are given. The theoretical results are supported by measurements.
