    Notizen: A new method for quantitating adenosine concentration by capillary gas chromatography-mass spectrometry-selected ion monitoring (GC-MS-SIM) has been developed and used as a reference method for evaluating a newly developed radioimmunoassay (RIA) for adenosine. Details of the GC-MS-SIM method are presented, along with the comparative results and uncertainties of both methods. General considerations in the statistical analysis of method comparison data are discussed with particular reference to studies using quantitative mass spectrometry as the standard method; the adenosine methods are used as specific examples in this discussion. Simultaneous estimation of the y-intercept and slope of the least squares regression line relating the results of the two methods using the 95% joint confidence ellipse demonstrated the absence of either constant or proportional error between the two methods. The relatively small uncertainty in the GC-MS-SIM measurements had no significant effect on the linear regression. Random error between the two methods was detected, and was estimated by the coefficient of variation in the RIA data as ten percent of the RIA value.

    Notizen: Diesel fuels from different parts of the UK have been analyzed for polycyclic aromatic hydrocarbons (PAH), nitrogen-containing PAH, and sulfur-containing PAH using capillary column GC with simultaneous parallel triple detection. The concentrations of polyaromatic compounds (PAC) were high and showed considerable variability amongst the fuels. The PAH are mainly naphthalene, fluorene, and phenanthrene and their alkylated homologs; the PANH are mainly carbazole and its methyl derivatives; the PASH are mainly dibenzothiophene and its methyl derivatives.

    Notizen: A benchtop SFC/MS system is described which utilizes supercritical carbon dioxide in a 50 micron diameter open tubular column interfaced directly to an unchanged commercially available benchtop capillary GC/MS system equipped with a chemical ionization (CI) source. A small amount of methane reagent gas was admitted co-axially to a capillary restrictor at the exit of the capillary chromatographic column. This make-up gas served as the CI reagent gas and appeared to optimize the sensitivity of the system while providing abundant (M+1) ions for the analytes investigated in this study. Good chromatographic intergrity was obtained for the GC/MS test compound, decafluorotriphenylphosphine (DFTPP), but the capillary restrictor appeared to cause some tailing of the ion current profiles resulting from low nanogram levels of caffeine and some fatty acid esters. Improvements in the SFC/MS capillary restrictor interface and the pumping system of the benchtop GC/MS system should increase the capability of this system for future applications.

    Notizen: For chromatography with supercritical fluids (SFC), the dependence of the capacity ratios k′ and of the mean resolution Rm on temperature and pressure is presented as a three-dimensional diagram. A sufficient number of test chromatograms were run for the diagram to lead to an isocratic network in form of a curved surface. The isocratic network possesses a characteristic shape and contains all information about the temperature and pressure dependence of k′ and Rm for a given volume flow rate and chromatographic system. The specific system studied comprised pentane as the mobile phase, unmodified silica as the stationary phase, and a set of four polycyclic hydrocarbons as the test mixture. The isocratic net of this system allows interpolation of k′ and Rm for any temperature and pressure. Together with similar experimental data from other systems, this allows qualitative forecasts about the isocratic nets of other systems.

    Notizen: Plasma samples from volunteers who had received an oral dose of acetanilide have been analysed by gas chromatography mass spectrometry and ultraviolet absorption techniques. The gas chromatography mass spectrometry method involved extraction of the plasma and analysis of the acetanilide using selected ion monitoring with a deuterated internal standard. In the ultraviolet method the plasma was hydrolysed with acid to convert the acetanilide to aniline, and this compound was diazotized and coupled with N-1-naphthylethylene-diamine. The absorbance of the resulting complex was read at 550 nm. Acetanilide levels in plasma determined by the selected ion monitoring method were significantly lower than those measured by spectrophotometry. Pharmacokinetic data calculated from the results obtained using these two assays are very different and illustrate the need for an accurate and specific method of analysis. The major metabolites of acetanilide are shown not to interfere with these assays and the results suggest the possible presence of a new metabolite of acetanilide.

    Notizen: The in vivo liver metabolism of cannabinol has been studied in the mouse and rat by combined gas chromatography and mass spectrometry. Cannabinol glucuronide was the major metabolite of cannabinol in the mouse and was accompanied by relatively large amounts of 7-hydroxycannabinol, cannabinol-7-oic acid and their corresponding glucuronide conjugates. Lower concentrations of glucuronides were found in the rat. Two series of disubstituted metabolites were found containing either a 7-hydroxyl or a 7-carboxylic acid group and a second hydroxyl group in the 1″-4″ positions of the sidechain. These were of low concentration in the mouse but higher in the rat; 1″-hydroxy metabolites were particularly abundant in the latter species. Also found in the rat livers were small amounts of sidechain monohydroxy metabolites and larger quantities of 4″,5″-bisnorcannabinol-3″-oic acid; these were absent in the mouse. The metabolites were identified using the trimethylsilyl (TMS),[2H9]TMS and methyl ester-TMS derivatives, and by reduction of acid metabolites with lithium aluminium deuteride to the corresponding alcohols.

    Notizen: A new method based on selected ion monitoring chemical ionization mass spectrometry was developed to measure the quaternary ammonium neuromuscular blocking agents, pancuronium and vecuronium, in serum. An ion-pair extraction procedure is utilized to separate the compounds of interest from biological fluids. The intensities of the ion currents produced by the bisamines formed from the drugs and the corresponding deuterated internal standards through thermolytic dequaternization are monitored. The assay shows good linearity over the range of 1-500 ng/ml. This assay has been utilized in a variety of clinical pharmacokinetic studies involving surgical pediatric, geriatric and obstetric patients requiring anesthesia.
