Notes: A natural microbial population of sewage origin was grown at four temperatures (10, 20, 30, 40°C) and five mean residence times (3, 6, 12, 18, 24 hr) in a continuous stirred tank reactor. Samples of cells were removed during steady-state operation and analyzed for chemical oxygen demand (COD), ash content, protein, carbohydrate, and nucleic acids. The oxygen equivalence (OE) of the cells was measured directly as g COD/g ash-free cells and was determined indirectly by calculating the calorific content from the macromolecular composition. The results indicate that OE is a function of growth conditions. Furthermore the COD test is a poor measureof OE because of a high level of variation associated with the test. Additional studies should be performed using bomb calorimetry to establish a quantitative relationship between growth conditions and OE.

Notes: The influence of both imposed anodic potential in Ringer's solution and surface finish on the fatigue lives of annealed 316 type stainless steel and annealed pure titanium were measured and statistically compared to fatigue data run in air. The applied potentials in simulated extracellular fluid approximated conditions existing within the body while also producing the types of surface defects actually found on removed long time implants within the time interval of the accelerated R. R. Moore fatigue tests. Differentiating tests were run at single levels of applied cyclic stress well above the endurance limits. In Ringer's solution, the fatigue life of the 316 stainless steel decreased with increasing applied potential, and at +500 mV was significantly shorter than when run in air. At each condition, the 316 stainless steel was independent of initial surface finish. In contrast, the fatigue life of titanium improved rapidly with increasingly fine surface finishes. Furthermore, compared to air, the application of +500 mV in Ringer's solution improved the life of the rough surface finished material and markedly increased the number of cycles to failure for the electropolished specimens.

Notes: Observations on the pattern of nuclear incorporation of 3H-TdR in long term (8-day) and short term (3-day) 3T3 cultures with local cell densities between 0.2 × 104 and 6.2 × 104 cells/cm2 are reported. Contrary to a number of previous studies our observations indicate that density dependent inhibition is exhibited in relatively sparse cultures, commencing at 0.5 × 104 cells/cm2. Various possible mechanisms which could have caused the observed pattern of density-dependent regression in labelling index are discussed.

Notes: Tetrahymena pyriformis ST (3 × 104 cells/ml) was treated with 0.1 mg/ml chloramphenicol (CAP). Cell division ceased after 1.5 divisions with no decreased viability. Total mitochondrial volume and succinic dehydrogenase (SDH) activity/liter increased 1.7-fold and 3-fold, respectively. SDH activity/cell decreased whereas malate dehydrogenase activity/cell and respiratory control ratios and P: O ratios of isolated mitochondria were unchanged in treated cells. During 12 hours of growth in CAP the total surface area of mitochondrial inner and outer membrane was essentially unchanged or increased 4-fold, respectively. Mitochondria from cells treated with chloramphenicol had decreased size, buoyant density and protein: lipid ratio in the membranes. The membrane ubiquinone: protein ratio was unchanged.Tetrahymena cells contained 3.6 × 10-12 g of mitochondrial DNA and 6,800 mitochondria in a volume of 41,000 μ3. A 4-hour treatment with CAP caused a 4-fold increase in the number of mitochondria/cell and a 10-fold increase in mitochondria/liter in contrast to a 4-fold increase in number of mitochondria/liter in control cells. Thus CAP stimulated division of mitochondria. Individual mitochondria of treated cells had one-tenth the volume of control mitochondria. The rate of increase of mitochondrial DNA/liter was the same in control and CAP-treated cultures. The amount of DNA/mitochondrion decreased 75% in CAP-treated cells due to the rapid division of mitochondria. The cell volume, cell protein content and mitochondrial DNA content/cell decreased with growth of control cultures.

Notes: The bianthraquinonic biological pigments luteoskyrin and rugulosin and five polyhydroxyanthraquinone derivatives are studied by carbon-13 NMR in DMSO solution. Peak assignment for the fourteen carbon atoms of these compounds is achieved by proton spin decoupling and by investigating the effect of ionisation of the hydroxyl groups upon the carbon chemical shifts. Carbon chemical shifts in the planar hydroxyanthraquinones can be rationalised in terms of conjugation and intramolecular hydrogen bonding. The latter is responsible for the relative acidity of the hydroxyl groups in the analogues, and for the different conformations proposed for luteoskyrin and rugulosin. Tautomeric equilibria occur in DMSO and water-DMSO solutions for the anionic species [LS]2- and [RG]2-. This can account for the binding of luteoskyrin and rugulosin to nucleic acids.

Description / Table of Contents: NMR spectra of several R3P/AlCl3 systems [with R3 = (CH3)3 and (CH3)2(C6H5)] in solution have been measured. The results suggest the occurrence of fast exchanges involving 1/1 and 2/1 adducts and free phosphine. The existence of the 2/1 compounds has been confirmed by low temperature (173 K) experiments.

Notes: The synthesis of new ternary silicate and germanate phases containing large alkali and alkaline-earth cations is described. They are made by solid-state reaction of mixtures of carbonates or oxalates with SiO2 or GeO2, or by fusion and subsequent recrystallization of the glass.Representatives of the cubic M4+M2+X3O9 family include Na4CaSi3O9 and the isostructural compounds K4CaGe3O9, K4SrGe3O9 and K4SrSi3O9. K4BaSi3O9 is pseudocubic: the symmetry of Na4SrSi3O9 is unknown. The rhombohedral M8+M2+X10O25 family includes K8CaSi10O25, K8SrSi10O25 and K8BaSi10O25.Na2CaGe2O6 and Na2SrGe2O6 are isostructural but both are structurally unrelated to Na2BaSi2O6. Na2Ba2Ge2O7 and Na2Ba2Si2O7 are structurally similar.

Notes: Interactions of glutaraldehyde with either n-butylamine, poly(α,L-lysine), or collagen resulted in a fast release of protons in dilute aqueous solutions at various pH values, followed by much slower changes. The latter reactions, which extended over hours and days, were followed spectrophotometrically and revealed the formation of distinct absorption bands in the visible and near-ultraviolet regions in all the above systems. The visible-range bands disappeared upon treatment with sodium borohydride. A qualitative relationship between oxygen uptake by the system n-butylamine-glutaraldehyde and the slow formation of colored products has been established, while the chemical nature of the reaction products has not been determined.Sedimentation velocity, viscosity, and optical rotation measurements on the products of interaction between poly(L-lysine) and glutaraldehyde in aqueous solution indicated large conformational changes in the polyamino acid present in excess (in residues) over the dialdehyde. In particular, the intrinsic viscosity dropped considerably after interaction, indicating intramolecular crosslinking. At molar ratios of 1:1 between polylsine residues and aldehyde groups, intermolecular crosslinking of polylysine was obtained at pH 8.6.Electron microscopic examinations of collagen samples treated by glutaraldehyde at various pH values indicated changes from unordered to more ordered structures upon treatment with glutaraldehyde, in particular at pH 10.The present structural and optical investigations are considered to be relevant to tanning processes of hides and to fixation procedures.
