    Notizen: Results of pilot plant studies using a glass airlift fermentation device (55 liter fermentation volume) have proven the relative merits of such a system in the fermentation of a filamentous mold, Monascus purpureus, on 4% (w/w) starch media. The resultant overall yield of cell mass (Yx/s) of 0.38 was an appreciable increase over the 0.32 obtained with a pilot scale stirred tank fermentor previously studied. Power requirements of the airlift fermentor were approximately 50% of those for the mechanically agitated system. The lack of mechanical shear in the airlift system provides a more gentle environment or the cultivation of organisms than does the high degree of shear prevalent in the mechanically agitated vessels. Mass transfer of oxygen to the aqueous phase of the fermentation volume is improved significantly through use of the airlift device. Mass transfer coefficients in the range of 200 reciprocal hr were obtained to approximately 80 reciprocal hr in the stirred tank fermentor.

    Notizen: The presence of microorganisms have been shown to increase by over 40% the mass transfer rates from small oxygen bubbles at low Reynolds number flow. This increase was found to be due only to the microbe cells as inert particles disrupting the quasi-static liquid surface film surrounding the gas bubble and thus decreasing the gas-liquid interfacial resistance. The observed increase in oxygen mass transfer rates was not dependent on cell viability, no effect was noted due to cell-liquid interfacial resistance, nor was the phenomenon due to altering the physical properties of the liquid during cell propagation. These results were obtained in a unique plexiglass apparatus designed for observing under a microscope a small (0.4 mm dia.) stationary oxygen bubble collapsing into a flowing fluid. The oxygen bubble was injected by a small hypodermic needle and the fluid was suspensions of the yeast Candida intermedia, the bacterium Pseudomonas ovalis, 0.3μ alumina, as well as base points of cell free broth and pure water. Several well-known chemical inhibitors of oxidative phosphorylation were used to limit cell oxygen uptake. Calculations of oxygen mass transfer rates were compared with the semi-empirical model of Frössling, the circulating sphere model of Levich, and the rigid sphere concentration boundary layer model of Fried-lander, the latter two showing strong Reynolds number dependence that may be due to radial fluid motion.

    Notizen: Relatively poor SCP production (4.2 mg/L h) was obtained using C. cellulolyticum and ground aspen wood treated with steam at atmospheric pressure for 1 h. The percentage of protein in the final product increased to 21.4% at a specific growth rate of 0.15 h-1 when the wood sample was treated with steam at a higher pressure (280 psig for 4 min) according to the "Stake" process. Alkali treatment (10% and 15% w/w at 121°C for 30 min), known to solubilize hemicelluloses and some of the lignin, gave intermediate results. More complete delignification of wood using NaClO2 increased the protein composition in the final product to 37.9%, at a specific growth rate of 0.19 h-1. Cellulose utilization was lowest (12.4%) in the case of the wood treated with steam at atmospheric pressure; it was higher at 75.3 and 78.5% for wood treated with NaOH at 10 and 15% w/w levels, respectively. The cellulose utilization was highest (90%) for wood treated with NaClO2.

    Notizen: Penicillin amidase was extracted from Escherichia coli ATCC 9637, grown on phenylacetic, acid and glutamate, and purified by fractional ion with streptomycin sulphate, ammonium sulphate and polyethylene glycol, followed by chromatography on DEAE-cellulose. The purification factor was 100-200 × and the overall yield was about 115%. The enzyme was chemically attached to derivatives of cellulose and the kinetics of these insolubilized penicillin amidase preparations was investigated.

    Notizen: The gases CO, CO2, and H2 were used as substrates in anaerobic fermentations producing organic acids. Various mixed bacterial sources were used, including sewage sludge digester effluent, rabbit feces, and soil. Nonsterile microorganism selection was carried out using CO2/H2 and CO/H2 as the primary carbon and energy sources. Cultures were grown in specially designed, high-pressure (to 70 psig) flasks. Methanogenic bacteria were eliminated from the cultures. Liquid products of the fermentations were acetic through caproic acids, with the even-numbered acids predominating. Carbon balances showed conclusively that acetic acid was formed from carbon contained in the CO or CO2 feed gas. Measurements made included rates of acid product formation, cell density, and degree of gas utilization. Limited characterization of the microorganisms was also performed. Production of organic acids by mixed culture inocula from CO2/H2 or CO/H2 had not been reported previously. Application of this work is to the production of organic chemicals from synthesis gas (SNG), produced by the gasification of fossil fuels (peat, lignite, and various ranks of coals), biomass (agricultural and forest residues, and various biomass crops grown expressly for energy recovery), and municipal solid waste.

    Notizen: With a chemostat culture, both the bacterial growth rate and the growth environment can be independently varied between wide limits. Changing the growth rate of Aerobacter aerogenes organisms (in either a glycerol-limited medium or a Mg2+-limited medium) affected the bacterial cell wall content; invariably slow growing organisms were smaller than faster growing ones and had a higher cell wall/biomass ratio. Changing the growth rate also influenced the composition of the walls but in this respect glycerol-limited organisms and Mg2+-limited organisms behaved differently. Thus, whereas increasing the growth rate of glycerol-limited cultures caused the cell wall 2-keto-3-deoxyoctonic acid (KDO) and heptose contents to increase progressively, with Mg2+-limited cultures they decreased. Furthermore, although KDO and heptose are both components of the lipopolysaccharide layer, their ratio varied with growth rate, and with the nature of the growth-limitation, indicating changes in the lipopolysaccharide composition. These results are discussed with particular reference to the influence of environment on cell wall content and composition, and the use of continuous culture for the production of bacterial vaccines.

    Notizen: A procedure for measuring the rate of heat production from a fermentation has been developed. The method is based on measuring the rate of temperature rise of the fermentation broth resulting from metabolism, when the temperature controller is turned off. The heat accumulation measured in this manner is then corrected for heat losses and gains. A sensitive thermistor is used to follow the temperature rise with time. This procedure is shown to be as accurate as previous methods but much simpler in execution. Using this technique, the rate of heat production during metabolism was found to correlate with the rate of oxygen consumption. Experiments were performed using bacteria (E. coli and B. subtilis), a yeast (C. intermedia), and a mold (A. niger). The substrates investigated included glucose, molasses, and soy bean meal. The proportionality constant for the correlation is independent of the growth rate, slightly dependent on the substrate, and possibly dependent On the type of organism growth. This correlation has considerable potential for predicting heat evolution from the metabolism of microorganisms on simple or complex substrates and providing quantitative parameters necessary for heat removal calculations.

    Notizen: The filamentous fungus, Rhizopus nigricans, was immobilized in polyacrylamide, alginate, and agar gels and its ability to 11α-hydroxylate progesterone was examined. No activity was detected using polyacrylamide gel but both agar and alginate gels have proved capable of hydroxylation. Agar gels displayed faster rates and higher yields. It was possible to induce hydroxylase synthesis within agar and alginate gels, and microscopical examination provided evidence for hyphal growth within these gels. The concept of increased biomass was used to explain the observed increase in the rates of hydroxylase activity of the immobilized cells. Conversely, hyphal overcrowding was postulated for the rapid inactivation observed under some operating conditions.

    Notizen: A range of cellulosic raw materials in the form of agricultural crop residue was analyzed for chemical composition and assessed for potential yields of sugars through chemical pretreatment and enzymatic hydrolysis of these materials. Corn stover was used as a representative raw material for a preliminary process design and economic assessment of the production of sugars and ethanol. With the process as presently developed, 24 gal ethanol can be obtained per ton of corn stover at a processing cost of about $1.80/gal exclusive of by-product credits. The analysis shows the cost of ethanol to be highly dependent upon: (1) the cost of the biomass, (2) the extent of conversion to glucose, (3) enzyme recovery and production cost, and (4) potential utilization of xylose. Significant cost reduction appears possible through further research in these directions.

    Notizen: The cyanobacterium Anabaena cylindrica B629 was suspended in small glass beds and incubated in a gas-tight glass vessel outdoors under a gas atmosphere comprising carbon monoxide (0.2%), acetylene (5%), oxygen (6.5%), and nitrogen. The solution phase initially contained sodium bicarbonate (10mM) at pH 7. Under these conditions the organism continuously produced hydrogen gas for over three weeks. The temperature of the culture was maintained below 30°C and minimum night temperatures were recorded. The vessel was covered by a shadecloth, which reduced the natural illumination by approximately 70%. The system is an alternative to those requiring the strict absence of oxygen and little nitrogen, and requires virtually no attention during the incubation period.
