    Notes: Intermediate subviral particles (ISVP) derived from reovirus represent a simple model system for the switch-on of transcriptase function. In such particles the endogenous transcriptase is present in a switched-off form, one step removed from the switched-on state. Switch-on of transcriptase function is an active process in this system and can be triggered by K+ ions. A variety of agents which affect gene expression in cells were tested for an effect on switch-on in ISVP. Marked effects on switch-on in ISVP were observed with a diverse group of test agents, including DMSO and other solvents, BUdR, TdR, caffeine, theophylline, and temperature. The correlation in response between ISVP and cells suggests that the ISVP system may be useful as a model for studying the biochemical mechanisms underlying the perturbative effects of such agents on gene expression in cells.

    Notes: The morphological and ultrastructural changes that occur during preparation of porcine, bovine, and murine spermatozoa for flow cytometric quantification of the relative DNA content of the X- and Y-chromosome-bearing sperm populations were examined. Ejaculated spermatozoa from the boar and bull were washed using a series of dimethyl sulfoxide (DMSO) solutions prior to fixation, whereas the epididymal mouse spermatozoa were washed only in phosphate-buffered saline (PBS). Spermatozoa from all three species were then fixed in ethanol and processed for fluorochrome staining by a treatment regimen consisting of sulfhydryl reduction and proteolysis. The processed sperm nuclei were stained for DNA with the fluorochrome, 4′-6-diamidino-2-phenylindole (DAPI) before quantification by flow cytometry. Scanning and transmission electron micrographs of sperm heads taken at various steps of the preparation and staining procedures show 1) that the rigorous washing procedure disrupted the plasma and outer acrosomal membranes, 2) that ethanol fixation resulted in removal of the outer membranes and disintegration of the nuclear envelope, and 3) that thiol and proteolysis treatment removed the remaining cellular organelles including the tail and rapidly induced partial decondensation of the tightly packed chromatin. Sequential micrographs showed that the nuclear matrix of all three species increased in thickness about twofold during the preparation and staining. Consequently, the harsh procedures currently used for quantitative staining of DNA for high-resolution flow cytometric analyses destroy most cellular organelles and thereby prevent simultaneous characterization of DNA content and other sperm cell constituents.

    Description: A liquid-helium-cooled grating spectrometer (CGS) is being developed as a facility instrument for the Kuiper Airborne Observatory (KAO), primarily to study for infrared lines originating in the interstellar medium. A maximum resolving power of approximately 6000 is achieved by means of a 45 cm long Echelle grating and is optically capable of operating in the spectral range from 25 to 300 microns. An array of detectors is used to simultaneously measure a line and the adjacent continuum from astronomical sources. Currently six detectors allow measurements in the 30 to 120 micron spectral band. The instrument, its operation, and its performance are described.

    Description: The Voyager IR investigation is described, which uses a Michelson interferometer in the 180-2500/cm range, and a single-channel radiometer for the visible and near-IR, sharing a 50-cm diameter telescope. Emphasis is placed on the differences between the Voyager and the previous designs, including reductions in the field of view and in the noise equivalent spectral radiance of the instrument. Attention is given to the optical layout, the electronics module, power supply placement, thermal control heaters and flash heaters, data reduction, and calibration. A sample spectrum of Jupiter is also discussed.

    Description: A decrease in the solar radius is determined using the technique of Dunham and Dunham (1973), in which timed observations are made just inside the path edges. When the method is applied to the solar eclipses of 1715, 1976, and 1979, the solar radius for 1715 is 0.34 + or - 0.2 arc second larger than the recent values, with no significant change between 1976 and 1979. The duration of totality is examined as a function of distance from the edges of the path. Corrections to the radius of the sun derived from observations of the 1976 and 1979 eclipses by the International Occultation Timing Association are also presented.

    Description: VLBI and CCD astrometry is presented which establishes the coincidence of the X-ray and radio object Cir X-1 with an object lying about one arcsec south of the object 'L' of Whelan et al. (1977), a conclusion reached previously by Argue and Sullivan (1982). The question whether the object L has any physical connection with Cir X-1 cannot yet be definitely answered.

    Description: Observations of the 691-GHz (J = 6-5) transition of CO in the BN/KL region of Orion obtained in February 1981, at the IR Telescope Facility at Mauna Kea are reported. The system employs a heterodyne receiver with an overall noise temperature of 3900 K DSB at 432 microns, 64 5-MHz IF-filter-bank channels, and a chopping secondary with 120-arcsec excursion, and has 35-arcsec resolution. Sample data are presented graphically and analyzed using a rate equation and a kinetic model. A 35 x 45-arcsec core with hot broad plateau emission (antenna temperature 180 + or - 36 K) surrounded by an area of strong narrow-line 120-K emission is found. It is suggested that the plateau emission originates in a wide thin sheet after a shock wave, with H2 density at least 10 to the 6th/cu cm and gas kinetic temperature greater than 500 K.

    Description: The catalog of X-ray sources detected during the NRL Large Area Sky Survey (LASS) with the HEAO 1 satellite is presented. The catalog is derived from the first six months of data from HEAO 1 and includes sources detected during one full scan. Positions and intensities for a total of 842 different sources are included, with a limiting flux of 250 nJy at 5 keV. The catalog is more than 90 percent complete at a flux level equivalent to 1.5 microjoules at 5 keV for a Crab-like spectrum. Cross-references with published literature are provided and coincidental identifications are proposed for some of the sources which have been never studied before. A cross-sectional line drawing of the sensor module of HEAO I is also provided.

    Description: The results of numerical models or of new observational programs are checked by comparing them with past observations. In view of the differing analysis techniques or differing data samples, the eddy diffusivities presented here agree remarkably well with past estimates. However, in the application of K-values to two-dimensional models, the actual magnitude of the diffusivities is no more important than their spatial patterns, i.e., their gradients with height and latitude. It should thus be noted that the present patterns are often much different from those of past results.

    Description: The light curve of 4U 2129+47 is presented showing that it almost certainly has a period of approximately 0.22 days. This is the same period as the optical candidate proposed by Thorstensen et al. (1979). Binary X-ray source models, consisting of an M dwarf plus either a neutron star or a white dwarf, are briefly discussed.
