    Notes: The effect of temperature, water content, and the type of reagent on the silylation of fused silica capillaries was studied by 29Si and 13C CP-MAS NMR. Fumed silica (Cab-O-Sil M5), which is essentially a highly dispersed vitreous quartz with a surface comparable to that of fused silica capillary columns, was selected as a model material.Hexamethyldisilazane (HMDS) and 1,2-diphenyl-1,1,3,3-tetraphenyldisilazane (DPTMDS), which were used as silylation reagents, yielded trimethyl- and dimethylphenylsilyl surface groups respectively at lower temperatures (〈 350°C and 〈250°C respectively). At higher temperatures, increasingly more dimethylsilyl groups are formed, with the silicon bound to two oxygen atoms. This process occurs for DPTMDS at a considerably lower temperature than for HMDS. The formation of silyl groups on the surface and the disappearance of hydroxyl groups are followed independently. The 13C NMR and GC-MS of the reaction products showed that with DPTMDS, the formation of two Si-O-Si links is accompanied by a loss of phenyl groups rather than of methyl groups.After the Cab-O-Sil had been dried over P2O5, the formation of these double links occurred for HMDS only at temperatures above 460°C and for DPTMDS at 400°C. Thus we concluded that water supplies oxygen atoms for double Si-O-Si links (possibly crosslinks) necessary for efficient deactivation. This may explain the less successful silanization of fused silica capillaries because their water content is lower than that of glass capillaries.

    Notes: The kinetics of formation of the 1:1 complexes of some dialkyl-2,2′-bipyridyl-5,5′-dicarboxylates with the ion of Ni(II) have been determined by the stopped-flow spectrophotometric method. Second-order formation rate constants have been correlated with the Taft modification of the Hammett equation. A linear regression has been found (P = 0.95 confidence) with the following characteristics: slope = -0.6 ± 0.1; intercept = 1.32 ± 0.02 (log k0). An inductive effect of the alkyl groups upon the reaction of the complexation is proposed.

    Notes: The relationship between published vicinal proton-proton coupling constants and the pseudorotation properties of the pyrrolidine ring in L-proline, 4-hydroxy-L-proline, 4-fluoro-L-proline, and several linear and cyclic model proline peptides is investigated. Compared to earlier studies, several important improvements are incorporated: (1) a new empirical generalization of the classical Karplus equation is utilized, which allows a valid correction for the effects of electronegativity and orientation of substitutents on 3JHH; (2) an empirical correlation between proton-proton torsion angles and the pseudorotational parameters P and τm is derived; and (3) the best fit of the conformational parameters to the experimental coupling constants is obtained by means of a computerized iterative least-squares procedure. Two pseudorotation ranges were considered, classified as type N (χ2 positive sign) and type S (χ2 negative sign). The conformational equilibrium is fully described in terms of four geometrical parameters (PN, τN, PS, τS) and the equilibrium constant K. The present results indicate that, in general, the geometrical properties found in x-ray studies of proline and hydroxyproline residues are well preserved in solution. Several novel features are encountered, however. It is demonstrated that the proline ring occurs in a practically 1:1 conformational equilibrium between well-defined N- and S-type forms. Introduction of an amide group at the C-terminal end has no observable effect on this equilibrium, but the formation of a peptide bond at the imino nitrogen site results in a pronounced, but not exclusive, preference for an S-type form which is roughly 1.1 kcal/mol more stable than its N-type counterpart. The hydroxyproline ring system in neutral or acidic medium displays a pure N-type state, but N-acetylation results in the appearance of a minor (S-type) conformation. Cyclic proline dipeptides similarly exist in a biased conformational equilibrium. The major form (77-88%) corresponds to the N-type conformer observed in the solid state; the minor S-form has not been observed before. In contrast, cyclic hydroxyproline dipeptides display complete conformational purity. Ranges of endocyclic torsion angles deduced for the various classes of pyrrolidine derivatives in solution are presented. Each torsion appears confined to a surprisingly narrow range, comprising about 4°-8° in most cases. In all, the proline ring is far less “floppy” than hitherto assumed.

    Notes: The reaction of α-bromoimmonium bromides 4 with potassium cyanide in dimethylformamide gave rise to β-bromo-α-(dialkylamino)nitriles 5, which were dehydrobrominated in various basesolvent systems to afford (E)- and (Z)-α-cyanoenamines 2. An adaption of a previously published preparation of α-cyanoenamines starting from aldehydes via enamines led to an improved, efficient and fast synthesis of the title compounds.

Dagegen trat mit Silber-hexafluoroantimonat ausschließlich die letztere Umlagerungsreaktion ein.
    Notes: Tertiary α-bromoalkyl aryl ketones 1 were converted into 2-alkoxy-2-aryloxiranes 4 exclusively by reaction with excess potassium carbonate in the corresponding dry alcohol. Silver carbonate in a dry alcohol with these α-bromo ketones yielded competitively formation of 2-alkoxyoxiranes and semi-benzilic Favorskii rearrangement (→5), while silver hexafluoroantimonate in the same medium afforded the latter rearrangement reaction exclusively.

    Notes: The reaction of a diisocyanate and an asymmetric aliphatic-aromatic diacid containing a preformed imide group was studied in order to obtain polyamide-imides. It is shown in this paper that combined 1H and 13C NMR spectrometry will permit the quantitative evaluation of the proportions of head-tail, head-head, tail-tail additions, provided the content of urea in the polymer is low. In order to obtain unequivocal assignments and to thoroughly characterize the NMR spectra of the polymer, several model compounds resembling the polymer backbone were synthesized. A model polymer was synthesized with the same purpose, and its 1H and 13C NMR spectra fully assigned. Unexpected chemical functions in the polymer, like urea and carboxylic acid, can also be easily detected and quantified by NMR.

    Notes: The polycondensation of 4,4′-methylenediphenylisocyanate and 6-(4-carboxyphthalimido)-hexanoic acid was studied under different conditions in order to establish the influence of factors such as temperature, catalyst, stoichiometric disbalance, and addition order of reactants on the molecular weight and the chemical composition of the final polyamide-imide. All these factors proved to influence the extent of possible side reactions that lead to undersirable functions, the main ones being urea functions that locate within the polymer backbone. Other side-reactions proposed to explain the formation of acylureas or biurets did not take place in appreciable extent since these functions could not be detected by NMR spectroscopy. Therefore, particular emphasis was made to prevent the formation of urea by changing the polycondensation conditions. A first attempt to clarify the regularity degree of the polymers (amounts of head to tail and head to head addition) by 1H NMR spectroscopy was also made.

    Notes: Copolymers of (ethylene glycol/diethylene glycol) terephthalates were prepared by melt polycondensation. The values of the glass transition temperatures of the copolyesters, determined by dilatometry are in fair agreement with those predicted by the Fox equation. It was also found that the conformational entropy can be used to correlate equilibrium flexibility and glass transition behaviour of the copolyesters.

    Notes: Quantitative lateral and dorsoventral cineradiography shows that the masticatory movements of the mandible, condyles, tongue, and hyoid of Pteropus giganteus (Chiroptera) move along highly regular paths that are characteristic for each of the three food types tested.Mandibular movements are predominantly orthal, although a small forward translation occurs early in opening and small lateral deflections occur in both opening and closing phases. These deflections are related to the existence of active (bolus bearing) and balancing sides of the jaws, chewing being not truly bilateral. The deflections are associated with a shift of both condyles toward one side. In consequence the active condyle is located in a lateral part of the associated fossa, the inactive condyle in a medial part. Food transfer from side to side involves a reversal of the chewing direction during opening. Such reversals are especially frequent near the end of a chewing sequence.The fore, middle, and hind parts of the tongue differ in their movement patterns. Movements of the fore part, and to a lesser extent of the middle part, follow the open-close movements of the lower jaw. The hind part of the tongue moves predominantly dorsally during slow closing and ventrally during fast opening and fast closing. All three parts move forward during slow closing and slow opening, and backward during fast opening and fast closing. Movements of the hyoid are closely synchronized with those of the hind part of the tongue. Furthermore, tongue and hyoid movements are synchronized with jaw movements. All cycles of Pteropus giganteus are transport cycles, and the synchrony appears to reflect the consistency of the food (soft pulp, juices). Food consistency also accounts for the high swallowing rate and the absence of any significant difference between nonswallowing and swallowing cycles.

    Notes: The reassembly of microtubules is described in mitotic cells after release from nocodazole-induced block. The formation of microtubules was followed by light microscopic immunocytochemical staining using the PAP method, combined with to-luidine blue staining of the chromatin. The light microscopic observations on whole cells were compared with ultrastructural observations on thin sections. This step is essential to ascertain complete destruction of microtubules during the nocodazole treatment and to correlate immunocytochemical staining with the presence of microtubules.Removal of nocodazole (10 or 1 μg/ml) after a sufficiently long incubation to induce a complete disappearance of microtubules resulted in the appearance of tubulin staining specifically associated with the centromeres and with one or two isolated points in the cytoplasm. Electron microscopy confirmed that the staining was due to the massive accumulation of small microtubules at the kinetochores and centrosomes. Kinetochore nucleation was seen only in association with condensed metaphase-stage chromosomes and not with the less-condensed prophase chromosomes.In a second type of experiment cells were allowed to enter mitosis in the presence of an incompletely active concentration of nocodazole (0.1 μg/ml). The construction of the mitotic spindle was arrested; however, short microtubules were assembled at the kinetochores and centrosomes.These experiments demonstrate that in living mitotic PTK2 cells the kinetochores, as well as the centrosomes, exert a nucleating action on tubulin assembly.The further elongation of microtubules after removal of nocodazole was seen to occur preferentially along axes between the centrosomes and the kinetochores. This resulted in the construction of normal metaphases that evolved through anaphase and telophase. We have attempted to formulate a hypothesis that may explain the oriented assembly that seems to be essential in the construction of the spindle.
