    Description: The purpose of this study is to develop the necessary algorithms to determine the bubble size distribution and velocity in the wake of a ventilated or cavitating hydrofoil utilizing background illumination. A simplified experiment was carried out to validate the automatic bubble detection algorithm at the Saint Anthony Falls Laboratory (SAFL) of the University of Minnesota. The experiment was conducted in the SAFL high-speed water tunnel. First, particle shadow velocimetry (PSV) images of a bubbly flow were collected. Bubbles were identified in the images using an edge detection method based on the Canny algorithm. The utilized algorithm was designed to detect partly overlapping bubbles and reconstruct missing parts. After all images were analyzed, the bubble velocity was determined by applying a tracking algorithm.     Description: Little is known about how pro-obesity diets regulate tissue stem and progenitor cell function. Here we show that high-fat diet (HFD)-induced obesity augments the numbers and function of Lgr5(+) intestinal stem cells of the mammalian intestine. Mechanistically, a HFD induces a robust peroxisome proliferator-activated receptor delta (PPAR-delta) signature in intestinal stem cells and progenitor cells (non-intestinal stem cells), and pharmacological activation of PPAR-delta recapitulates the effects of a HFD on these cells. Like a HFD, ex vivo treatment of intestinal organoid cultures with fatty acid constituents of the HFD enhances the self-renewal potential of these organoid bodies in a PPAR-delta-dependent manner. Notably, HFD- and agonist-activated PPAR-delta signalling endow organoid-initiating capacity to progenitors, and enforced PPAR-delta signalling permits these progenitors to form in vivo tumours after loss of the tumour suppressor Apc.     Notes: Abstract The small GTPase Ran is essential for nucleocytoplasmic transport of macromolecules. In the yeast Saccharomyces cerevisiae, Rna1p functions as a Ran-GTPase activating protein (RanGAP1). Strains carrying the rna1-1 mutation exhibit defects in nuclear transport and, as a consequence, accumulate precursor tRNAs. We have isolated two recessive suppressors of the rna1-1 mutation. Further characterization of one of the suppressor mutations, srn10-1, reveals that the mutation (i) can not bypass the need for Rna1p function and (ii) suppresses the accumulation of unspliced pre-tRNA caused by rna1-1. The SRN10 gene is not essential for cell viability and encodes an acidic protein (pI = 5.27) of 24.8 kDa. Srn10p is located in the cytoplasm, as determined by indirect immunofluorescence microscopy. Two-hybrid analysis reveals that there is a physical interaction between Srn10p and Rna1p in vivo. Our results identify a protein that interacts with the yeast RanGAP1.

    Notes: Abstract The effects of SK&F 96365, a blocker of the receptor-operated Ca2+ channel, on contractilities and the Na+ channel of mouse diaphragm were studied. SK&F 96365 (10–50 µM) reversibly inhibited twitches, tetanic contractions and muscle and nerve action potentials. The IC50 was 17–24 µM. The inward Na+ current was suppressed and its recovery from inactivations delayed. Crotamine, a peptide toxin that binds to neurotoxin receptor site 3 of the muscle Na+ channel, enhanced the inhibitory effects of SK&F 96365 and reduced the IC50 to about 4 µM. Veratridine had similar effects, although it was less effective than crotamine. On the other hand, the crotamine-induced membrane depolarizations and spontaneous discharges of muscle action potentials were inhibited by SK&F 96365 noncompetitively. The inhibitory effects of tetrodotoxin and tetracaine were additive with those of SK&F 96365 but were enhanced slightly by crotamine. The results suggested that SK&F 96365 acts on a distinct site and blocks the Na+ channel of excitable membranes at a concentration range that inhibits the receptor-operated calcium channel.

    Notes: Garg's approximate analytical solutions of Biot's equations for wave propagation in a fluid-saturated elastic porous solid of infinite extent subjected to a velocity boundary condition of a Heaviside function at one end are examined for small and large drag. Garg's approximations were apparently introduced to facilitate exact inversion of Laplace transforms of certain quantities. The approximate solutions are compared with carefully evaluated numerical inverses of the Laplace transform solutions for different soils with widely varying properties. It is seen that for most soils (clays, silts and, sands) the error in Garg's approximate solutions in insignificant, and the solutions can be used as benchmarks for verifying numerical analysis procedures such as finite element codes.
