    Beschreibung: Along with the rapid development of GNSS, not only BeiDou, but also Galileo, and the newly launched GPS satellites can provide signals on three frequencies at present. To fully take advantage of the multi-frequency multi-system GNSS observations on precise point positioning (PPP) technology, this study aims to implement the triple-frequency ambiguity resolution (AR) for GPS, Galileo, and BeiDou-2 combined PPP using the raw observation model. The processing of inter-frequency clock bias (IFCB) estimation and correction in the context of triple-frequency PPP AR has been addressed, with which the triple-frequency uncalibrated phase delay (UPD) estimation is realized for real GPS observations for the first time. In addition, the GPS extra-wide-line UPD quality is significantly improved with the IFCB correction. Because of not being contaminated by the IFCB, the raw UPD estimation method is directly employed for Galileo which currently has 24 satellites in operation. An interesting phenomenon is found that all Galileo satellites except E24 have a zero extra-wide-lane UPD value. With the multi-GNSS observations provided by MGEX covering 15 days, the positioning solutions of GPS + Galileo + BeiDou triple-frequency PPP AR have been conducted and analyzed. The triple-frequency kinematic GNSS PPP AR can achieve an averaged 3D positioning error of 2.2 cm, and an averaged convergence time of 10.8 min. The average convergence time can be reduced by triple-frequency GNSS PPP AR by 15.6% compared with dual-frequency GNSS PPP AR, respectively. However, the additional third frequency has only a marginal contribution to positioning accuracy after convergence.

    Beschreibung: Neutrosophy (1995) is a new branch of philosophy that studies triads of the form (〈A〉, 〈neutA〉, 〈antiA〉), where 〈A〉 is an entity {i.e. element, concept, idea, theory, logical proposition, etc.}, 〈antiA〉 is the opposite of 〈A〉, while 〈neutA〉 is the neutral (or indeterminate) between them, i.e., neither 〈A〉 nor 〈antiA〉.Based on neutrosophy, the neutrosophic triplets were founded, which have a similar form (x, neut(x), anti(x)), that satisfy several axioms, for each element x in a given set.This collective book presents original research papers by many neutrosophic researchers from around the world, that report on the state-of-the-art and recent advancements of neutrosophic triplets, neutrosophic duplets, neutrosophic multisets and their algebraic structures – that have been defined recently in 2016 but have gained interest from world researchers. Connections between classical algebraic structures and neutrosophic triplet / duplet / multiset structures are also studied.     Beschreibung: The role of the local microenvironment in influencing cell behavior is central to both normal development and cancer formation. Here, we show that sprouty 1 (SPRY1) modulates the microenvironment to enable proper mammary branching morphogenesis. This process occurs through negative regulation of epidermal growth factor receptor (EGFR) signaling in mammary stroma. Loss of SPRY1 resulted in up-regulation of EGFR–extracellular signal–regulated kinase (ERK) signaling in response to amphiregulin and transforming growth factor alpha stimulation. Consequently, stromal paracrine signaling and ECM remodeling is augmented, leading to increased epithelial branching in the mutant gland. By contrast, down-regulation of EGFR–ERK signaling due to gain of Sprouty function in the stroma led to stunted epithelial branching. Taken together, our results show that modulation of stromal paracrine signaling and ECM remodeling by SPRY1 regulates mammary epithelial morphogenesis during postnatal development.

    Beschreibung: Asparaginyl endopeptidases (AEPs) are cysteine proteases which break Asx (Asn/Asp)–Xaa bonds in acidic conditions. Despite sharing a conserved overall structure with AEPs, certain plant enzymes such as butelase 1 act as a peptide asparaginyl ligase (PAL) and catalyze Asx–Xaa bond formation in near-neutral conditions. PALs also serve as macrocyclases in the biosynthesis of cyclic peptides. Here, we address the question of how a PAL can function as a ligase rather than a protease. Based on sequence homology of butelase 1, we identified AEPs and PALs from the cyclic peptide-producing plants Viola yedoensis (Vy) and Viola canadensis (Vc) of the Violaceae family. Using a crystal structure of a PAL obtained at 2.4-Å resolution coupled to mutagenesis studies, we discovered ligase-activity determinants flanking the S1 site, namely LAD1 and LAD2 located around the S2 and S1′ sites, respectively, which modulate ligase activity by controlling the accessibility of water or amine nucleophile to the S-ester intermediate. Recombinantly expressed VyPAL1–3, predicted to be PALs, were confirmed to be ligases by functional studies. In addition, mutagenesis studies on VyPAL1–3, VyAEP1, and VcAEP supported our prediction that LAD1 and LAD2 are important for ligase activity. In particular, mutagenesis targeting LAD2 selectively enhanced the ligase activity of VyPAL3 and converted the protease VcAEP into a ligase. The definition of structural determinants required for ligation activity of the asparaginyl ligases presented here will facilitate genomic identification of PALs and engineering of AEPs into PALs.
