    Description: This study presents the results of the calculation of the ecological flows below the parameters base, maintenance and maximum of Ecuadorian basins that have hydrometric data, based on the new methodology called Slope Method to the Mass-Curve. For Ecuador to establish the flows is of vital importance for the preservation of the natural ecosystems, taking into account that currently the investment in hydraulic constructions such as dams, hydroelectric power stations and others is enormous. The results obtained allow to establish a baseline for all the projects that somehow affect the fluvial ecosystems, of the geographic basins of Ecuador, in agreement with the availability of the water. The values of the ecological flows are presented graphically according to the location of the hydrometric stations, allowing to appreciate the magnitude of variation of these flows.

    Description: The first years of operation of a project, especially of hydroelectric plants, requires the permanent monitoring of the critical parameters that determine the normal operation of the project, verifying or adjusting the conditions that have been established in the design and construction of the work. For the Manduriacu Project in Ecuador the critical parameter has been the volume of sediments that the Guayllabamba River carries and that reduces the useful volume of the reservoir. This study presents the evaluation of sedimentation of the reservoir for the spring 2016 to spring 2017, conducted with a variety of tools such as the marine drone Z-BOAT 1800 HS (USV), the dual frequency echo sounder ECHOTRAC CV100 DUAL, the Long Range Radio HYDROLINK, the equipment for reception of position data GNSS TRIMBLE R10, the Echosounder control software and eChart connections as well as the Hypack MAX device manager software and other technical support.     Description: Inactivation of the TNFAIP3 gene, encoding the A20 protein, is associated with critical inflammatory diseases including multiple sclerosis, rheumatoid arthritis and Crohn's disease. However, the role of A20 in attenuating inflammatory signalling is unclear owing to paradoxical in vitro and in vivo findings. Here we utilize genetically engineered mice bearing mutations in the A20 ovarian tumour (OTU)-type deubiquitinase domain or in the zinc finger-4 (ZnF4) ubiquitin-binding motif to investigate these discrepancies. We find that phosphorylation of A20 promotes cleavage of Lys63-linked polyubiquitin chains by the OTU domain and enhances ZnF4-mediated substrate ubiquitination. Additionally, levels of linear ubiquitination dictate whether A20-deficient cells die in response to tumour necrosis factor. Mechanistically, linear ubiquitin chains preserve the architecture of the TNFR1 signalling complex by blocking A20-mediated disassembly of Lys63-linked polyubiquitin scaffolds.     Description: Microtubules have pivotal roles in fundamental cellular processes and are targets of antitubulin chemotherapeutics. Microtubule-targeted agents such as Taxol and vincristine are prescribed widely for various malignancies, including ovarian and breast adenocarcinomas, non-small-cell lung cancer, leukaemias and lymphomas. These agents arrest cells in mitosis and subsequently induce cell death through poorly defined mechanisms. The strategies that resistant tumour cells use to evade death induced by antitubulin agents are also unclear. Here we show that the pro-survival protein MCL1 (ref. 3) is a crucial regulator of apoptosis triggered by antitubulin chemotherapeutics. During mitotic arrest, MCL1 protein levels decline markedly, through a post-translational mechanism, potentiating cell death. Phosphorylation of MCL1 directs its interaction with the tumour-suppressor protein FBW7, which is the substrate-binding component of a ubiquitin ligase complex. The polyubiquitylation of MCL1 then targets it for proteasomal degradation. The degradation of MCL1 was blocked in patient-derived tumour cells that lacked FBW7 or had loss-of-function mutations in FBW7, conferring resistance to antitubulin agents and promoting chemotherapeutic-induced polyploidy. Additionally, primary tumour samples were enriched for FBW7 inactivation and elevated MCL1 levels, underscoring the prominent roles of these proteins in oncogenesis.     Description: After a review of the existing formulas for the time of concentration of a basin, it has been established that there is no unique formula that may be applied to a wide range of basins with different geomorphological parameters. So far, the results obtained according to the equations of different authors are completely different and in some cases even inapplicable. Therefore, a new practical formula has been obtained based to the theoretical analysis of the Chezy equation in order to determine the velocity of a flow at a control point, using the single empirical coefficient k. With this formula, the average runoff velocity is reached, which, in turn, allows the calculation of the concentration time of a basin.     Notes: Abstract As part of the technical support CST-12 provides for a wide variety of defense and nondefense programs within Los Alamos National Laboratory (LANL) and the Department of Energy (DOE) complex, new waste minimization technique is under development for radiological volatile organic analysis (Hot VOA). Currently all Hot VOA must be run in a glovebox. Several types of samples contain TRU radiological waste in the form of particulates. By prefiltering the samples, through a 1.2 micron syringe and counting the radioactivity, it has been found that many of the samples can be analyzed outside of a glovebox. In the present investigation, the types of Hot VOA samples that can take advantage of this new technique, the volume and types of waste reduced, and the experimental parameters will be discussed. Overall, the radioactive waste generated is minimized.

    Description: The analysis of concentrations of circulating antibodies in serum (antibody repertoire) is a fundamental, yet poorly studied, problem in immunoinformatics. The two current approaches to the analysis of antibody repertoires [next generation sequencing (NGS) and mass spectrometry (MS)] present difficult computational challenges since antibodies are not directly encoded in the germline but are extensively diversified by somatic recombination and hypermutations. Therefore, the protein database required for the interpretation of spectra from circulating antibodies is custom for each individual. Although such a database can be constructed via NGS, the reads generated by NGS are error-prone and even a single nucleotide error precludes identification of a peptide by the standard proteomics tools. Here, we present the I g R epertoire C onstructor algorithm that performs error-correction of immunosequencing reads and uses mass spectra to validate the constructed antibody repertoires.
