    Notizen: Summary The effect of manganese on the development of vesicular-arbuscular (VA) mycorrhizae and on the population of Mn-oxidizing soil bacteria was studied using red clover as host plant and Glomus mosseae or G. aggregatum as VA-colonizing fungi. The addition of Mn to the substrate in which plants grew had a detrimental effect on root colonization, G. aggregatum being more susceptible than G. mosseae. Mn uptake was lower in mycorrhizal than in non-mycorrhizal plants, especially when colonized by G. mosseae. The development of mycorrhizae seems to favour the Mn-oxidizing bacteria population.

    Notizen: Summary Aqueous extracts of burnt soil, unburnt soil and oven-heated unburnt soil were tested as to their effects on vesicular-arbuscular mycorrhizal (VAM) fungi (spore germination, mycelial propagule activity and root colonization). The extracts of burnt or heated soil inhibited VAM spore germination and extrarrhizal mycelium activity.

    Notizen: Abstract Quantitative and reproducible information concerning the development of the extraradical mycelium of arbuscular mycorrhizal fungi (AMF) is lacking due to the difficulties in extracting, identifying and estimating hyphal lengths. In this study, using a rhizobox growth system, the lengths of hyphae of AMF estimated using an image analysis system were not significantly different from data obtained by a trained observer using a modified grid-line intersect method. The assessment of lengths of hyphae on membrane filters or slides was, however, much quicker using image analysis, and allowed the complete sample to be quantified, unlike the grid-line method where a limited number of fields of view are assessed. The image analysis procedure is objective, observer-independent and less laborious than the manual method of assessment. Of the four different methods of sample preparation compared, membrane filter methods were found to be the most appropriate for quantitative sampling from three non-soil substrates. Glomus monosporum (UKC M3) produced twice as much extraradical mycelium and hyphal length per centimetre of colonised root than G. geosporum (BEG 11) on both leek and linseed in a durite sand at final harvest (63 days). Both AMF also produced more hyphal length per centimetre of colonised root on linseed than on leek. The spatial distribution of both AMF, however, was similar in durite sand and no correlation with levels of NaHCO3-extractable phosphorus was noted. In a third experiment, with G. manihotis (UKC INDO-1) colonising a tropical forage legume, Pueraria phaseoloides, in two other growth substrates, a different pattern of development of the extraradical mycelium was observed. Because of a higher content of particulate matter, which collected on the membrane filters, the extraction technique had to be modified to give optimal performance of the image analysis system.

    Notizen: Summary Red clover was grown in soil previously treated with P at various rates, and growth, nutrient uptake, nutrient uptake in relation to phosphorus values, and levels of vesicular-arbuscular mycorrhizal (VAM) infection were determined. The soil was a silty clay loam and Glomus lacteum was the only fungus colonizing the plant roots. An examination of the effects of various rates of P application and of VAM colonization on nutrient (P, K, Ca, Mg, Mn, Fe, and Zn) uptake showed that the Mg : P ratio significantly increased and the Mn : P ratio significantly decreased with increasing VAM infection. It is concluded that in the Trifolium pratense-Glomus lacteum symbiosis, mycorrhizae improve Mg uptake and depress Mn uptake.

The length of external mycelium and inoculum potential increased with clipping, but decreased with plant burning.     Notizen: Abstract A greenhouse experiment was carried out to determine the growth, micronutrient content and vesicular-arbuscular mycorrhizal (VAM) colonization of fescue and clover plants growing on a composite lignite mine soil. Five VAM fungus inocula were used. Results show that mine spoils had a high potential acidity and poor physical and chemical conditions for plant growth. Toxic levels of Al, Mn and Zn were found in plant tissues, with fescue showing apical necrosis. Glomus deserticola was the only VAM fungus that succeeded in colonizing plant roots, although it failed to improve the establishment or development of either species.

For the soils employed in this study, it proved considerably more effective and more precise than either the original wet sieving and decanting method or differential water/sucrose centrifugation. The apparatus gave results that were less subject to interference from extraneous particles than the wet sieving and decanting method. The effect of prior soil dispersion is also reported.
