    Notizen: Abstract Profiles of diarrhetic shellfish poisoning (DSP) toxins produced throughout the growth cycle and the cell cycle of the toxigenic marine dinoflagellate Prorocentrum lima were studied in triplicate unialgal batch cultures. Cells were pre-conditioned at 18 ± 1 °C, under a photon flux density (PFD) of 90 ± 5 μmol m−2 s−1 on a 14 h light:10 h dark photoperiod. In exponential growth phase, cultures were synchronized in darkness for 17 d. After dark synchronization, cultures were transferred back to the original photoperiod regime. Cells were harvested for DSP toxin analysis by LC-MS (liquid chromatography with mass spectrometry), and double-stranded (nuclear) DNA was quantified by flow cytometry. The cell populations became asynchronous within approximately 3 d after transition from darkness to the 14 h light:10 h dark photoperiod. This may be due to the prolonged division cycle (5 to 7 d) that is not tightly phased by the photoperiod. Unlike other planktonic Prorocentrum spp., cytokinesis in P. lima occurred early in the dark and ceased by “midnight”. Cellular levels of the four principal DSP toxins, okadaic acid (OA), OA C8-diol-ester (OA-D8), dinophysistoxin-1 (DTX1) and dinophysistoxin-4 (DTX4), ranged from 0.37 to 6.6, 0.02 to 1.5, 0.04 to 2.6, and 1.8 to 7.8 fmol cell−1, respectively. No toxin production was evident during the extended period of dark synchronization nor during the initial period when NH4 was consumed as the major nitrogen source. Soon after the cells were returned to the 14 h light:10 h dark cycle and they began to take up NO3, cellular levels of all four toxins gradually increased. This increase in DSP toxins usually occurred in the light, marked by a rise in DTX4 levels that preceded an increase in the cellular concentration of OA and DTX1 (delayed by 3 to 6 h). Thus, DTX4 synthesis is initiated in the G1 phase of the cell cycle and persists into S phase (“morning” of the photoperiod), whereas OA and DTX1 production occurs later during S and G2 phases (“afternoon”). No toxin production was measured during cytokinesis, which happened early in the dark. The evidence indicates that toxin synthesis is restricted to the light period and is coupled to cell cycle events.

    Notizen: Domoic acid is a naturally occurring amino acid, formerly available in limited amounts as a natural product of some algae of the family Rhodomelaceae. As the result of a recent incidence of toxicity in cultured blue mussels (Mytilus edulis) from a highly localized region of Atlantic Canada, useful amounts of this substance are likely to become available in the near future. Domoic acid is an important substance for fundamental studies in neurobiology as it possesses the highest affinity amongst known substances for the kainate receptors of the central nervous system. The present work describes fast atom bombardment (FAB) mass spectra of domoic acid, and of some of its isomers present in minor quantities in contaminated mussel extracts. These FAB spectra are subject to interferences from beam-inducced reduction reactions associated with matrices such as glycerol, but not with others such as 3-nitrobenzyl alcohol. Tandem mass spectrometry of the MH+ ions from these compounds is also reported. As a more feasible approach to quantitative analysis at trace levels, formation of the volatile tert-butyldimethylsilyl derivatives has been investigated and shown to be highly promising.

    Notizen: The establishment of reliable values for concentrations of 16 priority pollutant polycyclic aromatic hydrocarbons (PAH) in a suite of marine sediment reference materials included an examination of the methods used to determine such compounds. Results from the five techniques used indicate large method biases. The biases arise from chemical interferences in methods which use non-selective measuring systems with separation techniques which do not first completely resolve the analytes; e.g., single-parameter optical detectors with high performance liquid chromatographic (HPLC) techniques or a flame ionisation detector (FID) with capillary column gas chromatographic (GC) methods. Measurement by a mass spectrometer with HPLC and GC removes much of the method bias. Results for a representative sediment are discussed to illustrate these conclusions.

    Notizen: The development of microbore packet-column supercritical fluid chromatography/atmospheric pressure chemical ionization mass spectrometry (SFC/APCI-MS) for the analysis of polycyclic aromatic compounds (PAC) is described. The SFC system was coupled to the mass spectrometer using an improved interface designed to accommodate commercially available fused silica restrictors. The atmospheric pressure chemical ionization characteristics of PAC are discussed and the use of chemical ionization reagents to modify the type of spectra obtained is described. With selected-ion monitoring techniques the interface provided detection limits in the low picogram range for individual PAC standards. Using a photoionization detector for the off-line optimization of chromatographic conditions, SFC/APCI-MS was applied to the analysis of complex mixtures of PAC in coal tar and tar sand oil extracts. Tandem mass spectrometry was used to provide structural information.

    Notizen: A flame-generated, fullerene-rich material (J. B. Howard et al., Nature 352, 139 (1991)) has been characterized by liquid chromatography coupled on-line to ionspray mass spectrometry. Charge-transfer complexation, using a post-column addition of either an electron donor or acceptor, was utilized as a pre-ionization technique. Analyses in the positive-ion mode showed that the thermally metastable constituents of the flame-generated material, tentatively identified previously (J. F. Anacleto et al., Rapid Commun. Mass Spectrom. 6, 214 (1992)) as fullerene isomers on the basis of atmospheric pressure chemical ionization muss spectrometry, yielded mass spectra consistent with their identification as fullerene adducts with highly uiuaturated moieties CnH2n-4 with n = 5, 6, and 7. The molecular radical cations of these adduct species were susceptible to facile fragmentation yielding the molecular ions of the corresponding fullerenes. The negative-ion mode experiments failed to provide any information regarding the identity of these metastable constituents.

    Notizen: Workers in ferrous foundries show increased risk of lung cancer. In the steel casting process hot metal is poured into sand moulds solidified with organic binders, producing a plume of smoke containing a variety of organic compounds and showing strong mutagenicity in the Salmonella/S9 assay. We have collected the emissions produced when steel is poured into an experimental sand mould solidified with oil, clay and cereal, a widely used binder system. The organic constituents of these emissions have been fractionated by preparative reverse-phase high perfomance liquid chromatography (HPLC) and mutagenic fractions have been analysed by capillary column gas chromatography/mass spectrometry (GC/MS). Of the 65 compounds for which mass spectra are reported, 54 have been tentatively identified as alkyl derivatives of polycyclic aromatic compounds. Many compounds of this class are known to be carcinogenic and mutagenic. In addition, several unsubstituted polycyclic aromatic hydrocarbons, including the carcinogenic benz[a]anthracene and benzo[a]pyrene, were found to be present.

    Notizen: Summary Domoic acid was recognized recently [1, 2] to be a marine neurotoxin associated with shellfish harvested or cultured for use as human food. Evidence about the occurrence of domoic acid and its importance to shellfish industries is reviewed. The preparation and certification of two reference materials for the determination of domoic acid, an instrument calibration solution (DACS-1, released by the Marine Analytical Chemistry Standards Program [MACSP] in May, 1989), and a tissue reference material (MUS-1, homogenized soft tissues of Mytilus edulis, released by the MACSP in August, 1989) are described. We believe these are the first certified standards or reference materials to be available for the determination of shellfish toxins, a problem of increasing importance to aquaculturists and the seafood industry, as well as to agencies concerned with the safety of food. The tissue homogenate preparation techniques we have developed and used may be of general interest for the preparation of other tissue reference materials for the determination of other organic compounds, since the sealed, fluid homogenate samples seem acceptably stable without being continuously frozen or refrigerated.

    Notizen: A combined liquid chromatography/mass spectrometry (LC/MS) method using ion-spray ionization is described for the sensitive determination of okadaic acid and dinophysistoxin-1, the principal toxins implicated in cases of diarrhetic shellfish poisoning. The method is used to confirm the presence of okadaic acid in a culture of the dinoflagellate species Prorocentrum concavum and in natural populations of dinoflagellates from eastern Canadian waters. Several analogues of the toxin have also been detected. Quantitative results are comparable to those obtained by high performance liquid chromatography with fluorescence detection. With slight modifications, the LC/MS method is also used to analyse the fluorescent anthrylmethyl ester of okadaic acid. Preliminary tandem mass spectrometric results, obtained using fast-atom bombardment ionization, are presented for okadaic acid.
