    Notes: Abstract The National Institute of Standards and Technology (NIST) has developed several Standard Reference Materials (SRMs) based on human serum. NIST SRM 909b, Human Serum, is a lyophilized human serum material with concentrations for seven organic and six inorganic analytes at two levels certified solely by definitive methods (DMs). This material provides the vehicle by which high precision, high accuracy measurements made with DMs at NIST can be transferred through the measurement hierarchy to other laboratories. Isotope dilution gas chromatographic-mass spectrometric (GC-IDMS) methods were applied to measure cholesterol, creatinine, glucose, urea, uric acid, triglycerides, and total glycerides. Thermal ionization isotope dilution mass spectrometry (TI-IDMS) was used for determination of lithium, magnesium, potassium, calcium, and chloride. In addition, chloride was determined by coulometry, providing a comparison between two DMs. Sodium, which lacks a stable isotope that would permit isotope dilution mass spectrometric (IDMS) measurement, was determined by gravimetry. SRM 909b includes certified values for total glycerides and triglycerides, which were not certified in the previous lot of this material (SRM 909a). Improvement in uniformity of vial fill weight in the production of SRM 909b resulted in smaller certified uncertainties over previous freeze-dried serum SRMs. Uncertainties at the 99% level of confidence for relative expanded uncertainty (%) for certification of the organic analytes on a mmol/L/g basis ranged from 0.44% for urea (level II) to 5.04% for glucose (level II). (In-house studies have shown glucose to be a relatively unstable analyte in similar lyophilized serum materials, degrading at about 1% per year.) Relative expanded uncertainties (99% C.I.) for certification of inorganic analytes on a mmol/L/g basis ranged from 0.25% for chloride (level I) to 0.49% for magnesium (level II).

    Notes: It is reported here that the rpr DNA repair gene of Serratia marcescens does not complement an Escherichia coli xth nfo AP endonuclease mutation for resistance to methyl methanesulphonate (MMS). Rather, rpr sensitized Escherichia coli wild-type, xth, and nfo strains to MMS. Also, it was found that rpr could not complement a triple tag alkA recA mutation in E. coli, indicating that there are limits to rpr complementing capabilities. It was determined that rpr gene dosage was not a factor in recA complementation. MMS sensitization of an E. coli wild-type strain, however, was directly related to rpr copy number. These data indicate that Rpr does not have an associated AP endonuclease activity, and that it is incapable of substituting for Tag I, Tag II, and RecA in a tag alkA recA background.

    Notes: Predator inspection behaviour at different levels of attack motivation (attack status of the predator) was investigated in European minnows Phoxinus phoxinus from a population sympatric with pike Esox lucius, during controlled laboratory experiments. Shoals of minnows performed more predator inspections and formed larger inspection group sizes shortly after an attack by a pike. After inspection, minnows returned to the safety of the main shoal, regardless of predator motivation. Minnows which inspected last-before and first-after a strike by a pike modified their behaviour after inspection; they reduced feeding, increased shoaling, flicked their dorsal and pectoral fins and skittered. This behaviour signified alarm and appeared to reflect the severity of the threat posed by the predator at the time of inspection. Fish that had inspected when the pike displayed low attack motivation did not modify their behaviour after inspection to reflect alarm. Information concerning the attack motivation of the predator is probably transferred passively throughout the shoal by changed inspector behaviour and by inspection rate. These data demonstrate that: (1) minnows modified their behaviour after inspection to reflect a predator's attack motivation; and (2) minnows which inspected immediately before a strike appeared to anticipate the future attack and modified their behaviour accordingly.

    Notes: We report here the molecular isolation of a DNA fragment which encodes Tag-like activity from the Gram-negative bacterium Serratia marcescens. A recombinant plasmid encoding Tag-like activity was isolated from a S. marcescens plasmid gene library by complementation of an Escherichia coli tag mutant, which is deficient in 3-methyladenine DNA glycosylase I. The clone complements E. coli tag, recA, alkA, but not alkB, mutants for resistance to the DNA-damaging agent methyl methanesulphonate (MMS). The coding region of the Tag activity, initially isolated on a 6.5kb BamHI fragment, was defined to a 1.8kb BglII-Smal fragment. Labelling of plasmid-encoded proteins using maxicells revealed that the 1.8kb fragment encodes two proteins of molecular weights 42000 and 16000. Data presented here suggest that the cloned fragment encodes a DNA repair protein(s) that has similar activity to the 3-methyladenine DNA glycosylase I of E. coli.

    Notes: Abstract Lead (Pb) and cadmium (Cd) have been determined in six new environmental standard reference materials (SRMs) using isotope dilution inductively coupled plasma mass spectrometry (ID ICP-MS). The SRMs are the following: SRM 1944, New York-New Jersey Waterway Sediment, SRMs 2583 and 2584, Trace Elements in Indoor Dust, Nominal 90 mg/kg and 10,000 mg/kg Lead, respectively, SRMs 2586 and 2587, Trace Elements in Soil Containing Lead from Paint, Nominal 500 mg/kg and 3000 mg/kg Lead, respectively, and SRM 2782, Industrial Sludge. The capabilities of ID ICP-MS for the certification of Pb and Cd in these materials are assessed. Sample preparation and ratio measurement uncertainties have been evaluated. Reproducibility and accuracy of the established procedures are demonstrated by determination of gravimetrically prepared primary standard solutions and by comparison with isotope dilution thermal ionization mass spectrometry (ID TIMS). Material heterogeneity was readily demonstrated to be the dominant source of uncertainty in the certified values.

    Notes: Abstract A cDNA expression library constructed from Culicoides variipennis sonorensis was screened using an antibody specific for Hsp60 of Heliothis virescens. A single clone encoding the complete heat shock protein (Hsp60) of C. variipennis was identified and its 2400-bp insert was sequenced. The encoded 62-kDa protein contains 581 amino acids and includes a 26-amino acid putative mitochondrial targeting sequence at its N terminus and a GGM motif at its carboxyl terminus. Deduced amino acid sequences are highly similar (67–78%) to Hsp60 of other species, including the fruit fly, the house mouse, the Norwegian rat, the Chinese hamster, the human, a nematode, and the tobacco budworm moth. This is the initial isolation of a coding sequence for a stress-induced protein in C. variipennis.

    Notes: Abstract Isotope dilution inductively coupled plasma mass spectrometry (ID-ICP-MS) was applied to the certification of Pb in four levels of NIST blood SRM, 955a. This standard reference material (SRM) represents a significant improvement over the previous blood reference material and will greatly aid method development. The lowest level, 47.76 ng/g Pb was determined with analysis uncertainty (95% CI, ID-ICP-MS uncertainties) of less than 1% and the highest level, 517.9 ng/g Pb to 0.3%. Uncertainty in the lowest level was due to sample inhomogeneity and variability in the analytical blank as the RSD on ratio measurements was typically better than 0.2%. Properly applied isotope dilution coupled with careful isotope ratio measurements on the ICP-MS offers precision and accuracy for blood Pb analyses beyond what is currently obtainable with routine methods.
