    Notes: The recycling of sewage sludge or biosolids to grassland is strategically important in the European Union (EU) and its use is tightly regulated to control the risk of pathogen transfer to animals and the food chain. Sewage sludges not only contain valuable concentrations of beneficial nutrients, but also elevated concentrations of potentially toxic metals (PTM) compared with average background concentrations in the soil. The EC and UK regulations refer to six PTM, Cd, Pb, Cu, Zn, Ni and Hg, with provisional regulations for Cr, that have to be controlled to prevent detrimental effects on soil and animal health. Despite these regulations, there is still a danger that grazing animals may ingest elevated concentrations of PTM. Biosolids may adhere to herbage after the surface application of sewage sludge to grassland. The repeated surface application of sewage sludge to grassland can lead to elevated concentrations of PTM at the soil surface that may be ingested, together with soil and herbage, by grazing ruminants. This may lead to accumulation of Cd or Pb in liver or kidney. The risk to the human food chain is considered to be low, but the impact on the environment is still unknown. There is little information, for example, on the amount of soil and PTMs that may become incorporated into conserved grass. At present EU and UK legislation and voluntary codes of practice have been developed to protect animal from pathogens in sewage sludge and to minimize any potential risks from accumulation of PTM. The background and implementation of the legislation are examined in this review, and the source and mechanisms of accumulation of PTM by the grazing animal are evaluated.

    Notes: Summary The elemental compositions of chloragosome “granules” in the earthworm Lumbricus rubellus living in non-polluted and heavily Zn-polluted soils were determined by fully quantitative electron probe X-ray microanalysis. P, Ca, S and Zn were the major elemental components of the chloragosomes. The in vivo accumulation of Zn by the chloragosomes was accompanied by diminished chloragosomal Ca concentrations. Zn was apparently bound by at least two ligand pools (Pool 1=uncharacterised; Pool 2=P-containing ligands, binding approximately 45% and 55% of the Zn, respectively) in the “control” chloragosomes. In Zn-contaminated chloragosomes, most (∼70%) was bound by P-containing ligand(s) but some (〈1%) was also bound by S-containing ligands. It is suggested that the sequestration of Zn in chloragosomes results in the detoxification of the metal by accumulative immobilisation.

High Pb concentrations were found in chloragosomes of Wemyss animals; Pb was not detected in chloragosomes of Dinas Powys animals. Partial correlation and regression analysis indicated that the in vivo accumulation of Pb by chloragosomes was accompanied by diminished chloragosomal Ca concentrations. Pb is bound by P-containing ligand(s) in the chloragosome matrix. The sequestration of Pb by chloragosomes results in the detoxification of the metal by accumulative immobilization.

    Notes: Summary A problem with the use of muscle grafting as a therapeutic procedure is to produce a graft functionally adequate to replace a muscle of complex architecture, such as a sphincter muscle. We thought it might be possible to use dead cadaver muscles, repopulated by the patient's own muscle precursor cells (mpc), to reconstruct muscles whose anatomy would be imposed by the framework of dead muscle and whose genetic constitution would be determined by the mpc. Here we show, in the mouse, that an extensor digitorum longus (EDL) muscle, killed by repeated freezing and thawing, repopulated with mpc and grafted into a nu/nu or tolerant AKR host mouse, is capable of supporting muscle formation. By using the allotypic isoenzyme forms of glucose-6-phosphate isomerase as markers, we have shown that the newly regenerated muscle in such grafts is derived mainly from the implanted mpc, but also to some extent from the host mouse's own mpc. By 50–70 days after grafting, new muscle fibres were found to constitute up to 70% of the graft. Many fibres had assumed diameters in the normal range for mouse muscle, often having peripherally placed nuclei. These findings raise the possibility of the therapeutic use of such grafts. To our surprise, dead EDL muscle grafts into which no mpc had been implanted were also the site of good muscle regeneration. New-formed muscle in these grafts was shown to be derived entirely from mpc which must have migrated into the graft from the host. Investigation of the mechanisms underlying this phenomenon should further our knowledge of factors which regulate the proliferation and movement of dormant mpc in adult animals.
