    Description: Under a tension of ~65 pN, double-stranded DNA undergoes an overstretching transition from its basic (B-form) conformation to a 1.7 times longer conformation whose nature is only recently starting to be understood. Here we provide a structural and thermodynamic characterization of the transition by recording the length transient following force steps imposed on the -phage DNA with different melting degrees and temperatures (10–25°C). The shortening transient following a 20–35 pN force drop from the overstretching force shows a sequence of fast shortenings of double-stranded extended (S-form) segments and pauses owing to reannealing of melted segments. The lengthening transients following a 2–35 pN stretch to the overstretching force show the kinetics of a two-state reaction and indicate that the whole 70% extension is a B-S transition that precedes and is independent of melting. The temperature dependence of the lengthening transient shows that the entropic contribution to the B-S transition is one-third of the entropy change of thermal melting, reinforcing the evidence for a double-stranded S-form that maintains a significant fraction of the interstrand bonds. The cooperativity of the unitary elongation (22 bp) is independent of temperature, suggesting that structural factors, such as the nucleic acid sequence, control the transition.

    Description: Spinal muscular atrophy (SMA) is a neurodegenerative disease caused by the loss of Survival Motor Neuron-1 ( SMN1 ) . In all SMA patients, a nearly identical copy gene called SMN2 is present, which produces low levels of functional protein owing to an alternative splicing event. To prevent exon-skipping, we have targeted an intronic repressor, Element1 (E1), located upstream of SMN2 exon 7 using Morpholino-based antisense oligonucleotides (E1 MO -ASOs). A single intracerebroventricular injection in the relatively severe mouse model of SMA (SMN7 mouse model) elicited a robust induction of SMN protein, and mean life span was extended from an average survival of 13 to 54 days following a single dose, consistent with large weight gains and a correction of the neuronal pathology. Additionally, E1 MO -ASO treatment in an intermediate SMA mouse (SMN RT mouse model) significantly extended life span by ~700% and weight gain was comparable with the unaffected animals. While a number of experimental therapeutics have targeted the ISS-N1 element of SMN2 pre-mRNA, the development of E1 ASOs provides a new molecular target for SMA therapeutics that dramatically extends survival in two important pre-clinical models of disease.

    Description: The function of the c-myc gene and its role in tumorigenesis are poorly understood. In order to elucidate the role of c-myc oncogene activation in B cell malignancy, the phenotypic changes caused by the expression of c-myc oncogenes in human B lymphoblastoid cells immortalized by Epstein-Barr virus were analyzed. C-myc oncogenes caused the down-regulation of lymphocyte function-associated antigen-1 (LFA-1) adhesion molecules (alpha L/beta 2 integrin) and loss of homotypic B cell adhesion in vitro. Down-regulation of LFA-1 occurred by (i) posttranscriptional modulation of LFA-1 alpha L-chain RNA soon after acute c-myc induction, and (ii) transcriptional modulation in cells that chronically express c-myc oncogenes. Analogous reductions in LFA-1 expression were detectable in Burkitt lymphoma cells carrying activated c-myc oncogenes.     Description: We present a strong lensing system, composed of four multiple images of a source at z  = 2.387, created by two lens galaxies, G1 and G2, belonging to the galaxy cluster MACS J1115.9+0129 at z  = 0.353. We use observations taken as part of the Cluster Lensing and Supernova survey with Hubble, and its spectroscopic follow-up programme at the Very Large Telescope, to estimate the total mass distributions of the two galaxies and the cluster through strong gravitational lensing models. We find that the total projected mass values within the half-light radii, R e , of the two lens galaxies are M T,G1 (〈 R e , G1 ) = (3.6 ± 0.4)  x  10 10 M and M T,G2 (〈 R e , G2 ) = (4.2 ± 1.6)  x  10 10 M . The effective velocity dispersion values of G1 and G2 are (122 ± 7) km s –1 and (137 ± 27) km s –1 , respectively. We remark that these values are relatively low when compared to those of 200–300 km s –1 , typical of lens galaxies found in the field by previous surveys. By fitting the spectral energy distributions of G1 and G2, we measure projected luminous over total mass fractions within R e of 0.11 ± 0.03, for G1, and 0.73 ± 0.32, for G2. The fact that the less massive galaxy, G1, is dark matter-dominated in its inner regions raises the question of whether the dark matter fraction in the core of early-type galaxies depends on their mass. Further investigating strong lensing systems will help us understand the influence that dark matter has on the structure and evolution of the inner regions of galaxies.

    Notes: Zusammenfassung Aus den gewonnenen Resultaten kann man folgende Schlüsse ziehen: Die magnetische Flußverteilung im Eisenkern des fünfschenkligen Dreiphasentransformators darf für praktische Zwecke auf Grund der normalen oder mittleren Permeabilitätskurve nach den Formeln von Küchler und Stallmann, oder einfacher nach unserer graphischen Methode, bestimmt werden. Hierzu können aus Symmetriegründen beiden Jochen und Rückschenkeln die gleichen Werte der magnetischen Permeabilitätμ 4 =μ 7 ;μ 5 =μ 6 zugeschrieben werden, wenn auch die entsprechenden Induktionsflüsse durch den Einfluß der Oberwellen dritter und höherer Ordnung nicht die gleiche Form und Amplitude besitzen werden. Um die Bedeutung dieser Oberwellen soweit wie möglich herunterzudrücken, benutzt man am besten die Dreieckschaltung einer besonderen, tertiären Wicklung, falls die primäre und sekundäre Wicklung aus anderen Gründen in Stern geschaltet werden. Eine direkte Verbindung der Nullpunkte des Generators und Transformators ist weniger wirksam, und wird in der Praxis seltener gebraucht. Unter gewöhnlichen Verhältnissen wird die Reluktanz der Rückschenkel bedeutend größer als die der Joche; aus diesem Grunde sind auch die ersten meistens magnetisch weniger ausgenutzt als die letzteren. Die Eisenverluste bei dem gleichen Gesamtgewicht werden demnach größer als die eines dreischenkligen Kerns. Um diesen Unterschied zu begrenzen, sind die Stoßfugen so klein wie möglich zu halten, oder das Eisenpaket überlappt wie bei der Manteltype aufzubauen. Die nicht ganz einfache Anordnung, die von Prof. Vidmar in dem eingangs erwähnten Aufsatz vorgeschlagen wurde: besondere Luftspalte in den eigentlichen Jochstrecken einstellbar zu machen, und somit die Trennung des ganzen Transformators in drei selbständige Teile, die bewickelt zum Versand kommen können, vorzunehmen, steht mit dem Zweck der neuen Bautype in einigem Widerspruch, und dürfte aus verschiedenen Gründen kaum empfohlen werden. Die Eisenverluste können jedenfalls nach der berechneten Flußverteilung mit genügender Annäherung vorgesehen werden, weil die zufälligen Fehler der Induktionsbestimmung in den Jochen und Rückschenkeln entgegengesetztes Vorzeichen besitzen und sich daher größtenteils aufheben.
