    Notizen: Summary The retention behaviour of some substituted benzoic acids was tested on reversed-phase pre-coated plates in the presence of ammonium bromide and various tetraalkylammonium compounds with different alkyl chain lengths. It could be shown that the Rf values of the test compounds decrease in the absence and in the presence of ion-pair reagents with increasing water content of the eluent. The Rf values of the strongly acidic substituted benzoic acids decrease with both increasing concentration of the ion-pair reagent and with increasing chain length of the tetraalkylammonium compounds. Changing the inorganic counter-ions had no real influence on the separation and the size of Rf values when using tetramethylammonium compounds as ion-pair reagents.

    Notizen: Summary Up to now in thin-layer chromatography a series of retention mechanisms for the determination of enantiomeric compositions have been applied. Subject of this paper is a corresponding survey with a focal point on ligand exchange chromatography. In connection with this retention mechanism most successfully used in TLC for the separation of optical antipodes, some additional findings of the authors are included. Retention mechanisms in question are: - Formation of inclusion complexes within the cavities of cyclodextrins. Substance classes separated by this system are e.g. dansyl amino acids or metallocenes. - Interaction with so-called “brush type” phases. In this case, chiral groups with π-acceptor of π-donor properties are chemically or physically bonded to an aminomodified silica gel matrix. Chiral compounds, like aromatic alcohols, were separated on these stationary phases. - Differences in retention of diastereomers formed previously or during the chromatographic process on an achiral stationary phase. Some examples for the manifold applicability of this principle are the separation of chiral amines from biological matrices, hydroxy-fatty acids, and amino acids. - Selective interactions with the helical structures of cellulose or cellulose triacetate. Referring to this technique, separations of several amino acids and of the analgesic agent Oxidanac in the R-and S-species are described. - Enantioselective ligand exchange, based on reversed phase silica gels impregnated with copper salts and optically active amino acid derivatives. In thin-layer chromatography this is the most important principle used mainly for the separation of racemic mixtures of free amino acids and amino acid derivatives. The recently developed HPTLC pre-coated plate CHIR with concentrating zone is also based on this retention mechanism. Aside from the applications mentioned above, dipeptides, hydroxy-carboxylic acids, α-halogenated carboxylic acids, amino lactones, and thiazolidine derivatives have been separated on this plate up to now. Furthermore, this new pre-coated layer is particularly suitable for an optical in situ evaluation and quantitative determinations of optical antipodes.

    Notizen: Summary A simple method for separating and detecting glucose and fructose is described. Merely heating the chromatogram under an IR radiator, or on a hotplate or in a drying cabinet, renders components separated on HPTLC plates NH2 visible under UV light. The detection limits are similar to those obtained with spray reagents and dipping techniques. Possible application of the method to other carbohydrates is briefly mentioned.

Chromatographic separation on NH2-modified silica gel layers is followed by in-situ visualization brought about by heating the plate.     Notizen: Summary In a manner analogous to that for surface-active silica gel, HPTLC pre-coated plates for nano TLC have also been developed from two inactive sorbents. The two materials are microcrystalline cellulose and a synthetically produced, porous silica (Silica 50000) with a very low specific surface area. The chromatographic properties of these inert sorbents and of the new HPTLC pre-coated plates prepared therefrom are examined in relation to separations of amino acid mixtures and carbohydrate mixtures and are related to the chromatographic properties of the inactive sorbents and TLC precoated plates used hitherto.

This is a moderately polar sorbent based on silica gel 60, which can be used both in adsorption chromatography and in reversed-phase chromatography. This new pre-coated layer is particularly suitable for separation of steroids, alkaloids and derivatized amino acids. The possibility of separating habitforming drugs and preservatives in the presence of ionpair reagents is also discussed.

    Notizen: Summary Based on experience in the purely thermally induced (no analytical reagents) fluorimetric detection of glucose and fructose after thin-layer chromatographic separation on NH2-layers, the application of this method to other sugars is described. After separation with mobile phases containing pyridine, this “dry”in situ derivatization permits a very simple qualitative and quantitative method of detection for the 20 carbohydrates studied.
