    Notes: Although the co-ordination of promotive root-sourced cytokinin (CK) and inhibitory shoot apex-sourced auxin (IAA) is central to all current models on lateral bud dormancy release, control by those hormones alone has appeared inadequate in many studies. Thus it was hypothesized that the IAA : CK model is the central control but that it must be considered within the relevant timeframe leading to lateral bud release and against a backdrop of interactions with other hormone groups. Therefore, IAA and a wide survey of cytokinins (CKs), were examined along with abscisic acid (ABA) and polyamines (PAs) in released buds, tissue surrounding buds and xylem sap at 1 and 4 h after apex removal, when lateral buds of chickpea are known to break dormancy. Three potential lateral bud growth inhibitors, IAA, ABA and cis-zeatin 9-riboside (ZR), declined sharply in the released buds and xylem following decapitation. This is in contrast to potential dormancy breaking CKs like trans-ZR and trans-zeantin 9-riboside 5′phosphate (ZRMP), which represented the strongest correlative changes by increasing 3.5-fold in xylem sap and 22-fold in buds. PAs had not changed significantly in buds or other tissues after 4 h, so they were not directly involved in the breaking of bud dormancy. Results from the xylem and surrounding tissues indicated that bud CK increases resulted from a combination synthesis in the bud and selective loading of CK nucleotides into the xylem from the root.

    Notes: Using two ecotypes of Stellaria longipes an alpine form with low plasticity and a prairie form with high plasticity, we investigated whether ethylene was involved in the response to wind stress and might be important in controlling plasticity of stem elongation. Stem growth inhibition was positively correlated with concentration of ethephon application and elevation in ambient ethylene in alpine ecotypes, whereas stem growth in prairie plants was stimulated by low ethephon concentrations. When treated with high AVG, the effects were reversed: alpine plant growth was promoted and prairie plant growth was inhibited. Prairie plants exhibited a daily rhythm in ethylene evolution which increased and peaked at 1500 h, and which was absent in alpine plants. Ethylene evolution did not change significantly during the first 2 weeks of growth in alpine plants, whereas ethylene in prairie plants increased significantly during periods of rapid stem elongation. Wind treatment inhibited growth in both ecotypes, but only alpine plants showed a recovery of growth to control levels when wind stressed plants were pretreated with STS. In addition, only alpine plants showed an increase in ethylene evolution in response to wind simulation, whereas prairie plant ethylene evolution did not deviate from rhythms observed in unstressed plants. We concluded that ethylene dwarfs stems in alpine S. longipes in response to wind stress. However, low levels of ethylene may stimulate growth in prairie ecotypes and act independently of wind stress intensity. The contrasting ability to synthesize and respond to ethylene can account for part of the difference in plasticity documented between the two ecotypes.

    Notes: The most widely accepted model of radial water entry from the soil into the xylem of roots is based on principles of ordinary passive diffusion. However, long-standing problems with this model remain unresolved, which concern variable intrinsic properties of conductivity, Lp, changing reflection coefficients, σ, and inaccurate resolution of osmotic differentials between the soil and xylem. Our study re-examined pressure flow relationships in isolated roots of tomato (Lycopersicon esculentum Mill. cv. Montfavet), pea (Pisum sativum cv. Baccara) and soybean (Glycine max L. Merryl cv. Essor) manipulated in a pressure chamber. In addition to problems previously recognized with the simple passive diffusion model, a new conflict, flow saturation, was observed at high pressures. Experiments revealed that the plateau in flow, Jmax seen at high pressures followed natural rhythms diurnally and developmentally, and was not due to root damage or unnatural flow restriction. Near the end of the photoperiod, Jmax closely correlated with root dry mass. The above inconsistencies between observations in pressure-flow kinetics and ordinary passive diffusion indicate that either the current model should be adjusted or a new model should be proposed.

    Notes: Although cytokinins (CKs) are widely thought to have a role in promoting shoot branching, there is little data supporting a causative or even a correlative relationship between endogenous CKs and timing of bud outgrowth. We previously showed that lateral bud CK content increased rapidly following shoot decapitation. However, it is not known whether roots are the source of this CK. Here, we have used shoot decapitation to instantaneously induce lateral bud release in chickpea seedlings. This treatment rapidly alters rate and direction of solvent and solute (including CK) trafficking, which may be a passive signalling mechanism central to initiation of lateral bud release. To evaluate changes in xylem transport, intact and decapitated plants were infiltrated with [3H]zeatin riboside ([3H]ZR), a water-soluble blue dye or [3H]H2O by injection into the hypocotyl. All three tracers were recovered in virtually all parts of the shoot within 1 h of injection. In intact plants, solute accumulation in the lateral bud at node 1 was significantly less than in the adjacent stipule and nodal tissue. In decapitated plants, accumulation of [3H]ZR and of blue dye in the same bud position was increased 3- to 10-fold relative to intact plants, whereas content of [3H]H2O was greatly reduced indicating an increased solvent throughput. The stipule and cut stem, predicted to have high evapotranspiration rates, also showed increased solute content accompanied by enhanced depletion of [3H]H2O. To assess whether metabolism modifies quantities of active CK reaching the buds, we followed the metabolic fate of [3H]ZR injected at physiological concentrations. Within 1 h, 80–95% of [3H]ZR was converted to other active CKs (mainly zeatin riboside-5′phosphate (ZRMP) and zeatin (Z)), other significant, but unconfirmed metabolites some of which may be active (O-acetylZR, O-acetylZRMP and a compound correlated with sites of high CK-concentrations) and inactive catabolites (adenosine, adenine, 5′AMP and water). Despite rapid metabolic degradation, the total active label, which was indicative of CK concentration in buds, increased rapidly following decapitation. It can be inferred that xylem sap CKs represent one source of active CKs appearing in lateral buds after shoot decapitation.

    Notes: Abstract Both methyl jasmonate (MJ) and ethylene have been implicated in promoting senescence, but the specific roles of each and the mechanisms by which they act are not well known. We tested the possibility that MJ and ethylene interact to promote senescence. In sunflower seedlings, the ability of MJ to affect ethylene metabolism was investigated in hypocotyls, cotyledons, and leaves. 1-aminocylcopropane-1-carboxylic acid (ACC)-dependent ethylene production was promoted to different extents depending on the organ and the age of the tissue. Newly emerged hypocotyls were sensitive to MJ, but became desensitized as the cotyledons emerged. The cotyledons increased and peaked in MJ sensitivity from emergence to the production of the primary leaves. Leaves were found to be somewhat insensitive to MJ treatment compared to cotyledons at all ages tested. In cotyledons, MJ also promoted ACC and ethylene production. However the changes in ACC, and ACC-dependent ethylene production were not directly correlated with those in ethylene production with respect to MJ concentration or tissue age. Moreover, changes in ACC-dependent ethylene production did not correlate with in vitro ACC oxidase activity. We hypothesized that MJ affects ethylene production by increasing the spatial access of ACC to ACC oxidase perhaps through increased membrane permeability. Ethylene was not involved in the MJ-induced loss of chlorophyll. But the breakdown of cell integrity and cell membranes (estimated by monitoring conductivity of the solution that bathed the cotyledons) was greatly and synergistically promoted by the combination of MJ and ethylene. Promotion of membrane breakdown by MJ and ethylene could be inhibited by treatments with ethylene inhibitors (STS or CoCl2), and neither MJ nor ACC treatment alone could induce as much membrane breakdown as both together. We suggest that MJ and ethylene interact to accelerate some aspects of senescence in specific organs for nutrient remobilization for the benefit of the whole plant.

    Notes: Abstract Morphological relationships were investigated among diploidStellaria porsildii, polyploidS. longipes, and diploidS. longifolia. Canonical discriminant analysis, based on a priori assumptions to maximize differences among groups, showed thatS. longipes clusters equally distant between the two diploid species along an axis connecting the diploids' centroids, but it differs along an axis perpendicular to this axis. The intermediacy along the former axis is evidence thatS. longipes is an amphiploid derived fromS. longifolia andS. porsildii. The divergence along the latter axis may be attributable to adaptively valuable heterotic traits which were retained following amphidiploidization. The only morphological discontinuity occurred between the two diploids, whereas the morphological range ofS. longipes overlapped the range of both diploids forming a continuum. The lack of discrete clusters is likely due to hybridization and introgression withS. longifolia on one hand, and convergence of traits betweenS. longipes var.monantha andS. porsildii on the other. High a posteriori assignments in classificatory discriminant analysis supports the separation ofS. longipes var.monantha from otherS. longipes specimens. AlthoughS. longipes var.monantha grouped close toS. porsildii, the two groups separate based on leaf shape traits. Overall results support, firstly, the hypothesis thatS. porsildii is a diploid parent species which by hybridizing withS. longifolia gave rise to polyploidS. longipes. Secondly, results suggest thatS. longipes var.monantha converged morphologically towardsS. porsildii relatively recently due to ecological specialization, and merits distinction at least as a variety ofS. longipes.
