    Description: At high ambient temperature, plants display dramatic stem elongation in an adaptive response to heat. This response is mediated by elevated levels of the phytohormone auxin and requires auxin biosynthesis, signaling, and transport pathways. The mechanisms by which higher temperature results in greater auxin accumulation are unknown, however. A basic helix-loop-helix transcription factor, PHYTOCHROME-INTERACTING FACTOR 4 (PIF4), is also required for hypocotyl elongation in response to high temperature. PIF4 also acts redundantly with its homolog, PIF5, to regulate diurnal growth rhythms and elongation responses to the threat of vegetative shade. PIF4 activity is reportedly limited in part by binding to both the basic helix-loop-helix protein LONG HYPOCOTYL IN FAR RED 1 and the DELLA family of growth-repressing proteins. Despite the importance of PIF4 in integrating multiple environmental signals, the mechanisms by which PIF4 controls growth are unknown. Here we demonstrate that PIF4 regulates levels of auxin and the expression of key auxin biosynthesis genes at high temperature. We also identify a family of SMALL AUXIN UP RNA (SAUR) genes that are expressed at high temperature in a PIF4-dependent manner and promote elongation growth. Taken together, our results demonstrate direct molecular links among PIF4, auxin, and elongation growth at high temperature.

    Description: Neurokinin-1 receptor (NK1R) and mu-opioid receptor (muOR) agonists affected respiratory rhythm when injected directly into the preBotzinger Complex (preBotC), the hypothesized site for respiratory rhythmogenesis in mammals. These effects were mediated by actions on preBotC rhythmogenic neurons. The distribution of NK1R+ neurons anatomically defined the preBotC. Type 1 neurons in the preBotC, which have rhythmogenic properties, expressed both NK1Rs and muORs, whereas type 2 neurons expressed only NK1Rs.     Description: In the developing brain, transcription factors (TFs) direct the formation of a diverse array of neurons and glia. We identifed 1445 putative TFs in the mouse genome. We used in situ hybridization to map the expression of over 1000 of these TFs and TF-coregulator genes in the brains of developing mice. We found that 349 of these genes showed restricted expression patterns that were adequate to describe the anatomical organization of the brain.     Description: Previous studies have shown a correspondence between the abundance of particular plant species and methane flux. Here we apply multivariate analyses, including a weighted averaging approach, to assess the suitability of vegetation composition as a predictor of methane flux. We developed a functional classification of the vegetation, in terms of a number of plant traits expected to influence methane production and transport, and compared this with a purely taxonomic classification at species-level and higher. We applied both weighted averaging and indirect and direct ordination approaches to six sites in the UK, and found good relationships between methane flux and vegetation composition (classified both taxonomically and functionally). Plant species and functional groups also showed meaningful responses to management and experimental treatments. In addition to the UK, we applied the functional group classification across different geographical regions (Canada and Netherlands) to assess the generality of the method. Again, the relationship appeared good at the site level, suggesting some general applicability of the functional classification. The method seems to have the potential for incorporation into large-scale (national) greenhouse gas accounting programmes (in relation to peatland condition/management) using vegetation mapping schemes. The results presented here strongly suggest that robust predictive models can be derived using plant species data (for use in national-scale studies). For trans-national-scale studies, where the taxonomic assemblage of vegetation differs widely between study sites, a functional classification of plant species data provide an appropriate basis for predictive models of methane flux. © 2012 Blackwell Publishing Ltd

    Description: Successful quantitative precipitation forecasts under convectively unstable conditions depend on the ability of the model to capture the location, timing and intensity of convection. Ensemble forecasts of two mesoscale convective outbreaks over the UK are examined with a view to understanding the nature and extent of their predictability. In addition to a control forecast, twelve ensemble members are run for each case with the same boundary conditions but with perturbations added to the boundary layer. The intention is to introduce perturbations of appropriate magnitude and scale so that the large-scale behaviour of the simulations is not changed. In one case, convection is in statistical equilibrium with the large-scale flow. This places a constraint on the total precipitation, but the location and intensity of individual storms varied. In contrast, the other case was characterised by a large-scale capping inversion. As a result, the location of individual storms was fixed, but their intensities and the total precipitation varied strongly. The ensemble shows case-to-case variability in the nature of predictability of convection in a mesoscale model, and provides additional useful information for quantitative precipitation forecasting.
