    Description: The tip is one of the critical factors to improve the efficiency in picking up individual DNA molecules from solid substrates based on atomic force microscope (AFM) nanomanipulation. We found that wearing AFM tips on certain solid substrates in advance to nanomanipulation operation would largely improve the pickup efficiency, which was ascribed to the increasing affinity of the tip to the DNA molecules along with the increase of the tip radius after wearing. It was demonstrated that bare mica was superior to APTES-modified mica to keep the tip clean while wearing, which was crucial for DNA pickup during AFM nanomanipulation. Microsc. Res.     Description: The mobile DOAS technique was used to quantify the NOx vehicle emission of the central urban area, enclosed by the Inner Ring Viaduct Road (IRVR), of Shanghai, China. Three field measurement campaigns were performed during the pre-EXPO period in October 2009, the EXPO opening ceremony, and the closing ceremony period in 2010. The spatial and temporal distributions of NO2 Vertical Column Densities were derived, and the effects of traffic and wind conditions were studied. The averaged NO2 emissions rates from the IRVR area were determined to be 2.1 ± 0.9 ton/h, 2.8 ± 1.4 ton/h and 2.7 ± 1.4 ton/h for these three campaigns. The annual vehicle NOx emissions for the central urban area were estimated to be (2.3 ± 1.0) × 104 ton in 2009 and (3.0 ± 1.5) × 104 ton in 2010, and those for the whole city were (13.4 ± 5.9) × 104 ton in 2009 and (17.6 ± 8.4) × 104 ton in 2010, by considering a typical NO/NO2 ratio, NOx lifetime, as well as the traffic emission share of central urban area with respect to the entire city. The vehicle NOx emissions quantified by mobile DOAS were found to exhibit an increasing trend in the central urban area of Shanghai, by 1.6 times and 2.1 times from 2006 to 2009 and 2010. Due to wind field variations, unknown NOx chemical transformation, and uncertainties in AMFs, the systematic derivation of the final NOx emission rates obtained was about 55% for averaged several consecutive days.

    Description: Groundwater flow systems and stagnant zones in drainage basins are critical to a series of geologic processes. Unfortunately, the difficulty of mapping flow system boundaries and no field example of detected stagnant zones restrict the application of the concept of nested flow systems. By assuming the variation in bulk resistivity of an aquifer with uniform porosity is mainly caused by groundwater salinity, the magnetotelluric technique is used to obtain the apparent resistivity of a profile across a groundwater-fed river in the Ordos Plateau, China. Based on the variations in apparent resistivity of the Cretaceous sandstone aquifer, the basin-bottom hydraulic trap below the river has been detected for the first time, and its size is found to be large enough for possible deposition of large ore bodies. The boundary between local and regional flows has also been identified, which would be useful for groundwater exploration and calibration of large-scale groundwater models.

    Description: The multi-walled (B, N)-codoped titanate nanotubes (TNTs) have been prepared via a simple hydrothermal route. Such (B, N)-codoped TNTs showed a decrease in band gap energy as compared with the undoped TNTs. The effect of the doping amounts of B and N on the photocatalytic activity of TNTs was studied. Among the as-prepared samples, (1% B, 1% N)-codoped TNTs exhibited the best photocatalytic activity for methyl orange decomposition under Ultraviolet–Visible (UV–Vis) light irradiation. The photocatalytic activity of (1% B, 1% N)-codoped TNTs is twice as high as that of the undoped TNTs.

    Description: Anoctamin1 (ANO1) encodes a Ca 2+ -activated chloride (Cl - ) channel (CaCC) in variety tissues of many species. Whether ANO1 expresses and functions as a CaCC in cardiomyocytes remain unknown. The objective of this study is to characterize the molecular and functional expression of ANO1 in cardiac myocytes and the role of ANO1-encoded CaCCs in ischemia-induced arrhythmias in the heart. Quantitative real-time RT-PCR, immunofluorescence staining assays and immunohistochemisrty identified the molecular expression, location and distribution of ANO1 in mouse ventricular myocytes (mVMs). Patch-clamp recordings combined with pharmacological analyses found that ANO1 was responsible for a Ca 2+ -activated Cl - current ( I Cl.Ca ) in cardiomyocytes. Myocardial ischemia led to a significant increase in the current density of I Cl.Ca , which was inhibited by a specific ANO1 inhibitor, T16A inh -A01, and an antibody targeting at the pore area of ANO1. Moreover, cardiomyocytes isolated from mice with ischemia-induced arrhythmias had an accelerated early phase 1 repolarization of action potentials (APs) and a deeper “spike and dome” compared to control cardiomyocytes from non-ischemia mice. Application of the antibody targeting at ANO1 pore prevented the ischemia-induced early phase 1 repolarization acceleration and caused a much shallower “spike and dome”. We conclude that ANO1 encodes CaCC and plays a significant role in the phase 1 repolarization of APs in mVMs. The ischemia-induced increase in ANO1 expression may be responsible for the increased density of I Cl.Ca in the ischemic heart and may contribute, at least in part, to ischemia-induced arrhythmias. © 2014 Wiley Periodicals, Inc.

    Description: Phase stability and macroscopic performances of ZrC are closely related to the behavior of native point defects. In this study, structures and stabilities of native point defects in ZrC as well as diffusion of C-related defects are investigated by first-principles calculations. It is shown that the carbon vacancy ( V C ) and interstitials (C i s) are the dominant native point defects in ZrC . Six types of C i configurations: two C-trimers, one C-tetrahedron, and three C-dimers are identified with low defect formation energies. The V C has a high migration energy (4.39 eV) which suggests its low mobility in ZrC . The C i s have low diffusion energy barriers (from 0.26 to 1.29 eV) which lead to their high mobility. In addition, the impact of defects ( V C , V Zr , Zr C , and C Zr ) on bonding strengths of neighboring Zr – C bonds is discussed. Especially near V C , the 1NN (nearest neighboring) and 2NN Zr – C bonds are strengthened but 4NN Zr – C bonds are weakened. Interestingly, the 3NN Zr – C bonds are almost not affected by the presence of V C . These results may be closely related to the short-range interactions and ordering of V C in nonstoichiometric ZrC .

    Description: The tropical-extratropical teleconnection during boreal summer is characterized by a circumglobal teleconnection (CGT) pattern in the northern hemisphere (NH). Around the late 1970s, there was an abrupt change in the coupled atmosphere-ocean system over the NH and global tropics. This study found that the CGT has experienced a significant change since late 1970s. During the recent epoch (1979-2010), the major CGT centers weakened with pronounced changes over the North Atlantic and Europe. Partial correlation analysis of 200-hPa geopotential height and the NH summer monsoon heat sources indicates that the change of Indian summer monsoon (ISM) rainfall has a global impact on the CGT, whereas other NH summer monsoons (West African, western North Pacific, and North American) have only a regional modification to the CGT. The weakening of the CGT pattern in the recent epoch is a consequence of weakened coupling between ISM rainfall and midlatitude circulation, which results from the reduced interannual variability of ISM rainfall due to changes in El Niño-Southern Oscillation properties. The large change of the CGT over the North Atlantic and Europe is attributed to a southward shift of upper-level westerlies and enhanced coupling to West African monsoon rainfall.

    Description: Forkhead (Fox) transcription factors display functional diversity and are involved in various metabolic and developmental processes. The Spodoptera exigua Fox ( SeFox ) encodes a protein of 353 amino acids with a theoretical molecular mass of approximately 38.99 kDa and an isoelectric point of 8.86. qPCR results revealed that SeFox was expressed mainly in the brain, fat body, epidermis, midgut, Malpighian tubules, and testis. SeFox was expressed, with some changes, throughout development in the fat body and whole body. Injection of dsSeFox ( SeFox dsRNA) into larvae resulted in incidences of albino plus molting deformity (4.8%), molting deformity (26.2%), and albino phenotypes (69.1%). dsSeFox injection resulted in approximately 50% knockdown of transcript levels at 36 h. Compared with control groups, hexokinase ( HK) expression was reduced to approximately 40% at 48 h postinjection. Chitin synthase A ( CHSA ) expression was reduced to two-thirds at 24 h, but increased at 72 h. Compared with untreated control and green fluorescent protein-treated groups, Chitin synthase B ( CHSB ) expression decreased to 33% following dsSeFox injection by 36 h. We infer from our results that forkhead transcription factors act in chitin synthesis in S. exigua .
