    Notes: Abstract. P-glycoprotein (P-gp), the product of human MDR1 gene, which functions as an ATP-dependent drug efflux pump, is N-linked glycosylated at asparagine residues 91, 94, and 99 located within the first extracellular loop. We report here the biochemical characterization of glycosylation-deficient (Gly−) P-gp using a vaccinia virus based transient expression system. The staining of HeLa cells expressing Gly− P-gp (91, 94, and 99N→Q), with P-gp specific monoclonal antibodies, MRK-16, UIC2 and 4E3 revealed a 40 to 50% lower cell-surface expression of mutant P-gp compared to the wild-type protein. The transport function of Gly− P-gp, assessed using a variety of fluorescent compounds indicated that the substrate specificity of the pump was not affected by the lack of glycosylation. Additional mutants, Gly− D (91, 94, 99N→D) and Gly−Δ (91, 94, 99 N deleted) were generated to verify that the reduced cell surface expression, as well as total expression, were not a result of the glutamine substitutions. Gly− D and Gly−Δ Pgps were also expressed to the same level as the Gly− mutant protein. 35S-Methionine/cysteine pulse-chase studies revealed a reduced incorporation of 35S-methionine/cysteine in full length Gly− P-gp compared to wild-type protein, but the half-life (∼3 hr) of mutant P-gp was essentially unaltered. Since treatment with proteasome inhibitors (MG-132, lactacystin) increased only the intracellular level of nascent, mutant P-gp, the decreased incorporation of 35S-methionine/cysteine in Gly− P-gp appears to be due to degradation of improperly folded mutant protein by the proteasome and endoplasmic reticulum-associated proteases. These results demonstrate that the unglycosylated protein, although expressed at lower levels at the cell surface, is functional and suitable for structural studies.

    Notes: Abstract Precursors for layer-structured perovskite thin films of Sr2Bi2Ta3O12.5 or SBT223, and Sr2Bi2Nb3O12.5 or SBN223 were prepared by the reactions of Sr-Bi (2 : 2) double methoxyethoxide and Ta or Nb methoxyethoxide, followed by partial hydrolysis. By several analytical techniques, such as 1H-, 13C- and 93Nb-NMR (nuclear magnetic resonance) and FT-IR (Fourier transform infrared) the molecular structure of the precursors was found to be similar to the layer-structured perovskite crystal sub-lattice. As a result, the onset of crystallization in the sol-gel derived SBT223 thin films was at a low temperature of 550°C. By rapid thermal annealing in an oxygen atmosphere, the single-phase perovskite films exhibited preferred (1 1 7) orientation. In contrast to the SBT system, the observed number of the pseudo-perovskite layers of TaO6 octahedral units between Bi2O 2 2 + layers in the SBT223 was three. Despite the elongation of the unit cell along the c-axis, 700°C-heated SBT223 thin film did not exhibit a saturated hysteresis loop due to the small crystallite size.

    Notes: Abstract In this investigation, several spectroscopic and analytical techniques were used to determine the chemical compositions and structures of the lead, zirconium, titanium, and Pb-(Zr, Ti) alkoxides involved in the sol-gel synthesis of PZT thin films. These techniques included 1H, 13C, and 207Pb NMR; FT-IR; gas chromatography; Karl Fischer titration; and number-average molecular weights (M n ) determined by cryoscopy. It was found that the titanium precursor had a M n of 548 and a formula of [Ti(OCH2CH2OCH3)4]1.6; the zirconium precursor had a M n of 1015 and a formula of [Zr(OCH2CH2OCH3)4]2.6; and the lead precursor had a formula Pb6(OOCCH3)5(OCH2CH2OCH3)7. 4 H2O and a molecular weight of 2131 (M n =2113). It was observed that residual water from the incomplete dehydration of lead acetate trihydrate coupled with released water due to the esterification of acetic acid caused M-O-M (M=Pb, Zr, Ti) bonds in the Pb-(Zr, Ti) alkoxide. Two possible isomeric structures of the Pb-(Zr, Ti) alkoxide have been proposed. They are both cyclic and have a formula of Pb2M′M″O2(OR)8(ROH)2, (M′M″=Zr and/or Ti) and a molecular weight of 1336 (M n =1386).

    Notes: Abstract The pyrolysis behavior of acid and base-catalyzed bulk and thin film Pb(Zr, Ti)O3, or PZT, gels as well as their components have been studied using Thermo-Gravimetric Analysis (TGA), Differential Thermal Analysis (DTA), and Dynamic Mass Spectrometric Analysis (DMSA). The TGA/DTA data reflected the structural differences of the acid and base-catalyzed gels. Gels obtained using an acid catalyst were less cross-linked, denser, and more homogeneous than base-catalyzed gels. Based on the understanding of structure evolution in the silica system, the various events in the TGA/DTA studies of acid-catalyzed PZT gels were attributable to specific mechanisms. The DMSA showed that primarily water, 2-methoxyethanol, acetone, and carbon dioxide were evolved for both acid and base catalyzed PZT gels. The presence of the latter two volatiles was associated with the decomposition of the acetate group via the carbonate route. Acetate and carbonate groups were determined by Fourier Transform Intrared Spectroscopy (FTIR) to be present in the gel structure prior to pyrolysis. Differences in the synthesis of the prehydroiyzed solution were found to affect the amount of residual alkoxy groups, gel structure, pyrolysis behavior, and therefore, the resulting microstructures of sol-gel derived PZT thin films. Finally, some suggestions for improving the processing of sol-gel PZT thin films are given.

    Notes: Abstract An attempt to reduce the production cost on tissue cultured plants, photoautotrophic culture of a high value orchid Dendrobium was established under CO2-enriched conditions. The shoot length and the number of leaves were almost equal in plantlets grown on medium with 2 % sucrose or without sucrose and under normal or enhanced (40 g m-3) CO2 concentration, whereas the fresh and dry masses were higher in cultures grown in sucrose containing media or under CO2 enrichment. Development of roots was observed only on media without sucrose, but CO2 enrichment did not have significant effects on in vitro rootings.

    Notes: Abstract Issues related to the automated identification and elimination of the adverse influence of small geometric model features on the quality of automatically generated meshes, using local mesh modification operators, are addressed. The definition of mesh validity with respect to the geometric model is extended to include multiple mesh entity classifications. Checks based on mesh topology are used to ensure no dimensional reductions in the locally modified mesh. Example geometric models of varied complexity containing small geometric features are used to demonstrate the ability of presented procedures to improve mesh quality in terms of aspect ratio and small angle metrics.

    Notes: Abstract By irradiation of the tunneling junction of a scanning tunneling microscope with intensity-modulated laser light a gap-width modulation due to thermal expansion of tip and sample was produced. Photothermal images were obtained by spatial mapping of the resulting modulation of the tunneling current or its logarithm. The various mechanisms responsible for the observed contrast are discussed quantitatively. In case of a highly corrugated gold film on mica the contrast arises mainly from either the current variations caused by the non-zero reaction time of the current control loop or from a geometry factor. In both cases the images reflect certain properties of the sample topography. On the other hand, for a liquid-crystal film adsorbed on graphite a contrast on a molecular scale was found which is attributed to variations of the effective barrier height.

    Notes: Abstract The compositional dependence of the glass transition temperature (T g), and the temperature of the maximum rate of crystallization (T p) of As-Se glasses containing Sb has been measured using non-isothermal DSC. The variation of these characteristic temperatures is found useful in interpreting the structural changes in As-Sb-Se glasses. The role of Sb in these glasses and its effect on the crystallization characteristics is analyzed.
