Building Units of Oligosaccharides, XLV. - Synthesis of a Branched Tetrasaccharide Unit of the O-Specific Side-Chain of the Lipopolysaccharide Obtained from Shigefla flexneri Serotype 6The synthesis of the branched tetrasaccharide β-D-GalNAc(1→2)-[β-L-Rha(1→4)]-β-L-Rha-(1→4)-L-Rha (28) is described. This structure has been proposed as a repeating unit of the O-specific side-chain of the lipopolysaccharide obtained from Shigella flexneri Serotype 6. The silver silicate method has been tested in order to achieve all the β-glycosidic linkages between L-rhamnose and 2-azido-2-deoxy-D-galactopyranose. The halide 13 which was assigned to be the central branching point has first been coupled to the rhamnoside 2. The resulting disaccharide 17 could be combined with a further L-rhamnose unit and 2-azido-2-deoxy-D-galactose leading to the tetrasaccharide 24 which could be deprotected to yield 28.

Es wird die Synthese der verzweigten Tetrasaccharid-Einheit β-D-GalNAc(1→2)-[β-L-Rha(1→4)] -β-L-Rha(1→4)-L-Rha (28) beschrieben, die als Struktur der Repeating-unit der O-spezifischen Seitenkette des Lipopolysaccharides von Shigella flexneri Serotype 6 vorgeschlagen wurde. Hierbei wurde in allen Verknüpfungsschritten das Silbersilicat-Verfahren zur Herstellung β-glycosidischer Bindungen von L-Rhamnose und 2-Azido-2-desoxy-D-galactose erprobt. Das als mittleres Verzweigungsstück vorgesehene Halogenid 13 wurde primär mit dem Rhamnosid 2 zum L-Rhamnose-Disaccharid 17 gekuppelt. Hieran ließen sich sukzessiv eine weitere L-Rhamnose-Einheit und 2-Azido-2-desoxy-D-galactose β-glycosidisch knüpfen zu 24, das zu 28 entblockiert wurde.
