Description / Table of Contents: Abstract A chromatographic system is described which will separate nucleosides on account of interactions between complementary purine and pyrimidine bases. Columns were prepared from a granular resin to which numerous molecules of a purine or a pyrimidine base had been bound covalently. When mixtures of nucleosides dissolved in aqueous buffer were allowed to pass through such columns, separations occurred as a result of dissimilar retentions of different nucleosides on the resin beds. The observed retentions can be accounted for by the formation of intermolecular complexes between the bases fixed to the resin and the nucleoside bases chromatographed in the mobile phase, in analogy to the pairing of bases in the two-stranded DNA molecule.
