Synthesis, Spectroscopic and Conformational Investigations of 10α-Methyl Steroid SpirolactonesThe 10α-methyl dienone 5, prepared from the mixture of the ketones 1 via the enone 4, reacts with methanethiol to give a mixture of the 7α-methylthio derivatives 6 and 7. The conformations of the A and B rings in the compounds 4-7 have been determined by means of CD investigations and compared to the energetically preferred conformations calculated on the basis of semiempiric quantum mechanical calculations (AM 1). The compounds 6 and 7, in contrast to comparable derivatives in the normal series, show no aldosterone antagonistic activity.

Das aus dem Ketongemisch 1 über das Enon 4 dargestellte 10α-Methyldienon 5 reagiert mit Methanthiol zum Gemisch der 7α-Methylthioderivate 6 und 7. Die Konformationen der A- und B-Ringe in den Verbindungen 4-7 wurden mit Hilfe von CD-Untersuchungen zugeordnet und mit den durch semiempirische quantenchemische Berechnungen (AM1) ermittelten, energetisch bevorzugten Konformationen verglichen. Die Verbindungen 6 und 7 zeigen, im Gegensatz zu vergleichbaren Strukturen in der Normalreihe, keine Aldosteron-antagonistischen Wirkungen.
