A possible use of phosphorylating mitochondrial electron transport particles (ETPH) has been investigated for ATP regeneration. The oxidative phosphorylation of ETPH was considerably inhibited by the hydrolytic activity of ATPase and adenylate kinase. The hydrolytic activity of ATPase and adenylate kinase were found to be intensively retarded in the presence of Mg2+. ETPH continuously regenerated ATP from ADP over 4 hr when suspended in an isotonic buffer containing ADP, succinate, and 100 mM MgSO4. Furthermore, repeated use of ETPH was possible for ATP regeneration primarily due to considerable stabilization of the electron transport system.
