    Notizen: . Micronuclear mitosis in living Spirostomum teres has been studied by sensitive polarization microscopy, and the dynamic aspects of micronuclear division are described. The small, spherical, interphase micronuclei lie in form-fitting depressions in the macronuclear surface. Nuclear division begins with the rounding and slight swelling of the macronucleus and, coincidentally, the micronuclei move out of the depressions and away from the macronucleus, increase in size, and become weakly birefringent. As mitosis proceeds, the micronuclei increase in uniaxial birefringence and elongate to form irregular ovoids that convert to angular structures displaying principal axes of positive birefringence so divergent as to appear oriented at a right angle to one another. Micronuclei maintain this appearance for as long as 60 min and then abruptly change to rectangular-shaped structures, increase in uniaxial birefringence, and begin anaphase elongation. The somewhat dumbbell-shaped micronuclei lengthen at the constant rate of 2.0 μm/min to reach lengths 〉70 μm. It appears that little half-spindle shortening occurs during spindle elongation. Accompanying the changes in micronuclear spindle length are changes in birefringence. Just before elongation begins, presumably metaphase, the micronucleus is uniformly and intensely birefringent. At the magnifications employed, a chromosome plate is not clearly visible as a region of reduced birefringence. As elongation begins, the putative half-spindles are more birefringent than is the interzone, a condition that is maintained until the spindles have achieved ∼30% elongation, at which time a region of increased birefringence develops at the center of the interzone. This pattern persists for a very short time, then gives way to a uniform birefringence of the entire separation spindle that is maintained until elongation is completed. The rate of micronuclear spindle elongation, changes in micronuclear dimensions, and corresponding changes in birefringence are discussed with respect to possible mechanisms of mitosis.

Studies performed with the basidiomycete Laccaria trullisata collected from the sandy beach at the Hempstead Lake State Park, Long Island, New York, during the growing seasons of 1979 and 1980, have demonstrated a carposphere (equivalent to rhizosphere) effect. This region exerts a positive influence on the population density of amoebae when numbers are compared with those obtained in the bare sand 5 cm away. Moreover, amoebae have been shown to exist in, and have been recovered from, internal tissue of the cap (72%) and stalk (91%) of these mushrooms. A partial characterization of three strains of amoebae isolated from the internal tissue of L. trullisata and established in clonal culture is presented.

The successive steps of morphogenesis are strongly correlated with nuclear processes. The comparative study of different types of morphogenesis (binary fission, sexual phenomena, starvation-induced reorganization) leads to the hypothesis that two morphogenetic fields (a ventral one and a dorsal one) depend on separate regulatory systems. From the viewpoint of evolution, the morphogenetic sequences of some hypotrichs have been compared. A general scheme of the evolution of cortical regulation is proposed, taking into account the extension of the area concerned with stomatogenic activity.

    Notizen: Mouse omentum was studied after intraperitoneal challenge with tachyzoites of Toxoplasma gondii. Parasites inhabit omental histiocytes, fibroblasts, mesothelial cells, and free peritoneal macrophages. Recently infected cells showed enhanced metabolic and functional activity. Villous projections of the parasitophorous vacuole wall appeared, usually opposite the anterior pole of the parasite. In mesothelial cells, projections formed terminal swellings not observed in other infected cells. Activation of host cells was followed by reduction of the density of the cytoplasmic matrix, autophagosome formation, and intracellular edema, indicating the damage. The wall of the parasitophorous vacuole loses the supporting host cell endoplasmic reticulum that was attached to the vacuole just after entrance of the parasite into the cell. Then lysis of the parasitophorous vacuole and complete cell destruction occurs. The growth of parasites in undamaged cells does not coincide with the inflammatory response. Inflammation of the peritoneum develops only after the start of mass destruction of infected cells. Thus tachyzoites of Toxoplasma exert significant pathogenic effects by their ability to activate the host cell, causing lysis of the parasitophorous vacuole and subsequent destruction of the entire cell.

    Notizen: Discophrya collini is a free-living suctorian with retractile tentacles covered by a thick fibrous cortex. The tentacles contain a microtubular central canal surrounded at the base by a fibrous collar. Electrical stimulation induces a reproducible tentacle retraction. With extracellular electrodes, the tentacles nearest the anode respond initially, contracting by up to 75% of their original length. There is an inverse relationship between voltage level and duration of stimulus in producing a threshold response, and at a set voltage, between duration and degree of retraction. With intracellular electrodes, the membrane potential has been measured as -30 mV, and tentacle retraction occurs in response to as little as 1.25 nA when the intracellular electrode is made the cathode of the circuit. SEM studies show that retracted tentacles have a wrinkled cortex, while TEM shows that the microtubular canal bends as it enters the cytoplasm. No consistent changes occur in the microtubule configuration of the canal on retraction, suggesting that the microtubules are not directly involved in the contractile mechanism.

    Notizen: A rapid in vitro prescreen for Fe-binding chelators has been developed with growth of Crithidia fasciculata and the sparing of its heme requirement in a defined medium as a test system. The prescreen functions as an index of chelator-mediated Fe transport and as an index of growth inhibition, presumably by the interference with Fe and/or heme metabolism at intracellular chelatable sites. Of 161 chelators examined, 84 were active heme-sparers; 32 of these inhibited growth at low chelator concentrations. Twenty-eight other chelators inhibited growth and another 49 were inactive. Such chelating activity directed at Fe and heme targets in hemoflagellates may provide leads for chemotherapy.

    Notizen: Viable merozoites of Plasmodium knowlesi were isolated and the proteins that were labeled on intact merozoites by lactoperoxidase-catalyzed radioiodination were identified. Sodium dodecyl sulfate polyacrylamide gel electrophoresis and autoradiography of Triton soluble extracts of labeled merozoites demonstrated eight major bands ranging in apparent molecular weight from 150,000 D to 22,000 D. Exposure of intact merozoites to trypsin (10 μg/ml) for 10 min resulted in the loss of the two highest molecular weight proteins (150,000 D and 105,000 D) and the appearance of two new bands at 70,000 D and 62,000 D. Trypsin treatment under these conditions also removed the receptor(s) for merozoite attachment to erythrocytes. Therefore, these high molecular weight proteins are candidates for the merozoite component that attaches to erythrocytes. There was no evidence that the labeled membrane components were serum or erythrocyte membrane components, two potential contaminants in the preparation. Anti-rhesus erythrocyte antibody did not precipitate labeled merozoite proteins. Furthermore, the immunoprecipitation of labeled merozoite proteins by rhesus anti-merozoite serum was not inhibited by erythrocyte ghosts.

    Notizen: Species of trypanosomatids without endosymbionts (Leptomonas seymouri, L. collosoma, L. samueli, Crithidia fasciculata, C. luciliae, C. acanthocephali, Herpetomonas megaseliae, H. mariadeanei, H. samuelpessoai, H. muscarum muscarum, Trypanosoma cruzi) and species of trypanosomatids with endosymbionts (Crithidia deanei, C. oncopelti, Blastocrithidia culicis) were comparatively studied by means of electron microscopy. Artificially aposymbiotic strains derived from species with symbiont were also included in the survey. Species with symbiont were found to differ in some ultrastructural aspects from the group of species without symbiont. Paraxial rods of flagella or intraflagellar structure were found exclusively in species without symbiont. Peripheral branching of mitochondria, accompanied by absence of subpellicular microtubules in sites where the mitochondrial branches are appressed to the cell membrane, were found exclusively in species with symbiont. Networks of kinetoplast DNA fibrils were found to be larger and looser in species with symbiont. Symbiont-free strains of species with symbiont retained the same morphological characteristics of their parental species.

    Notizen: During a freeze-fracture electron microscopical study of the plasma membrane of Tetrahymena, several different types of organized particle assemblies were observed. Three of these were found only on the protoplasmic face and were localized in the anterior-ventral region of the cell. These consisted of plate-like arrays composed of 4–25 triplet rows of small 3–4 nm particles; long, paired linear arrays localized at the tops of cortical ridges and composed of 7–8 nm particles; and elongated tetragonal arrays located in the grooves between ridges and composed of approximately 10 nm particles. The distribution of these arrays is consistent with roles in cellular morphogenesis, chemoreception, or cell-cell pairing during conjugation. In addition, a unique particle track associated with the cytoproct (anal pore) was observed in the external face of the plasma membrane. Furthermore, the protoplasmic face of the plasma membrane is characterized by a high density of particles organized into localized microarrays, consisting of small paracrystals or strings, which exhibit a loose higher-order patterning most evident toward the anterior end of the cell. Particle distributions on the protoplasmic face do not appear to be significantly altered by conditions that cause clumping of alveolar membrane particles. Taken together, these observations are consistent with the idea that the proteins of the plasma membrane are highly ordered and relatively immobile and that the structure of the plasma membrane is regionally differentiated.

A new species of Dactylosoma (Dactylosomatidae, Piroplasmia), for which the name Dactylosoma hannesi n. sp. is proposed, was discovered in blood erythrocytes of Mugil cephalus, Liza richardsoni, and L. dumerili (Mugilidae) from Swartkops estuary, located east of Port Elizabeth, South Africa. The life cycle of this species differs in some respects from that described for all other known species of Dactylosoma and Babesiosoma. Mature schizonts contain eight nuclei but undergo division only to two to four daughter cells. During cytoplasmic cleavage, schizonts assume triad, rosette, or cruciform shapes. Merozoites are finally produced through a series of binary fissions of these daughter cells which may also be involved in additional nuclear divisions.

    Notizen: Electron microscopy was used to examine the flagellar apparatus of Herpetomonas ampelophilae from the gut and malpighian tubules of Drosophila melanogaster. The flagellates attach to the microvilli either by weaving their flagella between the microvilli or by engulfing several microvilli with an external flagellar membrane. The first type predominated in the gut while the second type was limited to the malpighian tubules. Desmosomes were not involved in either type of attachment. A subpellicular collar with emerging microtubules was found to be adjacent to the desmosome of the flagellar pocket of herpetomonads in the gut.

    Notizen: Prorocentrum micans Ehrenberg, a free-living marine dinoflagellate, was used to test the intracellular toxic action of cadmium. The cells were cultivated in Erdschreiber medium, with Cd concentrations of 10–100 ppb. Thin sections of treated cells, examined ultramicroscopically, exhibited vacuolations, increased numbers of lysosomes, and severe mitochondrial damage. The first two alterations are a general response to toxicity; the third is Cd specific. Although some chloroplasts were affected by Cd, they were not very sensitive to its action. The nuclear apparatus was not morphologically affected.

    Notizen: Toxoplasma-like avian parasites inhabiting mononuclear phagocytes have been called Haemogregarina, Toxoplasma. avian Toxoplasma, Atoxoplasma, and Lankesterella by various authors. My attempts to transmit the parasites by bloodsucking mites or by transfer of blood and tissues of infected sparrows and canaries were unsuccessful. However, it was noted that the infection was exacerbated under conditions that favored transmission of coccidiosis: crowding and lack of cleanliness. Oral inoculation of sporulated oocysts of Isospora resulted in death from overwhelming macrophage infection with Toxoplasma-like organisms. Experiments using sparrows and canaries showed that the Isospora species involved was not cross infectious. Further investigations using canaries demonstrated that after oral oocyst inoculation, infection of macrophages spread from the submucosa of the duodenum to the liver. spleen, and lungs. After several generations in the internal organs, asexual multiplication, occured in the intestinal epithelium of the small intestinc. Fecal oocysts first appeared at the end of 9–10 days. Oocysts continued to be passed in the feces for months after infection. This chronicity may be explained by the relatively long life of the macrophages that serve as host cells for the asexual stages as compared to the intestinal epithelium which is the cell type parasitized by conventional coccidia.

    Notizen: Stages of mitosis of the micronuclei of Stentor coeruleus were described as seen by transmission electron microscopy. Cells in division and those regenerating new oral membranelles were studied. Microtubules were found in early prophase in the karyoplasm and interspersed between the condensing chromatin. A monaxial intranuclear spindle is formed by early metaphase, with kinetochore microtubule attachment sites on the chromosomes. The spindle elongates, separating the daughter nuclei at anaphase. A new nuclear envelope, consisting of two unit membranes, begins to form at late anaphase. Small segments of membrane found in the space between the newly forming and the old micronuclear envelopes appear to fuse to form the new nuclear envelope. No ultrastructural differences were found in the mitotic nuclei of cells in division or regeneration.

Strains of Tetrahymena thermophila were examined in an attempt to establish what role certain ions (Na+, K+, Li+, Ba++, Ca++, Mg++, Mn++, Al+++, Fe+++) play in influencing cell survival time in a culture medium. In short-term experiments (20–30 min), cell survival time in a 1% peptone medium is directly related to the valence of the ion employed. Long-term observations (lasting up to five days) in a 1% peptone medium containing lower ion concentrations revealed that the effects on cell-cycle time are not correlated with the valence state of the ion. Comparisons were made among the ionic resistances of strains of T. thermophila, of T. pyriformis sensu stricto, and of two subspecies of T. pigmentosa. Strains within a species are highly correlated in their patterns of ionic response, while marked differences between species occur. The most distinctive group of strains examined came from one of the subspecies (syngen 6) of T. pigmentosa.

A two-stage chemostat modified to accommodate the growth of adhesive organisms was used to determine the yield constant, Y, of a representative soil amoeba, Acanthamoeba polyphaga, utilizing as its prey Pseudomonas paucimobilis. The first stage consisted of a glucose-limited bacterial culture in steady state. The second stage consisted of a simplified predator-prey system, nongrowing bacteria serving as the limiting substrate for amoebae. A refined methodology to more accurately determine Y was developed, and Y for Acanthamoeba polyphaga in batch and continuous culture was determined to be 19.1%.

The swiftness of thermotaxis of Paramecium caudatum has been investigated for various populations of organisms by measuring the transient spatial distribution of the gathering process of organisms that are transferred to a temperature-gradient cell from the culture medium. The dispersion obtained from the spatial distribution for each population is found to decrease linearly with time and finally reach a steady state value. The gathering rate determined by the slope of the dispersion strongly depends on population; it increases with population.

    Notizen: . Blastocystis hominis, an anaerobic intestinal protozoan parasite of man, has a generation time (GT) in axenic culture of 8.5–19.4 h, depending on the strain tested. Average GT of the eight strains was 11.7 h. Zero growth time cell counts of 5.0 × 105/ml to 2.0 × 106/ml rose in 3–5 days to 1 × 107 or 1 × 108 cells/ml. The GT was determined for the 24-h period during which the most rapid growth occurred; about 2% of the B. hominis cells were in division during this time. Division under the culture conditions provided was by binary fission, the usual mode for B. hominis in vitro as well as in vivo. Division times were determined also by direct observation of individual dividing cells in slide cultures. These were usually ca. 40–60 min but sometimes as low as 20 min.

    Notizen: A trypanosomatid flagellate, Leptomonas sp., develops and multiplies in the macronucleus (only) of natural and laboratory-reared populations of the ciliate Euplotes. Up to 90% of the natural populations of Euplotes in our test pond had such nuclear infections. Laboratory infections were transmitted to this ciliate by feeding it liberated parasites. Paramecium resisted infections. All laboratory-induced infections were lethal to Euplotes, while control clones of the uninfected ciliates remained viable. This leptomonad, unlike Leptomonas karyophilus (found in Paramecium), shows no leishmanial forms in its several ciliate hosts and shows a varied pattern of locomotion.

    Notizen: Stocks of the Tetrahymena pyriformis complex have been collected in North America and their mating reactivity has been studied. In addition to stocks mating with Tetrahymena americanis, T. borealis, T. pigmentosa, T. hyperangularis, and T. australis, stocks belonging to old syngen 5 and three new mating groups, numbers 13, 14, and 15, were discovered. Syngen 5 and groups 13 and 14 are distinct “biological” species, based on their reproductive isolation from other groups and on the ability of withingroup crosses to produce immature progeny. These species have been named T. hegewischi n. sp., T. sonneborni n. sp., and T. nipissingi n. sp., respectively. The cross between the two group 15 stocks did not produce immature progeny, and there is not sufficient evidence to conclude that this pair of stocks represents a separate species. Temperature tolerance measurements have been made on stocks representing all known micronucleate members of “pyriformis” complex. Within each species, the range of temperature tolerances is narrow; the average within-species standard deviation is 0.63°C. The species averages range from 32.7 to 40.7°C. Using syngen numbers, the order from lowest to highest temperature tolerance is 9, 8, 10, 7, 6, 4, 13, 14, 12, 11, 5, 3, 2, 1. The large differences among species make temperature tolerance a useful aid in identification, but the origins of the differences among species are unknown.

    Notizen: Members of the family Sarcocystidae, as defined by Frenkel, have had a complicated history, principally due to the existence of both coccidial and cystic stages. This formerly clandestine relationship resulted in dual or partial designations of nomenclature for individual species. The problem was further compounded by the obligatory heteroxeny of several of the genera, making it impossible to transmit the parasites from one individual to another of a single host. As a result, oocysts similar in appearance, though from hosts separated taxonomically up to the familial level, were sometimes considered to be identical. Discoveries within the last decade have generated much interest and some understanding. Current studies of these and other coccidia should emphasize complete cyclical transmissions with cognizance of potential heteroxeny with the production of tissue cysts in intermediate hosts.

    Notizen: During spring and autumn, the total number of amoebae and the number of Acanthamoeba species able to grow at 37°C were determined in six thermally polluted factory discharges and the surrounding surface waters. The isolated Acanthamoeba strains were studied for growth in axenic medium, cytopathic effect in Vero cell cultures, and virulence in mice. Although more amoebae were isolated in autumn, the number of Acanthamoeba species was lower than in spring, when the percent of pathogenic strains among the isolates was highest. Higher concentrations of amoebae were found in warm discharges, and more virulent strains occurred in thermal discharges than in surface waters.

    Notizen: Eight species of loricate choanoflagellates (Acanthoecidae), Acanthoecopsis spiculifera Norris, Bicosta antennigera Moestrup, Bicosta spinifera Throndsen, Calliacantha multispina Manton & Oates, Calliacantha simplex Manton & Oates, Crinolina aperta Leadbeater, Diaphanoeca multiannulata n. sp., and Parvicorbicula socialis (Meunier) Deflandre, have been observed, by light and electron microscopy, in samples obtained from the Weddell Sea during the austral summer of 1977. Diaphanoeca multiannulata is described for the first time from these samples: the other organisms are discussed. The distribution of most species within the Weddell Sea was widespread. Habitats in which choanoflagellates were found included the water column, the edge of (or ponds on) ice floes, and the interior of ice floes. The distributional, environmental, habitat, and/or morphological range of all previously described species is expanded. Methods of variation of transverse costal diameters between genera may be potentially useful to the understanding of taxonomy and phylogeny of this family.

    Notizen: . Allantosoma intestinalis, a suctorian ciliate isolated from the intestine of the horse, was studied utilizing light and electron optical methods. These small sausage-shaped organisms have a varying number of tentacles (between one and 12) located at each extremity of the body. The microtubular axoneme of each tentacle in cross-section consists of two files of microtubules arranged in a daisy-like configuration. Haptocysts occur in the tentacle shaft, abutted to the plasma membrane of the knob of the tentacle, and in the cell body. The haptocysts are bottle-shaped, with prominent annular striations around their midportion. The cell is covered by three membranes, an outer plasma membrane, an outer alveolar, and an inner alveolar membrane. A thin epiplasmic layer is found beneath the inner alveolar membrane, and a single row of microtubules underlies the epiplasm. The subpellicular microtubules are arranged parallel to each other forming a corset around the cell along the long axis: such a system is not characteristic of suctorians. A field of diminutive kinetosomes (each 180 nm long, max. of 15 per field), lacking cilia, was found below the cortex. The function of these prokinetosomes is unknown. A ciliated swarmer has not been observed, only the nonciliated adult. The characteristics of Allantosoma are compared with those of other suctorian genera.

    Notizen: . Temperature shifts have been used to block critical points in the conjugation sequence of Paramecium tetraurelia. Increasing temperatures above 27°C reduced ciliary agglutination, pair formation, and nuclear exchange; a complete inhibition of these stages occurred at 37°C. Temperatures below 19°C had no effect on ciliary agglutination or nuclear exchange but completely inhibited pair formation. The bases for the cells’ inability to form pairs at 19°C and 37°C were sought. Cells placed below 19°C were unable to deciliate or fuse membranes in the holdfast region; at 37°C, membrane fusion in both the holdfast and paroral regions was prevented. Time course studies on cross-fertilization reveal that temperatures 35°C block all stages of the process up to the actual exchange of pronuclei. After the exchange has begun, the process continues despite the elevated temperature. Temperature shifts are discussed as a means of conditionally blocking critical points in the developmental program of conjugation.

[14C]chimyl and [3H]batyl alcohols were added to Crithidia fasciculata cultures during the mid-log phase of cell growth, and the lipid extracts of the cells were analyzed for degradation products. C. fasciculata cells were able to take up exogenous glyceryl ethers, and in amounts as high as the endogenous lipid content. The glyceryl ether taken up by the cells was incorporated into lipids either prior to the ether bond cleavage or after degradation to fatty acid. The extent of degradation and the degree of incorporation of degradation products into cellular lipid were higher for chimyl than for batyl alcohol. Batyl alcohol was not metabolized efficiently, leading to the formation of large intracellular pools of free substrate. One product of glyceryl ether degradation was identified as alkyl-dihydroxy acetone, and was detected inside and outside of the cells. The data strongly suggest that this product is the first stable intermediate in the degradation process and indicate that the extracellular formation of alkyl-dihydroxy acetone is due to the action of exocnzyme; ecteted by the cells. The constant detection of alk I cnyl glycerol among the degradation products indicates the existence of a second mechantsm in C. fasciculata for converting the alkyl-to alkenyl-glycerol.

    Notizen: . Sick budgerigars from a local aviary were found to be infected with Giardia. The trophozoites were of the Giardia duodenalis type as defined by Filice with elongate median bodies pointed on one or both ends and more or less perpendicular to the long axis of the body. Using three fixation-staining methods, and material from three birds, the length ranged from 10 to 18 μm and the width from 4.5 to 11 μm with a mean length to width ratio of 2. Attempts to culture the trophozoites in vitro from intestinal scrapings were unsuccessful. Also attempts to transmit the infection by fecal cysts to canaries and mice failed. It is proposed that the budgerigar form be called Giardia duodenalis, race psittaci.

    Notizen: Three species of Pseudocohnilembus were examined with respect to infraciliature and the silverline system. The freshwater species P. putrinus and P. pusillus exhibited no differences in general organization of the argyrophilic structures when compared with the marine P. marinus. A review of the Pseudocohnilembus species that have been described on the basis of silver preparations shows that at present four species have been well characterized: P. pusillus (Quennerstedt, 1869) nov. comb., P. putrinus (Kahl, 1928) nov. comb., P. hargisi Evans & Thompson, 1964, and P. marinus Thompson, 1966, P. persalinus Evans & Thompson, 1964 and P. longiseta Evans & Thompson, 1964 are regarded as identical with the older known P. pusillus, because with respect to morphology and argyrophilic structures they are within the range of variability of that species; and their names thus fall as synonyms of P. pusillus.

    Notizen: The structure of micronemata arising from the surface of the bloodstream form of Trypanosoma gambiense was studied by electron microscopy. In order to produce micronemata, trypanosomes were incubated in either 1) phosphate buffered saline supplemented with glucose (PBSG), 2) immune mouse serum or 3) PBSG after passage through a DEAE-cellulose column. Electron microscopic examination of the parasite revealed the presence of thread-like micronemata arising from the anterior end and from the flagellar pocket regardless of the incubation conditions. Negative staining revealed a distinct peripheral fringe layer with nodular protrusions covering the entire surface of the micronema. The distribution and number of intramembrane particles (IMP) on the P and E faces of the micronema were similar to those of the flagellum of T. gambiense, indicating a close relationship between the membrane structure of the micronema and the flagellum. Micronemata became fragmented and adhered to each other after incubation of the parasite in the media for 12 h. Since micronemata tend to have the characteristics of adhesiveness and fragmentation, fragments of these structures might adhere to various host organs. Dispersal of potential antigenic material might be responsible, in part, for the induction of the host immune response.

    Notizen: The motion of Paramecium caudatum has been investigated at various temperatures by measuring the transient behavior of spatial distribution in the diffusion process of organisms that, by electric stimulus, are initially gathered at a single place in the glass culture cell. The spatial distribution through the course of diffusion has a nearly Gaussian profile. Dispersion was obtained at 1 sec intervals and increased linearly with time. The time dependence of the dispersion gave a diffusion coefficient for the random motion of the organisms. The results show that the diffusion coefficient has a maximum at the temperature at which the paramecia were cultivated.

    Notizen: . Conjugating cells of Euplotes vannus (syngens Naples and Barcarès) were investigated for Con A-binding sites by means of fluorescence microscopy. Cells fixed with 0.4% paraformaldehyde and incubated with 15 μg/ml FITC-Con A showed a distinct region of strong fluorescence, the size, shape and localization of which characteristically changed during the course of conjugation, first appearing at courtship stage (cells contact without forming pairs) and vanishing soon after the migration pronuclei have been exchanged, but before cells have separated. Con A binding and conjugation were blocked by cyclohexamide and α-galactosidase. Con A-binding was also inhibited by α-mannosidase, α-glucosidase and β-galactosidase without affecting conjugation except a delay of pair formation. The results suggest an involvement of newly formed or translocated glycoconjugates in cell pairing during conjugation.

Uninucleate zoites were found in arteries of mesenteric lymph nodes but not in other organs. Zoites measured 4.9 × 3.0 (3.5–7.0 × 2.1–3.5) μm. Of the 36 zoites studied, 31 were in endothelial cells, four were in macrophages in the lumen of arteries, and one was free in the lumen of an artery. Infected endothelial cells were two to three times larger than uninfected cells. Zoites appeared structurally similar to sporozoites. The occurrence of zoites in macrophages suggests that sporozoites of Sarcocystis might use such cells to reach the site of their first merogony.

The enzyme that catalyzes the critical conversion of the proposed reductive thymidine catabolic end product, P-aminoisobutyric acid, to the initial anabolic reutilization substrate, probably methylmalonic semialdehyde, was investigated to un- ambiguously substantiate the operation of a reductive pyrimidine catabolic and reutilization pathway in Tetrahymena pyriformis strain GL. Although most of the studies on the further metabolism of P-aminoisobutyric acid in other organisms suggested transamination of p-aminoisobutyric acid to methylmalonic semialdehyde followed by its further oxidation to methylmalonic acid, such a transaminating system could not be demonstrated. By means of a sensitive fluorometric assay system, however, a low, but significant amount of P-aminoisobutyric acid oxidase activity was detected. This enzymatic activity exhibited the following characteristics in homogenates: good activity in alkaline 0.2 M Tris-HCI buffer with a rather broad pH optimum ranging from 7.8 to 9.0; optimum activity at a temperature of 37°C; stimulation on the addition of exogenous FAD; inhibition on the addition of divalent cations, EDTA, or PCMB; little stimulation on the addition of detergents; and no increase in activity on repeated freezing and thawing. In addition, crude preparations of this oxidase were found to be relatively stable when stored up to one week either refrigerated or frozen, to have a specific activity of 2.8 nmoles/min/mg of protein, and to have a K, of 3.6 × 10− M for d,l-β-aminoisobutyric acid. Whether this high Km, is physiologically significant or not, however, will have to await further investigation. Preliminary (NH4)2SO4 fractionation and affinity chromatography studies also indicated that this enzyme appears to be a unique and specific oxidase whose activity is separable from other marker enzymes, including other oxidases.

    Notizen: . Phosphatase activity in Trypanosoma rhodesiense has been examined histochemically by light and electron microscopy and by enzymatic assay in homogenate fractions. Using a method with lead as capture ion, acid phosphatase was found in lysosome-like vesicles and in the flagellar pocket. No alkaline adenosine triphosphatase (ATPase) was detectable by this method. Direct assay of p-nitrophenylphosphatase activity in homogenate fractions showed that acid phosphatase activity was strongly membrane-bound, but that activity at pH 9 was minimal in both soluble and particulate fractions. “Endogenous” ATPase activity was localized specifically and reproducibly in the mitochondrial membranes and under the plasma membrane of the flagellum. This nonenzymic reaction product could not be eradicated by glycerol extraction or glucose depletion. Unlike the membrane staining, which was manifest only after lead treatment, heat-resistant electron-dense material was found in the matrix of lysosomal vesicles in trypanosomes fixed in glutaraldehyde only and not subjected to further treatment with heavy metal reagents. X-ray emission analysis showed the presence of calcium and phosphorus, indicating that the matrix might have a phosphate storage function.

    Notizen: The longitudinal flagellum of Ceratium tripos moves in two dissimilar ways: undulation and retraction. The undulatory wave is planar and has a wavelength of 74.3 ± 9.6 μm and an amplitude of 14.2 ± 2.3 μm in sea water. The beat frequency is 30 Hz at 20°C, pH 8.0. The retractile motion is unique to Ceratium and is triggered by mechanical stimulation on the cell body, especially at the tip of the apical horn. When it retracts, the longitudinal flagellum folds every 4–5 μm along the flagellum. Cinematographic study showed that the flagellum folded from tip to base and was finally installed into the sulcus, a groove on the ventral side of the cell. This motion is completed in sea water within 28 msec. The retracted flagellum then re-extends and restores the undulation within a few seconds. The flagellum unfolds in the proximal portion first, then the distal, and finally the middle portion. Fixation always triggers the retraction. Scanning electron microscopy showed that the flagellum is folded and secondarily twisted in a helix. A new fiber in addition to the flagellar axoneme was found in the retracted flagellum by phase microscopy. This fiber (R-fiber) seems to contract during the retraction to fold the flagellum.

    Notizen: Phagocytosis of culture forms of Trypanosoma cruzi was assayed by a radioisotopic method. Purified polymorphonuclear leukocytes (PMN) were mixed with 3H-uridine-labeled T. cruzi epimastigotes in the presence or absence of anti-T. cruzi antibodies. The reaction was stopped by adding N-ethyl-maleimide, and noningested parasites were lysed by complement. The percentage of radioactivity incorporated into the PMN pellet was recorded. The phagocytosis reaction was rapid, yielding maximum incorporation at 30 min at which point the radioactivity associated with the PMN cells decreased through release of the isotope to the supernatant. The degree of incorporation of radio-labeled parasites was a function of the effector/target cell ratio and the antibody concentration. The method is suitable for the quantitative determination of phagocytosis of T. cruzi by normal PMN.

    Notizen: The effect of holothurin (a marine biotoxin) on the resistance of mice to Trypanosoma musculi was measured by studying changes in the parasite population in vivo. Swiss Webster (SW), Beige (BG), and Black (BL) mice treated with holothurin prior to and simultaneously with infection of trypanosomes had lower parasitemias than controls. Higher levels of parasitemia were observed in mice treated after infection with trypanosomes. The timing of administration of holothurin appeared to be an important factor in the observed effect. The minor variations in the parasitemia seemed to be related to the mouse strain.

    Notizen: The structure of the cyst wall of the heliozoon Echinosphaerium nucleofilum has been investigated using light microscopy, scanning and transmission electron microscopy, and X-ray microanalysis. The cyst wall is a composite structure of seven or eight layers. These are: an enveloping gelatinous layer; a layer of siliceous spheroidal bodies; an electron-dense supporting membrane; a broad electron-lucent zone; an electron-dense layer; a layer of helicoidally packed material; and one or two layers with a granular appearance lying next to the plasma membrane of the encysted organism. The structure of the cyst wall closely resembles that of Actinophrys sol, confirming the close relationship of these actinophryid heliozoa while emphasizing their distinctiveness from other amoeboid protista.

The cortical morphogenetic pattern of Diophrys is similar to that of other members of the family Euplotidae. The opisthe oral primordium, which develops in a subsurface pouch, forms posterior to the parental buccal cavity. The proter inherits the parental adoral zone of membranelles (AZM) apparently unchanged. The endoral membrane forms to the right of the posterior end of the AZM in the proter, in association with the developing AZM in the opisthe. The paroral cirrus and membrane develop from a single streak that first appears along the right edge of the buccal cavity in the proter to the right of the developing buccal structures of the opisthe. Frontal and transverse cirri develop in both proter and opisthe from five separate cirral primordia that form to the right of the buccal cavity. Left marginal cirri do not develop in association with the corresponding parental structures. Kinetosomes formed within the opisthe oral primordium, or kinetosomes that were part of any parental ciliary structure, do not appear to become part of any developing paroral structures, frontal, transverse, or left marginal cirri. Speciation within the genus Diophrys and evolution of the family Euplotidae as they relate to the morphogenesis of cortical structure are discussed.

    Notizen: Autecology (cellular organelles and secretions, encystment and dispersal abilities), environmental conditions (physiological tolerances and interaction with other organisms), and evolutionary history contribute to protist biogeography, which manifests ecological and historical aspects. Ecological biogeography is seen in the influence of geochemistry on the distribution of fresh-water phytoflagellates and algae, and of moisture and vegetation type on soil-litter protists. A temporal feature is often present because many protists encyst and respond only to certain ranges of temperature and organic content. Historical biogeography has occurred by radiative host evolution on symbiotic protozoa (e.g., termite flagellates and rumen ciliates), and by the isolating effects of water currents, temperature, and density gradients on oceanic protists (coccoliths, dinoflagellates, foraminifera, radiolaria, and tintinnines). These two aspects combine in protists living on animal surfaces. Humans affect protist biogeography by increasing parasite ranges through human migrations and by water pollution. They can diminish these situations by disease control and exploiting appropriate ciliates in sewage disposal. Many free-living protozoa appear to be cosmopolitan, but mating types and isoenzyme studies suggest that speciation with its geographic connotations may be more widespread than presently appreciated.

    Notizen: Available evidence indicates that many of the antiprotozoal drugs currently in use significantly modify the immune response of the host. The effect depends on both the drug and the host. Some drugs enhance the immune response, some are immuno-suppressants, and others enhance some types of immune mechanisms while suppressing others. Future efforts in the development of antiprotozoal drugs should consider their effects on both the parasite and the immune response of the host. Also in the chemotherapy of protozoal infections consideration should be given to the combined usage of immunoenhancing agents and antiprotozoal drugs.

    Notizen: A survey of protozoa polluting bottled mineral water in Mexico was carried out using samples obtained from the three best-selling brands of bottled mineral water in the country. The organisms were concentrated through filtration procedures and subsequently cultured in sterile media. The cultures were observed over four weeks, with identification to the level of genus and species. Most commonly found were the amoebae Naegleria gruberi, Acanthamoeba astronyxis, and Vahlkampfia vahlkampfi (trophic as well as cystic stages) plus one flagellate, Bodomorpha minima. No ciliates were detected. The public health importance of the findings is obvious, since some strains of Naegleria and Acanthamoeba have the potential to cause human disease that may lead to death.

Amphidinium carteri was unable to grow on nutrient-enriched seawater in the presence of 200 μg/ml fluoride (F) but could be adapted to grow successfully on this F concentration when repeatedly cultured with stepwise increases in sub-inhibitory F concentration. Electron microscopic investigation of the F-adapted dinoflagellate cells showed abnormal ultrastructural features in the chloroplast (especially the pyrenoid), mitochondria, and nucleus. Simultaneous comparison with the F-inhibited dinoflagellate cells showed that thylakoid formation was extremely disorganized by fluoride and that F-adaptation conferred a prolamellar-like configuration on the thylakoids in the center of the pyrenoid. This unexpected appearance of lamellae formation in the F-adapted cells suggested that the pyrenoid may be a center for thylakoid assembly. Such cells also showed large, intensely osmiophilic inclusions in the mitochondria. Microbodies are found in close juxtaposition to the mitochondria and chloroplast, suggesting an increased metabolic dependence on photorespiration. The F-adapted nucleus showed dark and light concentric rings in the nucleolus region, accompanied by other signs of mitotic activity, which were not observed in the F-inhibited cells. It was inferred that the F-adaptation may have required some form of genetic change resulting presumably in the development of a phenotype mutant.

    Notizen: . The large fresh-water microaerobic amoeba Pelomyxa palustris does not contain mitochondria, but three types of bacterial endosymbionts are always present. Thus, it is of interest in the discussion of the possible origin of mitochondria from primitive prokaryotes. Gas exchanges (O2, CO2) and concentration of endosymbionts were determined in individual amoebae, in which the life cycle stage was noted. Grey type (stationary phase) amoebae had a lower O2 uptake and lower endosymbiont concentration than light type (growth phase) amoebae, and highest O2 uptake was found in centrifugal pieces of light type Pelomyxa, centrifuged in vivo, which contained nearly all of the endosymbionts. In light type amoebae, the respiratory activity was independent of O2 concentration between 1 and 21%, and, when compared on the basis of dry weight and protein, of the same order as that of other free-living protozoa. The R.Q. was slightly higher than 1, indicating that glycolysis does not play a significant role in energy metabolism. It is concluded that P. palustris is fully aerobic, and suggestions are presented as to the role of the endosymbionts in its respiratory metabolism.

    Notizen: . A series of new in vitro systems for the cultivation of bloodstream forms of Trypanosoma (Trypanozoon) brucei brucei, T. (T.) b. rhodesiense, and T. (T.) b. gambiense was developed. The standard system consists of a feeder layer of fibroblast-like cells derived from embryos of New Zealand White rabbits (REF) or a mountain vole, Microtus montanus (MEF), with HEPES-buffered Minimum Essential Medium (MEM), with Earle's salts, supplemented with 15% inactivated rabbit serum. These two and other feeder layers were cross-checked with different sera to test for growth support of bloodstream forms of the three trypanosome subspecies studied. Cultures could be initiated with bloodstream forms from mammalian hosts or from cryopreserved stabilates. Metacyclic forms from infected Glossina m. morsitans could also be used as inoculum; they transformed within 6 h to bloodstream forms. Maintenance of cultures and growth properties are described in detail. Experiments were undertaken to confirm that the cultivated bloodstream forms still possess some of the characteristic features of pleomorphic bloodstream populations. Cultivated bloodstream forms were always infective for mice, and a surface coat could be demonstrated by electron microscopy. They could also be cyclically transmitted through tsetse flies, and the metacyclic forms from these flies could be brought back into culture. In vitro cloning with single bloodstream forms and metacyclic forms could be achieved with high cloning efficiency. The consumption of glucose and the production of pyruvate and lactate were determined.

    Notizen: Fathead minnows, Pimephales promelas, raised from eggs in the laboratory, were experimentally infected with oocysts of Eimeria iroquoina from either P. promelas or the common shiner, Notropis cornutus. Within intestinal epithelial cells, trophozoites thought to be derived from the sporozoites contained a prominent electron-dense refractile body. Merozoites dedifferentiated into trophic forms by losing components of their apical complex and pellicle. The inner membrane components of the pellicle appeared discontinuous, and the micronemes became enclosed within vacuoles. Prior to merozoite formation, multinucleate meronts were limited by a single membrane. Golgi complexes were associated with the nuclei of this stage. Merozoites were formed by ectomerogony in one generation and by endomerogony in the final generation. In both forms of merogony the final nuclear division was coupled with the onset of differentiation of the merozoites and featured eccentric mitotic spindles associated with centrocones located within the nuclear envelope and with the precursors of the apical complex. A Golgi complex was closely associated with the nucleus and apical tip of the forming merozoite. Unlike other Eimeria species, the complete pellicle of the merozoites of the final asexual generation of E. iroquoina was formed within the cytoplasm of the meront, without association with the limiting membrane, thus, all pellicular components are synthesized de novo. The inner membranes of the pellicle initially appeared as longitudinal strips, each of which was associated with a pair of the 22–24 subpellicular microtubules. Mature meronts of the final asexual generation averaged 9 μm in diameter and produced 13–16 merozoites. With the exception of the internal completion of the pellicle of the final generation merozoites, the basic processes of merogony in fish Eimeria species are similar to those recorded in terrestrial hosts.

    Notizen: The glycerophospholipids of the protozoon Tetrahymena pyriformis W are unique in that the polyunsaturated fatty acid γ-linolenate (18:3Δ6,9,12) is a major component of both the sn-C-1 and sn-C-2 positions. [1-14C]γ-linolenate was found at both carbons of the three lipids, in general agreement with the mass distribution of γ-linolenate, except for markedly greater labeling at the sn-C-2 position of phosphatidylcholine. We hypothesize that an acyltransferase exists in Tetrahymena that can esterify γ-linolenate at both carbons during glycerophospholipid biosynthesis.

    Notizen: Promastigotes of Leismania donovani cultured for either 3 or 10 days in vitro and inoculated intracardially into golden hamsters with an equal number of organisms from either population showed a 7-fold difference in infectivity when compared at both 10 to 16 days post-infection. Reproducible histochemical staining for the promastigote enzymes glucose-6-phosphate dehydrogenase (G6PDH) and peptidase after polyacrylamide gel electrophoresis showed two isoelectric variants of G6PDH (Bands 1 and 2) that displayed a 45% decrease (Band 1) and a 60% increase (Band 2) in total activity when 3- and 10-day-old promastigores were compared. Peptidase activity, present in a single band, increased 7-fold in 10-day-old promastigotes. A decrease in the lectin-induced agglutination of promastigotes by castor bean agglutinin (RCA60), specific for D-galactose and N-acetyl-D-galactosamine, was seen when 3- and 10-day-old promastigotes are compared. Antisera raised against sonicated 10-day-old promastigotes showed a unique precipitin band between the antiserum and sonicated 10-day-old promastigotes not found between the antiserum and sonicated 3-day-old promastigotes.

    Notizen: The compound 6-(L-erythro-1,2′,3′-trihydroxypropyl)pterin, at a concentration of 50 pg/ml (“L-erythro-neopteria”), supports half-maximal growth of Crithidia fasciculata; biopterin at a concentration of 30 pg/ml is shown to yield similar growth. N2-dimethyl-6-(L-erythro-1′,2′,3′-trihydroxypropyl)pterin (A) was inactive even at 100 ng/ml. Synergism was observed with the N2-dimethylamino derivative (A) in the presence of suboptimal biopterin, its activity then being of the order of L-erythro-neopterin. In contrast, the stereoisomeric N2-dimethyl-6-(D-erythro-1′,2′,3′-trihydroxypropyl)pterin (“dimethyl-D-erythro-neopterin”) and its 3′-mono-phosphate only slightly enhanced growth under similar conditions but both threo-isomers had no supplementary activity. Biopterin-induced growth was slowed by 6-(D-erythro1′,2′,3′-trihydroxypropyl)pterin (D-neopterin); the threo-isomers had no such effect. An adaptive demethylation capacity by growing cultures and competition of biopterin uptake by D-neopterin seems likely. The report of the occurrence in Euglena of N2-dimethyl-6-(L-threo-1′,2′,3′-trihydroxypropyl)pterin and its 3′-mono-phosphate adds further interest to our observations.

    Notizen: We investigated the macronuclear DNA genomes of several marine and fresh-water ciliates. The marine forms studied were: Uronema nigricans, Parauronema virginianum, Parauronema acutum, and two strains of Miamiensis avidus; the fresh-water ciliates included: Tetrahymena pyriformis, Paramecium octaurelia, and P. caudatum. The organisms were cultured axenically and the DNA extracted from isolated and purified macronuclear preparations. Reassociation rate constants of purified DNA preparations used to calculate kinetic complexity were determined both optically and by hydroxyapatite chromatography. Analytical complexity was determined chemically. Ciliate macronuclear DNA appeared to reassociate as a single unique sequence, except for a small fraction (4% of the total DNA) that was repetitive and renatured rapidly. Values for the kinetic complexities of macronuclear DNA in these forms varied over a relatively narrow range, from 1.5 to 3.8 times 1010 daltons, and were only 7–15x larger than that of the bacterium Escherichia coli. On the other hand, values for analytical complexities of macronuclear DNA of marine and fresh-water ciliates varied over two orders of magnitude and were related to the size of the animals. It is suggested that ploidy levels of macronuclear DNA in these ciliates may represent a functionally permanent amplification of the genome.

However, when Ca2+ was removed by EGTA to negate the effects of the Ca2+-binding protein (calmodulin), which is required for the full activity of guanylate cyclase in this organism, no significant change in the enzymatic activity was observed throughout the cell cycle. On the other hand, the reduced guanylate cyclase activity at division phase was associated with a decreased level of calmodulin content. These results suggest that fluctuations in guanylate cyclase activity during the cell cycle would be dependent on the concentration of calmodulin.

    Notizen: Identification and characterization of kDNA is described in the naturally occurring totally dyskinetoplastic species Trypanosoma equinum. Fluorescence microscopy of live cells, using the highly sensitive and specific probe DAPI (4,6,-diamidino-2-phenyl-indole), showed the presence of a diversity of extranuclear fluorescent bodies scattered along the length of the organism. Transmission electron microscopic studies revealed a close similarity between the distribution of these DAPI-fluorescing particles and of dense aggregates of nonfibrillar material resembling the kDNA of dyskinetoplastic strains of other species. Variable sized remnants of kDNA, occurring singly or in clusters, were found scattered throughout the mitochondrion. Analytical cesium chloride ultracentrifugation of total cellular DNA extracts showed a kDNA banding profile at a buoyant density equal to 1.691 gm/cm3, representing approximately 11% of the total cellular DNA content. Molecular spreads of isolated kDNA revealed a population of open circular molecules ranging in contour length from 0.11–9.69 μm.

    Notizen: Morphogenesis in Conchophthirus curtus has been investigated by the use of protargol silver impregnation supplemented by selective use of scanning electron microscopy. Events are assigned to nine sequential stages; the last, stage 9, which involves maturation of the somatic ciliature and infraciliature as well as the final development of the buccal cavities of both proter and opisthe, occurs following cytokinesis. Stomatogenesis involves both the parental haplokinety and the deep kinetosomal unit (DKU). In perhaps a unique phenomenon, phylogenetically, the ciliated parental haplokinety forms the early oral primordium of the opisthe and is replaced in the proter by the DKU. The DKU then acts as a formative center for new kinetosomes that migrate into the developing opisthe primordium. Late in the process, the haplokinetal primordia of both proter and opisthe give rise to their respective DKU's. Some events of nuclear activity and somatic development are also described. The high degree of structural differentiation of this ciliate has provided the opportunity to examine temporal relationships during morphogenesis. We have found that somatic, buccal, and nuclear events proceed in a tightly coupled sequence. Hence, somatic and nuclear development can be directly correlated with the nine stages of stomatogenesis.

Owls fed sporulated oocysts became patent on days 8–13, and peak oocyst production occurred between days 14 and 18. Owls fed infected mice became patent on days 8–10, and peak oocyst production occurred between days 9 and 13. The reduction in length of the life cycle in owls fed infected mice provides evidence that both indirect and direct life cycles can occur in this species and suggests that parallels exist in the lives of various isosporid, eimeriid, and other coccidia that may not have been sufficiently appreciated in the past.

    Notizen: Histological examination of gills of the rock crab (Cancer irroratus Say, 1917) taken from specimens collected in polluted and nonpolluted areas indicated that gill discoloration was not directly related to the degree of microbial fouling. Light microscope studies demonstrated that bacteria, peritrich and suctorian ciliates, and some naviculoid diatoms were the principal members of the fouling community. Examination of the bacteria by electron microscopy showed that several distinct types were present, and that some of these were attached to the gill by holdfast structures while others were enmeshed in a filamentous slime layer. An unusual small marine flagellate and an unidentified amoeba were also examined ultrastructurally.

    Notizen: The endogenous development of Eimeria christenseni was studied in 10 two- to four-week-old kids inoculated with 106-107 sporulated oocysts. They were killed at intervals from two to 26 days after inoculation, and their tissues were examined for endogenous stages of the coccidian by light microscopy. Such stages were found in the small intestine and mesenteric lymph nodes. In the sexual cycle, two generations of meronts were found. The first generation developed in endothelial cells of lacteals in the jejunun and ileum and mesenteric lymph nodes, and mature meronts were first seen 14 days after inoculation. The second generation developed in epithelial cells of the glands of Lieberkuehn in the jejunum and ileum and in mesenteric lymph nodes, and its mature meronts were first seen by 16 days. Sexual stages were present mostly in epithelial cells of the tips and sides of the villi and less frequently in crypt cells of the jejunum and ileum. Mature macrogametes and microgamonts and oocysts were also first seen by 16 days. The prepatent period was 17 (14-23) days; the patent ranged from 8 to more than 30 days. Sporulation time was 3-4 days at 30°C. E. christenseni was found to be pathogenic, kids inoculated with 1-5 × 105 sporulated oocysts exhibited the following signs: severe diarrhea, anorexia, polydipsia, poor hair coat, and extreme weakness. They recovered about a month later, but their growth rates appeared to be lower than those of uninoculated animals kept under the same conditions. One kid died 20 days after inoculation with 107 oocysts.

    Notizen: The ultrastructure of Rhodomonas lacustris was examined using scanning and transmission electron microscopy, and the pigments were scanned in vivo by the opal plate technique. The periplast is composed of hexagonal plates, with trichocysts at the corners. This cryptomonad lacks a gullet but has a deep ventral furrow, in which more than 20 large ejectosomes are located. The chloroplast has mainly three thylakoids per lamella, and it is structurally reminiscent of that of certain haptophycean flagellates. The pigments appear to be as usual within the photosynthetic cryptomonads, with phycoerythrin type PE I (phycocyanin was not detected). Other cellular inclusions (e.g., lysosome-like vesicles and the contractile vacuolar complex) are also discussed.

    Notizen: Following conjugation in ciliates, the usual fate of the old pre-conjugant macronucleus is resorption. In some species, however, old macronuclei, or their fragments, have the ability to reform functional vegetative macronuclei when new macronuclear anlagen are defective. The present work on Euplotes shows that if anlagen are allowed to carry out their essential roles in early exconjugant development, including influence on cortical reorganization such that feeding can resume, they can then be permanently damaged by UV-microbeam irradiation and regeneration of old macronuclear fragments can occur. E. aediculatus exconjugants were anlage-irradiated at 40–60 hr of development and the irradiated cells cultured individually and fed. Squashes revealed enlargement and anteriorward migration of the persistent (posterior) macronuclear fragments. The first post-conjugant fission of such cells was delayed (times ranged 6–43 days) and did not seem to involve the damaged anlagen, which remained rudimentary, did not divide along with the cells, and were subsequently resorbed. It appeared that cell fission was supported by the fragments of the old macronuclei, which either divided or partitioned themselves between the two daughter cells. Mating tests performed on early clones derived from irradiated exconjugants revealed ample conjugation competence; intraclonal conjugation in such clones was also apparent. The absence of the immature period seen in normal exconjugants provides further evidence that the clones arose from cells with regenerated macronuclei.

    Notizen: Cell free preparations of Tetrahymena thermophila contain an enzyme that catalyzes the direct desaturation of stearoyl CoA to octadecenoic acid. The enzyme is associated with the microsomal fraction of the ciliate. Substrate for the enzyme consists of either free stearic acid or stearoyl CoA. Both ATP and CoA are required when free stearate is the substrate and are also highly stimulatory when stearoyl CoA is the substrate. With stearoyl CoA as the substrate, either NADH or NADPH are required for desaturase activity. In the presence of ATP and CoA, either NAD or NADP can replace NADH and NADPH. Desaturase activity is optimal when the enzyme is incubated at a pH of 7.2 and a temperature of 30–35°C. Highest levels of the stearoyl CoA desaturase are found in stationary phase ciliates grown at 35°C.

    Notizen: . Sporocysts of the coccidian Sarcocystis tenella were originally isolated in the feces of a coyote. Sporocysts used for inoculation of lambs were obtained from experimentally infected dogs. At 14, 16, and 19 days postinoculation (DPI) of lambs with the sporocysts, various developmental stages of first-generation meronts were found within cells located between the endothelium and internal elastic membrane of mesenteric arteries. At 19, 21, and 25 DPI, second-generation merogony occurred in cells associated with capillaries and arterioles of kidney glomeruli and convoluted tubules. Meronts of both generations were bounded by a double pellicular membrane and were situated free in the host cell cytoplasm. Merozoites formed by endopolygeny that involved multiple intranuclear spindles of a single, large irregular nucleus. First-generation meronts measured 22.6 × 17.1 μm (19–28.7 × 7.5–24 μm) and contained 120–240 merozoites, which measured 7.1 × 1.6 μm (4.8–7.5 × 1.3–1.8 μm). Corresponding values for second-generation meronts were 13.2 × 9.2 μm (8.3–15 × 7–13.5 μ), 32–80, and 5.8 × 1.7 μm (5.6–6.2 × 1.4–2.2 μm).

M. bhadrensis n. sp. was noticed in the musculature of the fry of Labeo rohita, collected from Bhadra Fish Seed Farm, in September 1979; and M. shettii, M. vedavatiensis, M. venkateshi, and M. hosadurgensis n. spp. were found in the gills of Cirrhina mrigala sampled from Vanivilasa Sagar Fish Farm in April 1980.     Notizen: The cell surface of Tritrichomonas foetus was characterized by using 18 highly purified lectins with specificities for N-acetyl glucosamine, N-acetyl galactosamine, galactose, mannose, and sialic acid. The specificity of the lectin-induced cell agglutination was verified by inhibition of the agglutination with the specific sugars. By using cytochemical techniques associated with electron microscopy, carbohydrates were detected on the cell surface of T. foetus. The following techniques were used: periodic acid-thiosemicarbazide-silver proteinate, concanavalin A-horseradish peroxidase, and ruthenium red. Anionic sites were detected on the cell surface of the protozoan at pH's 1.8 and 7.2 with the use of colloidal iron hydroxide and cationized ferritin particles, respectively. The binding of colloidal iron particles, as well as the agglutination induced by the lectin from Limulus polyphemus, indicated the presence of sialic acid on the cell surface of T. foetus.

    Notizen: A simple technique for plating trypanosomatids includes use of plates with lower agar concentrations than those usually prescribed for plating bacteria and a simple system to prevent dehydration due to the long incubation time needed for formation of visible colonies. Consistently high plating efficiency was thus achieved. Colonies from Herpetomonas samuelpessoai and Crithidia deanei were clearly distinguishable from each other; their individual characteristics varied with plating conditions.

    Notizen: By using fluorescent isothiocyanate-conjugated concanavalin A (FITC-Con A), cell surface events were examined under a light microscope during the early period of the conjugation process in Tetrahymena thermophila. Until the two complementary mating types (D-III and IV) were mixed, Con A-binding activities were hardly detected on the cell surface of ciliates. After mixing, however, the FITC-Con A (25 μg/ml) bound especially to the anterior cell surface at the early stage of conjugation, followed by characteristic changes of the Con A-binding pattern and, subsequently, by formation of a bright fluorescent ring around the area of contact between conjugants. Such alterations of FITC-Con A-binding pattern were found to be interrupted or eliminated by cycloheximide (2 μg/ml). These findings are related to the onset and subsequent conjugation in T. thermophila.

    Notizen: In a few homoiothermic host species, Eimeria spp. in a schizogonic stage, a gamogonic stage, or as resting sporozoites infect nonepithelial tissues. However, epithelium serves as a critical site in the cycle of these and all other species known from warm-blooded hosts. Eimeria funduli, infecting at least four different killifishes, undergoes both schizogony and gamogony in non-epithelial hepatic and pancreatic cells and requires an invertebrate host to complete its cycle. Oocysts are not released from the living fish into the environment. The cycle for this species, or aspects of it, may exemplify those found in several piscine species infecting nonepithelial (and possibly epithelial) cells. Eimeriu funduli differs from most species of Eimeria infecting homoiotherms in other respects, some of which may also characterize traits for piscine and other poikilotherm eimerians in general. Endogenous development is affected by temperature and host-age, sporogony occurs in the host, and infections occur in several related fishes. The presence of an intermediate host in at least E. funduli and the presence of sporozoites in macrophages of several eimerians of homoiotherms, as well as other features, suggest a closer relationship between eimeriids and Lankesterella, Schellackia, and even the Haemosporina than previously assumed.

    Notizen: Phytomonas davidi (Trypanosomatidae) possesses typical trypanosomatid organelles: subpellicular microtubules, kinetoplast-mitochondrial complex, K-DNA, and four subflagellar pocket microtubules. A greater concentration of subpellicular microtubules was observed in the latex forms than in those found in the salivary glands of its insect vector. Only in the latex flagellates (the stage with postnuclear torsion) were subpellicular microtubules interconnected by crossbridges observed. Morphology and development of mitochondrial aristae varied according to the source of the flagellates. Organisms taken from culture medium had extensively developed plate-like cristae; sparse tubular cristae were observed in the latex forms; and highly developed tubular cristae were seen in flagellates from the lumen of the vector's salivary glands, though organisms in the salivary gland channels had few or none.

    Notizen: In contrast with isosporoid species of coccidia that have established extraintestinal phases of development, the eimeriids, except for a few species, generally have been considered inhabitants of the intestinal tract. Eimeria infection in sandhill cranes (Grus canadensis) and whooping cranes (G. americana) may result in disseminated visceral coccidiosis. Nodules were observed in the oral cavity of 33% (n = 95) of the G. canadensis at the Patuxent Wildlife Research Center (PWRC) in Laurel, MD. Necropsy of six of the afflicted cranes revealed granulomatous nodules in many tissues and organs. Histologic studies disclosed protozoan organisms morphologically resembling schizonts in the granulomas, and endogenous stages of coccidia were present in the intestines of four birds. Fecalysis of three of four sandhill cranes yielded oocysts of E. reichenowi and E. gruis. Only E. reichenowi-type oocysts were recovered from a dead whooping crane sample. Domestic broiler chicks each intubated with about 1 times 106 pooled sporulated oocysts of E. reichenowi and E. gruis were not infected. Exposure of six incubator-hatched and hand-reared sandhill crane chicks to oocysts artificially (two chicks) and naturally (four chicks) resulted in typical infection of intestinal epithelium with invasion of subepithelial tissues extending to the muscular layer and widespread extraintestinal development. Asexual and sexual stages occurred primarily in macrophages in the liver, spleen, heart, and lung. In the lung, oocysts were found in bronchial exudate and epithelial lining cells. Six of ten G. canadensis chicks, one adult G. americana, and three of five G. americana chicks that died naturally at PWRC had disseminated visceral coccidiosis.

    Notizen: The somatic cortex of Spathidium spathula is described ultrastructurally. The pellicle consists of an outer membrane and an underlying alveolar system. Numerous membrane-bound mucocysts and spherical electron-opaque bodies have similar circular sites of attachment to the outer membrane. Below these are a microfibrillar zone and an underlying region of rough ER. Mitochondria are lined up under the rough ER in longitudinal cortical ridges. Parasomal sacs are found near the basal bodies and are associated with cytoplasmic membranous sacs. Various microtubular and fiber systems are associated with single basal bodies: (1) a short kinetodesmal fiber; (2) two transverse microtubular ribbons and a transverse fiber; (3) a postciliary microtubular ribbon, initially sandwiched by two fibers, which gives rise to longitudinal subpellicular microtubules extending posteriorly for a distance of some four or five basal bodies; and (4) a system of overlapping subkinetal microtubules. A three-dimensional reconstruction is included. The somatic cortex of S spathula is similar to that reported for other Haptorida of the ciliate subclass Gymnostomata.

    Notizen: The species-specific lethal factor obtained from Amoeba indica, a freeliving mononucleate amoeba, can be partially purified by differential centrifugation, gel exclusion chromatography, and elution from Cibacron blue-agarose. The factor is unstable in the partially purified form, but activity is correlated with a heat-labile protein having an apparent molecular weight of 186,000. The factor causes inhibition of division and death when injected into A. proteus. Purification of the lethal factor indicates that aggregation of the factor with amoebal proteins is not a hindrance to further investigation. The natural role of the factor is discussed.

Electron-microscopic observation of the stabilized third cell revealed that the two cell membranes at the uniting region were partially fused to form cytoplasmic bridges. Chemically treated cells began to cases feeding before the cell union and fully ceased such activity at ∼130 min after the treatment.

    Notizen: In its amoeboid stage, Protacanthamoeba caledonica n. g., n. sp. closely resembles the genus Acanthamoeba, on both light- and electron-microscopical levels, including possession of a centrosphere with a plaque-shaped centriole-like body. The cyst wall differs from that of Acanthamoeba in lack of preformed exit pores and in fine structure; the occasional apparent division into exocyst and endocyst is due to irregular splitting. The strain isolated from a Scottish estuary did not grow at 37°C and did not grow normally on agar made with 25% sea water, but cysts remained viable after a week in full-strength sea water. Protacanthamoeba n. g. is distinguished from Acanthamoeba on the basis of cyst structure, but it is assigned to the family Acanthamoebidae.

    Notizen: Blastocrithidia triatomae, a species very resistant to cultivation in conventional media, was successfully grown in association with cultured lepidopteran cells.     Notizen: Leptomonas lactosovorans n. sp. is described from the gut of the reduviid hemipteran Zelurus martinsi. It can be grown in a defined medium. The requirements for amino acids, vitamins, purine, and heme were similar to those established for Crithida fasciculata. Growth is stimulated by aeration. Generation time is longer than for any other insect trypanosomatid described; its utilization of lactose as a carbon source is a unique feature among trypanosomatids.

    Notizen: Anti-malarial gamete antibodies prevent the fertilization of gametes in the mosquito midgut and prevent transmission of malaria. Recently, hybridoma cell lines secreting monoclonal antibodies (10G3 and 11C7) against gametes of the malarial parasite have been developed. These antibodies act synergistically to mediate 80–90% suppression of the infectivity of gametocytes, although neither monoclonal antibody alone has a significant effect on gametocyte infectivity. We performed immuno-electron microscopy to characterize the interactions of these monoclonal antibodies with gametes of Plasmodium gallinaceum. Male gametes exposed to either 10G3 or 11C7 agglutinated into loose clusters, while those exposed to a mixture of 10G3 and 11C7 agglutinated into long, rope-like bundles. This difference appears to be related to the distribution of the antibodies on the surface of the gametes. When 10G3 or 11C7 labeled with a ferritin-conjugated anti-mouse Ig were used singly, the ferritin particles were distributed in focal areas over the surface of the parasites. By contrast, when the male gametes were exposed to a mixture of 10G3 and 11C7, the ferritin particles were distributed over their entire surface. Female gametes reacted similarly to these antibodies. These observations indicate that combinations of antibody specificities that reduce fertilization efficiency coat the entire surface of the gametes. On the other hand, focal interactions resulting from a single antibody are unable to block fertilization.

    Notizen: Inhibition of cell division in Euglena gracilis by visible light is pH-dependent, being most severe in the pH range 3.5–5.0. Transfer of phototrophic cells from pH 6.8 to pH 4.2 can lead to cell death at modest light intensities (500 ft-cd) and inhibition of division at lower intensities (300 ft-cd). Inhibition is preceded by a large influx of phosphate, most of which remains in the cold PCA pool. It is suggested that light may act to reduce control over phosphate entry at these intermediate pH levels, and excess phosphate leads to inhibition of cell division or death.
