    Description / Table of Contents: Zusammenfassung Männlichen weißen Mäusen wurde 30 Tage lang täglich einmal Metopiron intraperitoneal injiziert, um eine etwaige Wirkung der Steroidsynthese in der Nebennierenrinde auf hypothalamo-hypophysäre Systeme zu verfolgen. Die histologische und autoradiographische Untersuchung hatte folgende Ergebnisse: 1. Das supraoptico-neurohypophysäre und das tubero-infundibuläre System reagieren auf die experimentelle Störung der Hormonsynthese. 2. In allen Abschnitten des Tractus supraoptico-neurohypophyseus kommt es zu einer Verminderung Paraldehyd-Fuchsin-färbbaren Neurosekretes. Dieser Neurosekretabnahme entspricht ein unverminderter Einbau von Cysteinschwefel in die großzelligen Kerngebiete und ein beschleunigter Transport radioaktiver Substanzen durch die Zona interna infundibuli. Die Neurohypophyse gibt das am Schwefel markierte Produkt vorzeitig ab. 3. Im tubero-infundibulären System lassen sich keine Paraldehyd-Fuchsin-positiven Substanzen nachweisen, obwohl das Auftreten PAF-positiven Neurosekretes in der Zona externa infundibuli nach beiderseitiger Adrenalektomie erwiesen ist. Dagegen zeigt das Autoradiogramm des kleinzelligen Hypothalamus einen vermehrten Einbau des Cysteinschwefels im Bereich des Nucleus infundibularis und eine beschleunigte Abgabe markierten Materials an die Zone externa infundibuli. Gleichzeitig ist die Aufnahme der Schwefelaktivität in der Adenohypophyse gesteigert. 4. Es ergibt sich also eine unterschiedliche Wirkung von Metopiron und Adrenalektomie auf den Hypothalamus. Der Vergleich der färberischen mit der autoradiographischen Darstellung zeigt, daß die abgeschwächte Farbreaktion bei den Versuchstieren Ausdruck einer Aktivitätssteigerung im neurosekretorischen System ist. Es konnte nicht entschieden werden, ob die radioaktiv markierten Stoffe im tubero-infundibulären System in Nervenzellen des Nucleus infundibularis oder in Gliazellen, z. B. dem Tanycytenependym, gebildet werden. 5. Der erhöhte Gehalt des Isotops in den Ganglienzellen des Nervus trigeminus zeigt, daß die Wirkung von Metopiron nicht auf den Hypothalamus beschränkt bleibt, sondern darüber hinaus andere Bereiche des Nervensystems beeinflußt.
    Notes: Summary Male albino mice were subjected to single daily intraperitoneal injections of Metopiron over a period of 30 days. The effects of inhibition of adrenocortical steroid-synthesis on the hypothalamo-hypophyseal system were then studied, histologically and autoradiographically. The findings are as follows: 1. The supraoptico-neurohypophyseal system and the tubero-infundibular system are both affected by experimental interference with the hormone synthesis. 2. In all parts of the supraoptico-neurohypophyseal tract there is a decrease of PAF-positive neurosecretory substance. This corresponds to an undiminished incorporation of cysteine sulphur in the magnocellular nuclear areas and an accelerated transport of radioactive substances through the inner layer of the median eminence. The neurohypophysis releases the sulphur-labelled substance prematurely. 3. In the tubero-infundibular system PAF-positive material could not be demonstrated although the occurrence of PAF-positive neurosecretory substance in the external layer of the median eminenc following bilateral adrenalectomy has been established. On the other hand, the autoradiogram of the parvocellular hypothalamus shows a strongly increased incorporation of the cysteine sulphur in the region of the nucleus infundibularis and a more rapid release of tracer substance into the external layer of the median eminence. Simultaneously the uptake of the sulphur in the adenohypophysis is intensified. 4. It is thus clear that Metopiron exerts a dissimilar effect from adrenalectomy on the hypothalamus. A comparison of sections stained with PAF and autoradiographs shows that, following treatment with Metopiron, the weakened staining reaction is the expression of an increased activity in the neurosecretory system. It could not be decided whether the radioactively labelled substances in the tubero-infundibular system are produced in the nerve cells of the nucleus infundibularis or in glial cells, for instance in the tanycyte-ependyma. 5. The increased content of the isotope in the ganglion cells of the trigeminus nerve demonstrates that the effect of Metopiron is not limited to the hypothalamus but also involves other parts of the nervous system.

    Notes: Summary The primary plexus of the toad hypothalamic-adenohypophysial portal system has two types of loops. The short loops are localized in the external region of the median eminence and surrounded by nerve endings and glial cells. The long loops approach the ependymal lining of the median eminence. The ascending and descending branches of these loops are surrounded by nerve and ependymal endings and glial cells. The actual subependymal portion of the long loops is virtually in contact with ependymal processes only, which form a “cuff” interposed between this portion of the long loops and the fibres of the hypothalamic-neurohypophysial tract. Many of the vascular endings of the ependymal processes have electron dense granules whose diameter ranges between 700 and 1400 Å. The ultrastructure of the ependymal cells suggests that these granules are transport material and not secretory material. This anatomical arrangement linking the ependyma of the median eminence and the long loops of the primary plexus of the hypothalamic-adenohypophysial portal system makes the possibility of an interrelationship between the cerebrospinal fluid and the portal blood very considerable.

    Description / Table of Contents: Zusammenfassung Der Nucleus supraopticus der Ratte, die einer Dehydratation ausgesetzt war, wurde ultrastrukturell-morphometrisch analysiert. Dabei zeigte sich, daß die relativen Volumenanteile der einzelnen Zellkompartimente während der fünftägigen Durstperiode eine auffallende Konstanz aufweisen. Hingegen läßt sich eine absolute Zunahme der Einzelzellvolumina und somit auch der an der Synthese und Sekretion der Neurohormone beteiligten Zellkompartimente feststellen. Die vorliegenden Befunde sprechen für einen beschleunigten Abtransport des neurosekretorischen Materials bei gesteigerter Synthese. Auf eine optimale Standardisierung der Perfusionsmethode bei Untersuchungen am neurosekretorischen Zwischenhirnsystem wird hingewiesen.
    Notes: Summary The supraoptic nucleus of the dehydrated rat has been analysed by electron microscopy and morphometry. With that it appears, that the relative volumes of the different cell compartments are striking constant. Otherwise one can see an absolute increase of the cell volume together with the cell compartments which take part at the synthesis and secretion of the neurohormones. These results are expression of an accelerated move of the neurosecretory material during increased synthesis. The importance of an optimal standardization of the perfusion-method in investigations of the neurosecretory system is demonstrated.

    Description / Table of Contents: Zusammenfassung Der Hypophysenhinterlappen von Ratten, die einer Dehydratation unterworfen waren, wurde ultrastrukturell-morphometrisch untersucht. Als wichtigster Befund wurde bereits nach 36stündiger Dehydratation eine massive Abnahme der Neurosekretgranula festgestellt. Gleichzeitig ist ein signifikanter Anstieg der Fettkörper in den Pituicyten zu verzeichnen. Die Fettkörper können nach Ansicht der Autoren als Endprodukt der Membranreste oder der Trägersubstanz des Neurohormons interpretiert werden. Eine Mitbeteiligung der Lysosomen bei ihrer Verarbeitung wird diskutiert.
    Notes: Summary The neurohypophysis of the rat after dehydration has been studied by electron microscopic and morphometric methods. As the main result we found a massive decrease of the neurosecretory granules already at a dehydration of 36 hours. At the same time one can note a significant increase of fat vacuoles in the pituicytes. These fat vacuoles could be the final product of residual membranes or of the carrier substance of the neurohormones. The possibility of a functional role of lysosomes in the catabolism is discussed.

    Notes: Summary The SPX-organ in Boreomysis arctica (Krøyer) (Crustacea Malacostraca Mysidacea) was investigated light and electron microscopically. The organ consists of a group of cells (the SPX-cells) and a vesicle surrounded by a connective tissue sheath. It is situated near the base of the sensory papilla of the eye-stalk. Neurosecretory material is produced in the SPX-cells and transported in axon-like projections from these cells into the vesicle. These processes contain no neurotubuli. Numerous fibres from an afferent nerve emanating from the medulla terminalis also enter the vesicle, where they form a dense irregular meshwork. This nerve transports no neurosecretory material. There are numerous synaptic contacts between the afferent nerve fibres and the neurosecretory processes from the SPX-cells. The neurosecretory material released from them accumulates in haemocoelic spaces in the vesicle. Release is most probably effected by the afferent nerve.
