    Beschreibung: Carbon nanotubes (CNTs) used as a probe for scanning probe microscopy has become one of the many potential usages of CNTs that is finding real applications in scientific research and industrial communities. It has been proposed that the unique mechanical buckling properties of the CNT would lessen the imaging force exerted on the sample and, thus, make CNT scanning probes ideal for imaging soft materials, including biological samples in liquid environments. The hydrophobic nature of the CNT graphitic sidewall is clearly chemically incompatible with the aqueous solution requirements in some biological imaging applications. In this paper, we present electron micrograph results demonstrating the instability of CNT scanning probes when submerged in aqueous solution. Moreover, we also introduce a novel approach to resolve this chemical incompatibility problem. By coating the CNT probe with ethylenediamine, thus rendering the CNT probe less hydrophobic, we demonstrate the liquid imaging capability of treated CNT probes. Experimental data for imaging in aqueous solutions are presented, which include an ultrathin Ir film and DNA molecules on a mica surface.

    Beschreibung: Ethylene production by 10 or 20 m2 stands of wheat, soybean, lettuce, potato, and tomato was monitored throughout growth and development in an atmospherically closed plant chamber. Chamber ethylene levels varied among species and rose during periods of canopy expansion and rapid growth for all species. Following this, ethylene levels either declined during seed fill and maturation for wheat and soybean, or remained relatively constant for potato and tomato (during flowering and early fruit development). Lettuce plants were harvested during rapid growth and peak ethylene production. Chamber ethylene levels increased rapidly during tomato ripening, reaching concentrations about 10 times that measured during vegetative growth. The highest ethylene production rates during vegetative growth ranged from 1.6 to 2.5 nmol m-2 d-1 during rapid growth of lettuce and wheat stands, or about 0.3 to 0.5 nmol g-1 fresh weight per hour. Estimates of stand ethylene production during tomato ripening showed that rates reached 43 nmol m-2 d-1 in one study and 93 nmol m-2 d-1 in a second study with higher lighting, or about 50x that of the rate during vegetative growth of tomato. In a related test with potato, the photoperiod was extended from 12 to 24 hours (continuous light) at 58 days after planting (to increase tuber yield), but this change in the environment caused a sharp increase in ethylene production from the basal rate of 0.4 to 6.2 nmol m-2 d-1. Following this, the photoperiod was changed back to 12 h at 61 days and ethylene levels decreased. The results suggest three separate categories of ethylene production were observed with whole stands of plants: 1) production during rapid vegetative growth, 2) production during climacteric fruit ripening, and 3) production from environmental stress.

    Beschreibung: The productivity of lettuce in a combination of high light, high temperature, and elevated CO2 has not been commonly studied because rapid growth usually causes a calcium deficiency in meristems called tipburn, which greatly reduces quality and marketability. We eliminated tipburn by blowing air directly onto the meristem, which allowed us to increase the photosynthetic photon flux (PPF) to 1000 micromoles m-2 s-1 (57.6 mol m-2 d-1); two to three times higher than normally used for lettuce. Eliminating tipburn doubled edible yield at the highest PPF level. In addition to high PPF, CO2 was elevated to 1200 micromoles m-2 mol-1, which increased the temperature optimum from 25 to 30 degrees C. The higher temperature increased leaf expansion rate, which improved radiation capture and more than doubled yield. Photosynthetic efficiency, measured as canopy quantum yield in a whole-plant gas exchange system, steadily increased up to the highest temperature of 32 degrees C in high CO2. The highest productivity was 19 g m-2 d-1 of dry biomass (380 g d-1 fresh mass) averaged over the 23 days the plants received light. Without the limitation of tipburn, the combination of high PPF, high temperature, and elevated CO2 resulted in a 4-fold increase in growth rate over productivity in conventional environments.

    Beschreibung: Nanomaterials are part of an industrial revolution to develop lightweight but strong materials for a variety of purposes. Single-wall carbon nanotubes are an important member of this class of materials. They structurally resemble rolled-up graphite sheets, usually with one end capped; individually they are about 1 nm in diameter and several microns long, but they often pack tightly together to form rods or ropes of microscopic sizes. Carbon nanotubes possess unique electrical, mechanical, and thermal properties and have many potential applications in the electronics, computer, and aerospace industries. Unprocessed nanotubes are very light and could become airborne and potentially reach the lungs. Because the toxicity of nanotubes in the lung is not known, their pulmonary toxicity was investigated. The three products studied were made by different methods and contained different types and amounts of residual catalytic metals. Mice were intratracheally instilled with 0, 0.1, or 0.5 mg of carbon nanotubes, a carbon black negative control, or a quartz positive control and euthanized 7 d or 90 d after the single treatment for histopathological study of the lungs. All nanotube products induced dose-dependent epithelioid granulomas and, in some cases, interstitial inflammation in the animals of the 7-d groups. These lesions persisted and were more pronounced in the 90-d groups; the lungs of some animals also revealed peribronchial inflammation and necrosis that had extended into the alveolar septa. The lungs of mice treated with carbon black were normal, whereas those treated with high-dose quartz revealed mild to moderate inflammation. These results show that, for the test conditions described here and on an equal-weight basis, if carbon nanotubes reach the lungs, they are much more toxic than carbon black and can be more toxic than quartz, which is considered a serious occupational health hazard in chronic inhalation exposures.

    Beschreibung: Rapid microbial monitoring technologies are invaluable in assessing contamination of spacecraft and associated environments. Universal and widespread elements of microbial structure and chemistry are logical targets for assessing microbial burden. Several biomarkers such as ATP, LPS, and DNA (ribosomal or spore-specific), were targeted to quantify either total bioburden or specific types of microbial contamination. The findings of these assays were compared with conventional, culture-dependent methods. This review evaluates the applicability and efficacy of some of these methods in monitoring the microbial burden of spacecraft and associated environments. Samples were collected from the surfaces of spacecraft, from surfaces of assembly facilities, and from drinking water reservoirs aboard the International Space Station (ISS). Culture-dependent techniques found species of Bacillus to be dominant on these surfaces. In contrast, rapid, culture-independent techniques revealed the presence of many Gram-positive and Gram-negative microorganisms, as well as actinomycetes and fungi. These included both cultivable and noncultivable microbes, findings further confirmed by DNA-based microbial detection techniques. Although the ISS drinking water was devoid of cultivable microbes, molecular-based techniques retrieved DNA sequences of numerous opportunistic pathogens. Each of the methods tested in this study has its advantages, and by coupling two or more of these techniques even more reliable information as to microbial burden is rapidly obtained.     Beschreibung: The study of Earth's radiation belts is one of the oldest subjects in space physics. Despite the tremendous progress made in the last four decades, we still lack a complete understanding of the radiation belts in terms of their configurations, dynamics, and detailed physical accounts of their sources and sinks. The static nature of early empirical trapped radiation models, for examples, the NASA AP-8 and AE-8 models, renders those models inappropriate for predicting short-term radiation belt behaviors associated with geomagnetic storms and substorms. Due to incomplete data coverage, these models are also inaccurate at low altitudes (e.g., 〈1000 km) where many robotic and human space flights occur. The availability of radiation data from modern space missions and advancement in physical modeling and data management techniques have now allowed the development of new empirical and physical radiation belt models. In this paper, we will review the status of modern radiation belt modeling. Published by Elsevier Ltd on behalf of COSPAR.

    Beschreibung: The latest process for producing large quantities of single-walled carbon nanotubes (SWNTs) to emerge from the Rice University, dubbed HiPco, is living up to its promise. The current production rates approach 450 mg/h (or 10 g/day), and nanotubes typically have no more than 7 mol % of iron impurities. Second-generation HiPco apparatus can run continuously for 7-10 days at a time. In the HiPco process nanotubes grow in high-pressure, high-temperature flowing CO on catalytic clusters of iron. Catalyst is formed in situ by thermal decomposition of iron pentacarbonyl, which is delivered intact within a cold CO flow and then rapidly mixed with hot CO in the reaction zone. Upon heating, the Fe(CO)5 decomposes into atoms that condense into larger clusters. SWNTs nucleate and grow on these particles in the gas phase via CO disproportionation: CO + CO --〉 CO2 + C (SWNT), catalyzed by the Fe surface. The concentration of CO2 produced in this reaction is equal to that of carbon and can therefore serve as a useful real-time feedback parameter. It was used to study and optimize SWNT production as a function of temperature, pressure, and Fe(CO)5 concentration. The results of the parametric study are in agreement with current understanding of the nanotube formation mechanism.

    Beschreibung: Chemical kinetic models for the nucleation and growth of clusters and single-walled carbon nanotube (SWNT) growth are developed for numerical simulations of the production of SWNTs. Two models that involve evaporation and condensation of carbon and metal catalysts, a full model involving all carbon clusters up to C80, and a reduced model are discussed. The full model is based on a fullerene model, but nickel and carbon/nickel cluster reactions are added to form SWNTs from soot and fullerenes. The full model has a large number of species--so large that to incorporate them into a flow field computation for simulating laser ablation and arc processes requires that they be simplified. The model is reduced by defining large clusters that represent many various sized clusters. Comparisons are given between these models for cases that may be applicable to arc and laser ablation production. Solutions to the system of chemical rate equations of these models for a ramped temperature profile show that production of various species, including SWNTs, agree to within about 50% for a fast ramp, and within 10% for a slower temperature decay time.

    Beschreibung: The cWINNOWER algorithm detects fuzzy motifs in DNA sequences rich in protein-binding signals. A signal is defined as any short nucleotide pattern having up to d mutations differing from a motif of length l. The algorithm finds such motifs if a clique consisting of a sufficiently large number of mutated copies of the motif (i.e., the signals) is present in the DNA sequence. The cWINNOWER algorithm substantially improves the sensitivity of the winnower method of Pevzner and Sze by imposing a consensus constraint, enabling it to detect much weaker signals. We studied the minimum detectable clique size qc as a function of sequence length N for random sequences. We found that qc increases linearly with N for a fast version of the algorithm based on counting three-member sub-cliques. Imposing consensus constraints reduces qc by a factor of three in this case, which makes the algorithm dramatically more sensitive. Our most sensitive algorithm, which counts four-member sub-cliques, needs a minimum of only 13 signals to detect motifs in a sequence of length N = 12,000 for (l, d) = (15, 4).     Beschreibung: Different types of lasers are now routinely used to prepare single-walled carbon nanotubes. The original method developed by researchers at Rice University used a "double-pulse laser oven" process. Several researchers have used variations of the lasers to include one-laser pulse (green or infrared), different pulse widths (ns to micros as well as continuous wave), and different laser wavelengths (e.g., CO2, or free electron lasers in the near to far infrared). Some of these variations are tried with different combinations and concentrations of metal catalysts, buffer gases (e.g., helium), oven temperatures, flow conditions, and even different porosities of the graphite targets. This article is an attempt to cover all these variations and their relative merits. Possible growth mechanisms under these different conditions will also be discussed.

    Beschreibung: BACKGROUND: Reducing cost and time is the major concern in clinical diagnostics, particularly in molecular diagnostics. Miniaturization technologies have been recognized as promising solutions to provide low-cost microchips for diagnostics. With the recent advancement in nanotechnologies, it is possible to further improve detection sensitivity and simplify sample preparation by incorporating nanoscale elements in diagnostics devices. A fusion of micro- and nanotechnologies with biology has great potential for the development of low-cost disposable chips for rapid molecular analysis that can be carried out with simple handheld devices. APPROACH: Vertically aligned multiwalled carbon nanotubes (MWNTs) are fabricated on predeposited microelectrode pads and encapsulated in SiO2 dielectrics with only the very end exposed at the surface to form an inlaid nanoelectrode array (NEA). The NEA is used to collect the electrochemical signal associated with the target molecules binding to the probe molecules, which are covalently attached to the end of the MWNTs. CONTENT: A 3 x 3 microelectrode array is presented to demonstrate the miniaturization and multiplexing capability. A randomly distributed MWNT NEA is fabricated on each microelectrode pad. Selective functionalization of the MWNT end with a specific oligonucleotide probe and passivation of the SiO2 surface with ethylene glycol moieties are discussed. Ru(bpy)2+ -mediator-amplified guanine oxidation is used to directly measure the electrochemical signal associated with target molecules. SUMMARY: The discussed MWNT NEAs have ultrahigh sensitivity in direct electrochemical detection of guanine bases in the nucleic acid target. Fewer than approximately 1000 target nucleic acid molecules can be measured with a single microelectrode pad of approximately 20 x 20 microm2, which approaches the detection limit of laser scanners in fluorescence-based DNA microarray techniques. MWNT NEAs can be easily integrated with microelectronic circuitry and microfluidics for development of a fully automated system for rapid molecular analysis with minimum cost.

    Beschreibung: A detailed study was made of the biological cleaning effectiveness, defined in terms of the ability to remove bacterial spores, of a number of methods used to clean hardware surfaces. Aluminum (Al 6061) and titanium (Ti 6Al-4V) were chosen for the study as they were deemed the two materials most likely to be used in spacecraft extraterrestrial sampler construction. Metal coupons (1 cm x 2.5 cm) were precleaned and inoculated with 5.8 x 10(3) cultivable Bacillus subtilis spores, which are commonly found on spacecraft surfaces and in the assembly environments. The inoculated coupons were subsequently cleaned using: (1) 70% isopropyl alcohol wipe; (2) water wipe; (3) multiple-solvent flight-hardware cleaning procedures used at the Jet Propulsion Laboratory (JPL); (4) Johnson Space Center-developed ultrapure water rinse; and (5) a commercial, semi-aqueous, multiple-solvent (SAMS) cleaning process. The biological cleaning effectiveness was measured by agar plate assay, sterility test (growing in liquid media), and epifluorescent microscopy. None of the cleaning protocols tested completely removed viable spores from the surface of the aluminum. In contrast, titanium was capable of being cleaned to sterility by two methods, the JPL standard and the commercial SAMS cleaning process. Further investigation showed that the passivation step employed in the JPL standard method is an effective surface sterilant on both metals but not compatible with aluminum. It is recommended that titanium (Ti 6Al-4V) be considered superior to aluminum (Al 6061) for use in spacecraft sampling hardware, both for its potential to be cleaned to sterilization and for its ability to withstand the most effective cleaning protocols.

    Beschreibung: Zoological data and a growing mythology contributed to ancient Western knowledge about large serpents. Yet little modern attention has been paid to the sources, transmission, and receipt in the early Middle Ages of the ancients' information concerning "dragons" and "sea serpents." Real animals--primarily pythons and whales--lie behind the ancient stories. Other animals, conflations of different animals, simple misunderstandings, and willful exaggerations are found to account for the fanciful embellishments, but primitive myths played no significant role in this process during classical times. The expedition of Alexander the Great into India (327-325 B.C.) and the Bagradas River incident in North Africa (256 B.C.) had enormous repercussions on the development of serpent lore. Credible evidence is found for the presence of ancient populations of pythons living along the North African coast west of Egypt and along the coast of the Arabian Sea between the Indus River and the Strait of Hormuz--places where they no longer exist today. The maximum sizes of ancient pythons may have been greater than those of today's specimens.

    Beschreibung: Robert Barany won the 1914 Nobel Prize in medicine for his convection hypothesis for caloric stimulation. Microgravity caloric tests aboard the 1983 SpaceLab 1 mission produced nystagmus results that contradicted the basic premise of Barany's convection theory. In this paper, we present a fluid structural analysis of the caloric stimulation of the lateral semicircular canal. Direct numerical simulations indicate that on earth, natural convection is the dominant mechanism for endolymphatic flow. However, in the microgravity environment of orbiting spacecraft, where buoyancy effects are mitigated, an expansive convection becomes the sole mechanism for producing endolymph motion and cupular displacement. Transient 1 g and microgravity case studies are presented to delineate the different dynamic behaviors of the 1 g and microgravity endolymphatic flows. The associated fluid-structural interactions are also analyzed based on the time evolution of cupular displacements.

    Beschreibung: Plants will be an important component of future long-term space missions. Lighting systems for growing plants will need to be lightweight, reliable, and durable, and light-emitting diodes (LEDs) have these characteristics. Previous studies demonstrated that the combination of red and blue light was an effective light source for several crops. Yet the appearance of plants under red and blue lighting is purplish gray making visual assessment of any problems difficult. The addition of green light would make the plant leave appear green and normal similar to a natural setting under white light and may also offer a psychological benefit to the crew. Green supplemental lighting could also offer benefits, since green light can better penetrate the plant canopy and potentially increase plant growth by increasing photosynthesis from the leaves in the lower canopy. In this study, four light sources were tested: 1) red and blue LEDs (RB), 2) red and blue LEDs with green fluorescent lamps (RGB), 3) green fluorescent lamps (GF), and 4) cool-white fluorescent lamps (CWF), that provided 0%, 24%, 86%, and 51% of the total PPF in the green region of the spectrum, respectively. The addition of 24% green light (500 to 600 nm) to red and blue LEDs (RGB treatment) enhanced plant growth. The RGB treatment plants produced more biomass than the plants grown under the cool-white fluorescent lamps (CWF treatment), a commonly tested light source used as a broad-spectrum control.

    Beschreibung: ALTEA-MICE will supplement the ALTEA project on astronauts and provide information on the functional visual impairment possibly induced by heavy ions during prolonged operations in microgravity. Goals of ALTEA-MICE are: (1) to investigate the effects of heavy ions on the visual system of normal and mutant mice with retinal defects; (2) to define reliable experimental conditions for space research; and (3) to develop animal models to study the physiological consequences of space travels on humans. Remotely controlled mouse setup, applied electrophysiological recording methods, remote particle monitoring, and experimental procedures were developed and tested. The project has proved feasible under laboratory-controlled conditions comparable in important aspects to those of astronauts' exposure to particle in space. Experiments are performed at the Brookhaven National Laboratories [BNL] (Upton, NY, USA) and the Gesellschaft fur Schwerionenforschung mbH [GSI]/Biophysik (Darmstadt, FRG) to identify possible electrophysiological changes and/or activation of protective mechanisms in response to pulsed radiation. Offline data analyses are in progress and observations are still anecdotal. Electrophysiological changes after pulsed radiation are within the limits of spontaneous variability under anesthesia, with only indirect evidence of possible retinal/cortical responses. Immunostaining showed changes (e.g. increased expression of FGF2 protein in the outer nuclear layer) suggesting a retinal stress reaction to high-energy particles of potential relevance in space. c2004 COSPAR.     Beschreibung: The ALTEA project investigates the risks of functional brain damage induced by particle radiation in space. A modular facility (the ALTEA facility) is being implemented and will be operated in the International Space Station (ISS) to record electrophysiological and behavioral descriptors of brain function and to monitor their time dynamics and correlation with particles and space environment. The focus of the program will be on abnormal visual perceptions (often reported as "light flashes" by astronauts) and the impact on retinal and brain visual structures of particle in microgravity conditions. The facility will be made available to the international scientific community for human neurophysiological, electrophysiological and psychophysics experiments, studies on particle fluxes, and dosimetry. A precursor of ALTEA (the 'Alteino' project) helps set the experimental baseline for the ALTEA experiments, while providing novel information on the radiation environment onboard the ISS and on the brain electrophysiology of the astronauts during orbital flights. Alteino was flown to the ISS on the Soyuz TM34 as part of mission Marco Polo. Controlled ground experiments using mice and accelerator beams complete the experimental strategy of ALTEA. We present here the status of progress of the ALTEA project and preliminary results of the Alteino study on brain dynamics, particle fluxes and abnormal visual perceptions. c2004 COSPAR.     Beschreibung: The main goal of PUR experiment (phage and uracil response) is to examine and quantify the effect of specific space conditions on nucleic acid models. To achieve this an improved method was elaborated for the preparation of DNA and bacteriophage thin films. The homogeneity of the films was controlled by UV spectroscopy and microscopy. To provide experimental evidence for the hypothesis that interplanetary transfer of the genetic material is possible, phage T7 and isolated T7 DNA thin films have been exposed to selected space conditions: intense UVC radiation (lambda=254 nm) and high vacuum (10(-4) Pa). The effects of DNA hydration, conformation and packing on UV radiation damage were examined. Characteristic changes in the absorption spectrum, in the electrophoretic pattern of DNA and the decrease of the amount of PCR products have been detected indicating the photodamage of isolated and intraphage DNA. c2004 COSPAR.     Beschreibung: Anionic (sodium laureth sulfate, SLES), amphoteric (cocamidopropyl betaine, CAPB) and nonionic (alcohol polyethoxylate, AE) surfactants were added to separate nutrient film technique (NFT) hydroponic systems containing dwarf wheat (Triticum aestivum cv. USU Apogee) in a series of 21 day trials. Surfactant was added either in a (1). temporally dynamic mode (1-3 g surfactant m(-2) growing area d(-1)) as effected by automatic addition of a 300 ppm surfactant solution to meet plant water demand, or (2). continuous mode (2 g surfactant m(-2) growing area d(-1)) as effected by slow addition (10 mLh(-1)) of a 2000 ppm surfactant solution beginning at 4d after planting. SLES showed rapid primary degradation in both experiments, with no accumulation 24 h after initial addition. CAPB and AE were degraded less rapidly, with 30-50% remaining 24 h after initial addition, but CAPB and AE levels were below detection limit for the remainder of the study. No reductions in vegetative growth of wheat were observed in response to SLES, but biomass was reduced 20-25% with CAPB and AE. Microbial communities associated with both the plant roots and wetted hardware surfaces actively degraded the surfactants, as determined by monitoring surfactant levels following pulse additions at day 20 (with plants) and day 21 (after plant removal). In order to test whether the biofilm communities could ameliorate phytotoxicity by providing a microbial community acclimated for CAPB and AE decay, the continuous exposure systems were planted with wheat seeds after crop removal at day 21. Acclimation resulted in faster primary degradation (〉90% within 24h) and reduced phytotoxicity. Overall, the studies indicate that relatively small areas (3-5m(2)) of hydroponic plant systems can process per capita production of mixed surfactants (5-10 g x person(-1)d(-1)) with minimal effects on plant growth.

    Beschreibung: Mutations in BRCA1 are characterized by predisposition to breast cancer, ovarian cancer and prostate cancer as well as colon cancer. Prognosis for this cancer survival depends upon the stage at which cancer is diagnosed. Reliable and rapid mutation detection is crucial for the early diagnosis and treatment. We developed an electronic assay for the detection of a representative single nucleotide polymorphism (SNP), deletion and insertion in BRCA1 gene by the microelectronics microarray instrumentation. The assay is rapid, and it takes 30 minutes for the immobilization of target DNA samples, hybridization, washing and readout. The assay is multiplexing since it is carried out at the same temperature and buffer conditions for each step. The assay is also highly specific, as the signal-to-noise ratio is much larger than recommended value (72.86 to 321.05 vs. 5) for homozygotes genotyping, and signal ratio close to the perfect value 1 for heterozygotes genotyping (1.04).

    Beschreibung: According to D. E. Broadbent's (1958) selective filter theory, people do not process unattended stimuli beyond the analysis of basic physical properties. This theory was later rejected on the basis of numerous findings that people identify irrelevant (and supposedly unattended) stimuli. A careful review of this evidence, however, reveals strong reasons to doubt that these irrelevant stimuli were in fact unattended. This review exposed a clear need for new experiments with tight control over the locus of attention. The authors present 5 such experiments using a priming paradigm. When steps were taken to ensure that irrelevant stimuli were not attended, these stimuli produced no priming effects. Hence, the authors found no evidence that unattended stimuli can be identified. The results support a modern version of Broadbent's selective theory, updated to reflect recent research advances. 2004 APA.

    Beschreibung: We use spatially continuous satellite data as a correlate of precipitation within tropical Africa and show that the majority of documented Ebola hemorrhagic fever outbreaks were closely associated with sharply drier conditions at the end of the rainy season. We propose that these trigger events may enhance transmission of Ebola virus from its cryptic reservoir to humans. These findings suggest specific directions to help understand the sylvatic cycle of the virus and may provide early warning tools to detect possible future outbreaks of this enigmatic disease.

    Beschreibung: In this paper we analyze the effects of the following phenomena associated with the thermocapillary migration of a drop. The first is the influence of Newtonian surface rheology of the interface and the second is that of the energy changes associated with stretching and shrinkage of the interfacial area elements, when the drop is in motion. The former occurs because of dissipative processes in the interfacial region, such as when surfactant molecules are adsorbed at the interface in sufficient concentration. The interface is typically modeled in this instance by ascribing to it a surface viscosity. This is a different effect from that of interfacial tension gradients arising from surfactant concentration gradients. The stretching and shrinkage of interfacial area elements leads to changes in the internal energy of these elements that affects the transport of energy in the fluids adjoining the interface. When an element on the interface is stretched, its internal energy increases because of the increase in its area. This energy is supplied by the neighboring fluids that are cooled as a consequence. Conversely, when an element on the interface shrinks, the adjoining fluids are warmed. In the case of a moving drop, elements of interfacial area are stretched in the forward half of the drop, and are shrunk in the rear half. Consequently, the temperature variation on the surface of the drop and its migration speed are modified. The analysis of the motion of a drop including these effects was first performed by LeVan in 1981, in the limit when convective transport of momentum and energy are negligible. We extend the analysis of LeVan to include the convective transport of momentum by demonstrating that an exact solution of the momentum equation is obtained for an arbitrary value of the Reynolds number. This solution is then used to calculate the slightly deformed shape of the drop from a sphere.

    Beschreibung: In order to determine the effect of the number of sonic boom occurrences on annoyance, a computer-based system was developed for studying the subjective response of people to the occurrence of simulated sonic booms in their homes. The system provided a degree of control over the noise exposure not found in community surveys and a degree of situational realism not available in the laboratory. A system was deployed for eight weeks in each of 33 homes. Each day from 4 to 63 sonic booms were played as the test subject went about his or her normal activities. At the end of the day, the test subjects rated their annoyance to the sonic booms heard during the day. The sonic booms consisted of different combinations of waveforms, levels, and occurrence rates. The experiment confirmed that the increase in annoyance resulting from multiple occurrences can be modeled by the addition of the term "10 * log(number of occurrences)" to the sonic boom level. Of several noise metrics considered, perceived level was the best annoyance predictor. Comparisons of the subjective responses to the different sonic boom waveforms found no differences that were not accounted for by the noise metrics.

    Beschreibung: A new optical scheme to detect glucose concentration in the aqueous humor of the eye is presented. The ultimate aim is to apply this technique in designing a new instrument for, routinely and frequently, noninvasively monitoring blood glucose levels in diabetic patients without contact (no index matching) between the eye and the instrument. The optical scheme exploits the Brewster reflection of circularly polarized light off of the lens of the eye. Theoretically, this reflected linearly polarized light on its way to the detector is expected to rotate its state of polarization, owing to the presence of glucose molecules in the aqueous humor of a patient's eye. An experimental laboratory setup based on this scheme was designed and tested by measuring a range of known concentrations of glucose solutions dissolved in water.     Beschreibung: A round-spore-forming Bacillus species that produces an exosporium was isolated from the surface of the Mars Odyssey spacecraft. This novel species has been characterized on the basis of phenotypic traits, 16S rDNA sequence analysis and DNA-DNA hybridization. According to the results of these analyses, this strain belongs to the genus Bacillus and is a Gram-positive, aerobic, rod-shaped, endospore-forming eubacterium. Ultrathin sections of the spores showed the presence of an exosporium, spore coat, cortex and core. 16S rDNA sequence similarities between this strain, Bacillus fusiformis and Bacillus silvestris were approximately 96% and DNA-DNA reassociation values with these two bacilli were 23 and 17%, respectively. Spores of the novel species were resistant to desiccation, H2O2 and UV and gamma radiation. Of all strains tested, the spores of this strain were the most consistently resistant and survived all of the challenges posed, i.e. exposure to conditions of desiccation (100% survival), H2O2 (26% survival), UV radiation (10% survival at 660 J m(-2)) and gamma radiation (0.4% survival). The name proposed for this novel bacterium is Bacillus odysseyi sp. nov.; the type strain is 34hs-1T (=ATCC PTA-4993T=NRRL B-30641T=NBRC 100172T).

    Beschreibung: The spectrum and light curve of the bright X-ray source CG X-1 in the field of the Circinus galaxy are reexamined. Previous analyses have concluded that the source is an accreting black hole of mass 〉 or approx. 50 solar masses although it has been noted that the light curve resembles that of an AM Herculis system. Here we show that the short period and an assumed main-sequence companion constrain the mass of the companion to less than 1 solar mass. Furthermore, a possible eclipse seen during one of the Chandra observations and a subsequent XMM-Newton observation constrain the mass of the compact object to less than 60 solar masses. If such a system lies in the Circinus galaxy, then the accreting object must either radiate anisotropically or strongly violate the Eddington limit. Even if the emission is beamed, then the companion star that intercepts this flux during eclipse will be driven out of thermal equilibrium and evaporate within approx. 10(exp 3) yr. We find that the observations cannot rule out an AM Herculis system in the Milky Way and that such a system can account for the variations seen in the light curve.

    Beschreibung: Orbital debris impacts on the International Space Station occur frequently. To date, none of the impacting particles has been large enough to penetrate manned pressurized volumes. We used the Manned Spacecraft Crew Survivability code to evaluate the risk to crew of penetrations of pressurized modules at two assembly stages: after Flight 1J, when the pressurized elements of Kibo, the Japanese Experiment Module, are present, and after Flight 1E, when the European Columbus Module is present. Our code is a Monte-Carlo simulation of impacts on the Station that considers several potential event types that could lead to crew loss. Among the statistics tabulated by the program is the probability of death of one or more crew members in the event of a penetration, expressed as the risk factor, R. This risk factor is dependent on details of crew operations during both ordinary circumstances and decompression emergencies, as well as on details of internal module configurations. We conducted trade studies considering these procedure and configuration details to determine the bounds on R at the 1J and 1E stages in the assembly sequence.     Beschreibung: Obtaining uniform mechano-dwarfing of Arabidopsis thaliana (L.) Heynh. seedlings within dense plantings is problematic. Alternative forms of mechano-stimulation were applied to seedlings in effort to obtain uniform growth reduction compared with undisturbed controls in both greenhouse and controlled growth environments. Arabidopsis grown under low photosynthetic photon flux (PPF) artificial light grew upright with limited leaf expansion, which enhanced mechano-responsiveness compared to that of rosette-growing plants under filtered sunlight or high PPF artificial light. Hypocotyls of seedlings grown at PPFs 〉 60 micromoles m-2 s-1 elongated less and had 6% less sensitivity to mechanical stress than seedlings grown at PPFs 〈 60 micromoles m-2 s-1. Fluorescent lamps alone (F) or fluorescent plus incandescent (F+I) lamps were compared for seedling responses to mechanical stress. Under F lighting, hypocotyl elongation was reduced 25% to 40% by twice-daily brush or plate treatments, and brushed seedlings exhibited more growth reduction than did plate treatments. Seedlings grown under F+I lamps exhibited similar stress-induced growth reduction compared to seedlings grown under F only, but stressed F+I seedlings lodged to a greater extent due to excessive hypocotyl elongation. Temperature-response studies using standardized F-only lighting indicated increased hypocotyl elongation but decreased leaf expansion, and decreased mechano-responsivity to brushing over the temperature range from 20 to 28 degrees C. Daylength studies indicated similar degrees of mechano-inhibition of hypocotyl elongation over the daylength range of 12, 16, 20, and 24 hours, whereas fresh weight of stressed seedling shoots declined compared to controls. A combination of environmental growth parameters that give repeatable, visual mechanical dwarfing of Arabidopsis include low-PPF fluorescent lighting from 55 to 60 micromoles m-2 s-1, ambient temperatures from 22 to 25 degrees C, and twice-daily brush treatments.

    Beschreibung: Described is the Liulin-5 experiment and instrumentation, developed for investigation of the space radiation doses depth distribution in a human phantom on the Russian Segment of the International Space Station (ISS). Liulin-5 experiment is a part of the international project MATROSHKA-R on ISS. The experiment MATROSHKA-R is aimed to study the depth dose distribution at the sites of critical organs of the human body, using models of human body-anthropomorphic and spherical tissue-equivalent phantoms. The aim of Liulin-5 experiment is long term (4-5 years) investigation of the radiation environment dynamics inside the spherical tissue-equivalent phantom, mounted in different places of the Russian Segment of ISS. Energy deposition spectra, linear energy transfer spectra, flux and dose rates for protons and the biologically-relevant heavy ion components of the galactic cosmic radiation will be measured simultaneously with near real time resolution at different depths of the phantom by a telescope of silicon detectors. Data obtained together with data from other active and passive dosimeters will be used to estimate the radiation risk to the crewmembers, verify the models of radiation environment in low Earth orbit, validate body transport model and correlate organ level dose to skin dose. Presented are the test results of the prototype unit. The spherical phantom will be flown on the ISS in 2004 year and Liulin-5 experiment is planned for 2005 year. c2004 COSPAR.     Beschreibung: Space flight results in the exposure of astronauts to a mixed field of radiation composed of energetic particles of varying energies, and biological indicators of space radiation exposure provides a better understanding of the associated long-term health risks. Current methods of biodosimetry have employed the use of cytogenetic analysis for biodosimetry, and more recently the advent of technological progression has led to advanced research in the use of genomic and proteomic expression profiling to simultaneously assess biomarkers of radiation exposure. We describe here the technical advantages of the Luminex(TM) 100 system relative to traditional methods and its potential as a tool to simultaneously profile multiple proteins induced by ionizing radiation.     Beschreibung: In cells irradiation by heavy ions has been hypothesized to produce microlesions, regions of local damage. In cell membranes this damage is thought to manifest itself in the form of holes. The primary evidence for microlesions comes from morphological studies of cell membranes, but this evidence is still controversial, especially since holes also have been observed in membranes of normal, nonirradiated, cells. However, it is possible that damage not associated with histologically discernable disruptions may still occur. In order to resolve this issue, we developed a system for detecting microlesions based on liposomes filled with fluorescent dye. We hypothesized that if microlesions form in these liposomes as the result of irradiation, then the entrapped dye will leak out into the surrounding medium in a measurable way. Polypropylene vials containing suspensions of vesicles composed of either dipalmitoyl phosphatidylcholine, or a combination of egg phosphatidylcholine and cholesterol were irradiated at the Brookhaven National Laboratory using 56Fe ions at 1 GeV/amu. In several cases we obtained a significant loss of the entrapped dye above the background level. Our results suggest that holes may form in liposomes as the result of heavy ion irradiation, and that these holes are large enough to allow leakage of cell internal contents that are at least as large as a 1 nm diameter calcein molecule. c2004 COSPAR.     Beschreibung: Radiation-induced chromosome translocations remain in peripheral blood cells over many years, and can potentially be used to measure retrospective doses or prolonged low-dose rate exposures. However, several recent studies have indicated that some individuals possess clones of cells with balanced chromosome abnormalities, which can result in an overestimation of damage and, therefore, influence the accuracy of dose calculations. We carefully examined the patterns of chromosome damage found in the blood lymphocytes of twelve astronauts, and also applied statistical methods to screen for the presence of potential clones. Cells with clonal aberrations were identified in three of the twelve individuals. These clonal cells were present in samples collected both before and after space flight, and yields are higher than previously reported for healthy individuals in this age range (40-52 years of age). The frequency of clonal damage appears to be even greater in chromosomes prematurely condensed in interphase, when compared with equivalent analysis in metaphase cells. The individuals with clonal aberrations were followed-up over several months and the yields of all clones decreased during this period. Since clonal aberrations may be associated with increased risk of tumorigenesis, it is important to accurately identify cells containing clonal rearrangements for risk assessment as well as biodosimetry.     Beschreibung: Because of the variability of collagen crosslinks, their use as markers for bone resorption is often criticized. We hypothesized that the variability could be reduced by collecting urine for 24 hours (or longer) instead of using single voids, and by not normalizing to creatinine. Urine samples were collected from 22 healthy subjects during two or more 24-hour periods. Each 24-hour pool and each 2nd void of the day were analyzed for N-telopeptide (NTX), pyridinium (PYD), and deoxypyridinoline (DPD) crosslinks. Data were analyzed by using linear regression. For NTX, R2 for the two, 2nd-void samples (n = 38) was 0.55, whereas R2 for the two 24-hour pools was 0.51 or 0.52, expressed per day or per creatinine. For PYD and DPD, R2 for the 2nd-void samples was 0.26 and 0.18, R2 for the 24-hour pools expressed per day was 0.58 and 0.74, and R2 for the 24-hour pools expressed per creatinine was 0.65 and 0.76, respectively. Regression of the 2nd void and the corresponding 24-hour pool, expressed per day, yielded R2 = 0.19, 0.19, and 0.08, for NTX, PYD, and DPD, respectively (n = 76 each). For the 2nd-void sample and its corresponding 24-hour pool, expressed per creatinine, R2 = 0.24, 0.33, and 0.08, respectively. In a separate study, the coefficient of variation for NTX was reduced (P 〈 0.05) when data from more than one 24-hour collection were combined. Thus, the variability inherent in crosslink determinations can be reduced by collecting urine for longer periods. In research studies, the high variability of single-void collections, compounded by creatinine normalization, may alter or obscure findings.

    Beschreibung: Air passenger miles will likely double by year 2020. The altered and restrictive environment in an airliner cabin can influence haematological homeostasis in passengers and crew. Flight-related deep venous thromboemboli (DVT) have been associated with at least 577 deaths on 42 of 120 airlines from 1977 to 1984 (25 deaths/million departures), whereas many such cases go unreported. However, there are four major factors that could influence formation of possible flight-induced DVT: sleeping accommodations (via sitting immobilisation); travellers' medical history (via tissue injury); cabin environmental factors (via lower partial pressure of oxygen and lower relative humidity); and the more encompassing chair-rest deconditioning (C-RD) syndrome. There is ample evidence that recent injury and surgery (especially in deconditioned hospitalised patients) facilitate thrombophlebitis and formation of DVT that may be exacerbated by the immobilisation of prolonged air travel.In the healthy flying population, immobilisation factors associated with prolonged (〉5 hours) C-RD such as total body dehydration, hypovolaemia and increased blood viscosity, and reduced venous blood flow (pooling) in the legs may facilitate formation of DVT. However, data from at least four case-controlled epidemiological studies did not confirm a direct causative relationship between air travel and DVT, but factors such as a history of vascular thromboemboli, venous insufficiency, chronic heart failure, obesity, immobile standing position, more than three pregnancies, infectious disease, long-distance travel, muscular trauma and violent physical effort were significantly more frequent in DVT patients than in controls. Thus, there is no clear, direct evidence yet that prolonged sitting in airliner seats, or prolonged experimental chair-rest or bed-rest deconditioning treatments cause DVT in healthy people.

    Beschreibung: Temperature, pressure, and time have been thought to control the smectite-to-illite (S-I) reaction, an important diagenetic process used for petroleum exploration. We demonstrated that microorganisms can promote the S-I reaction by dissolving smectite through reduction of structural Fe(III) at room temperature and 1 atmosphere within 14 days. This reaction typically requires conditions of 300 degrees to 350 degrees C, 100 megapascals, and 4 to 5 months in the absence of microbial activity. These results challenge the conventional concept of the S-I reaction and of reaction kinetic models.

    Beschreibung: The role of two binocular cues to motion in depth-changing disparity (CD) and interocular velocity difference (IOVD)- was investigated by measuring stereomotion speed discrimination and static disparity discrimination performance (stereoacuity). Speed discrimination thresholds were assessed both for random dot stereograms (RDS), and for their temporally uncorrelated equivalents, dynamic random dot stereograms (DRDS), at relative disparity pedestals of -19, 0, and +19 arcmin. While RDS stimuli contain both CD and IOVD cues, DRDS stimuli carry only CD information. On average, thresholds were a factor of 1.7 higher for DRDS than for RDS stimuli with no clear effect of relative disparity pedestal. Results were similar for approaching and receding targets. Variations in stimulus duration had no significant effect on thresholds, and there was no observed correlation between stimulus displacement and perceived speed, confirming that subjects responded to stimulus speed in each condition. Stereoacuity was equally good for our RDS and DRDS stimuli, showing that the difference in stereomotion speed discrimination performance for these stimuli was not due to any difference in the precision of the disparity cue. In addition, when we altered stereomotion stimulus trajectory by independently manipulating the speeds and directions of its monocular half-images, perceived stereomotion speed remained accurate. This finding is inconsistent with response strategies based on properties of either monocular half-image motion, or any ad hoc combination of the monocular speeds. We conclude that although subjects are able to discriminate stereomotion speed reliably on the basis of CD information alone, IOVD provides a precise additional cue to stereomotion speed perception.

    Beschreibung: To investigate effects of low dose heavy particle radiation to CNS system, we adopted mouse neonatal brain cells in culture being exposed to heavy ions by HIMAC at NIRS and NSRL at BNL. The applied dose varied from 0.05 Gy up to 2.0 Gy. The subsequent biological effects were evaluated by an induction of apoptosis and neuron survival focusing on the dependencies of the animal strains, SCID, B6, B6C3F1, C3H, used for brain cell culture, SCID was the most sensitive and C3H the least sensitive to particle radiation as evaluated by 10% apoptotic criterion. The LET dependency was compared with using SCID and B6 cells exposing to different ions (H, C, Ne, Si, Ar, and Fe). Although no detectable LET dependency was observed in the high LET (55-200 keV/micrometers) and low dose (〈0.5 Gy) regions. The survivability profiles of the neurons were different in the mouse strains and ions. In this report, a result of memory and learning function to adult mice after whole-body and brain local irradiation at carbon ion and iron ion.

    Beschreibung: Measurements of chromosome aberrations in peripheral blood lymphocytes are currently the most sensitive and reliable indicator of radiation exposure that can be used for biological dosimetry. This technique has been implemented recently to study radiation exposures incurred by astronauts during space flight, where a significant proportion of the dose is delivered by high-LET particle exposure. Traditional methods for the assessing of cytogenetic damage in mitotic cells collected at one time point after exposure may not be suitable for measuring high-LET radiation effects due to the drastic cell cycle perturbations and interphase cell death induced by this type of exposure. In this manuscript we review the recent advances in methodology used to study high-LET induced cytogenetic effects and evaluate the use of chemically-induced Premature Chromosome Condensation (PCC) as an alternative to metaphase analysis. Published data on the cytogenetic effects of in vitro exposures of high-LET radiation is reviewed, along with biodosimetry results from astronauts after short or long space missions.

    Beschreibung: We examined the apparent dissociation of perceived length and perceived position with respect to the Muller-Lyer (M-L) illusion. With the traditional (two-chevron) figure, participants made accurate open-loop pointing responses at the endpoints of the shaft, despite the presence of a strong length illusion. This apparently non-Euclidean outcome replicated that of Mack, Heuer, Villardi, and Chambers (1985) and Gillam and Chambers (1985) and contradicts any theory of the M-L illusion in which mislocalization of shaft endpoints plays a role. However, when one of the chevrons was removed, a constant pointing error occurred in the predicted direction, as well as a strong length illusion. Thus, with one-chevron stimuli, perceived length and location were no longer completely dissociated. We speculated that the presence of two opposing chevrons suppresses the mislocalizing effects of a single chevron, especially for figures with relatively short shafts.

    Beschreibung: The National Aeronautics and Space Administration (NASA) has been conducting controlled environment research with potatoes (Solanum tuberosum L.) in recirculating nutrient film technique (NFT)-hydroponic systems as a human life support component during long-duration spaceflight. Standard nutrient solution management approaches include constant pH regulation with nitric acid (HNO3) and daily adjustment of electrical conductivity (EC) equivalent to half-strength modified Hoagland's solution, where nitrate (NO3-) is the sole nitrogen (N) source. Although tuber yields have been excellent with such an approach, N use efficiency indices are expected to be low relative to tuber biomass production. Furthermore, the high amount of N used in NFT-hydroponics, typically results in high inedible biomass, which conflicts with the need to minimize system mass, volume, and expenditure of resources for long-duration missions. More effective strategies of N fertilization need to be developed to more closely match N supply with demand of the crop. Hence, the primary objective of this study was to identify the optimal N management regime and plant N requirement to achieve high yields and to avoid inefficient use of N and excess inedible biomass production. In separate 84-day cropping experiments, three N management protocols were tested. Treatments which decreased NO3(-)-N supply indirectly through lowering nutrient solution EC (Expt. I), or disabling pH control, and/or supplying NH4(+)-N (Expt. III) did not significantly benefit tuber yield, but did influence N use efficiency indices. When supplied with an external 7.5 mM NO3(-)-N for the first 42 days after planting (DAP), lowered to 1.0 mM NO3(-)-N during the final 42 days (Expt. II), plants were able to achieve yields on par with plants which received constant 7.5 mM NO3(-)-N (control). By abruptly decreasing N supply at tuber initiation in Expt. II, less N was taken up and accumulated by plants compared to those which received high constant N (control). However, proportionately more plant accumulated N was used (N use efficiency) to produce tuber biomass when N supply was abruptly lowered at tuber initiation in Expt. II. Hence, a hydroponic nutrient solution N management system may be modified to elicit greater plant N-use while maintaining overall high tuber yield as opposed to achieving high tuber yields through excess N supply and shoot growth.

    Beschreibung: Conducting cell biology experiments in microgravity can be among the most technically challenging events in a biologist's life. Conflicting events of spaceflight include waiting to get manifested, delays in manifest schedules, training astronauts to not shake your cultures and to add reagents slowly, as shaking or quick injection can activate signaling cascades and give you erroneous results. It is important to select good hardware that is reliable. Possible conflicting environments in flight include g-force and vibration of launch, exposure of cells to microgravity for extended periods until hardware is turned on, changes in cabin gases and cosmic radiation. One should have an on-board 1-g control centrifuge in order to eliminate environmental differences. Other obstacles include getting your funding in a timely manner (it is not uncommon for two to three years to pass between notification of grant approval for funding and actually getting funded). That said, it is important to note that microgravity research is worthwhile since all terrestrial life evolved in a gravity field and secrets of biological function may only be answered by removing the constant of gravity. Finally, spaceflight experiments are rewarding and worth your effort and patience.

    Beschreibung: The present work tested whether the relationship between functional traits and inoculum density reflected structural diversity in bacterial communities from a land-use intensification gradient applying a mathematical model. Terminal restriction fragment length polymorphism (T-RFLP) analysis was also performed to provide an independent assessment of species richness. Successive 10-fold dilutions of a soil suspension were inoculated onto Biolog GN(R) microplates. Soil bacterial density was determined by total cell and plate counts. The relationship between phenotypic traits and inoculum density fit the model, allowing the estimation of maximal phenotypic potential (Rmax) and inoculum density (KI) at which Rmax will be half-reduced. Though Rmax decreased with time elapsed since clearing of native vegetation, KI remained high in two of the disturbed sites. The genetic pool of bacterial community did not experience a significant reduction, but the active fraction responding in the Biolog assay was adversely affected, suggesting a reduction in the functional potential. c2004 Federation of European Microbiological Societies. The BTF will support continuous operation, which will allow performance of long-duration experiments and will significantly increase the on-orbit science throughput.

    Beschreibung: Expressions of the gibberellin biosynthesis gene were investigated in a normal upright type and a gravi-response mutant, a weeping type of Japanese flowering cherry (Prunus spachiana), that is unable to support its own weight and elongates downward. A segment of the gibberellin 3 beta-hydroxylase cDNA of Prunus spachiana (Ps3ox), which is responsible for active gibberellin synthesis, was amplified by using real-time RT-PCR. The content of Ps3ox mRNA in the weeping type was much greater than that in the upright type, while the endogenous gibberellin level was much higher in the elongating zone of the weeping type. These results suggest that the amount and distribution of synthesized gibberellin regulate secondary xylem formation, and the unbalanced distribution of gibberellin affects the gravi-response of the Prunus tree.

    Beschreibung: Interfacial thermal resistance results of various nanotube and nanofiber coatings, prepared by chemical vapor deposition (CVD) methods, are reported at relatively low clamping pressures. The five types of samples examined include multi-walled and single-walled nanotubes growth by CVD, multi-walled nanotubes grown by plasma enhanced CVD (PECVD) and carbon nanofibers of differing aspect ratio grown by PECVD. Of the samples examined, only high aspect ratio nanofibers and thermally grown multi-walled nanotubes show an improvement in thermal contact resistance. The improvement is approximately a 60% lower thermal resistance than a bare Si-Cu interface and is comparable to that attained by commercially available thermal interface materials.

    Beschreibung: Molecular analyses were carried out on four preflight and six postflight International Space Station (ISS)-associated potable water samples at various stages of purification, storage, and transport, to ascertain their associated microbial diversities and overall microbial burdens. Following DNA extraction, PCR amplification, and molecular cloning procedures, rDNA sequences closely related to pathogenic species of Acidovorax, Afipia, Brevundimonas, Propionibacterium, Serratia, and others were recovered in varying abundance. Retrieval of sequences arising from the iodine (biocide)-reducing Delftia acidovorans in postflight waters is also of concern. Total microbial burdens of ISS potable waters were derived from data generated by an ATP-based enumeration procedure, with results ranging from 0 to 4.9 x 10(4) cells/ml. Regardless of innate biases in sample collection and analysis, such circumstantial evidence for the presence of viable, intact pathogenic cells should not be taken lightly. Implementation of new cultivation approaches and/or viability-based assays are requisite to confirm such an occurrence.

    Beschreibung: Phase-contrast X-ray diffraction imaging and high-angular-resolution diffraction combined with phase-contrast radiographic imaging were employed to characterize defects and perfection of a uniformly grown tetragonal lysozyme crystal in the symmetric Laue case. The full-width at half-maximum (FWHM) of a 4 4 0 rocking curve measured from the original crystal was approximately 16.7 arcsec and imperfections including line defects, inclusions and other microdefects were observed in the diffraction images of the crystal. The observed line defects carry distinct dislocation features running approximately along the 〈1 1 0〉 growth front and have been found to originate mostly in a central growth area and occasionally in outer growth regions. Inclusions of impurities or formations of foreign particles in the central growth region are resolved in the images with high sensitivity to defects. Slow dehydration led to the broadening of a fairly symmetric 4 4 0 rocking curve by a factor of approximately 2.6, which was primarily attributed to the dehydration-induced microscopic effects that are clearly shown in X-ray diffraction images. The details of the observed defects and the significant change in the revealed microstructures with drying provide insight into the nature of imperfections, nucleation and growth, and the properties of protein crystals.

    Beschreibung: This study examined how task switching is affected by hierarchical task organization. Traditional task-switching studies, which use a constant temporal and spatial distance between each task element (defined as a stimulus requiring a response), promote a flat task structure. Using this approach, Experiment 1 revealed a large switch cost of 238 ms. In Experiments 2-5, adjacent task elements were grouped temporally and/or spatially (forming an ensemble) to create a hierarchical task organization. Results indicate that the effect of switching at the ensemble level dominated the effect of switching at the element level. Experiments 6 and 7, using an ensemble of 3 task elements, revealed that the element-level switch cost was virtually absent between ensembles but was large within an ensemble. The authors conclude that the element-level task repetition benefit is fragile and can be eliminated in a hierarchical task organization.

    Beschreibung: Ground-based analogs of spaceflight are an important means of studying physiologic and nutritional changes associated with space travel, and the NASA Extreme Environment Mission Operations V (NEEMO) is such an analog. To determine whether saturation diving has nutrition-related effects similar to those of spaceflight, we conducted a clinical nutritional assessment of the NEEMO crew (4 men, 2 women) before, during, and after their 14-d saturation dive. Blood and urine samples were collected before, during, and after the dive. The foods consumed by the crew were typical of the spaceflight food system. A number of physiologic changes were observed, during and after the dive, that are also commonly observed during spaceflight. Hemoglobin and hematocrit were lower (P 〈 0.05) after the dive. Transferrin receptors were significantly lower immediately after the dive. Serum ferritin increased significantly during the dive. There was also evidence indicating that oxidative damage and stress increased during the dive. Glutathione peroxidase and superoxide dismutase decreased during and after the dive (P 〈 0.05). Decreased leptin during the dive (P 〈 0.05) may have been related to the increased stress. Subjects had decreased energy intake and weight loss during the dive, similar to what is observed during spaceflight. Together, these similarities to spaceflight provide a model to use in further defining the physiologic effects of spaceflight and investigating potential countermeasures.

However, the accuracy of ultrasound by physicians with minimal ultrasound training is unknown. METHODS: After one hour of standardized training, physicians with minimal US experience clinically evaluated patients presenting with pain and trauma to the upper arm or leg. The investigators then performed a long-bone US evaluation, recording their impression of fracture presence or absence. Results of the examination were compared with routine plain or computer aided radiography (CT). RESULTS: 58 patients were examined. The sensitivity and specificity of US were 92.9% and 83.3%, and of the physical examination were 78.6% and 90.0%, respectively. US provided improved sensitivity with less specificity compared with physical examination in the detection of fractures in long bones. CONCLUSION: Ultrasound scans by minimally trained clinicians may be used to rule out a long-bone fracture in patients with a medium to low probability of fracture.

    Beschreibung: This paper considers the updating of Bayesian posterior densities for pharmacokinetic models associated with patients having changing parameter values. For estimation purposes it is proposed to use the Interacting Multiple Model (IMM) estimation algorithm, which is currently a popular algorithm in the aerospace community for tracking maneuvering targets. The IMM algorithm is described, and compared to the multiple model (MM) and Maximum A-Posteriori (MAP) Bayesian estimation methods, which are presently used for posterior updating when pharmacokinetic parameters do not change. Both the MM and MAP Bayesian estimation methods are used in their sequential forms, to facilitate tracking of changing parameters. Results indicate that the IMM algorithm is well suited for tracking time-varying pharmacokinetic parameters in acutely ill and unstable patients, incurring only about half of the integrated error compared to the sequential MM and MAP methods on the same example.

    Beschreibung: Transient mixing driven by buoyancy occurs through the birth of a symmetric Rayleigh-Taylor morphology (RTM) structure for large length scales. Beyond its critical bifurcation the RTM structure exhibits self-similarity and occurs on smaller and smaller length scales. The dynamics of the RTM structure, its nonlinear growth and internal collision, show that its genesis occurs from an explosive bifurcation which leads to the overlap of resonance regions in phase space. This event shows the coexistence of regular and chaotic regions in phase space which is corroborated with the existence of horseshoe maps. A measure of local chaos given by the topological entropy indicates that as the system evolves there is growth of uncertainty. Breakdown of the dissipative RTM structure occurs during the transition from explosive to catastrophic bifurcation; this event gives rise to annihilation of the separatrices which drives overlap of resonance regions. The global bifurcation of explosive and catastrophic events in phase space for the large length scale of the RTM structure serves as a template for which mixing occurs on smaller and smaller length scales.     Beschreibung: BACKGROUND AND AIMS: The objective of this research was to examine the effects of differences in light spectrum on the stomatal conductance (Gs) and dry matter production of lettuce plants grown under a day/night cycle with different spectra, and also the effects on Gs of short-term exposure to different spectra. METHODS: Lettuce (Lactuca sativa) plants were grown with 6 h dark and 18 h light under four different spectra, red-blue (RB), red-blue-green (RBG), green (GF) and white (CWF), and Gs and plant growth were measured. KEY RESULTS AND CONCLUSIONS: Conductance of plants grown for 23 d under CWF rose rapidly on illumination to a maximum in the middle of the light period, then decreased again before the dark period when it was minimal. However, the maximum was smaller in plants grown under RB, RGB and GF. This demonstrates that spectral quality during growth affects the diurnal pattern of stomatal conductance. Although Gs was smaller in plants grown under RGB than CWF, dry mass accumulation was greater, suggesting that Gs did not limit carbon assimilation under these spectral conditions. Temporarily changing the spectral quality of the plants grown for 23 d under CWF, affected stomatal responses reversibly, confirming studies on epidermal strips. This study provides new information showing that Gs is responsive to spectral quality during growth and, in the short-term, is not directly coupled to dry matter accumulation.

    Beschreibung: We have developed a new method based on the sublimation of adenine from Escherichia coli to estimate bacterial cell counts in natural samples. To demonstrate this technique, several types of natural samples, including beach sand, seawater, deep-sea sediment, and two soil samples from the Atacama Desert, were heated to a temperature of 500 degrees C for several seconds under reduced pressure. The sublimate was collected on a cold finger, and the amount of adenine released from the samples was then determined by high-performance liquid chromatography with UV absorbance detection. Based on the total amount of adenine recovered from DNA and RNA in these samples, we estimated bacterial cell counts ranging from approximately 10(5) to 10(9) E. coli cell equivalents per gram. For most of these samples, the sublimation-based cell counts were in agreement with total bacterial counts obtained by traditional DAPI (4,6-diamidino-2-phenylindole) staining.

    Beschreibung: High frequency electrocardiography analyzing the entire QRS complex in the frequency range of 150 to 250 Hz may prove useful in the detection of coronary artery disease, yet the long-term stability of these waveforms has not been fully characterized. Therefore, we prospectively investigated the reproducibility of the root mean squared voltage, kurtosis, and the presence versus absence of reduced amplitude zones in signal averaged 12-lead high frequency QRS recordings acquired in the supine position one month apart in 16 subjects and one year apart in 27 subjects. Reproducibility of root mean squared voltage and kurtosis was excellent over these time intervals in the limb leads, and acceptable in the precordial leads using both the V-lead and CR-lead derivations. The relative error of root mean squared voltage was 12% month-to-month and 16% year-to-year in the serial recordings when averaged over all 12 leads. Reduced amplitude zones were also reproducible up to a rate of 87% and 81%, respectively, for the month-to-month and year-to-year recordings. We conclude that 12-lead high frequency QRS electrocardiograms are sufficiently reproducible for clinical use.

    Beschreibung: Results of a parametric study of carbon nanotube production by the double-pulse laser oven process are presented. The effect of various operating parameters on the production of single-wall carbon nanotubes (SWCNTs) is estimated by characterizing the nanotube material using analytical techniques, including scanning electron microscopy, transmission electron microscopy, thermo gravimetric analysis and Raman spectroscopy. The study included changing the sequence of the laser pulses, laser energy, pulse separation, type of buffer gas used, operating pressure, flow rate, inner tube diameter, as well as its material, and oven temperature. It was found that the material quality and quantity improve with deviation from normal operation parameters such as laser energy density higher than 1.5 J/cm2, pressure lower than 67 kPa, and flow rates higher than 100 sccm. Use of helium produced mainly small diameter tubes and a lower yield. The diameter of SWCNTs decreases with decreasing oven temperature and lower flow rates.

    Beschreibung: We report on experiments in which a more viscous fluid displaces a less viscous one in a vertical cylindrical tube. These experiments were performed using silicone oils in a vertical pipette of small diameter. The more viscous fluid also had a slightly larger density than the less viscous fluid. In the initial configuration, the fluids were at rest, and the interface was nominally flat. A dye was added to the more viscous fluid for ease of observation of the interface between the fluids. The flow was initiated by pumping the more viscous fluid into the less viscous one. The displacement velocity was such that the Reynolds number was smaller than unity and the Peclet number for mass transfer between the fluids was large compared to unity. For upward displacement of the more viscous fluid from an initially stable configuration, an axisymmetric finger was observed under all conditions. However, a needle-shaped spike was seen to propagate from the main finger in many cases, similar to that observed by Petitjeans and Maxworthy for the displacement of a more viscous fluid by a less viscous one.

    Beschreibung: This work studies pressurization and pressure control of a large liquid hydrogen storage tank. A finite element model is developed that couples a lumped thermodynamic formulation for the vapor region with a complete solution of the Navier-Stokes and energy equations for the flow and temperature fields in the liquid. Numerical results show that buoyancy effects are strong, even in microgravity, and can reposition a vapor bubble that is initially at the center of the tank to a region near the tank wall in a relatively short time. Long-term tank pressurization with the vapor bubble at the tank wall shows that after an initial transient lasting about a week, the final rate of pressure increase agrees with a purely thermodynamic analysis of the entire tank. However, the final pressure levels are quite different from thermodynamic predictions. Numerical results also show that there is significant thermal stratification in the liquid due to the effects of natural convection. A subcooled jet is used to provide simultaneous cooling and mixing in order to bring the tank pressure back down to its initial value. Three different jet speeds are examined. Although the lowest jet speed is ineffective at controlling the pressure because of insufficient penetration into the liquid region, the highest jet speed is shown to be quite effective at disrupting thermal stratification and reducing the tank pressure in reasonable time.

    Beschreibung: Richtmyer-Meshkov (RM) instability occurs when a planar interface separating two fluids of different density is impulsively accelerated in the direction of its normal. It is one of the most fundamental fluid instabilities and is of importance to the fields of astrophysics and inertial confinement fusion. Because RM instability experiments are normally carried out in shock tubes, where the generation of a sharp, well-controlled interface between gases is difficult, there is a scarcity of good experimental results. The experiments presented here use a novel technique that circumvents many of the experimental difficulties that have previously limited the study of RM instability in shock tubes. In these experiments, the instability is generated incompressibly, by bouncing a rectangular tank containing two liquids off of a fixed spring. These experiments, which utilize PLIF flow visualization, yield time-motion image sequences of the nonlinear development and transition to turbulence of the instability that are of a quality unattainable in shock tube experiments. Measurements obtained from these images, therefore, provide benchmark data for the evaluation of nonlinear models for the late-time growth of the instability. Because the run time in these experiments is limited, new experiments in the NASA Glenn 2.2 second drop tower, capable of achieving longer run times, are currently under way.

    Beschreibung: Asthma is a chronic disease that can result in exacerbations leading to urgent care in emergency departments (EDs) and hospitals. We examined seasonal and temporal trends in pediatric asthma ED (1997-1999) and hospital (1986-1999) admission data so as to identify periods of increased risk of urgent care by age group, gender, and race. All pediatric ED and hospital admission data for Maryland residents occurring within the state of Maryland were evaluated. Distinct peaks in pediatric ED and hospital asthma admissions occurred each year during the winter-spring and autumn seasons. Although the number and timing of these peaks were consistent across age and racial groups, the magnitude of the peaks differed by age and race. The same number, timing, and relative magnitude of the major peaks in asthma admissions occurred statewide, implying that the variables affecting these seasonal patterns of acute asthma exacerbations occur statewide. Similar gross seasonal trends are observed worldwide. Although several environmental, infectious, and psychosocial factors have been linked with increases in asthma exacerbations among children, thus far they have not explained these seasonal patterns of admissions. The striking temporal patterns of pediatric asthma admissions within Maryland, as described here, provide valuable information in the search for causes.

    Beschreibung: Physiological and molecular phylogenetic approaches were used to investigate variation among 12 cyanobacterial strains in their tolerance of sulfide, an inhibitor of oxygenic photosynthesis. Cyanobacteria from sulfidic habitats were found to be phylogenetically diverse and exhibited an approximately 50-fold variation in photosystem II performance in the presence of sulfide. Whereas the degree of tolerance was positively correlated with sulfide levels in the environment, a strain's phenotype could not be predicted from the tolerance of its closest relatives. These observations suggest that sulfide tolerance is a dynamic trait primarily shaped by environmental variation. Despite differences in absolute tolerance, similarities among strains in the effects of sulfide on chlorophyll fluorescence induction indicated a common mode of toxicity. Based on similarities with treatments known to disrupt the oxygen-evolving complex, it was concluded that sulfide toxicity resulted from inhibition of the donor side of photosystem II.

    Beschreibung: Bacillus anthracis, the causative agent of the human disease anthrax, Bacillus cereus, a food-borne pathogen capable of causing human illness, and Bacillus thuringiensis, a well-characterized insecticidal toxin producer, all cluster together within a very tight clade (B. cereus group) phylogenetically and are indistinguishable from one another via 16S rDNA sequence analysis. As new pathogens are continually emerging, it is imperative to devise a system capable of rapidly and accurately differentiating closely related, yet phenotypically distinct species. Although the gyrB gene has proven useful in discriminating closely related species, its sequence analysis has not yet been validated by DNA:DNA hybridization, the taxonomically accepted "gold standard". We phylogenetically characterized the gyrB sequences of various species and serotypes encompassed in the "B. cereus group," including lab strains and environmental isolates. Results were compared to those obtained from analyses of phenotypic characteristics, 16S rDNA sequence, DNA:DNA hybridization, and virulence factors. The gyrB gene proved more highly differential than 16S, while, at the same time, as analytical as costly and laborious DNA:DNA hybridization techniques in differentiating species within the B. cereus group.

    Beschreibung: The three-dimensional, atomic-resolution protein structures produced by X-ray crystallography over the past 50+ years have led to tremendous chemical understanding of fundamental biochemical processes. The pace of discovery in protein crystallography has increased greatly with advances in molecular biology, crystallization techniques, cryocrystallography, area detectors, synchrotrons and computing. While the methods used to produce single, well-ordered crystals have also evolved over the years in response to increased understanding and advancing technology, crystallization strategies continue to be rooted in trial-and-error approaches. This review summarizes the current approaches in protein crystallization and surveys the first results to emerge from the structural genomics efforts.

    Beschreibung: Spatial relationships between clinical data for pediatric asthmatics (hospital and emergency department utilization rates), and socioeconomic and urban characteristics in Baltimore City were analyzed with the aim of identifying factors that contribute to increased asthma rates. Socioeconomic variables and urban characteristics derived from satellite data explained 95% of the spatial variation in hospital rates. The proportion of families headed by a single female was the most important variable accounting for 89% of the spatial variation. Evidence suggests that the high rates of hospital admissions and emergency department (ED) visits may partially be due to the difficulty of single parents with limited resources managing their child's asthma condition properly. This knowledge can be used for education towards mitigating ED and hospital events in Baltimore City.

    Beschreibung: Humans reach peak bone mass at age 30. After this point, we lose 1 to 2 percent of bone mass each decade. In the microgravity environment of space, astronauts lose bone mass at an accelerated rate of 1 to 2 percent each month. When astronauts travel to Mars, they may be in space for as long as 3 years. During this time, they may lose about half of their bone mass from weight-bearing bones. This loss may be irreversible. The drastic loss in bone that astronauts experience in space makes them much more vulnerable to fractures. In addition, the corresponding removal of calcium from bone results in higher levels of calcium in the blood, which increases the risk of developing kidney stones. Currently, studies are being conducted which investigate factors governing bone adaptation and mechanotransduction. Bone is constantly adapting in response to mechanical stimuli. Increased mechanical loading stimulates bone formation and suppresses bone resorption. Reduction in mechanical loading caused by bedrest, disuse, or microgravity results in decreased bone formation and possibly increased bone resorption. Osteoblasts and osteoclasts are the two main cell types that participate in bone remodeling. Osteoblasts are anabolic (bone-forming) cells and osteoclasts are catabolic (bone-resorbing) cells. In microgravity, the activity of osteoblasts slows down and the activity of osteoclasts may speed up, causing a loss of bone density. Mechanotransduction, the molecular mechanism by which mechanical stimuli are converted to biochemical signals, is not yet understood. Exposure of cells to fluid flow imposes a shear stress on the cells. Several studies have shown that the shear stress that results from fluid flow induces a cellular response similar to that induced by mechanical loading. Thus, fluid flow can be used as an in vitro model to simulate the mechanical stress that bone cells experience in vivo. Previous in vitro studies have shown that fluid flow induces several responses in osteoblasts, including increased proliferation, osteoblastic differentiation, alkaline phosphatase activity, and production of nitric oxide, prostaglandins, and osteopontin. Several proteins have been implicated in osteoblastic mechanotransduction including Bone Morphogenetic Protein-2 (BMP-2), parathyroid hormone, 1,25-dihydroxyvitamin D3 receptor, osteopontin (OPN), osteoprotegerin (OPG), and alkaline phosphatase (AP). We will characterize relative levels of each protein in mineralizing or non-mineralizing MC3T3 osteoblastic cells that have been exposed to fluid flow compared to non-fluid flow using immunofluorescent staining and two- photon laser microscopy as well as western blotting. Because calcium-mediated pathways are important in osteoblastic signaling, we will transfect MC3T3 cells with cameleon probes for Ca2+ containing YFP and CFP. Results will be analyzed using FRET/FLIM to study differential release of intracellular Ca(2+) in response to fluid flow and conditions inducing matrix mineralization. In addition, we plan to conduct several microarray experiments to determine differential gene expression in MC3T3 cells in response to fluid flow and conditions inducing mineralization.

    Beschreibung: Comets, fine-grained matrices of chondrites, and chondritic interplanetary dust particles (IDPs) are each composed of both crystalline and amorphous silicates. The primitive solar nebula, in which comets and asteroids accreted, was formed from the collapsed core of a Giant Molecular Cloud, that, in turn, condensed from materials present in the interstellar medium (ISM). Despite observations that reveal the presence of crystalline magnesium silicate minerals in the shells of very high mass-loss-rate stars [1,2], typical silicate grains in the ISM are most likely to be amorphous, given their relatively long residence time in such a high radiation environment. An upper limit of ~3% crystalline grains can be derived from their non-detection in spectra of ISM solids [3]. If the vast majority of grains that enter the primitive solar nebula are amorphous, then the observation of crystalline dust in comets and primitive chondrite matrices indicates the action of specific processes required to transform the amorphous starting materials into the crystals that are observed.

    Beschreibung: We discovered a low-mass companion to the M dwarf GJ 164 with the CCD-based imaging system of the Stellar Planet Survey astrometric program. The existence of GJ 164B was confirmed with Hubble Space Telescope NICMOS imaging observations. A high-dispersion spectral observation in V sets a lower limit of Deltam 〉 2.2 mag between the two components of the system. Based on our parallax value of 82 +/- 8 mas, we derive the following orbital parameters: P = 2.04 +/- 0.03 yr, a = 103 +/- 0.03, and M-total 0.265 +/- 0.020 M-circle dot. The component masses are M-A = 0.170 +/- 0.015 M-circle dot and M-B = 0.095 +/- 0/015 M-circle dot. Based on its mass, colors, and spectral properties, GJ 164B has spectral type M6-M8 V.

The time-of-flight spectrometer registered 29 spectra during flyby and measured the first negative ion mass spectra of cometary particles. The dust detectors recorded particles over a broad mass range, 10(exp -11) to 10(exp -4) g. Unexpectedly, the dust distribution along Stardust's flight path was far from uniform, but instead occurred in short 'bursts', suggesting in-flight breakup of fragments ejected from the nucleus. High-resolution, stunning images of the Wild 2 surface show a diverse and complex variety of landforms not seen from comets 1P/Halley and 19P/Borrelly or icy satellites of the outer solar system. Longer-exposure images reveal large numbers of jets projected nearly around the entire perimeter of the nucleus, many of which appear to be highly collimated. A triaxial ellipsoidal fit of the Wild 2 nucleus images yields the principal nucleus radii of 1.65 X 2.00 X2.75 km (+/- 0.05 km). The orientations and source locations on the nucleus surface of 20 highly collimated and partially overlapping jets have been traced. There is every indication that the expected samples were successfully collected from the Wild 2 coma and are poised for a return to Earth on 15 January 2006.

    Beschreibung: The Terrestrial Planet Finder Coronagraph will rely heavily on modeling and analysis throughout its mission lifecycle. Optical modeling is especially important, since the tolerances on the optics as well as scattered light suppression are critical for the mission's success. The high contrast imaging necessary to observe a planet orbiting a distant star requires new and innovative technologies to be developed and tested, and detailed optical modeling provides predictions for evaluating design decisions. It also provides a means to develop and test algorithms designed to actively suppress scattered light via deformable mirrors and other techniques. The optical models are used in conjunction with structural and thermal models to create fully integrated optical/structural/thermal models that are used to evaluate dynamic effects of disturbances on the overall performance of the coronagraph. The optical models we have developed have been verified on the High Contrast Imaging Testbed. Results of the optical modeling verification and the methods used to perform full three-dimensional near-field diffraction analysis are presented.

    Beschreibung: The first high-dynamic-range interferometric mode planned to come on line at the Keck Observatory is mid-infrared nulling. This observational mode, which is based on the cancellation of the on-axis starlight arriving at the win Keck telescopes, will be used to examine nearby stellar systems for the presence of circumstellar exozodiacal emission. This paper describes the system level layout of the Keck Interferometer Nuller (KIN), as well as the final performance levels demonstrated in the laboratory integration and test phase at the Jet Propulsion Laboratory prior to shipment of the nuller hardware to the Keck Observatory in mid-June 2004. On-sky testing and observation with the mid-infrared nuller are slated to begin in August 2004.

    Beschreibung: We studied horizontal eye movements induced by en-bloc yaw rotation, over a frequency range 0.2 - 2.8 Hz, in 10 normal human subjects as th ey monocularly viewed a target located at their near point of focus. We measured gain and phase relationships between eye-in-head velocity and head velocity when the near target was either earth-fixed or head-fixed. During viewing of the earth-fixed near target,median gain was 1.49 (range 1.24 - 1.87) at 0.2 Hz for the group of subjects, but decl ined at higher frequencies, so that at 2.8 Hz median gain was 1.08 (r ange 0.68 - 1.67). During viewing of the head-fixed near target, median gain was 0.03 (range 0.01 - 0.10) at 0.2 Hz for the group of subjec ts, but increased at higher frequencies, so that at 2.8 Hz median gai n was 0.71 (range 0.28 - 0.94). We estimated the vestibular contribution to these responses (vestibulo-ocular reflex gain, Gvor) by applyin g transient head perturbations (peak acceleration〉 1,000 deg's(exp 2) ) during sinusoidal rotation under the two viewing conditions. Median Gvor, estimated 〈 70m after the onset of head perturbation, was 0.98 (range 0.39 - 1.42) while viewing the earth-fixed near target, and 0. 97 (range 0.37 - 1.33) while viewing the head-fixed near target. For the group of subjects, 9 out of 10 subjects showed no sigificant diff erence of Gvor between the two viewing conditions ( p 〉 0.053 ) at all test frequencies. Since Gvor accounted for only approximately 73% of the overall response gain during viewing of the earth-fixed target, we investigated the relative contributions of non-vestibular factors. When subjects viewed the earth-fixed target under strobe illumination , to eliminate retinal image slip information, the gain of compensato ry eye movements declined compared with viewing in ambient room light . During sum-of-sine head rotations, while viewing the earth-fixed target, to minimize contributions from predictive mechanisms, gain also declined Nonetheless, simple superposition of smooth-pursuit tracking of sinusoidal target motion could not fully account for the overall r esponse at higher frequencies, suggesting other non-vestibular contributions. During binocular viewing conditions when vergence angle was s ignificantly greater than monocular viewing (p 〈 0.001), this gain of compensatory eye movements did not show proportional change; indeed, gain could not be correlated with vergence angle during monocular or binocular viewing. We conclude that several separate factors contribute to generate eye rotations during sinusoidal yaw head rotations whi le viewing a near target; these include the VOR, visual-tracking eye movements that utilize retinal image motion, predictive eye movements and, possibly, other unidentified nonvestibular factors. For these experiments, vergence was not an important determinant of response gain .

    Beschreibung: Changes in high frequency QRS components of the electrocardiogram (HF QRS ECG) (150-250 Hz) are more sensitive than changes in conventional ST segments for detecting myocardial ischemia. We investigated the accuracy of 12-lead HF QRS ECG in detecting ischemia during adenosine tetrofosmin myocardial perfusion imaging (MPI). 12-lead HF QRS ECG recordings were obtained from 45 patients before and during adenosine technetium-99 tetrofosmin MPI tests. Before the adenosine infusions, recordings of HF QRS were analyzed according to a morphological score that incorporated the number, type and location of reduced amplitude zones (RAZs) present in the 12 leads. During the adenosine infusions, recordings of HF QRS were analyzed according to the maximum percentage changes (in both the positive and negative directions) that occurred in root mean square (RMS) voltage amplitudes within the 12 leads. The best set of prospective HF QRS criteria had a sensitivity of 94% and a specificity of 83% for correctly identifying the MPI result. The sensitivity of simultaneous ST segment changes (18%) was significantly lower than that of any individual HF QRS criterion (P less than 0.00l). Analysis of 12-lead HF QRS ECG is highly sensitive and specific for detecting ischemic perfusion defects during adenosine MPI stress tests and significantly more sensitive than analysis of conventional ST segments.

    Beschreibung: The recent successful rendezvous of the Stardust spacecraft with comet Wild-2 will be followed by its return of cometary dust to Earth in January 2006. Results from two separate dust impact detectors suggest that the spacecraft collected approximately the nominal fluence of at least 1,000 particles larger than 15 micrometers in size. While constituting only about one microgram total, these samples will be sufficient to answer many outstanding questions about the nature of cometary materials. More than two decades of laboratory studies of stratospherically collected interplanetary dust particles (IDPs) of similar size have established the necessary microparticle handling and analytical techniques necessary to study them. It is likely that some IDPs are in fact derived from comets, although complex orbital histories of individual particles have made these assignments difficult to prove. Analysis of bona fide cometary samples will be essential for answering some fundamental outstanding questions in cosmochemistry, such as (1) the proportion of interstellar and processed materials that comprise comets and (2) whether the Solar System had a O-16-rich reservoir. Abundant silicate stardust grains have recently been discovered in anhydrous IDPs, in far greater abundances (200 5,500 ppm) than those in meteorites (25 ppm). Insight into the more subtle O isotopic variations among chondrites and refractory phases will require significantly higher precision isotopic measurements on micrometer-sized samples than are currently available.

It is about the natural connections that exist between various parts of the system. Therefore, it is mainly cast in terms of those mechanisms and phenomena rather than individual missions in space. However, to give credit, this has only been possible because of the existence of a fleet of deep space missions such as Ulysses, SOHO, and the Voyagers. It is only with them working in concert that a real understanding of the physics can be, and has been achieved. There are fourteen chapters in the book, written by top scientists from around the world. The level of presentation is very high but the authors were given enough space to present understandable introductions, physical discussions, and extensive bibliographies. The book can be of use to average scientists and academicians as well as to specialists.

    Beschreibung: RX J0806.3+1527 is a candidate double degenerate binary with possibly the shortest known orbital period. The source shows an approximately equal to 100% X-ray intensity modulation at the putative orbital frequency of 3.11 mHz (321.5 s). If the system is a detached, ultracompact binary gravitational radiation should drive spin-up with a magnitude of nu(sup dot) approximately 10(exp -16) Hz per second. Efforts to constrain the X-ray frequency evolution to date have met with mixed success, principally due to the sparseness of earlier observations. Here we describe the results of the first phase coherent X-ray monitoring campaign on RX J0806.3+1527 with Chandra. We obtained a total of 70 ksec of exposure in 6 epochs logarithmically spaced over 320 days. With these data we conclusively show that the X-ray frequency is increasing at a rate of 3.77 plus or minus 0.8 x 10(exp -16) Hz per second. Using the ephemeris derived from the new data we are able to phase up all the earlier Chandra and ROSAT data and show they are consistent with a constant nu(sup dot) = 3.63 plus or minus 0.06 x 10(exp -16) Hz per second over the past decade. This value appears consistent with that recently derived by Israel et al. largely from monitoring of the optical modulation, and is in rough agreement with the solutions reported initially by Hakala et al., based on ground-based optical observations. The large and stable nu(sup dot) over a decade is consistent with gravitational radiation losses driving the evolution. An intermediate polar (IP) scenario where the observed X-ray period is the spin period of an accreting white dwarf appears less tenable because the observed nu(sup dot) requires an m(sup dot) approximately equal to 4 x 10 (exp -8) solar mass yr(sup -l), that is much larger than that inferred from the observed X-ray luminosity (although this depends on the uncertain distance and bolometric corrections), and it is difficult to drive such a high m(sup dot) in a binary system with parameters consistent with all the multiwavelength data. If the ultracompact scenario is correct, then the X-ray flux cannot be powered by stable accretion which would drive the components apart, suggesting a new type of energy source (perhaps electromagnetic) may power the X-ray flux.

    Beschreibung: Currently, the best available probe of the early phase of gamma-ray burst (GRB) jet attributes is the prompt gamma-ray emission, in which several intrinsic and extrinsic variables determine GRB pulse evolution. Bright, usually complex bursts have many narrow pulses that are difficult to model due to overlap. However, the relatively simple, long spectral lag, wide-pulse bursts may have simpler physics and are easier to model. In this work we analyze the temporal and spectral behavior of wide pulses in 24 long-lag bursts, using a pulse model with two shape parameters - width and asymmetry - and the Band spectral model with three shape parameters. We find that pulses in long-lag bursts are distinguished both temporally and spectrally from those in bright bursts: the pulses in long spectral lag bursts are few in number, and approximately 100 times wider (10s of seconds), have systematically lower peaks in vF(v), harder low-energy spectra and softer high-energy spectra. We find that these five pulse descriptors are essentially uncorrelated for our long-lag sample, suggesting that at least approximately 5 parameters are needed to model burst temporal and spectral behavior. However, pulse width is strongly correlated with spectral lag; hence these two parameters may be viewed as mutual surrogates. We infer that accurate formulations for estimating GRB luminosity and total energy will depend on several gamma-ray attributes, at least for long-lag bursts. The prevalence of long-lag bursts near the BATSE trigger threshold, their predominantly low vF(v) spectral peaks, and relatively steep upper power-law spectral indices indicate that Swift will detect many such bursts.

    Beschreibung: We present imaging results and source structure analysis of extragalactic radio sources observed using the Very Long Baseline Array (VLBA) at 24 GHz and 43 GHz as part of an ongoing NASA, USNO, NRAO and Bordeaux Observatory collaboration to extend the International Celestial Reference Frame (ICRF) to higher radio frequencies. The K/Q-band image database now includes images of 108 sources at 43 GHz (Q-braid) and images of 230 sources at 24 GHz (K-band). Preliminary analysis of the observations taken to date shows that the sources are generally more compact as one goes from the ICRF frequency of 8.4 GHz to 24 GHz. This result is consistent with the standard theory of compact extragalactic radio sources and suggests that reference frames defined at these higher radio frequencies will be less susceptible to the effects of intrinsic source structure than those defined at lower frequencies.

    Beschreibung: In this paper, we present an optimal open-loop slew trajectory algorithm developed at GSFC for the so-called "Yardstick design" of the James Webb Space Telescope (JWST). JWST is an orbiting infrared observatory featuring a lightweight, segmented primary mirror approximately 6 meters in diameter and a sunshield approximately the size of a tennis court. This large, flexible structure will have significant number of lightly damped, dominant flexible modes. With very stringent requirements on pointing accuracy and image quality, it is important that slewing be done within the required time constraint and with minimal induced vibration in order to maximize observing efficiency. With reaction wheels as control actuators, initial wheel speeds as well as individual wheel torque and momentum limits become dominant constraints in slew performance. These constraints must be taken into account when performing slews to ensure that unexpected reaction wheel saturation does not occur, since such saturation leads to control failure in accurately tracking commanded motion and produces high frequency torque components capable of exciting structural modes. A minimum-time constraint is also included and coupled with reaction wheel limit constraints in the optimization to minimize both the effect of the control torque on the flexible body motion and the maneuver time. The optimization is on slew command parameters, such as maximum slew velocity and acceleration, for a given redundant reaction wheel configuration and is based on the dynamic interaction between the spacecraft and reaction wheel motion. Analytical development of the slew algorithm to generate desired slew position, rate, and acceleration profiles to command a feedback/feed forward control system is described. High-fidelity simulation and experimental results are presented to show that the developed slew law achieves the objectives.

    Beschreibung: We introduce a million-second observation of the supernova remnant Cassiopeia A with the Chandra X-ray Observatory. The bipolar structure of the Si-rich ejecta (NE jet and SW counterpart) is clearly evident in the new images, and their chemical similarity is confirmed by their spectra. These are most likely due to jets of ejecta as opposed to cavities in the circumstellar medium, since we can reject simple models for the latter. The properties of these jets and the Fe-rich ejecta will provide clues to the explosion of Cas A.

    Beschreibung: We report on our Chandra and RXTE observations of the bright old nova, V603 Aql, performed in 2001 April, supplemented by our analysis of archival X-ray data on this object. We find that the RXTE data are contaminated by the Galactic Ridge X-ray emission. After accounting for this effect, we find a high level of aperiodic variability in the RXTE data, at a level consistent with the uncontaminated Chandra data. The Chandra HETG spectrum clearly originates in a multi-temperature plasma. We constrain the possible emission measure distribution of the plasma through a combination of global and local fits. The X-ray luminosity and the spectral shape of V603 Aql resemble those of SS Cyg in transition between quiescence and outburst. The fact that the X-ray flux variability is only weakly energy dependent can be interpreted by supposing that the variability is due to changes in the maximum temperature of the plasma. The plasma density is likely to be high, and the emission region is likely to be compact. Finally, the apparent overabundance of Ne is consistent with V603 Aql being a young system.

    Beschreibung: Ultraluminous X-ray sources (ULXs) with apparent luminosities up to hundreds of times the Eddington luminosity for a neutron star have been discovered in external galaxies. The existence of intermediate-mass black holes has been proposed to explain these sources. We present evidence for an intermediate-mass black hole in the ULX M82 X-1 based on the spectral features and timing (quasi-periodic oscillation [QPO]) properties of the X-radiation from this source. We revisited XMM-Newton and Rossi X-Ray Timing Explorer (RXTE) data for M82 X-1 obtained in 2001 and 1997 for XMM and RXTE, respectively. We show for these observations that the source is either in transition or in a high/soft state with photon spectral indices 2.1 and 2.7, respectively. We confirm the early determination of the QPO frequency nu approx. = 55 mHz in this source by Strohmayer & Mushotzky and identify this as the low-frequency QPO for the source. We apply a new method to determine the black hole mass of M82 X-1. The method uses the index-QPO low-frequency correlation that has been recently established in Galactic black hole candidates GRS 1915+105, XTE JI550-564, 4U 1630-47, and others. Using scaling arguments and the correlation derived from the consideration of Galactic black holes, we conclude that M82 X-1 is an intermediate black hole with a mass of the order of 1000 solar mass,.

    Beschreibung: Interstellar glycolaldehyde (CH20HCHO) has been detected with the 100-m Green Bank Telescope (GBT) toward the star-forming region Sagittarius B2(N) by means of the 1(sub 10)-1(sub 01),2(sub 11)-2(sub 02),3(sub 12)-3(sub 0), and 4(sub 13)-4(sub 04) rotational transitions at 13.48, 15.18, 17.98, and 22.14 GHz, respectively. An analysis of these four high signal- to-noise rotational transitions yields a glycolaldehyde state temperature of ~8 K. Previously reported emission line detections of glycolaldehyde with the NRAO 12-m telescope at mm-wavelengths (71 GHz to 103 GHz) are characterized by a state temperature of -50 K. By comparison the GBT detections are surprisingly strong and seen in emission at 13.48 GHz, emission and absorption at 15.18 GHz, and absorption at 17.98 GHz and 22.14 GHz. We attribute the strong absorption observed by the GBT at the higher frequencies to the correspondingly smaller GBT beams coupling better to the continuum source(s) in Sagittarius B2(N). A possible model for the two-temperature regions of glycolaldehyde is discussed.

    Beschreibung: An XMM-Newton observation of the luminous Seyfert 1 galaxy 1H 0419-577 in September 2002, when the source was in an extreme low-flux state, found a very hard X-ray spectrum at 1-10 keV with a strong soft excess below approximately 1 keV. Comparison with an earlier XMM-Newton observation when 1H 0419-577 was X-ray bright indicated the dominant spectral variability was due to a steep power law or cool Comptonized thermal emission. Four further XMM-Newton observations, with 1H 0419-577 in intermediate flux states, now support that conclusion, while we also find the variable emission component in intermediate state difference spectra to be strongly modified by absorption in low ionisation matter. The variable soft excess is seen to be an artefact of absorption of the underlying continuum while the core soft emission is attributed to recombination in an extended region of more highly ionised gas. This new analysis underlines the importance of fully accounting for absorption in characterizing AGN X-ray spectra.

    Beschreibung: We have developed a new method based on the sublimation of adenine from Escherichia coli to estimate bacterial cell counts in natural samples. To demonstrate this technique, several types of natural samples including beach sand, seawater, deep-sea sediment, and two soil samples from the Atacama Desert were heated to a temperature of 500 C for several seconds under reduced pressure. The sublimate was collected on a cold finger and the amount of adenine released from the samples then determined by high performance liquid chromatography (HPLC) with UV absorbance detection. Based on the total amount of adenine recovered from DNA and RNA in these samples, we estimated bacterial cell counts ranging from approx. l0(exp 5) to l0(exp 9) E. coli cell equivalents per gram. For most of these samples, the sublimation based cell counts were in agreement with total bacterial counts obtained by traditional DAPI staining. The simplicity and robustness of the sublimation technique compared to the DAPI staining method makes this approach particularly attractive for use by spacecraft instrumentation. NASA is currently planning to send a lander to Mars in 2009 in order to assess whether or not organic compounds, especially those that might be associated with life, are present in Martian surface samples. Based on our analyses of the Atacama Desert soil samples, several million bacterial cells per gam of Martian soil should be detectable using this sublimation technique.

    Beschreibung: High-energy gamma rays are a valuable tool for studying particle acceleration and radiation in the magnetospheres of energetic pulsars. The seven or more pulsars seen by instruments on the Compton Gamma Ray Observatory (CGRO) show that: the light curves usually have double-peak structures (suggesting a broad cone of emission); gamma rays are frequently the dominant component of the radiated power; and all the spectra show evidence of a high-energy turnover. For all the known gamma-ray pulsars, multiwavelength observations and theoretical models based on such observations offer the prospect of gaining a broad understanding of these rotating neutron stars. The Gamma-ray Large Area Space Telescope (GLAST), now in planning for a launch in 2006, will provide a major advance in sensitivity, energy range, and sky coverage.

    Beschreibung: An important component of Milankovitch's astronomical theory of climate change is the precession index. The precession index, along with the Earth's tilt and orbital eccentricity, are believed to be the major controlling factors of climate change in the last few million years. The precession index is e sin omega(sub s) where e is the Earth's orbital eccentricity and omega(sub s) measures how close the Sun is to the Earth at midsummer. When omega(sub s) = 90deg the Sun is close to the Earth during northern summer, and at 270deg it is far from the Earth during northern summer. The precession index varies with time, because both the eccentricity e and the parameter omega(sub s) are constantly changing due to disturbances in the Earth's orbit by other planets, and due to the precession of the Earth, The change is largely periodic, with a period of about 23,000 years.

    Beschreibung: A puzzling feature of the Chandra-detected quasar jets is that their X-ray emission decreases faster along the jet than their radio emission, resulting from an outward-increasing radio-to-X-ray ratio. In some sources this behavior is so extreme that the radio emission peak is located clearly downstream of that of the X-rays. This is a rather unanticipated behavior given that the inverse Compton nature of the X-rays and the synchrotron radio emission are attributed to roughly the same electrons of the jet's nonthermal electron distribution. In this letter we show that this morphological behavior can result from the gradual deceleration of a relativistic flow and that the offsets in peak emission at different wavelengths carry the imprint of this deceleration. This notion is consistent with another recent finding, namely, that the jets feeding the terminal hot spots of powerful radio galaxies and quasars are still relativistic with Lorentz factors GAMMA approximately 2-3. The picture of the kinematics of powerful jets emerging from these considerations is that they remain relativistic as they gradually decelerate from kiloparsec scales to the hot spots, where, in a final collision with the intergalactic medium, they slow down rapidly to the subrelativistic velocities of the hot spot advance speed.
