    Beschreibung: Fluorescence microscopy allows us to observe fluorescently labeled molecules in diverse biological processes and organelle structures within living cells. However, the diffraction limit restricts its spatial resolution to about half of its wavelength, limiting the capability of biological observation at the molecular level. Structured-illumination microscopy (SIM), a type of super-resolution microscopy, doubles the spatial resolution in all three dimensions by illuminating the sample with a patterned excitation light, followed by computer reconstruction. SIM uses a relatively low illumination power compared with other methods of super-resolution microscopy and is easily available for multicolor imaging. SIM has great potential for meeting the requirements of live-cell imaging. Recent developments in diverse types of SIM have achieved higher spatial (~50 nm lateral) and temporal (~100 Hz) resolutions. Here, we review recent advancements in SIM and discuss its application in noninvasive live-cell imaging.

    Beschreibung: One of the most popular super-resolution microscopies that breaks the diffraction barrier is stimulated emission depletion (STED) microscopy. As the optical set-up of STED microscopy is based on a laser scanning microscopy (LSM) system, it potentially has several merits of LSM like confocal or two-photon excitation LSM. In this article, we first describe the principles of STED microscopy and then describe the features of our newly developed two-photon excitation STED microscopy. On the basis of our recent results and those of other researchers, we conclude by discussing future research and new technologies in this field.

    Beschreibung: The electrically automated ultrathin sectioning apparatus, which has been developed in recent years, can produce consecutive ultrathin sections with a diamond knife and a gallium ion beam. These newly developed apparatuses, however, have several shortcomings, such as the limited block cutting area, thermal damage to the sample by the focused ion beam and a sample electronic charge. To overcome these faults and for easier scanning electron microscopy three-dimensional fine structural reconstruction, we have developed a new cutting method using a deep ultraviolet laser, which we have named the ‘LANTome (Light Ablation Nanotome)’. Using this method, we confirmed the widening of sectioning areas, shortening of the sectioning time, automatic smoothing of rough surfaces, no sample electronic charge and minimal heat effects on the sample tissue, such as thermal denaturation.

    Beschreibung: Recent advances in nanoscopy, which breaks the diffraction barrier and can visualize structures smaller than the diffraction limit in cells, have encouraged biologists to investigate cellular processes at molecular resolution. Since nanoscopy depends not only on special optics but also on ‘smart’ photophysical properties of photocontrollable fluorescent probes, including photoactivatability, photoswitchability and repeated blinking, it is important for biologists to understand the advantages and disadvantages of fluorescent probes and to choose appropriate ones for their specific requirements. Here, we summarize the characteristics of currently available fluorescent probes based on both proteins and synthetic compounds applicable to nanoscopy and provide a guideline for selecting optimal probes for specific applications.

    Beschreibung: As one of the most powerful tools in the biological investigation of cellular structures and dynamic processes, fluorescence microscopy has undergone extraordinary developments in the past decades. The advent of super-resolution techniques has enabled fluorescence microscopy – or rather nanoscopy – to achieve nanoscale resolution in living specimens and unravelled the interior of cells with unprecedented detail. The methods employed in this expanding field of microscopy, however, are especially prone to the detrimental effects of optical aberrations. In this review, we discuss how super-resolution microscopy techniques based upon single-molecule switching, stimulated emission depletion and structured illumination each suffer from aberrations in different ways that are dependent upon intrinsic technical aspects. We discuss the use of adaptive optics as an effective means to overcome this problem.

    Beschreibung: The acrosome is a Golgi-derived sperm cell organelle enclosed by a continuous acrosomal membrane. The acrosomal membrane complexes with surrounding matrices containing molecules necessary for fertilization; however, the complex of acrosomal membrane and associating matrices (CAMAM) has not been visualized in detail under living conditions. Here, we analyzed the CAMAM at the nanometer level using super-resolution stimulated emission depletion (STED) fluorescence microscopy and equatorin-enhanced green fluorescent protein transgenic mice. The STED images were compared with the corresponding images taken by immunoelectron microscopy. Consequently, the substructure of CAMAM could be differentiated at nanometer-scale resolution by STED microscopy without the need for sectioning. The information obtained in this study will be beneficial not only for understanding the molecular mechanism of fertilization but also for cell imaging under living conditions.

    Beschreibung: Large parts of the northern hemisphere are covered by snow and seasonal frost. Climate warming is affecting spatiotemporal variations of snow and frost, hence influencing snowmelt infiltration, aquifer recharge and river runoff patterns. Measurement difficulties have hampered progress in properly assessing how variations in snow and frost impact snowmelt infiltration. This has led to contradicting findings. Some studies indicate that groundwater recharge response is scale dependent. It is thus important to measure snow and soil frost properties with temporal and spatial scales appropriate to improve infiltration process knowledge. The main aim with this paper is therefore to review ground based methods to measure snow properties (depth, density, water equivalent, wetness, and layering) and soil frost properties (depth, water and ice content, permeability, and distance to groundwater) and to make recommendations for process studies aiming to improve knowledge regarding infiltration in regions with seasonal frost. Ground-based radar (GBR) comes in many different combinations and can, depending on design, be used to assess both spatial and temporal variations in snow and frost so combinations of GBR and tracer techniques can be recommended and new promising methods (auocostics and self potential) are evolving, but the study design must be adapted to the scales, the aims and the resources of the study.

    Beschreibung: Papua New Guinea is blessed with a plethora of enviable natural resources, but at the same time it is also cursed by quite a few natural disasters like volcanic eruptions, earthquakes, landslide, floods, droughts etc. Floods happen to be a natural process of maintaining the health of the rivers and depth of its thalweg; it saves the river from becoming morbid while toning up the fertility of the riverine landscape. At the same time, from human perspective, all these ecological goodies are nullified when flood is construed overwhelmingly as one of the most devastating events in respect to social and economic consequences. The present investigation was tailored to assess the use of multi-criteria decision approach (MCDA) in inland flood risk analysis. Categorization of possible flood risk zones was accomplished using geospatial data sets, like elevation, slope, distance to river, and land use/land cover, which were derived from digital elevation model (DEM) and satellite image, respectively. A pilot study area was selected in the lower part of Markham River in Morobe Province, Papua New Guinea. The study area is bounded by 146°31′ to 146°58′ east and 6°33′ to 6°46′ south; covers an area of 758.30 km2. The validation of a flood hazard risk map was carried out using past flood records in the study area. This result suggests that MCDA within GIS techniques is very useful in accurate and reliable flood risk analysis and mapping. This approach is convenient for the assessment of flood in any region, specifically in no-data regions, and can be useful for researchers and planners in flood mitigation strategies.

    Beschreibung: A pair of hydro-meteorological modeling systems were calibrated and evaluated for the Ayalon basin in central Israel to assess the advantages and limitations of one-way versus two-way coupled modeling systems for flood prediction. The models used included the Hydrological Engineering Center-Hydrological Modeling System (HEC-HMS) model and the Weather Research and Forecasting (WRF) Hydro modeling system. The models were forced by observed, interpolated precipitation from rain-gauges within the basin, and with modeled precipitation from the WRF atmospheric model. Detailed calibration and evaluation was carried out for two major winter storms in January and December 2013. Then, both modeling systems were executed and evaluated in an operational mode for the full 2014/2015 rainy season. Outputs from these simulations were compared to observed measurements from the hydrometric station at the Ayalon basin outlet. Various statistical metrics were employed to quantify and analyze the results: correlation, Root Mean Square Error (RMSE) and the Nash–Sutcliffe (NS) efficiency coefficient. Foremost, the results presented in this study highlight the sensitivity of hydrological responses to different sources of simulated and observed precipitation data, and demonstrate improvement, although not significant, at the Hydrological response, like simulated hydrographs. With observed precipitation data both calibrated models closely simulated the observed hydrographs. The two-way coupled WRF/WRF-Hydro modeling system produced improved both the precipitation and hydrological simulations as compared to the one-way WRF simulations. Findings from this study, as well as previous studies, suggest that the use of two-way atmospheric-hydrological coupling has the potential to improve precipitation and, therefore, hydrological forecasts for early flood warning applications. However, more research needed in order to better understand the land-atmosphere coupling mechanisms driving hydrometeorological processes on a wider variety precipitation and terrestrial hydrologic systems.

    Beschreibung: Polysaccharides account for more than 90% of the content of fungal cell walls, but the mechanism underlying the formation of the architecture of the cell walls, which consist of microfibrils embedded in an amorphous wall matrix, remains unknown. We used electron microscopy to investigate ten different fungal cell-wall polysaccharides to determine whether they could self-assemble into the fibrillar or amorphous component of fungal cell walls in a test tube without enzymes. The ultrastructures formed by precipitating β-1,3-glucan and β-1,6-glucan are different depending on the existence of branching in the molecule. Linear β-1,3-glucan and linear β-1,6-glucan precipitate into a fibrillar ultrastructure. Branched β-1,6-glucan, mannan and glycogen precipitates are amorphous. Branched β-1,3-glucan forms a fibrillar plus amorphous ultrastructure. Self-assembly among combinations of different linear and branched cell-wall polysaccharides results in an ultrastructure that resembles that of a yeast cell wall, which suggests that self-assembly of polysaccharides may participate in the development of the three-dimensional architecture of the yeast cell wall.

    Beschreibung: When considering drug delivery, the amount of drug that can be carried at a particular time and how the drug is incorporated efficiently into cells are important parameters. Transferrin (Tf)-conjugated nanocarriers have been used for the targeted delivery of drugs to cancer cells due to the availability of receptor-mediated clathrin-dependent endocytosis. In general, however, endocytosis seems to differ according to the size and shape of carriers. Large substances are generally internalized into cells by phagocytosis. We studied the internalization mechanism of Tf-conjugated nanoparticles (diameter, 522 nm). Tf-conjugated polystyrene particles were incorporated into cells by receptor-mediated endocytosis with large clathrin-coated vesicles even though their diameter was 〉500 nm and despite that fact that clathrin-coated vesicles have a diameter of 100 nm. This finding suggests that signals for internalization generated by stimulated Tf receptors (TfRs) activate clathrin-mediated endocytosis preferentially. Whether these larger particles could deliver drugs more efficiently than smaller particles was then examined. The toxicity of larger Tf-conjugated biodegradable nanoparticles (poly(lactic-co-glycolic acid)) encapsulating doxorubicin (diameter, 216 ± 38 nm) was appreciably dependent on the number of Tf molecules conjugated on a particle and the number of TfRs expressed on the cell membrane. Larger Tf-conjugated particles delivered drugs to cancer cells expressing many TfRs more selectively than their smaller counterparts (diameter, 56 ± 9 nm) if they were decorated with an appropriate number of Tf molecules.

    Beschreibung: Structural biology relies on good-quality protein crystals in order for structure determination. Many factors affect the growth process of a protein crystal including the way it nucleates and the types of damage and contamination during its growth. Although the nucleation process and quality of a crystal is vital to structure determination, they are both under-studied areas of research. Our research begins to explore ways of measuring the quality of protein crystals, using TEM, thus overcoming the problems associated with viewing wet specimens in a vacuum. Our current understanding of nucleation is that it is a two-step mechanism involving the formation of nuclei from dense liquid clusters; however; it is still unclear whether nuclei first start as amorphous aggregates or as crystalline lattices. Potentially, electron diffraction may be capable of uncovering this process. Using TEM imaging and diffraction of lysozyme as a model protein crystal, we report the internal two-dimensional strain and the density of crystallites in a protein crystal, at a resolution never seen before. The TEM diffraction shows unique features of crystal mosaicity that can be directly correlated to TEM images.

    Beschreibung: The cotyledon of legume seeds is a storage organ that provides nutrients for seed germination and seedling growth. The spatial and temporal control of the degradation processes within cotyledons has not been elucidated. Calcium oxalate (CaOx) crystals, a common calcium deposit in plants, have often been reported to be present in legume seeds. In this study, micro-computed tomography (micro-CT) was employed at the SPring-8 facility to examine the three-dimensional distribution of crystals inside cotyledons during seed maturation and germination of Lotus miyakojimae (previously Lotus japonicus accession Miyakojima MG-20). Using this technique, we could detect the outline of the embryo, void spaces in seeds and the cotyledon venation pattern. We found several sites that strongly inhibited X-ray transmission within the cotyledons. Light and polarizing microscopy confirmed that these areas corresponded to CaOx crystals. Three-dimensional observations of dry seeds indicated that the CaOx crystals in the L. miyakojimae cotyledons were distributed along lateral veins; however, their distribution was limited to the abaxial side of the procambium. The CaOx crystals appeared at stage II (seed-filling stage) of seed development, and their number increased in dry seeds. The number of crystals in cotyledons was high during germination, suggesting that CaOx crystals are not degraded for their calcium supply. Evidence for the conservation of CaOx crystals in cotyledons during the L. miyakojimae germination process was also supported by the biochemical measurement of oxalic acid levels.

    Beschreibung: The electron irradiation damage of MFI-type zeolite was estimated under various accelerating voltages of 100, 200 and 300 kV from successively captured high-resolution transmission electron microscopy (HRTEM) images. To determine the optimal accelerating voltage for HRTEM imaging of electron-sensitive MFI zeolite, the critical dose was estimated from the disappearance of a specific fast Fourier transform spot calculated from experimental images. Based only on the electron dose, a higher voltage was more advantageous. However, taking into account the minimum dose for imaging with a CCD camera, the optimal accelerating voltage for imaging MFI zeolite was 200 kV. The minimum dose for image detection with a CCD camera was surmised from the output/input signal ratio dependence on the accelerating voltage and the contrast range in simulated HRTEM images.

    Beschreibung: The magnetic field generated by a magnetic recording head is evaluated using electron holography. A magnetic recording head, which is connected to an electric current source, is set on the specimen holder of a transmission electron microscope. Reconstructed phase images of the region around the magnetic pole show the change in the magnetic field distribution corresponding to the electric current applied to the coil of the head. A simulation of the magnetic field, which is conducted using the finite element method, reveals good agreement with the experimental observations.

    Beschreibung: A new multilayer-coated varied line-spaced grating, JS4000, was fabricated and tested for extending the upper limit of a grating X-ray spectrometer for electron microscopy. This grating was designed for 2–3.8 keV at a grazing incidence angle of 1.35°. It was revealed that this new multilayer structure enables us to take soft-X-ray emission spectra continuously from 1.5 to 4.3 keV at the same optical setting. The full-width at half maximum of Te-L α1,2 (3.8 keV) emission peak was 27 eV. This spectrometer was applied to indium tin oxide particles and clearly resolved Sn-L α (3444 eV) and In-L β1 (3487 eV) peaks, which could not be resolved by a widely used energy-dispersive X-ray spectrometer.

    Beschreibung: For development of advanced materials, characterization using a scanning transmission electron microscope (STEM) including analysis via X-ray energy-dispersive spectrometry and electron energy-loss spectrometry is essential. Recent advances in aberration-corrected instruments have offered large-scale data acquisition at a high resolution for limited acquisition times both in imaging and in analysis. Further advanced procedures are required to analyze such large-scale datasets more efficiently including quantification. In addition, more simplified tuning procedures are crucial to the best possible resolution in the latest aberration-corrected instruments. In this review article, several approaches to perform advanced electron microscopy, which the author has been developing with his colleague, are described as ‘Microscopy Hacks’. These are (i) quantification and elemental/chemical-imaging procedures, (ii) advanced statistical approaches to handle large-scale datasets and (iii) instrument characterization and tuning procedures including the latest development of an ad hoc autotuning procedure for aberration-corrected STEM imaging.

    Beschreibung: The crystal structure of a new type of molybdenum oxide crystal encapsulated in a single-walled carbon nanotube (CNT) was examined via diffraction and spectroscopic techniques using both X-rays and electron beams. This new type of molybdenum oxide crystal has a chemical bonding state of MoO 3 , as confirmed by X-ray absorption spectroscopy, and the MoO 3 units exhibit axial symmetry, as clarified by electron diffraction from bundled and individual CNTs encapsulating the crystal. To obtain three-dimensional information on the structure, a cross-sectional sample was prepared using a conventional dimple and ion-mill method. High-resolution transmission electron microscopy images exhibit ring-like shapes that originated from the arrangement of the MoO 3 units inside the CNTs, as observed along the tube axis. The units are spaced 0.36 nm from each other in a ring arrangement and the distance between each ring is 0.391 nm.

    Beschreibung: This review summarizes the recent advances in three-dimensional (3D) imaging techniques and their application to polymer nanostructures, for example, microphase-separated structures of block copolymers. We place particular emphasis on the method of transmission electron microtomography (electron tomography for short; hereafter abbreviated as ET). As a result of recent developments in ET, truly quantitative 3D images of polymer nanostructures can now be obtained with subnanometer resolution. The introduction of scanning optics in ET has made it possible to obtain large amounts of 3D data from micrometer-thick polymer specimens by using conventional electron microscopes at a relatively low accelerating voltage, 200 kV. Thus, ET covers structures over a wide range of thicknesses, from a few nanometers to several hundred nanometers, which corresponds to quite an important spatial range for hierarchical polymer nanostructures. ET provides clear 3D images and a wide range of new structural information that cannot be obtained using other methods. Information traditionally derived from conventional microscopy or scattering methods can be directly acquired from 3D volume data. ET is a versatile technique that is not restricted to only polymer applications; it can also be used as a powerful characterization tool in energy applications such as fuel cells.

    Beschreibung: The performances of a newly developed 80–200 kV cold field emission gun (CFEG) transmission electron microscope (TEM) integrating a spherical aberration corrector for a TEM image-forming lens have been evaluated. To begin, we show that the stability of both emission and probe currents makes use of this new CFEG much friendlier. The energy spread of electrons emitted from the CFEG has been measured as a function of emission current and shows a very last 0.26 eV energy resolution at 200 kV and even 0.23 eV at 80 kV. The combination of the CFEG and the CEOS™ aberration corrector, associated with enhanced mechanical and electrical stabilities of this new microscope, allows reaching an information transfer below 75 pm at 200 and 80 pm at 80 kV. This unseen resolution at 200 kV has allowed us to study the structure of CoPt nanoparticles by observing direct images of their atomic arrangement along the high indexes zone axis. We have evidenced the presence of defects in these nanostructures that are not parallel to the electron beam. The precise stoichiometry of two iron oxides, FeO and Fe 2 O 3 , has been determined from an analysis of iron valence state that was obtained from a direct analysis of EELS fine structures spectrum of the two oxides.

    Beschreibung: Data mining from noisy data/images is one of the most important themes in modern science and technology. Statistical image processing is a promising technique for analysing such data. Automation of particle pickup from noisy electron micrographs is essential, especially when improvement of the resolution of single particle analysis requires a huge number of particle images. For such a purpose, reference-based matching using primary three-dimensional (3D) model projections is mainly adopted. In the matching, however, the highest peaks of the correlation may not accurately indicate particles when the image is very noisy. In contrast, the density and the heights of the peaks should reflect the probability distribution of the particles. To statistically determine the particle positions from the peak distributions, we have developed a density-based peak search followed by a peak selection based on average peak height, using multi-reference alignment (MRA). Its extension, using multi-reference multiple alignment (MRMA), was found to enable particle pickup at higher accuracy even from extremely noisy images with a signal-to-noise ratio of 0.001. We refer to these new methods as stochastic pickup with MRA (MRA-StoPICK) or with MRMA (MRMA-StoPICK). MRMA-StoPICK has a higher pickup accuracy and furthermore, is almost independent of parameter settings. They were successfully applied to cryo-electron micrographs of Rice dwarf virus. Because current computational resources and parallel data processing environments allow somewhat CPU-intensive MRA-StoPICK and MRMA-StoPICK to be performed in a short period, these methods are expected to allow high-resolution analysis of the 3D structure of particles.

    Beschreibung: The sensory nerve endings of the rat tongue, cheek and palate were studied using immunohistochemical staining and transmission electron microscopy analysis. The specimens were fixed in modified Karnovsky solution and embedded in Spurr resin. Substance P, calcitonin gene-related peptide (CGRP)- and protein gene product 9.5 (PGP b9.5)-containing nerve fibers in the rat tongue, cheek and palate were examined by electronic microscopical analysis and immunohistochemical localization. These fibers run very close to the basal lamina of the epithelium and extend into the filliform and fungiform papillae. Numerous plexiform fibers immunoreactive for substance P, CGRP and PGP 9.5 were found in the connective tissue of mucosa. Electron microscopic observations showed clearly immunostained nerve fibers, which are located very close to the basal lamina of epithelial cells. Some electron-dense granules may be observed in the axoplasms of both substance P and CGRP immunoreactive fibers. Several lamellar corpuscles into the subepithelial connective tissue papillae, Merkel corpuscles and numerous thin unmyelinated and myelinated axons were observed. The terminal axons revealed numerous mitochondria, neurofilaments, microtubules and clear vesicles in the base of axoplasmic protrusions. The lamellar cells showed caveolae and interlamelar spaces filled by amorphous substance. Between the lamellar cells and axoplasmic membrane, and in the adjacent lamellae region, desmosome-type junctions were observed. The quantitative and morphometric analysis showed nerve endings with an average area of 4.83 ± 3.4 μm 2 and 19.4 internal mitochondria in this site and the organized corpuscles with an average area of 79.24 ± 27.24 μm 2 and 24.23 internal mitochondria in this place. All the structures observed are involved in the transmission of pain and mechanoreceptors stimulus of these oral mucosae.

    Beschreibung: The F-actin cytoskeleton of Cryptococcus neoformans is known to comprise actin cables, cortical patches and cytokinetic ring. Here, we describe a new F-actin structure in fungi, a perinuclear F-actin collar ring around the cell nucleus, by fluorescent microscopic imaging of rhodamine phalloidin-stained F-actin. Perinuclear F-actin rings form in Cryptococcus neoformans treated with the microtubule inhibitor Nocodazole or with the drug solvent dimethyl sulfoxide (DMSO) or grown in yeast extract peptone dextrose (YEPD) medium, but they are absent in cells treated with Latrunculin A. Perinuclear F-actin rings may function as ‘funicular cabin’ for the cell nucleus, and actin cables as intracellular ‘funicular’ suspending nucleus in the central position in the cell and moving nucleus along the polarity axis along actin cables.

    Beschreibung: The low-energy Ar-ion milling method was used to prepare ultrathin specimens for transmission electron microscope observation. The samples were thinned initially by a usual focused ion beam technique or typical Ar-ion milling with a high energy of 2–10 keV and were thinned additionally by an Ar-ion beam with an energy less than 1 keV, typically 500–900 eV. This low-energy ion beam was scanned over the specimen, and secondary electrons induced by the ion beam could be detected to form secondary electron images with a resolution of a few micrometre. Because a desired area can be selected and thinned by the low-energy ion beam, redeposition or cross contamination from irradiation of a metal grid that supports the sample can be prevented. It was shown that the low-energy Ar-ion beam thins a surface amorphous damage layer preferentially and effectively rather than a crystal specimen. Images from ultrathin specimens of two different materials revealed a detailed structure.

    Beschreibung: Most models evaluated by the Intergovernmental Panel for Climate change estimate projected increases in temperature and precipitation with rising atmospheric CO2 levels. Researchers have suggested that increases in CO2 and associated increases in temperature and precipitation may stimulate vegetation growth and increase evapotranspiration (ET), which acts as a cooling mechanism, and on a global scale, may slow the climate-warming trend. This hypothesis has been modeled under increased CO2 conditions with models of different vegetation-climate dynamics. The significance of this vegetation negative feedback, however, has varied between models. Here we conduct a century-scale observational analysis of the Eastern US water balance to determine historical evapotranspiration trends and whether vegetation greening has affected these trends. We show that precipitation has increased significantly over the twentieth century while runoff has not. We also show that ET has increased and vegetation growth is partially responsible.

    Beschreibung: Ptychographic iterative engine (PIE), a form of coherent diffractive imaging in which the phase of both low-angle and high-angle diffraction data is used to form an image without a lens, can in principle deliver wavelength-limited resolution. Working at low accelerating voltages (30 keV) reduces specimen knock-on damage, increases high-resolution contrast and allows a scanning electron microscope to be used as a transmission electron microscopy. However, electrons are very strongly scattered at low energy. To that end, the effect of dynamical scattering on PIE at low accelerating voltages is calculated to illustrate the possibility of generating high-resolution images with low energy electrons.

    Beschreibung: The application of multispectral imaging to discriminate myelinated and demyelinated areas of neural tissue is herein presented. The method is applied through a custom-made, multispectral imaging monochromator, coupled to a commercially available microscope. In the present work, a series of spinal cord sections were analysed derived from mice with experimental autoimmune encephalomyelitis (EAE), an experimental model widely used to study multiple sclerosis (MS). The multispectral microscope allows imaging of local areas with loss of myelin without the need of tissue labelling. Imaging with the aforementioned method and system is compared in a parallel way with conventional methods (wide-field and confocal fluorescence microscopies). The diagnostic sensitivity of our method is 90.4% relative to the ‘gold standard’ method of immunofluorescence microscopy. The presented method offers a new platform for the possible future development of an in vivo , real-time, non-invasive, rapid imaging diagnostic tool of spinal cord myelin loss-derived pathologies.

    Beschreibung: Advancement of microscopic technologies established significant progress in our understanding of the brain. In the recent effort to elucidate the complete wiring map of the brain circuitry termed ‘connectome’, the different modalities of imaging technology, including those of light and electron microscopy, have started providing essential contribution in multiple organisms. The contribution would be impossible without the recent innovation in both acquisition and analyses of the big connectomic data. The current data demonstrated complicated networks with unidirectional and reciprocal connections of the cerebral circuits at the macroscopic and light microscopic (‘mesoscopic’) levels, and the unimaginable complexity of synaptic connections between axons and dendrites at the electron microscopic (‘microscopic’) level. At the same time, the data highlighted the necessity to make substantial advancement in methodology of the connectomic studies, including efficient handling and automated analyses of the acquired dataset. Further understanding about structural and functional connectome seems to be facilitated by combinations of the different imaging modalities. Such multidisciplinary approaches will give us the clues to address whether the complete connectome can elucidate fundamental mechanisms processing the basic and higher functions of human brains.

    Beschreibung: The occupation sites and the occupancies of trace dopants in La/Co co-doped Sr-M-type ferrite, SrFe 12 O 19 , were quantitatively and precisely determined by beam-rocking energy-dispersive X-ray spectroscopy (EDXS) on the basis of electron-channeling effects. Because the Co atoms, in particular, should be partially substituted for the five crystallographically inequivalent sites, which could be key parameters in improving the magneto-crystalline anisotropy, it is difficult yet intriguing to discover their occupation sites and occupancies without using the methods of large-scale facilities, such as neutron diffraction and synchrotron radiation. In the present study, we tackled this problem by applying an extended statistical atom location by channeling enhanced microanalysis method, using conventional transmission electron microscopy, EDXS and dynamical electron elastic/inelastic scattering theories. The results show that the key occupation sites of Co were the 2a, 4f 1 and 12k sites. The quantified occupancies of Co were consistent with those of the previous study, which involved a combination of neutron diffraction and extended X-ray absorption fine structure analysis, as well as energetics considerations based on by first-principles calculations.

    Beschreibung: Needle-shaped precipitates in an aged Al–0.62Mg–0.93Si (mass%) alloy were identified using a compositional threshold method, an isoconcentration surface, in atom probe tomography (APT). The influence of thresholds on the morphological and compositional characteristics of the precipitates was investigated. Utilizing optimum parameters for the concentration space, a reliable number density of the precipitates is obtained without dependence on the elemental concentration threshold in comparison with evaluation by transmission electron microscopy (TEM). It is suggested that careful selection of the concentration space in APT can lead to a reasonable average Mg/Si ratio for the precipitates. It was found that the maximum length and maximum diameter of the precipitates are affected by the elemental concentration threshold. Adjustment of the concentration threshold gives better agreement with the precipitate dimensions measured by TEM.

    Beschreibung: Acetazolamide (AZA) reduces the water permeability of aquaporin-4, the predominant water channel in the brain. We determined the structure of aquaporin-4 in the presence of AZA using electron crystallography. Most of the features of the 5-Å density map were consistent with those of the previously determined atomic model. The map showed a protruding density from near the extracellular pore entrance, which most likely represents the bound AZA. Molecular docking simulations supported the location of the protrusion as the likely AZA-binding site. These findings suggest that AZA reduces water conduction by obstructing the pathway at the extracellular entrance without inducing a large conformational change in the protein.

    Beschreibung: To understand the bainitic transformation behavior in Cu–17Al–11Mn (at.%) alloys, dynamic in situ observation during heating was carried out in a scanning electron microscope (SEM). In this study, after optimizing the sample preparation method and observation conditions, we successfully observed the transformation process with sufficient resolution and contrast. From the observation results, bainite is first formed preferentially at the grain boundaries of the parent phase. Bainite is also formed inside the grains to relax the elastic strain generated by the initial bainite. Regarding the growth mode, in the early stage of the transformation, bainite grows along the longitudinal direction, and in the late stage, it grows along the lateral direction. The growth rate of the bainite was also evaluated by continuous observation of the same plate. Dynamic in situ observation of a martensitic transformation in the same alloy was also performed to compare the growth mode with that of bainite, and it was found that the behavior is considerably different between bainitic and the martensitic transformations.

    Beschreibung: Although many studies of the Golgi apparatus structure have been performed by light and electron microscopy, the full shape of the Golgi apparatus remained unclear due to the technical limitations of the previously applied microscopy techniques. In this study, we used serial section scanning electron microscopy (SEM) for the morphological study of the Golgi apparatus. This method is useful for three-dimensional (3D) reconstruction of cellular structures without requiring specialized instruments, unlike focused ion beam SEM (FIB-SEM) and serial block face SEM (SBF-SEM). Using the serial section SEM method developed by our laboratory, we investigate the 3D shape of the osmium-impregnated Golgi apparatus in rat epididymal cells, pancreatic acinar cells and gonadotropes. The combination of serial section SEM and a 3D reconstruction technique enabled us to elucidate the entire shape of the Golgi apparatus in these cells. The full shape of the Golgi apparatus in epididymal cells formed a basket-like structure with oval-shaped cisterns, while the Golgi apparatus in an acinar cell from the pancreas was composed of elongated ribbon-like structures that were connected to each other, making a coarse network. The overall image of the Golgi apparatus cisterns from a gonadotrope looked like a spherical cage. This study has clearly shown that entire 3D shape of the Golgi apparatus varies depending on the cell type and that the Golgi cisterns network appears as a single mass located in the large region of the cytoplasm.

    Beschreibung: The initial determination of left–right asymmetry is an essential process in embryonic development. In mouse embryo, cilia in the node play an important role generating the nodal flow that subsequently triggers left–right determination in the embryo. Although nodal cilia have historically been thought to have a 9 + 0 axonemal configuration, the existence of 9 + 2 cilia has been reported so far. Because the distribution of those two types of cilia within the node has not yet been reported, we assessed the arrangement of 9 + 0 and 9 + 2 cilia in the node. In this study, we concluded that most of the nodal cilia were 9 + 0 in structure and there were much fewer 9 + 2 cilia than 9 + 0 cilia. Furthermore, the two types of cilia were randomly distributed in the node with no regularity. In addition, we studied the embryonic origin of the crown cells surrounding the node to better understand their identity.

    Beschreibung: The ultrastructure of melanin granules in human hair was studied using 1,000 kV high-voltage scanning transmission electron microscopy to successfully reconstruct three-dimensional images of the whole melanin granule. It was revealed that the melanin granule was composed of a membrane-like outer structure that included many spherical vesicles, and an inner matrix containing a sheet-like structure in the elongated direction of the melanin granule and a sheet-like arrays structure in the cross direction. The outer structure of the melanin granule was maintained even after exposure to hair-bleaching agents to decompose the melanin granule, suggesting that the outer structure was a highly robust structure and composition compared with the inner matrix .

    Beschreibung: The study presents a hydrology concept developed to include lateral water flow in the biogeochemical model ForSAFE. The hydrology concept was evaluated against data collected at Svartberget in the Vindeln Research Forest in Northern Sweden. The results show that the new concept allows simulation of a saturated and an unsaturated zone in the soil as well as water flow that reaches the stream comparable to measurements. The most relevant differences compared to streamflow measurements are that the model simulates a higher base flow in winter and lower flow peaks after snowmelt. These differences are mainly caused by the assumptions made to regulate the percolation at the bottom of the simulated soil columns. The capability for simulating lateral flows and a saturated zone in ForSAFE can greatly improve the simulation of chemical exchange in the soil and export of elements from the soil to watercourses. Such a model can help improve the understanding of how environmental changes in the forest landscape will influence chemical loads to surface waters.

    Beschreibung: The dependence of the microstructure of Ge 2 Sb 2 Te 5 (GST) on Ti diffusion into GST by annealing in GST/Ti/TiN phase-change random access memory stack was studied by various transmission electron microscopy (TEM) techniques. The microstructure and crystal structure of GST were identified with high-resolution TEM (HRTEM) and image simulation technique, and the Ti diffusion into GST was revealed by scanning transmission electron microscope-energy-dispersive X-ray spectroscopy analysis. It was observed that Ti atoms of Ti/TiN thin layers were incorporated into GST cell through several thermal annealing steps and they could retard the phase transition from face-centered cubic (FCC) phase into hexagonal close-packed (HCP) phase partially and restrain the increase in grain size. Thus, it is concluded that Ti diffusion can affect the microstructure of GST including the type of the crystal phase and grain size of GST. It was shown that the insertion of diffusion barrier between TiN and GST could block Ti diffusion into GST and make it possible for FCC phase to completely transform into HCP phase.

    Beschreibung: In this study, we introduce scanning electron microscopy (SEM) of semithin resin sections. In this technique, semithin sections were adhered on glass slides, stained with both uranyl acetate and lead citrate, and observed with a backscattered electron detector at a low accelerating voltage. As the specimens are stained in the same manner as conventional transmission electron microscopy (TEM), the contrast of SEM images of semithin sections was similar to TEM images of ultrathin sections. Using this technique, wide areas of semithin sections were also observed by SEM, without the obstruction of grids, which was inevitable for traditional TEM. This study also applied semithin section SEM to correlative light and electron microscopy. Correlative immunofluorescence microscopy and immune-SEM were performed in semithin sections of LR white resin-embedded specimens using a FluoroNanogold-labeled secondary antibody. Because LR white resin is hydrophilic and electron stable, this resin is suitable for immunostaining and SEM observation. Using correlative microscopy, the precise localization of the primary antibody was demonstrated by fluorescence microscopy and SEM. This method has great potential for studies examining the precise localization of molecules, including Golgi- and ER-associated proteins, in correlation with LM and SEM.

    Beschreibung: All-solid-state Li-ion batteries (LIBs) with solid electrolytes are expected to be the next generation devices to overcome serious issues facing conventional LIBs with liquid electrolytes. However, the large Li-ion transfer resistance at the electrode/solid-electrolyte interfaces causes low power density and prevents practical use. In-situ -formed negative electrodes prepared by decomposing the solid electrolyte Li 1+ x +3 z Al x (Ti,Ge) 2– x Si 3 z P 3– z O 12 (LASGTP) with an excess Li-ion insertion reaction are effective electrodes providing low Li-ion transfer resistance at the interfaces. Prior to our work, however, it had still been unclear how the negative electrodes were formed in the parent solid electrolytes. Here, we succeeded in dynamically visualizing the formation by in situ spatially resolved electron energy-loss spectroscopy in a transmission electron microscope mode (SR-TEM-EELS). The Li-ions were gradually inserted into the solid electrolyte region around 400 nm from the negative current-collector/solid-electrolyte interface in the charge process. Some of the ions were then extracted in the discharge process, and the rest were diffused such that the distribution was almost flat, resulting in the negative electrodes. The redox reaction of Ti 4+ /Ti 3+ in the solid electrolyte was also observed in situ during the Li insertion/extraction processes. The in situ SR-TEM-EELS revealed the mechanism of the electrochemical reaction in solid-state batteries.

    Beschreibung: We have investigated how accurately atomic-resolution annular dark-field (ADF) images match between experiments and simulations to conduct more reliable crystal structure analyses. Quantitative ADF imaging, in which the ADF intensity at each pixel represents the fraction of the incident probe current, allows us to perform direct comparisons with simulations without the use of fitting parameters. Although the conventional comparison suffers from experimental uncertainties such as an amorphous surface layer and specimen thickness, in this study we eliminated such uncertainties by using a single-layer graphene as a specimen. Furthermore, to reduce image distortion and shot noises in experimental images, multiple acquisitions with drift correction were performed, and the atomic ADF contrast was quantitatively acquired. To reproduce the experimental ADF contrast, we used three distribution functions as the effective source distribution in simulations. The optimum distribution function and its full-width at half-maximum were evaluated by measuring the residuals between the experimental and simulated images. It was found that the experimental images could be explained well by a linear combination of a Gaussian function and a Lorentzian function with a longer tail than the Gaussian function.

    Beschreibung: Mechanical properties of complex steels are affected by their multi-phase structure. Scanning electron microscopy (SEM) is routinely used for characterizing dual-phase (DP) steels, although the identification of steel constituents is not straightforward. In fact, there are several ways of enabling the ferrite–martensite segmentation by SEM, and a wide range of electron energies can be utilized. This study demonstrates the phase identification of DP steels at high, low and extremely low landing energies of the primary electrons from tens of keV to tens of eV. Visualization of the specimen surface at very low landing energies has been achieved by inserting an earthed detector between the pole piece and the negatively biased specimen. This ‘cathode lens mode’ enables the use of the full energy range up to the primary electron energies. It has been found that extremely slow electrons (〈100 eV) are exceptionally suitable for separation of the martensite from the ferrite matrix due to high surface sensitivity, enabling visualization of very fine features. Moreover, the channelling contrast is significantly suppressed at the landing energy of tens of eV of the primary electrons, which enables separation of the phases clearly even in the images acquired at low magnification. The contrast between the phases at tens of eV can be explained by the different thickness of native oxide covering the martensite and the ferrite phase.

    Beschreibung: ‘Tubular aggregates’ are morphologically distinct cytoplasmic structures that have been linked to a variety of pathological conditions. This report documents the presence of tubular aggregates in an insect cell line (C6/36 cells derived from Aedes albopictus ) following inoculation of the cells with material derived from cell culture passaged homogenized Culex australicus mosquitoes. The tubular aggregates were detected in ~2% of treated cells and had three morphological forms that were termed primary, secondary and tertiary, with progressively greater levels of structural complexity. The findings indicate that tubular aggregates can be induced in an insect cell culture system by an unidentified agent present in some mosquitoes.

    Beschreibung: In this paper, the water level fluctuations of eight Ethiopian Rift Valley lakes were analyzed for their hydrological stability in terms of water level dynamics and their controlling factors. Long-term water balances and morphological nature of the lakes were used as bases for the analyses. Pettit’s homogeneity test and Mann–Kendall trend analysis were applied to test temporal variations of the lake levels. It is found that the hydrological stability of most of the Ethiopian Rift Valley lakes is sensitive to climate variability. In terms of monotonic trends, Lake Ziway, Hawassa, Abaya and Beseka experienced significant increasing trend, while Ziway, Langano and Chamo do not. In addition, homogeneity test revealed that Lake Hawassa and Abaya showed significant upward shift around 1991/1992, which was likely caused by climate anomalies such as the El Niño / Southern Oscillation (ENSO) phenomena. Lake Abiyata is depicted by its significant decreasing monotonic trend and downward regime shift around 1984/1985, which is likely related to the extended water abstraction for industrial consumption.

    Beschreibung: Trends in high, moderate, and low streamflow conditions from United States Geological Survey (USGS) gauging stations were evaluated for a period of 1951–2013 for 18 selected watersheds in South Dakota (SD) using a modified Mann-Kendall test. Rainfall trends from 21 rainfall observation stations located within 20-km of the streamflow gauging stations were also evaluated for the same study period. The concept of elasticity was used to examine sensitivity of streamflow to variation in rainfall and land cover (i.e., grassland) in the study watersheds. Results indicated significant increasing trends in seven of the studied streams (of which five are in the east and two are located in the west), nine with slight increasing trends, and two with decreasing trends for annual streamflow. About half of the streams exhibited significant increasing trends in low and moderate flow conditions compared to high flow conditions. Ten rainfall stations showed slight increasing trends and seven showed decreasing trends for annual rainfall. Streamflow elasticity analysis revealed that streamflow was highly influenced by rainfall across the state (five of eastern streams and seven of western streams). Based on this analysis, a 10% increase in annual rainfall would result in 11%–30% increase in annual streamflow in more than 60% of SD streams. While streamflow appears to be more sensitive to rainfall across the state, high sensitivity of streamflow to rapid decrease in grassland area was detected in two western watersheds. This study provides valuable insight into of the relationship between streamflow, climate, and grassland cover in SD and would support further research and stakeholder decision making about water resources.

    Beschreibung: This study presents a comprehensive assessment of the variability and trends of the precipitation and temperature along with the trends in drought indices over the State of California. The non-parametric Mann–Kendall trend test is applied with a trend-free pre-whitening procedure in trend identification. A dataset containing 120-year (water years 1896–2015) monthly precipitation, average temperature, maximum temperature, minimum temperature and the Palmer Index for seven climatic regions of the state is used for this purpose. The results confirm previous work indicating that no clear trends are observed in precipitation, while a distinct warming trend is evident in temperature over the state. New findings of this study include: (1) in general, the variability of annual, winter (December–February) and spring (March–May) precipitation shows an increasing tendency, implying intensified frequency of the occurrence of dry or wet extremes; (2) on the annual scale and in the summer, statewide meteorological, hydrological and agricultural drought indices all have decreasing trends, indicating the more frequent occurrence of drought events; and (3) among seven regions, the South Coast Drainage region generally has the most significant warming trend, as well as the most significant declining trends in drought indices. Overall, these findings are highly meaningful from both theoretical and practical perspectives, in the context of providing critical information in developing prediction models and guiding water resources management practices, respectively.

    Beschreibung: In this article, we propose a smart image-analysis method suitable for extracting target features with hierarchical dimension from original data. The method was applied to three-dimensional volume data of an all-solid lithium-ion battery obtained by the automated sequential sample milling and imaging process using a focused ion beam/scanning electron microscope to investigate the spatial configuration of voids inside the battery. To automatically fully extract the shape and location of the voids, three types of filters were consecutively applied: a median blur filter to extract relatively larger voids, a morphological opening operation filter for small dot-shaped voids and a morphological closing operation filter for small voids with concave contrasts. Three data cubes separately processed by the above-mentioned filters were integrated by a union operation to the final unified volume data, which confirmed the correct extraction of the voids over the entire dimension contained in the original data.

    Beschreibung: Polyelectrolyte brushes are polyelectrolyte polymers with one end fixed to a substrate. In this study, direct nano-scale visualization of polyelectrolyte brushes was carried out under ‘aqueous conditions’ by atmospheric scanning electron microscopy. The thickness of the polyelectrolyte brush layer was measured under both dry and aqueous conditions, experimentally confirming the swollen state of the brushes. These experimental findings qualitatively agreed with the results from previous neutron reflectivity experiments using similar polyelectrolyte brushes. Such direct visualization of polymer brushes in real space opens up a new route for better understanding their surface properties, such as friction, adhesion and wettability.

    Beschreibung: The time synchronization protocol is indispensable in various applications of wireless sensor networks, such as scheduling, monitoring, and tracking. Numerous protocols and algorithms have been proposed in recent decades, and many of them provide micro-scale resolutions. However, designing and implementing a time synchronization protocol in a practical wireless network is very challenging compared to implementation in a wired network; this is because its performance can be deteriorated significantly by many factors, including hardware quality, message delay jitter, ambient environment, and network topology. In this study, we measure the performance of the Flooding Time Synchronization Protocol (FTSP) and Gradient Time Synchronization Protocol (GTSP) in terms of practical network conditions, such as message delay jitter, synchronization period, network topology, and packet loss. This study provides insights into the operation and optimization of time synchronization protocols. In addition, the performance evaluation identifies that FTSP is highly affected by message delay jitter due to error accumulation over multi-hops. We demonstrate that the proposed extended version of the FTSP (E-FTSP) alleviates the effect of message delay jitter and enhances the overall performance of FTSP in terms of error, time, and other factors.

    Beschreibung: The Internet of Things (IoT) will feature pervasive sensing and control capabilities via the massive deployment of machine-type communication devices in order to greatly improve daily life. However, machine-type communications can be illegally used (e.g., by criminals or terrorists) which is difficult to monitor, and thus presents new security challenges. The information exchanged in machine-type communications is usually transmitted in short packets. Thus, this paper investigates a legitimate surveillance system via proactive eavesdropping at finite blocklength regime. Under the finite blocklength regime, we analyze the channel coding rate of the eavesdropping link and the suspicious link. We find that the legitimate monitor can still eavesdrop the information sent by the suspicious transmitter as the blocklength decreases, even when the eavesdropping is failed under the Shannon capacity regime. Moreover, we define a metric called the effective eavesdropping rate and study the monotonicity. From the analysis of monotonicity, the existence of a maximum effective eavesdropping rate for a moderate or even high signal-to-noise (SNR) is verified. Finally, numerical results are provided and discussed. In the simulation, we also find that the maximum effective eavesdropping rate slowly increases with the blocklength.

    Beschreibung: GNSS receiver data crowdsourcing is of interest for multiple applications, e.g., weather monitoring. The bottleneck in this technology is the quality of the GNSS receivers. Therefore, we lay out in an introductory manner the steps to estimate the performance of an arbitrary GNSS receiver via the measurement errors related to its instrumentation. Specifically, we do not need to know the position of the receiver antenna, which allows also for the assessment of smartphone GNSS receivers having integrated antennas. Moreover, the method is independent of atmospheric errors so that no ionospheric or tropospheric correction services provided by base stations are needed. Error models for performance evaluation can be calculated from receiver RINEX (receiver independent exchange format)data using only ephemeris corrections. For the results, we present the quality of different receiver grades through parametrized error models that are likely to be helpful in stochastic modeling, e.g., for Kalman filters, and in assessing GNSS receiver qualities for crowdsourcing applications. Currently, the typical positioning precision for the latest smartphone receivers is around the decimeter level, while for a professional-grade receiver, it is within a few millimeters.

    Beschreibung: The power transmission lines are the link between power plants and the points of consumption, through substations. Most importantly, the assessment of damaged aerial power lines and rusted conductors is of extreme importance for public safety; hence, power lines and associated components must be periodically inspected to ensure a continuous supply and to identify any fault and defect. To achieve these objectives, recently, Unmanned Aerial Vehicles (UAVs) have been widely used; in fact, they provide a safe way to bring sensors close to the power transmission lines and their associated components without halting the equipment during the inspection, and reducing operational cost and risk. In this work, a drone, equipped with multi-modal sensors, captures images in the visible and infrared domain and transmits them to the ground station. We used state-of-the-art computer vision methods to highlight expected faults (i.e., hot spots) or damaged components of the electrical infrastructure (i.e., damaged insulators). Infrared imaging, which is invariant to large scale and illumination changes in the real operating environment, supported the identification of faults in power transmission lines; while a neural network is adapted and trained to detect and classify insulators from an optical video stream. We demonstrate our approach on data captured by a drone in Parma, Italy.

    Beschreibung: The number of connected sensors and devices is expected to increase to billions in the near future. However, centralised cloud-computing data centres present various challenges to meet the requirements inherent to Internet of Things (IoT) workloads, such as low latency, high throughput and bandwidth constraints. Edge computing is becoming the standard computing paradigm for latency-sensitive real-time IoT workloads, since it addresses the aforementioned limitations related to centralised cloud-computing models. Such a paradigm relies on bringing computation close to the source of data, which presents serious operational challenges for large-scale cloud-computing providers. In this work, we present an architecture composed of low-cost Single-Board-Computer clusters near to data sources, and centralised cloud-computing data centres. The proposed cost-efficient model may be employed as an alternative to fog computing to meet real-time IoT workload requirements while keeping scalability. We include an extensive empirical analysis to assess the suitability of single-board-computer clusters as cost-effective edge-computing micro data centres. Additionally, we compare the proposed architecture with traditional cloudlet and cloud architectures, and evaluate them through extensive simulation. We finally show that acquisition costs can be drastically reduced while keeping performance levels in data-intensive IoT use cases.

    Beschreibung: One of the most significant challenges in Internet of Things (IoT) environments is the protection of privacy. Failing to guarantee the privacy of sensitive data collected and shared over IoT infrastructures is a critical barrier that delays the wide penetration of IoT technologies in several user-centric application domains. Location information is the most common dynamic information monitored and lies among the most sensitive ones from a privacy perspective. This article introduces a novel mechanism that aims to protect the privacy of location information across Data Centric Sensor Networks (DCSNs) that monitor the location of mobile objects in IoT systems. The respective data dissemination protocols proposed enhance the security of DCSNs rendering them less vulnerable to intruders interested in obtaining the location information monitored. In this respect, a dynamic clustering algorithm is that clusters the DCSN nodes not only based on the network topology, but also considering the current location of the objects monitored. The proposed techniques do not focus on the prevention of attacks, but on enhancing the privacy of sensitive location information once IoT nodes have been compromised. They have been extensively assessed via series of experiments conducted over the IoT infrastructure of FIT IoT-LAB and the respective evaluation results indicate that the dynamic clustering algorithm proposed significantly outperforms existing solutions focusing on enhancing the privacy of location information in IoT.

    Beschreibung: There is strong demand for real-time suspicious tracking across multiple cameras in intelligent video surveillance for public areas, such as universities, airports and factories. Most criminal events show that the nature of suspicious behavior are carried out by un-known people who try to hide themselves as much as possible. Previous learning-based studies collected a large volume data set to train a learning model to detect humans across multiple cameras but failed to recognize newcomers. There are also several feature-based studies aimed to identify humans within-camera tracking. It would be very difficult for those methods to get necessary feature information in multi-camera scenarios and scenes. It is the purpose of this study to design and implement a suspicious tracking mechanism across multiple cameras based on correlation filters, called suspicious tracking across multiple cameras based on correlation filters (STAM-CCF). By leveraging the geographical information of cameras and YOLO object detection framework, STAM-CCF adjusts human identification and prevents errors caused by information loss in case of object occlusion and overlapping for within-camera tracking cases. STAM-CCF also introduces a camera correlation model and a two-stage gait recognition strategy to deal with problems of re-identification across multiple cameras. Experimental results show that the proposed method performs well with highly acceptable accuracy. The evidences also show that the proposed STAM-CCF method can continuously recognize suspicious behavior within-camera tracking and re-identify it successfully across multiple cameras.

    Beschreibung: The paper proposes a sensors platform to control a barrier that is installed for vehicles entrance. This platform is automatized by image-based license plate recognition of the vehicle. However, in situations where standardized license plates are not used, such image-based recognition becomes non-trivial and challenging due to the variations in license plate background, fonts and deformations. The proposed method first detects the approaching vehicle via ultrasonic sensors and, at the same time, captures its image via a camera installed along with the barrier. From this image, the license plate is automatically extracted and further processed to segment the license plate characters. Finally, these characters are recognized with the help of a standard optical character recognition (OCR) pipeline. The evaluation of the proposed system shows an accuracy of 98% for license plates extraction, 96% for character segmentation and 93% for character recognition.

    Beschreibung: Wireless sensor networks are widely used in many fields. Nodes in the network are typically powered by batteries. Because the energy consumption of wireless communication is related to the transmission distance, the energy consumption of nodes in different locations is different, resulting in uneven energy distribution of nodes. In some special applications, all nodes are required to work at the same time, and the uneven energy distribution makes the effective working time of the system subject to the node with the largest energy consumption. The commonly used clustering protocol can play a role in balancing energy consumption, but it does not achieve optimal energy consumption. This paper proposes to use the power supply line to connect the nodes to fully balance the energy. The connection scheme with the shortest power line length is also proposed. On the basis of energy balance, the method of transmitting data with the best hop count is proposed, which fully reduces the power consumption of the data transmission. The simulation results show that the proposed method can effectively reduce the energy consumption and prolong the lifetime of wireless sensor networks.

    Beschreibung: High frequency guided-waves offer a trade-off between the high sensitivity of local bulk ultrasonic thickness measurements and the large area scanning of lower frequency guided-waves, so it has been a growing interest for corrosion inspection with the dispersive SH1 mode. However, according to the dispersive curve, it is hard to generate the pure SH1 mode since the non-dispersive SH0 mode will be excited simultaneously. Thus, this paper investigates a transducer design method to generate a pure SH1 guided-wave, where the dual periodic-permanent-magnet electromagnetic acoustic transducers (PPM EMATs) are placed on exactly opposite positions either side of the plate symmetrically. The suppression effect for SH0 and the enhancement effect for SH1 of the dual PPM EMATs are mainly discussed by theoretical analysis and simulation analysis, and the influence of positioning errors of PPM EMATs placed on opposite sides of the plate on its performances are analyzed. Employing the proposed dual PPM EMATs, some experiments are performed to verify the reliability of finite element simulation. The results indicate that the dual PPM EMATs can suppress the SH0 mode and generate the pure SH1 mode effectively. Moreover, the longitudinal and lateral positioning errors can affect the dual PPM EMATs performances significantly.

    Beschreibung: In this paper, we report a wireless gas sensor based on surface acoustic waves (SAW). For room temperature detection of oxygen gas, a novel nanostructured ZnxFeyO gas-sensitive film was deposited on the surface of a SAW resonator by an oblique magnetron co-sputtering method. The measurements of X-ray diffraction (XRD) and a scanning electron microscope (SEM) showed that the crystal phase composition and the microstructures of ZnxFeyO films were significantly affected by the content of Fe. The experimental results showed that the sensors had a good response to O2 at room temperature. The max frequency shift of the sensors reached 258 kHz as the O2 partial pressure was 20%. Moreover, X-ray photoelectron spectroscopy (XPS) was performed to analyze the role of Fe in the sensitization process of the ZnxFeyO film. In addition, the internal relationship between the Fe content of the film and the sensitivity of the sensor was presented and discussed. The research indicates that the nanostructured ZnxFeyO film has a good potential for room temperature O2 gas detection applications.

    Beschreibung: According to the survey on various health centres, smart log-based multi access physical monitoring system determines the health conditions of humans and their associated problems present in their lifestyle. At present, deficiency in significant nutrients leads to deterioration of organs, which creates various health problems, particularly for infants, children, and adults. Due to the importance of a multi access physical monitoring system, children and adolescents’ physical activities should be continuously monitored for eliminating difficulties in their life using a smart environment system. Nowadays, in real-time necessity on multi access physical monitoring systems, information requirements and the effective diagnosis of health condition is the challenging task in practice. In this research, wearable smart-log patch with Internet of Things (IoT) sensors has been designed and developed with multimedia technology. Further, the data computation in that smart-log patch has been analysed using edge computing on Bayesian deep learning network (EC-BDLN), which helps to infer and identify various physical data collected from the humans in an accurate manner to monitor their physical activities. Then, the efficiency of this wearable IoT system with multimedia technology is evaluated using experimental results and discussed in terms of accuracy, efficiency, mean residual error, delay, and less energy consumption. This state-of-the-art smart-log patch is considered as one of evolutionary research in health checking of multi access physical monitoring systems with multimedia technology.

    Beschreibung: Smart grids incorporating internet-of-things are emerging solutions to provide a reliable, sustainable and efficient electricity supply, and electric vehicle drivers can access efficient charging services in the smart grid. However, traditional electric vehicle charging systems are vulnerable to distributed denial of service and privileged insider attacks when the central charging server is attacked. The blockchain-based charging systems have been proposed to resolve these problems. In 2018, Huang et al. proposed the electric vehicle charging system using lightning network and smart contract. However, their system has an inefficient charging mechanism and does not guarantee security of key. We propose a secure charging system for electric vehicles based on blockchain to resolve these security flaws. Our charging system ensures the security of key, secure mutual authentication, anonymity, and perfect forward secrecy, and also provides efficient charging. We demonstrate that our proposed system provides secure mutual authentication using Burrows–Abadi–Needham logic and prevents replay and man-in-the-middle attacks using automated validation of internet security protocols and applications simulation tool. Furthermore, we compare computation and communication costs with previous schemes. Therefore, the proposed charging system efficiently applies to practical charging systems for electric vehicles.

    Beschreibung: Haiyang-2A (HY-2A) has been working in-flight for over seven years, and the accuracy of HY-2A calibration microwave radiometer (CMR) data is extremely important for the wet troposphere delay correction (WTC) in sea surface height (SSH) determination. We present a comprehensive evaluation of the HY-2A CMR observation using the numerical weather model (NWM) for all the data available period from October 2011 to February 2018, including the WTC and the precipitable water vapor (PWV). The ERA(ECMWF Re-Analysis)-Interim products from European Centre for Medium-Range Weather Forecasts (ECMWF) are used for the validation of HY-2A WTC and PWV products. In general, a global agreement of root-mean-square (RMS) of 2.3 cm in WTC and 3.6 mm in PWV are demonstrated between HY-2A observation and ERA-Interim products. Systematic biases are revealed where before 2014 there was a positive WTC/PWV bias and after that, a negative one. Spatially, HY-2A CMR products show a larger bias in polar regions compared with mid-latitude regions and tropical regions and agree better in the Antarctic than in the Arctic with NWM. Moreover, HY-2A CMR products have larger biases in the coastal area, which are all caused by the brightness temperature (TB) contamination from land or sea ice. Temporally, the WTC/PWV biases increase from October 2011 to March 2014 with a systematic bias over 1 cm in WTC and 2 mm in PWV, and the maximum RMS values of 4.62 cm in WTC and 7.61 mm in PWV occur in August 2013, which is because of the unsuitable retrieval coefficients and systematic TB measurements biases from 37 GHz band. After April 2014, the TB bias is corrected, HY-2A CMR products agree very well with NWM from April 2014 to May 2017 with the average RMS of 1.68 cm in WTC and 2.65 mm in PWV. However, since June 2017, TB measurements from the 18.7 GHz band become unstable, which led to the huge differences between HY-2A CMR products and the NWM with an average RMS of 2.62 cm in WTC and 4.33 mm in PWV. HY-2A CMR shows high accuracy when three bands work normally and further calibration for HY-2A CMR is in urgent need. Furtherly, 137 global coastal radiosonde stations were used to validate HY-2A CMR. The validation based on radiosonde data shows the same variation trend in time of HY-2A CMR compared to the results from ECMWF, which verifies the results from ECMWF.

    Beschreibung: Crop yield estimation at a regional scale over a long period of time is of great significance to food security. In past decades, the integration of remote sensing observations and crop growth models has been recognized as a promising approach for crop growth monitoring and yield estimation. Optical remote sensing data are susceptible to cloud and rain, while synthetic aperture radar (SAR) can penetrate through clouds and has all-weather capabilities. This allows for more reliable and consistent crop monitoring and yield estimation in terms of radar sensor data. The aim of this study is to improve the accuracy for winter wheat yield estimation by assimilating time series soil moisture images, which are retrieved by a water cloud model using SAR and optical data as input, into the crop model. In this study, SAR images were acquired by C-band SAR sensors boarded on Sentinel-1 satellites and optical images were obtained from a Sentinel-2 multi-spectral instrument (MSI) for Hengshui city of Hebei province in China. Remote sensing data and ground data were all collected during the main growing season of winter wheat. Both the normalized difference vegetation index (NDVI), derived from Sentinel-2, and backscattering coefficients and polarimetric indicators, computed from Sentinel-1, were used in the water cloud model to derive time series soil moisture (SM) images. To improve the prediction of crop yields at the field scale, we incorporated remotely sensed soil moisture into the World Food Studies (WOFOST) model using the Ensemble Kalman Filter (EnKF) algorithm. In general, the trend of soil moisture inversion was consistent with the ground measurements, with the coefficient of determination (R2) equal to 0.45, 0.53, and 0.49, respectively, and RMSE was 9.16%, 7.43%, and 8.53%, respectively, for three observation dates. The winter wheat yield estimation results showed that the assimilation of remotely sensed soil moisture improved the correlation of observed and simulated yields (R2 = 0.35; RMSE =934 kg/ha) compared to the situation without data assimilation (R2 = 0.21; RMSE = 1330 kg/ha). Consequently, the results of this study demonstrated the potential and usefulness of assimilating SM retrieved from both Sentinel-1 C-band SAR and Sentinel-2 MSI optical remote sensing data into WOFOST model for winter wheat yield estimation and could also provide a reference for crop yield estimation with data assimilation for other crop types.

    Beschreibung: Urban areas globally are vulnerable to warming climate trends exacerbated by their growing populations and heat island effects. The Local Climate Zone (LCZ) typology has become a popular framework for characterizing urban microclimates in different regions using various classification methods, including a widely adopted pixel-based protocol by the World Urban Database and Access Portal Tools (WUDAPT) Project. However, few studies to date have explored the potential of object-based image analysis (OBIA) to facilitate classification of LCZs given their inherent complexity, and few studies have further used the LCZ framework to analyze land cover changes in urban areas over time. This study classified LCZs in the Salt Lake Metro Region, Utah, USA for 1993 and 2017 using a supervised object-based analysis of Landsat satellite imagery and assessed their change during this time frame. The overall accuracy, measured for the most recent classification period (2017), was equal to 64% across 12 LCZs, with most of the error resulting from similarities among highly developed LCZs and non-developed classes with sparse or low-stature vegetation. The observed 1993–2017 changes in LCZs indicated a regional tendency towards primarily suburban, open low-rise development, and large low-rise and paved classes. However, despite the potential for local cooling with landscape transitions likely to increase vegetation cover and irrigation compared to pre-development conditions, summer averages of Landsat-derived top-of-atmosphere brightness temperatures showed a pronounced warming between 1992–1994 and 2016–2018 across the study region, with a 0.1–2.9 °C increase among individual LCZs. Our results indicate that future applications of LCZs towards urban change analyses should develop a stronger understanding of LCZ microclimate sensitivity to changes in size and configuration of urban neighborhoods and regions. Furthermore, while OBIA is promising for capturing the heterogeneous and multi-scale nature of LCZs, its applications could be strengthened by adopting more generalizable approaches for LCZ-relevant segmentation and validation, and by incorporating active remote sensing data to account for the 3D complexity of urban areas.

    Beschreibung: An important application of airborne- and satellite-based hyperspectral imaging is the mapping of the spatial distribution of vegetation biophysical and biochemical parameters in an environment. Statistical models, such as Gaussian processes, have been very successful for modeling vegetation parameters from captured spectra, however their performance is highly dependent on the amount of available ground truth. This is a problem because it is generally expensive to obtain ground truth information due to difficulties and costs associated with sample collection and analysis. In this paper, we present two Gaussian processes based approaches for improving the accuracy of vegetation parameter retrieval when ground truth is limited. The first is the adoption of covariance functions based on well-established metrics, such as, spectral angle and spectral correlation, which are known to be better measures of similarity for spectral data owing to their resilience to spectral variabilities. The second is the joint modeling of related vegetation parameters by multitask Gaussian processes so that the prediction accuracy of the vegetation parameter of interest can be improved with the aid of related vegetation parameters for which a larger set of ground truth is available. We experimentally demonstrate the efficacy of the proposed methods against existing approaches on three real-world hyperspectral datasets and one synthetic dataset.

    Beschreibung: Spatial features retrieved from satellite data play an important role for improving crop classification. In this study, we proposed a deep-learning-based time-series analysis method to extract and organize spatial features to improve parcel-based crop classification using high-resolution optical images and multi-temporal synthetic aperture radar (SAR) data. Central to this method is the use of multiple deep convolutional networks (DCNs) to extract spatial features and to use the long short-term memory (LSTM) network to organize spatial features. First, a precise farmland parcel map was delineated from optical images. Second, hundreds of spatial features were retrieved using multiple DCNs from preprocessed SAR images and overlaid onto the parcel map to construct multivariate time-series of crop growth for parcels. Third, LSTM-based network structures for organizing these time-series features were constructed to produce a final parcel-based classification map. The method was applied to a dataset of high-resolution ZY-3 optical images and multi-temporal Sentinel-1A SAR data to classify crop types in the Hunan Province of China. The classification results, showing an improvement of greater than 5.0% in overall accuracy relative to methods without spatial features, demonstrated the effectiveness of the proposed method in extracting and organizing spatial features for improving parcel-based crop classification.

    Beschreibung: The classification of very-high-resolution (VHR) remote sensing images is essential in many applications. However, high intraclass and low interclass variations in these kinds of images pose serious challenges. Fully convolutional network (FCN) models, which benefit from a powerful feature learning ability, have shown impressive performance and great potential. Nevertheless, only classification results with coarse resolution can be obtained from the original FCN method. Deep feature fusion is often employed to improve the resolution of outputs. Existing strategies for such fusion are not capable of properly utilizing the low-level features and considering the importance of features at different scales. This paper proposes a novel, end-to-end, fully convolutional network to integrate a multiconnection ResNet model and a class-specific attention model into a unified framework to overcome these problems. The former fuses multilevel deep features without introducing any redundant information from low-level features. The latter can learn the contributions from different features of each geo-object at each scale. Extensive experiments on two open datasets indicate that the proposed method can achieve class-specific scale-adaptive classification results and it outperforms other state-of-the-art methods. The results were submitted to the International Society for Photogrammetry and Remote Sensing (ISPRS) online contest for comparison with more than 50 other methods. The results indicate that the proposed method (ID: SWJ_2) ranks #1 in terms of overall accuracy, even though no additional digital surface model (DSM) data that were offered by ISPRS were used and no postprocessing was applied.

    Beschreibung: Low Earth orbit (LEO) satellites play an important role in human space activities, and market demands for commercial uses of LEO satellites have been increasing rapidly in recent years. LEO satellites mainly consist of Earth observation satellites (EOSs), the major commercial applications of which are various sorts of Earth observations, such as map making, crop growth assessment, and disaster surveillance. However, the success rates of observation tasks are influenced considerably by uncertainties in local weather conditions, inadequate sunlight, observation dip angle, and other practical factors. The available time windows (ATWs) suitable for observing given types of targets and for transmitting data back to ground receiver stations are relatively narrow. In order to utilize limited satellite resources efficiently and maximize their commercial benefits, it is necessary to evaluate the overall effectiveness of satellites and planned tasks considering various factors. In this paper, we propose a method for determining the ATWs considering the influence of sunlight angle, elevation angle, and the type of sensor equipped on the satellite. After that, we develop a satellite effectiveness evaluation (SEE) model for satellite observation and data-downlink scheduling (SODS) based on the Availability–Capacity–Profitability (ACP) framework, which is designed to evaluate the overall performance of satellites from the perspective of time resource utilization, the success rate of tasks, and profit return. The effects of weather uncertainties on the tasks’ success are considered in the SEE model, and the model can be applied to support the decision-makers on optimizing and improving task arrangements for EOSs. Finally, a case study is presented to demonstrate the effectiveness of the proposed method and verify the ACP-based SEE model. The obtained ATWs by the proposed method are compared with those by the Systems Tool Kit (STK), and the correctness of the method is thus validated.

    Beschreibung: In real-time onboard hyperspectral-image(HSI) anomalous targets detection, processing speed and accuracy are equivalently desirable which is hard to satisfy at the same time. To improve detection accuracy, deep learning based HSI anomaly detectors (ADs) are widely studied. However, their large scale network results in a massive computational burden. In this paper, to improve the detection throughput without sacrificing the accuracy, a pruning–quantization–anomaly–detector (P-Q-AD) is proposed by building an underlying constraint formulation to make a trade-off between accuracy and throughput. To solve this formulation, multi-objective optimization with nondominated sorting genetic algorithm II (NSGA-II) is employed to shrink the network. As a result, the redundant neurons are removed. A mixed precision network is implemented with a delicate customized fixed-point data expression to further improve the efficiency. In the experiments, the proposed P-Q-AD is implemented on two real HSI data sets and compared with three types of detectors. The results show that the performance of the proposed approach is no worse than those comparison detectors in terms of the receiver operating characteristic curve (ROC) and area under curve (AUC) value. For the onboard mission, the proposed P-Q-AD reaches over 4.5 × speedup with less than 0.5 % AUC loss compared with the floating-based detector. The pruning and the quantization approach in this paper can be referenced for designing the anomalous targets detectors for high efficiency.

    Beschreibung: To date, several algorithms for the retrieval of cyanobacterial phycocyanin (PC) from ocean colour sensors have been presented for inland waters, all of which claim to be robust models. To address this, we conducted a comprehensive comparison to identify the optimal algorithm for retrieval of PC concentrations in the highly optically complex waters of Lake Balaton (Hungary). MEdium Resolution Imaging Spectrometer (MERIS) top-of-atmosphere radiances were first atmospherically corrected using the Self-Contained Atmospheric Parameters Estimation for MERIS data v.B2 (SCAPE-M_B2). Overall, the Simis05 semi-analytical algorithm outperformed more complex inversion algorithms, providing accurate estimates of PC up to ±7 days from the time of satellite overpass during summer cyanobacteria blooms (RMSElog 〈 0.33). Same-day retrieval of PC also showed good agreement with cyanobacteria biomass (R2 〉 0.66, p 〈 0.001). In-depth analysis of the Simis05 algorithm using in situ measurements of inherent optical properties (IOPs) revealed that the Simis05 model overestimated the phytoplankton absorption coefficient [aph(λ)] by a factor of ~2. However, these errors were compensated for by underestimation of the mass-specific chlorophyll absorption coefficient [a*chla(λ)]. This study reinforces the need for further validation of algorithms over a range of optical water types in the context of the recently launched Ocean Land Colour Instrument (OLCI) onboard Sentinel-3.

Due to its ideal scanning and acquisition conditions for low cloud coverage imagery, Namibia aims to make use of this new development and integrate Earth Observation data into its national monitoring system of sustainable development goals (SDG). The purpose of this study is to assess the potential of open source tools and global datasets to estimate the national SDG indicators on Change of water-related ecosystems (6.6.1), Rural population with access to roads (9.1.1), Forest coverage (15.1.1) and Land degradation (15.3.1). The results are set into perspective of existing information in each particular sector. The study shows that, in the absence of in-situ measurements or data collected through surveys, the Earth Observation-based results represent a high potential to supplement the national statistics for Namibia or to serve as primary data sources once validated through ground-truthing. Furthermore, examples are given for the limitations of the assessed Earth Observation solutions in the context of Namibia. Hence, the study also serves as valuable input for discussions on a consensus on national definitions and standards by all stakeholders responsible for releasing official statistics.

    Beschreibung: This paper presents a mathematical model of measuring blood flow based on electromagnetic induction for predicting the rate of arterial stenosis. Firstly, an electrode sensor was used to collect the induced potential differences from human skin surface in a uniform magnetic field. Then, the inversion matrix was constructed by the weight function theory and finite element method. Next, the blood flow volume inversion model was constructed by combining the induction potential differences and inversion matrix. Finally, the rate of arterial stenosis was predicted based on mathematical relationship between blood flow and the area of arterial stenosis. To verify the accuracy of the model, a uniform magnetic field distribution of Helmholtz coil and a 3D geometric model of the ulnar artery of the forearm with different rates of stenosis were established in COMSOL, a finite element analysis software. Simulation results showed that the inversion model had high accuracy in the measurement of blood flow and the prediction of rate of stenosis, and is of great significance for the early diagnosis of arterial stenosis and other vessel diseases.

    Beschreibung: Magnetic Induction Tomography (MIT) is a non-invasive imaging technique that has been widely applied for imaging materials with high electrical conductivity contrasts. Steel production is among an increasing number of applications that require a contactless method for monitoring the casting process due to the high temperature of hot steel. In this paper, an MIT technique is proposed for detecting defects and deformations in the external surfaces of metal, which has the potential to be used to monitor the external surface of hot steel during the continuous casting process. The Total Variation (TV) reconstruction algorithm was developed to image the conductivity distributions. Nonetheless, the reconstructed image of the deformed square metal obtained using the TV algorithm directly does not yield resonable images of the surface deformation. However, differential images obtained by subtracting the image of a perfect square metal with no deformations from the image obtained for a deformed square metal does provide accurate and repeatable deformation information. It is possible to obtain a more precise image of surface deformation by thresholding the differential image. This TV-based threshold-differencing method has been analysed and verified from both simulation and experimental tests. The simulation results reported that 0.92 % of the image region can be detected, and the experimental results indicated a 0.57 % detectability. Use of the proposed method was demonstareted in a MIT device which was used in continuous casting set up. The paper shows results from computer simulation, lab based cold tests, and real life data from continoeus cating demonstating the effectiveness of the proposed method.

    Beschreibung: Gas mixture quantification is essential for the recording and reproducing odors, because an odor consists of multiple chemical compounds. Gas mixture quantification using field asymmetric ion mobility spectrometry (FAIMS) was studied. Acetone, ethanol, and diethyl ether were selected as components of a ternary gas mixture sample as representatives of the ketone, alcohol, and ether chemical classes, respectively. One hundred and twenty-five points with different concentrations were measured. The results were evaluated by error hypersurface, variance, and the coefficient of variation. The error hypersurface showed that it is possible to reach the target composition by following the error-hypersurface gradient. Successful convergence was achieved with the gradient descent method in a simulation based on the measurement data. This result verified the feasibility of the quantification of a gas mixture using FAIMS.

    Beschreibung: In this study, a novel strain gauge arrangement and error reduction techniques were proposed to minimize crosstalk reading and simultaneously increase sensitivity on a decoupled six-axis force–moment (F/M) sensor. The calibration process that comprises the least squares method and error reduction techniques was implemented to obtain a robust decoupling matrix. A decoupling matrix is very crucial for minimizing error and crosstalk. A novel strain gauge arrangement that comprised double parallel strain gauges in the decoupled six-axis force–moment sensor was implemented to obtain high sensitivity. The experimental results revealed that the maximum calibration error, F/M sensor measurement error, and crosstalk readings were reduced to 3.91%, 1.78%, and 4.78%, respectively.

    Beschreibung: Conventional eddy current testing (ECT) using a pickup coil probe is widely employed for the detection of structural cracks. However, the inspection of conventional ECT for steel structures is difficult because of the magnetic noise caused by the nonuniform permeability of steel. To combat this challenge, we have developed a small magnetic sensor probe with a dual-channel tunneling magnetoresistance sensor that is capable of reducing magnetic noise. Applying this probe to a complicated component of steel structures—such as the welds joining a U-shaped rib and deck plate together—requires the reduction of signal fluctuation caused by the distance (liftoff) variations between the sensor probe and the subject. In this study, the fundamental crack signal and the liftoff signal were investigated with the dual-channel sensor. The results showed that the liftoff signals could be reduced and differentiated from the crack signals by the differential parameters of the dual-channel sensor. In addition, we proposed an extraction technique for the crack signal using the Lissajous curve of the differential parameters. The extraction technique could be applied to the inspections not only for flat plates but also for welded angles to detect cracks without the influence of the liftoff signal.

    Beschreibung: The objective of this paper is to study the ability of polymer optical fiber (POF) to be inserted in a knitted fabric and to measure both pressure and friction when walking. Firstly, POF, marketed and in development, have been compared in terms of the required mechanical properties for the insertion of the fiber directly into a knitted fabric on an industrial scale, i.e. elongation, bending rigidity, and minimum bending radius before plastic deformation. Secondly, the chosen optical fiber was inserted inside several types of knitted fabric and was shown to be sensitive to friction and compression. The knitted structure with the highest sensitivity has been chosen for sock prototype manufacturing. Finally, a feasibility study with an instrumented sock showed that it is possible to detect the different phases of walking in terms of compression and friction.

    Beschreibung: Dielectric grating-based sensors are usually based on the guided mode resonance (GMR) obtained using a thin planar waveguide layer (PWL) adjacent to a thin subwavelength grating layer. In this work, we present a detailed investigation of thick subwavelength dielectric grating structures that exhibit reflection resonances above a certain thickness without the need for the waveguide layer, showing great potential for applications in biosensing and tunable filtering. Analytic and numerical results are thoroughly discussed, as well as an experimental demonstration of the structure as a chemical sensor in the SWIR (short wave infrared) spectral range (1200–1800 nm). In comparison to the GMR structure with PWL, the thick grating structure has several unique properties: (i) It gives higher sensitivity when the spaces are filled, with the analyte peaking at certain space values due to an increase in the interaction volume between the analyte and the evanescent optical field between the grating lines; (ii) the TM (transverse magnetic) resonance, in certain cases, provides a better figure of merit; (iii) the sensitivity increases as the grating height increases; (iv) the prediction of the resonance locations based on the effective medium approximation does not give satisfactory results when the grating height is larger than a certain value, and the invalidity becomes more severe as the period increases; (v) a sudden increase in the Q-factor of the resonance occurs at a specific height value accompanied by the high local field enhancement (~103) characteristic of a nano-antenna type pattern. Rigorous numerical simulations of the field distribution are presented to explain the different observed phenomena.

    Beschreibung: In 2016, the IEEE task group ah (TGah) released a new standard called IEEE 802.11ah, and industrial Internet of Things (IoT) is one of its typical use cases. The restricted access window (RAW) is one of the core MAC mechanisms of IEEE 802.11ah, which aims to address the collision problem in the dense wireless networks. However, in each RAW period, stations still need to contend for the channel by Distributed Coordination Function and Enhanced Distributed Channel Access (DCF/EDCA), which cannot meet the real-time requirements of most industrial applications. In this paper, we propose a channel-aware contention window adaption (CA-CWA) algorithm. The algorithm dynamically adapts the contention window based on the channel status with an external interference discrimination ability, and improves the real-time performance of the IEEE 802.11ah. To validate the real-time performance of CA-CWA, we compared CA-CWA with two other backoff algorithms with an NS-3 simulator. The results illustrate that CA-CWA has better performance than the other two algorithms in terms of packet loss rate and average delay. Compared with the other two algorithms, CA-CWA is able to support industrial applications with higher deadline constraints under the same channel conditions in IEEE 802.11ah.

    Beschreibung: Existing correlations between features extracted from Electroencephalography (EEG) signals and emotional aspects have motivated the development of a diversity of EEG-based affect detection methods. Both intra-subject and inter-subject approaches have been used in this context. Intra-subject approaches generally suffer from the small sample problem, and require the collection of exhaustive data for each new user before the detection system is usable. On the contrary, inter-subject models do not account for the personality and physiological influence of how the individual is feeling and expressing emotions. In this paper, we analyze both modeling approaches, using three public repositories. The results show that the subject’s influence on the EEG signals is substantially higher than that of the emotion and hence it is necessary to account for the subject’s influence on the EEG signals. To do this, we propose a data transformation that seamlessly integrates individual traits into an inter-subject approach, improving classification results.

    Beschreibung: Depth cameras play a vital role in three-dimensional (3D) shape reconstruction, machine vision, augmented/virtual reality and other visual information-related fields. However, a single depth camera cannot obtain complete information about an object by itself due to the limitation of the camera’s field of view. Multiple depth cameras can solve this problem by acquiring depth information from different viewpoints. In order to do so, they need to be calibrated to be able to accurately obtain the complete 3D information. However, traditional chessboard-based planar targets are not well suited for calibrating the relative orientations between multiple depth cameras, because the coordinates of different depth cameras need to be unified into a single coordinate system, and the multiple camera systems with a specific angle have a very small overlapping field of view. In this paper, we propose a 3D target-based multiple depth camera calibration method. Each plane of the 3D target is used to calibrate an independent depth camera. All planes of the 3D target are unified into a single coordinate system, which means the feature points on the calibration plane are also in one unified coordinate system. Using this 3D target, multiple depth cameras can be calibrated simultaneously. In this paper, a method of precise calibration using lidar is proposed. This method is not only applicable to the 3D target designed for the purposes of this paper, but it can also be applied to all 3D calibration objects consisting of planar chessboards. This method can significantly reduce the calibration error compared with traditional camera calibration methods. In addition, in order to reduce the influence of the infrared transmitter of the depth camera and improve its calibration accuracy, the calibration process of the depth camera is optimized. A series of calibration experiments were carried out, and the experimental results demonstrated the reliability and effectiveness of the proposed method.

    Beschreibung: Two types of urea biosensors were integrated with a wireless measurement system and microfluidic measurement system. The two biosensors used were (i) a magnetic beads (MBs)-urease/graphene oxide (GO)/titanium dioxide (TiO2)-based biosensor and (ii) an MBs-urease/GO/ nickel oxide (NiO)-based biosensor, respectively. The wireless measurement system work exhibited the feasibility for the remote detection of urea, but it will require refinement and modification to improve stability and precision. The microchannel fluidic system showed the measurement reliability. The sensing properties of urea biosensors at different flow rates were investigated. From the measurement results, the decay of average sensitivity may be attributed to the induced vortex-induced vibrations (VIV) at the high flow rate. In the aspect of wireless monitoring, the average sensitivity of the urea biosensor based on MBs-urease/GO/NiO was 4.780 mV/(mg/dl) and with the linearity of 0.938. In the aspect of measurement under dynamic conditions, the average sensitivity of the urea biosensor based on MBs-urease/GO/NiO were 5.582 mV/(mg/dl) and with the linearity of 0.959. Both measurements performed NiO was better than TiO2 according to the comparisons.

    Beschreibung: Radio frequency (RF) based wireless power transfer provides an attractive solution to extend the lifetime of power-constrained wireless sensor networks. Through harvesting RF energy from surrounding environments or dedicated energy sources, low-power wireless devices can be self-sustaining and environment-friendly. These features make the RF energy harvesting wireless communication (RF-EHWC) technique attractive to a wide range of applications. The objective of this article is to investigate the latest research activities on the practical RF-EHWC design. The distribution of RF energy in the real environment, the hardware design of RF-EHWC devices and the practical issues in the implementation of RF-EHWC networks are discussed. At the end of this article, we introduce several interesting applications that exploit the RF-EHWC technology to provide smart healthcare services for animals, wirelessly charge the wearable devices, and implement 5G-assisted RF-EHWC.

    Beschreibung: Complex environments pose great challenges for autonomous mobile robot navigation. In this study, we address the problem of autonomous navigation in 3D environments with staircases and slopes. An integrated system for safe mobile robot navigation in 3D complex environments is presented and both the perception and navigation capabilities are incorporated into the modular and reusable framework. Firstly, to distinguish the slope from the staircase in the environment, the robot builds a 3D OctoMap of the environment with a novel Simultaneously Localization and Mapping (SLAM) framework using the information of wheel odometry, a 2D laser scanner, and an RGB-D camera. Then, we introduce the traversable map, which is generated by the multi-layer 2D maps extracted from the 3D OctoMap. This traversable map serves as the input for autonomous navigation when the robot faces slopes and staircases. Moreover, to enable robust robot navigation in 3D environments, a novel camera re-localization method based on regression forest towards stable 3D localization is incorporated into this framework. In addition, we utilize a variable step size Rapidly-exploring Random Tree (RRT) method which can adjust the exploring step size automatically without tuning this parameter manually according to the environment, so that the navigation efficiency is improved. The experiments are conducted in different kinds of environments and the output results demonstrate that the proposed system enables the robot to navigate efficiently and robustly in complex 3D environments.

    Beschreibung: In this work, we isolate individual wurtzite InAs nanowires and fabricate electrical contacts at both ends, exploiting the single nanostructures as building blocks to realize two different architectures of conductometric sensors: (a) the nanowire is drop-casted onto—supported by—a SiO2/Si substrate, and (b) the nanowire is suspended at approximately 250 nm from the substrate. We test the source-drain current upon changes in the concentration of humidity, ethanol, and NO2, using synthetic air as a gas carrier, moving a step forward towards mimicking operational environmental conditions. The supported architecture shows higher response in the mid humidity range (50% relative humidity), with shorter response and recovery times and lower detection limit with respect to the suspended nanowire. These experimental pieces of evidence indicate a minor role of the InAs/SiO2 contact area; hence, there is no need for suspended nanostructures to improve the sensing performance. Moreover, the sensing capability of single InAs nanowires for detection of NO2 and ethanol in the ambient atmosphere is reported and discussed.

    Beschreibung: Displacement is crucial for structural health monitoring, although it is very challenging to measure under field conditions. Most existing displacement measurement methods are costly, labor-intensive, and insufficiently accurate for measuring small dynamic displacements. Computer vision (CV)-based methods incorporate optical devices with advanced image processing algorithms to accurately, cost-effectively, and remotely measure structural displacement with easy installation. However, non-target-based CV methods are still limited by insufficient feature points, incorrect feature point detection, occlusion, and drift induced by tracking error accumulation. This paper presents a reference frame-based Deepflow algorithm integrated with masking and signal filtering for non-target-based displacement measurements. The proposed method allows the user to select points of interest for images with a low gradient for displacement tracking and directly calculate displacement without drift accumulated by measurement error. The proposed method is experimentally validated on a cantilevered beam under ambient and occluded test conditions. The accuracy of the proposed method is compared with that of a reference laser displacement sensor for validation. The significant advantage of the proposed method is its flexibility in extracting structural displacement in any region on structures that do not have distinct natural features.

    Beschreibung: Convenient indoor positioning has become an urgent need due to the improvement it offers to quality of life, which inspires researchers to focus on device-free indoor location. In areas covered with Wi-Fi, people in different locations will to varying degrees have an impact on the transmission of channel state information (CSI) of Wi-Fi signals. Because space is divided into several small regions, the idea of classification is used to locate. Therefore, a novel localization algorithm is put forward in this paper based on Deep Neural Networks (DNN) and a multi-model integration strategy. The approach consists of three stages. First, the local outlier factor (LOF), the anomaly detection algorithm, is used to correct the abnormal data. Second, in the training phase, 3 DNN models are trained to classify the region fingerprints by taking advantage of the processed CSI data from 3 antennas. Third, in the testing phase, a model fusion method named group method of data handling (GMDH) is adopted to integrate 3 predicted results of multiple models and give the final position result. The test-bed experiment was conducted in an empty corridor, and final positioning accuracy reached at least 97%.

    Beschreibung: Due to the recent rise in the use of lower-limb exoskeletons as an alternative for gait rehabilitation, gait phase detection has become an increasingly important feature in the control of these devices. In addition, highly functional, low-cost recovery devices are needed in developing countries, since limited budgets are allocated specifically for biomedical advances. To achieve this goal, this paper presents two gait phase partitioning algorithms that use motion data from a single inertial measurement unit (IMU) placed on the foot instep. For these data, sagittal angular velocity and linear acceleration signals were extracted from nine healthy subjects and nine pathological subjects. Pressure patterns from force sensitive resistors (FSR) instrumented on a custom insole were used as reference values. The performance of a threshold-based (TB) algorithm and a hidden Markov model (HMM) based algorithm, trained by means of subject-specific and standardized parameters approaches, were compared during treadmill walking tasks in terms of timing errors and the goodness index. The findings indicate that HMM outperforms TB for this hardware configuration. In addition, the HMM-based classifier trained by an intra-subject approach showed excellent reliability for the evaluation of mean time, i.e., its intra-class correlation coefficient (ICC) was greater than 0.75 . In conclusion, the HMM-based method proposed here can be implemented for gait phase recognition, such as to evaluate gait variability in patients and to control robotic orthoses for lower-limb rehabilitation.

    Beschreibung: Traditional compensation methods based on temperature-related parameters are not effective for complex total reflection prism laser gyro (TRPLG) bias variation. Because the high frequency oscillator voltage (UHFO) fundamentally affects the TRPLG bias, and the UHFO has a stronger correlation with the TRPLG bias when compared with the temperature, an introduction of UHFO into the TRPLG bias compensation can be evaluated. In consideration of the limitations of least squares (LS) regression and multivariate stepwise regression, we proposed a compensation method for TRPLG bias based on iterative re-weighted least squares support vector machine (IR-LSSVM) and compared with LS regression, stepwise regression, and LSSVM algorithm in large temperature cycling experiments. When temperature, slope of temperature variation, and UHFO were selected as inputs, the IR-LSSVM based on myriad weight function improved the TRPLG bias stability by 61.19% to reach the maximum and eliminated TRPLG bias drift. In addition, the UHFO proved to be the most important parameter in the process of TRPLG bias compensation; accordingly, it can alleviate the shortcomings of traditional compensation based on temperature-related parameters and can greatly improve the TRPLG bias stability.

    Beschreibung: A convolutional neural network (CNN) algorithm was developed to retrieve the land surface temperature (LST) from Advanced Microwave Scanning Radiometer 2 (AMSR2) data in China. Reference data were selected using the Moderate Resolution Imaging Spectroradiometer (MODIS) LST product to overcome the problem related to the need for synchronous ground observation data. The AMSR2 brightness temperature (TB) data and MODIS surface temperature data were randomly divided into training and test datasets, and a CNN was constructed to simulate passive microwave radiation transmission to invert the surface temperature. The twelve V/H channel combinations (7.3, 10.65, 18.7, 23.8, 36.5, 89 GHz) resulted in the most stable and accurate CNN retrieval model. Vertical polarizations performed better than horizontal polarizations; however, because CNNs rely heavily on large amounts of data, the combination of vertical and horizontal polarizations performed better than a single polarization. The retrievals in different regions indicated that the CNN accuracy was highest over large bare land areas. A comparison of the retrieval results with ground measurement data from meteorological stations yielded R2 = 0.987, RMSE = 2.69 K, and an average relative error of 2.57 K, which indicated that the accuracy of the CNN LST retrieval algorithm was high and the retrieval results can be applied to long-term LST sequence analysis in China.

    Beschreibung: This paper investigates mobile relaying in wireless powered communication networks (WPCN), where an unmanned aerial vehicle (UAV) is employed to help information delivery from multiple sources to destination with communication channels severely blocked. The sources are low-power without energy supply. To support information transmission, the UAV acts as a hybrid access point (AP) to provide wireless power transfer (WPT) and information reception for sources. We set the issue of system throughput maximization as the optimization problem. On the one hand, the system is subject to the information causality constraint due to the dependent processes of information reception and transmission for the UAV. On the other hand, the sources are constrained by a so-called neutrality constraints due to the dependent processes of energy harvesting and energy consumption. In addition, we take account of the access delay issue of all ground nodes. Specifically, two paradigms of delay-tolerant case and delay-sensitive case are presented. However, the formulated problem including optimizations for time slot scheduling, power allocation and UAV trajectory is non-convex and thus is difficult to obtain its optimal solution. To tackle this problem, we apply the successive convex approximation (SCA) technique and propose an iterative algorithm by which a suboptimal solution can be achieved. Simulation results validate our proposed design, and show that the obtained suboptimal solution is high-quality, as compared to benchmark scheme.
