    Description: Oncogenes are mutated and/or overexpressed at high levels in tumor cells. Tumors of the lung, breast, pancreas, and colon may display specific oncogenetic features. These tumors have been largely associated with exposure to environmental carcinogens and a variety of biological agents, including viruses. These carcinogens can induce specific genetic and epigenetic alterations in these tissues, leading to aberrant functioning of oncogenes and tumor suppressor genes. On the microRNAs (miRNAs) there are significant modifiers of both transcription and translation of oncogenes in carcinogenesis. Half of melanomas have an activating mutation in the BRAF gene, prompting development of novel drugs, vemurafenib and dabrafenib, specifically targeting mutated BRAF. Trametinib and cobimetinib, which block MEK, a BRAF effector protein, have been used in combination with BRAF inhibitors. A promising new melanoma treatment is immunotherapy, approach that boosts patient's own immune system to attack cancer. Pembrolizumab and nivolumab inhibit PD-1, whereas Ipilimumab targets CTLA-4, another immunity check point, to boost the immune response. Here we focus on pathways, mechanisms, targets and treatments of human skin cancers, with particular emphasis on the new developments in the research on melanomas.

    Description: This book deals with the rapid progress in the area of myelodysplastic syndromes (MDS). MDS are a group of age-associated heterogeneous malignant bone marrow stem cell disorders. MDS are characterized by ineffective hematopoiesis, which leads to refractory cytopenias and to clonal instability. Patients with MDS have myeloid dysplasia, intramedullary apoptosis and an increased risk of transformation to acute myeloid leukemia (AML). The use of next generation sequencing has allowed for the identification of molecular mutations in several genes in about 90% of MDS patients. Several mutations will likely be incorporated into future prognostic scoring systems for MDS. About 50% of MDS cases are characterized by the presence of cytogenetic abnormalities. The correct morphological and cytogenetic analysis interpretation plays an important role in diagnosis and prognosis of these disorders. Cell death and an inflammatory gene signature are associated with MDS. Better understanding of the genetic and molecular mechanisms of MDS pathogenesis provides an opportunity for new treatment strategies to be developed. Promising novel therapies targeting pathophysiological mechanisms of MDS are being studied but the drugs currently used in MDS therapy remain limited. The only curative therapy for MDS is allogeneic hematopoietic stem cell transplantation. Recent advances in strategies to minimize transplant-related toxicity make this treatment possible for more MDS patients who are sufficiently fit.

Two challenges were selected for the RoCKIn competitions due to their high relevance and impact on Europe's societal and industrial needs: domestic service robots (RoCKIn@Home) and innovative robot applications in industry (RoCKIn@Work). Along the book chapters the reader will find details on RoCKIn@Home and RoCKIn@Work, and about the activities carried out during the project lifetime, namely the developed open domain test beds for competitions targeting the two challenges and usable by researchers worldwide; the scoring and benchmarking methods to assess the performance of robot systems and subsystems; and the building up of a community of new teams. The book ends with an assessment by the project industrial partner about the impact of RoCKIn and other robot competitions on the industrial robot markets. Here we discuss the epigenetic changes in DNA and chromatin, which are increasingly associated with melanoma. In the meantime, robotics research has been expanding from fix based robots to mobile robots at a stunning pace. There have been significant milestones that are worth noting in recent decades. Examples are the octopus-like Tentacle Arm developed by Marvin Minsky in 1968, the Stanford Cart crossing a chair-filled room without human assistance in 1979, and most recently, humanoid robots developed by Honda. Despite rapid technological developments and extensive research efforts in mobility, perception, navigation and control, mobile robots still fare badly in comparison with human abilities. For example, in physical interactions with subjects and objects in an operational environment, a human being can easily relies on his/her intuitively force-based servoing to accomplish contact tasks, handling and processing materials and interacting with people safely and precisely. The intuitiveness, learning ability and contextual knowledge, which are natural part of human instincts, are hard to come by for robots. The above observations simply highlight the monumental works and challenges ahead when researchers aspire to turn mobile robots to greater benefits to humankinds. This book is by no means to address all the issues associated mobile robots, but reports current states of some challenging research projects in mobile robotics ranging from land, humanoid, underwater, aerial robots, to rehabilitation.
