    Description: Oncogenes are mutated and/or overexpressed at high levels in tumor cells. Tumors of the lung, breast, pancreas, and colon may display specific oncogenetic features. These tumors have been largely associated with exposure to environmental carcinogens and a variety of biological agents, including viruses. These carcinogens can induce specific genetic and epigenetic alterations in these tissues, leading to aberrant functioning of oncogenes and tumor suppressor genes. On the microRNAs (miRNAs) there are significant modifiers of both transcription and translation of oncogenes in carcinogenesis. In the last 50 years, several oncogenes and microRNAs related to these oncogenes have been identified in different types of human cancers. It is now clear that high expression of oncogenes, DNA damage response, and regulation of the cell cycle are related to the circadian clock. This book will mainly focus on the expressions of different oncogenes in breast, colon, and lung cancers. Moreover, readers will gain qualified scientific knowledge of the alterations in miRNAs in different types of cancers and the effects of the circadian clock on the expression of oncogenes in carcinogenesis.

    Description: Human skin cancers, the most common type of tumors, represent a significant health burden. The deadliest is unquestionably melanoma. Half of melanomas have an activating mutation in the BRAF gene, prompting development of novel drugs, vemurafenib and dabrafenib, specifically targeting mutated BRAF. Trametinib and cobimetinib, which block MEK, a BRAF effector protein, have been used in combination with BRAF inhibitors. A promising new melanoma treatment is immunotherapy, approach that boosts patient's own immune system to attack cancer. Pembrolizumab and nivolumab inhibit PD-1, whereas Ipilimumab targets CTLA-4, another immunity check point, to boost the immune response. Here we focus on pathways, mechanisms, targets and treatments of human skin cancers, with particular emphasis on the new developments in the research on melanomas.

    Description: This book deals with the rapid progress in the area of myelodysplastic syndromes (MDS). MDS are a group of age-associated heterogeneous malignant bone marrow stem cell disorders. MDS are characterized by ineffective hematopoiesis, which leads to refractory cytopenias and to clonal instability. Patients with MDS have myeloid dysplasia, intramedullary apoptosis and an increased risk of transformation to acute myeloid leukemia (AML). The use of next generation sequencing has allowed for the identification of molecular mutations in several genes in about 90% of MDS patients. Several mutations will likely be incorporated into future prognostic scoring systems for MDS. About 50% of MDS cases are characterized by the presence of cytogenetic abnormalities. The correct morphological and cytogenetic analysis interpretation plays an important role in diagnosis and prognosis of these disorders. Cell death and an inflammatory gene signature are associated with MDS. Better understanding of the genetic and molecular mechanisms of MDS pathogenesis provides an opportunity for new treatment strategies to be developed. Promising novel therapies targeting pathophysiological mechanisms of MDS are being studied but the drugs currently used in MDS therapy remain limited.     Description: Skin cancers, basal and squamous cell carcinomas, malignant melanomas, and Merkel cell carcinomas, constitute arguably the most common and increasingly prevalent human neoplasms. Here we discuss the epigenetic changes in DNA and chromatin, which are increasingly associated with melanoma. Several chapters focus on the posttranscriptional modification of the proteins at the melanocyte cell surface, their role in tumorigenesis, and their potential as therapeutic targets. Specifically, extracellular modifications of integrins, glycosylation of cell surface proteins, and changes of cadherins are presented. In a very interesting approach, a potential to target the mitochondria of melanoma cells is investigated. In conclusion, this volume presents various aspects of human skin cancers, components of the large worldwide effort to combat and eradicate this growing health concern.

    Description: Reactive oxygen species (ROS) and DNA double-strand breaks can result from mitochondrial defects and external sources, such as ionizing radiation. If not repaired properly, pathogenic mutations are generated. Human diseases resulting from inherited mitochondrial defects manifest in organs that physiologically require a high level of ATP synthesis. These diseases are clinically challenging, but new experimental clinical therapies include gene editing and mitochondrial transplants. Pathogenic ROS-associated cellular damage includes DNA double-strand breaks, and mouse models are now available to study multiple repair pathways.
