    Description: Most of the ancient town of Tindari (NE, Sicily) was settled on a plateau the most surficial layer of which was made of unconsolidated material. Ongoing excavations at the archaeological site at Tindari uncovered a large portion of the decumanus which suffered deformations preliminarily assigned to coseismic effects. An analysis of the local dynamic response through the simulation of strong seismic shaking to the bedrock and modelling of spectral ratios of the bedrock-soft soil was carried out to verify the susceptibility of superficial terrains of the promontory to coseismic deformations. To perform this simulation the finite element method (FEM) was used. Four accelerometric recordings of three earthquakes of medium-high magnitude, recorded on rocky sites, were chosen to simulate the seismic shaking, using a constitutive law for the materials composing the promontory layers both of linear-elastic type and of elastoplastic type. The analysis of the linear-elastic field allowed the definition of the frequencies for which the spectral ratios of the accelerations recorded the highest amplifications; in particular the frequency range 31.5–37.2 Hz can be combined with deformation of the paved floor of the decumanus. The analysis in the elastoplastic field highlighted the zones of promontory more susceptible to suffer plasticization process. The results show that the topmost layer of the decumanus is the most susceptible to suffer plasticization. Therefore, the performed analysis lends greater support to the hypothesis that the deformations were produced by seismic shaking.

    Description: The tectonic deformation of the Lipari-Vulcano complex, one of the most important active volcanic areas of Mediterranean region, is studied here through the analysis of ten years (1996-2006) of GPS data from both 3 permanent and 13 non-permanent stations. This area can be considered crucial for the understanding of the Eurasia-Africa plates interaction in the Mediterranean area, and, in general, this work emphasize a methodological approach, already applied in other areas worldwide (e.g. Shen et al., 1996, El-Fiki and Kato, 1999) where geodetic data and strain parameters maps of critical areas can help to improve our understanding of their geodynamical aspects. In this framework, this study is aimed at providing a kinematic deformation model on the basis of the dense geodetically estimated velocities of the Lipari-Vulcano complex. In particular, the observed deformation pattern can be described by a mix between 1) the main N-S regional compression and 2) a NNE-SSW compression with a small right-lateral strike slip component acting along a tectonic structure N°40W trending located between the two islands. This pattern was inspected through a simplified synthetic model.

    Description: We analyse P-wave traveltimes for the Mediterranean area, using both teleseismic and regional arrivals for shallow earthquakes reported in the Bulletins of the International Seismological Centre. We model delays between pairs of 0.5° × 0.5° cells, obtaining a detailed representation of the P traveltime heterogeneities. Examination of these anomalies shows the clear presence of geographically coherent patterns—consistent with known geological features—due to significant structure in the upper mantle. We present a scheme, based on an empirical heterogeneity correction (EHC) to P-wave traveltimes, to improve earthquake location. This method provides similar benefits to those of a location procedure based on ray tracing in a 3-D model, but it is simpler and computationally more efficient. The definition of the traveltime heterogeneity model, being based on a statistical procedure, bypasses most of the critical points and possible instabilities involved in model inversion. EHC relocation, applied to Mediterranean earthquakes, allows one to predict about 70 per cent of the estimated signal due to heterogeneity and produces epicentral and origin time-shifts of, respectively, 4.22 km and 0.35 s (rms). From a synthetic experiment, in which we use the proposed algorithm to retrieve known source locations, we estimate that the rms improvement achieved by the EHC relocation over a simpler, standard, 1-D location is more than 20 per cent for both epicentral mislocation and origin time-shifts.

    Description: Recent geological studies performed at Etna allow reassessing the stratigraphic frame of the volcano where distinct evolutionary phases are defined. This stratigraphic reconstruction was chronologically constrained on the basis of a limited number of U–Th and K–Ar age determinations whose uncertainty margins are sometimes too wide. For this reason, we successfully adopted at Etna the 40Ar/39Ar technique that allowed obtaining more precise age determinations. The incremental heating technique also gives information on sample homogeneity, and potential problems of trapped argon. Five samples were collected from stratigraphically well-controlled volcanic units in order to chronologically define the transition between the fissure-type volcanism of the Timpe phase to the central volcanism of the Valle del Bove Centers. Isotopic ages with an uncertainty margin of 2–4% have been obtained emphasizing that this transition occurred (130– 126 ka) without significant temporal hiatus.

    Description: The paper describes a case of a natural emission of methane from soil in an urban development area, generating a significant risk for the local population and buildings, due to gas explosiveness and asphyxiation potential. The site is located on the south-western margin of the East-European Platform in eastern Romania, in a hydrocarbon-prone area crossed by the Pericarpathian lineament and regional faults. Molecular composition of gas and stable isotopic analyses of methane (CH4〉90%, δ to the power of 13 C1: -49.4‰, δD1: -173.4‰) indicate a dominant thermogenic origin, with significant amounts of C2-C5 alkanes (~5%), likely migrating through faults from a deep reservoir. Possible candidates are the Saucesti and Secuieni gas fields, located in the same petroleum system. Two surface geochemical surveys, based on closed-chamber flux measurements, were performed to assess the degassing intensity and the extent of the affected area. Methane fluxes from soil reach orders of 10 to the power of 4 mg m to the power of -2 day to the power of -1. Gas seepage mainly occurs in one zone 30 000 m2 wide, and it is likely controlled by channeling along a fault and gas accumulation in permeable sediments and shallow subsoil. The estimated total CH4 emission is about 40 t year to the power of -1 CH4, of which 8–9 t year to the power of -1 are naturally released from soil and 30–35 t year to the power of -1 are emitted from shallow boreholes. These wells have likely channeled the gas accumulated in shallow alluvial sediment but gas flux from soil is still high and mitigation measures are needed to reduce the risk for humans and buildings.

    Description: The Chimaera gas seep, near Antalya (SW Turkey), has been continuously active for thousands of years and it is known to be the source of the first Olympic fire in the Hellenistic period. New and thorough molecular and isotopic analyses including methane (approximately 87% v/v; δ to the power of 13 C1 from -7.9‰ to -12.3‰; δ to the power of 13 D1 from -119‰ to -124‰), light alkanes (C2 + C3 + C4 + C5 = 0.5%; C6+: 0.07%; δ to the power of 13 C2 from -24.2‰ to -26.5‰; δ to the power of 13 C3 from -25.5‰ to -27‰), hydrogen (7.5–11%), carbon dioxide (0.01–0.07%; δ to the power of 13 CCO2: -15‰), helium (approximately 80 ppmv; R/Ra: 0.41) and nitrogen (2–4.9%; δ to the power of 15 N from -2‰ to -2.8‰) converge to indicate that the seep releases a mixture of organic thermogenic gas, related to mature type III kerogen occurring in Palaeozoic and Mesozoic organic-rich sedimentary rocks, and abiogenic gas produced by low-temperature serpentinization in the Tekirova ophiolitic unit. Methane is not related to mantle or magma degassing. The abiogenic fraction accounts for about half of the total gas released, which is estimated to be well beyond 50 ton year to the power of -1. Ophiolites and limestones are in contact along a tectonic dislocation leading to gas mixing and migration to the Earth’s surface. Chimaera represents the biggest emission of abiogenic methane on land discovered so far. Deep and pressurized gas accumulations are necessary to sustain the Chimaera gas flow for thousands of years and are likely to have been charged by an active inorganic source.

    Description: In eastern Elba Island (Tuscany, Italy), a shallow crustal level felsic, tourmaline-bearing, dyke-sill swarm of Late Miocene age is associated with abundant tourmaline-quartz hydrothermal veins and metasomatic masses. Development of these veins and masses in the host rocks demonstrates multiple hydro-fracturing by magmatic, boron-rich saline fluid. Tourmalines in felsic dykes are schorl, whereas in veins and metasomatic masses, tourmaline composition ranges from schorl-dravite through dravite to uvite. This compositional shift is evidence for an increasing contribution to the magmatic boron-rich fluids by a Mg-Ca-Ti-rich external component represented by biotite-rich and amphibolite host rocks. This system can be envisaged as an exposed proxy of the high temperature hydrothermal system presently active in the deepest part of the Larderello-Travale geothermal field (Tuscany).

    Notes: This article broadly positions the successful establishment of the field of French tropical geography in the immediate postwar period against developments stemming from a longer history of French colonial engagement in Africa, Asia and South America, and clarifies the seemingly late timing of, and paradoxes involved in, the creation of a body of French scientific knowledge about the tropics. Colonial scientific research did not develop in France until the end of the nineteenth century. However, the colonial geography appearing at this time did not rely on fieldwork but, rather, catered to the demands of the business class for overseas expansion and to public curiosity. Even while the medical geography of tropical areas and knowledge of tropical soils and ecology progressed greatly between 1900 and 1940, there were still only a few French geographers working in the tropics. With the advent of the Second World War, when “big science” appeared in France and its colonial empire, the number of French geographers involved in tropical research grew rapidly. The field of tropical geography built up by Pierre Gourou was a synthesis of approaches developed in South America, Africa and Indochina. Although it soon came under strong criticism for its pessimistic view of prospects for industrialisation and urbanisation in the tropics, it seduced French geographers because it matched the contemporary interest in zonality and relied on a genre de vie analysis of, typically, rural areas. Thus, the postwar blossoming of tropical geography shaped by Gourou was more a response to various internal dynamics within French geography than an exercise in imperialism. Its demise was not due to the eclipse of French colonialism but, rather, its inability to deal with the modernisation of tropical societies.

    Notes: This paper discusses four key phases in the study of the tropical milieu of monsoon Asia by French geographers and colonial actors over the last 150 years. First, it sees how the natural milieu initially occupied a central and determining place in French colonial and scientific assessments of Indochina, and how the notion of monsoon Asia appeared within the framework of an academic geography (and formatively in the two regional theses produced by Charles Robequain and Pierre Gourou). It then shows how, after the Second World War, francophone geographers developed a “tropicalist” approach to monsoon Asia, one that was inextricably linked with the culturalist approach that was pioneered by Gourou, which later evolved into new, model-building and interdisciplinary approaches to the study of tropical systems. It argues that these successive phases have marked a shift from a centred tropicality - a system of knowledge anchored in French colonialism and centring on debate about the determinist influence of the natural milieu - to a decentred tropicality that rejects environmental determinism, questions the ethnocentric character of tropical geography and integrates the study of tropical geography into the wider social sciences. The paper concludes with the suggestion that tropicality has had an epistemologically stronger and more institutionalised relationship with francophone geography than was the case in anglophone geography.

    Notes: In important respects, the disciplinary field of “tropical geography” is a uniquely French field of study, and Pierre Gourou is conventionally seen as its founder and doyen. Yet Gourou did not see himself as the creator of a new paradigm or research school, and geographers were generally more influenced by his writings than by his teaching or any personal connection. With particular reference to French geographical research in and on tropical Africa during the second half of the twentieth century, it is suggested that the development of tropical geography as a subfield - and tropicalism as a research orientation - can be put down to a variety of factors and circumstances. Geographical research on Africa was pivotal, as was the rise to prominence in French research institutes of some of Gourou's disciples. But African academics also played a part, as did criticism of tropical geography for its marginalisation of issues of development and geopolitics. The paper examines this postwar intellectual history and attempts to draw from it a positive and forward looking legacy - a reinvigorated and interdisciplinary “tropicalism”, the main axis of which would be the analysis of the specific characteristics of tropical ecology, and its use and transformation by the societies that live from it. Such a project may help us to confront the contemporary world ecological crisis, and forge critical research projects on globalisation (altermondialisme) that can discern and deal with the complex local-global, and rural-urban, articulations of this phenomenon.

    Notes: This article discusses the relevance of the work of Pierre Gourou for Vietnamese and non-Vietnamese scholarship on colonial and postcolonial rural society in Vietnam. The term “tropicality” is used to situate Gourou's work within the framework of both French and Vietnamese regimes of truth. It is argued that Gourou was aware of the complex human geographies of the tropics and monsoon Asia and the challenges this posed to both Western and “tropical” peoples. Gourou and the issue of tropicality is used to show that Vietnamese scholars did not completely reject French colonial systems of knowledge, and that decolonisation did not herald a complete shift in knowledge about rural Vietnam. Rather, since the 1940s there has been antagonism and accommodation between colonial and postcolonial, French and Vietnamese modes of knowledge production. While Gourou underscored the otherness of the tropics, and there are colonial overtones in his work, he had an immense influence on indigenous ethnography and geography in Vietnam and elsewhere in the formerly colonised world. The article traces this important influence and how it has been both questioned and affirmed since independence in the Vietnamese context. It is suggested that the humanistic approach that Gourou pioneered in his 1936 study of the Red River Delta of northern Vietnam has outlived and been able to overcome the setbacks and drawbacks of both colonial and revolutionary/Vietnamese politico-intellectual projects.

    Notes: The paper examines how land and forest management policies were elaborated in French Indochina circa 1900–40. It places their development in the context of a scientific and economic discourse about the value of land and forest resources, the most appropriate ways in which they might be exploited and the relationship between colonial science and indigenous knowledge. By focusing on debates and laws relating to the development of small-scale and plantation farming systems (Land Code legislations) and forest management and exploitation (Forest Code legislations) the paper seeks to ground arguments about Western conceptions of the “tropics” within a discussion of national policy development and impacts. Focusing primarily on Cochinchina and Annam (southern and central Vietnam) and drawing on materials from French archives, the paper shows how changes in both attitudes and legislation have had lasting consequences on systems of property rights in forest management and on the place and status of indigenous peoples in Indochina.

    Notes: Despite recent interest in sacred (fetish) groves as remnant forests, few studies have investigated their sustainability and conservation role in West Africa. This article employs a Geographical Information Systems (GIS) analysis of time series images (1960-98), comprehensive social surveys and ecological field methods to evaluate four sacred groves and eight unprotected tree stands in the coastal savanna of Ghana and compare these with vegetation in the distant forested hinterland. There were strong similarities and substantial differences in tree species between different sacred groves, and between these and the unprotected stands and proximate deciduous forests. In addition, far fewer tree losses were documented in the sacred groves than in the local unprotected stands. Although these sacred groves were on average only partially representative of deciduous forest vegetation, their stronger sustainability compared with unprotected tree stands may be important to consider in detail for conservation.

    Notes: This paper aims to examine how seafarers from the Pacific Republic of Kiribati cope with the experience of working with crews of different nationalities, and, further, how the exposure to different cultures during their journeys through international waters influences both their own identity as well as their perceptions of I-Kiribati culture. Based on examples from open and semi-structured interviews with seafarers working on German merchant ships and Japanese fishing vessels, the paper questions the application of concepts of “hybridity” in the case of these I-Kiribati men in favour of the idea of “cultural flexibility”. It further considers to what degree seafarers strongly rooted in the clearly confined cultural values of Kiribati have adapted the values received through their training and employment by German or Japanese and Korean companies and officers.     Notes: This paper focuses on the large-scale land use and land cover changes that have taken place in Sarawak state, East Malaysia over the three decades of 1972-2002. Results are presented from a detailed land use and cover change (LUCC) study in the Niah River catchment using satellite imagery, questionnaire surveys and interviews. Successive waves of land cover changes have taken place. Large forest areas have been logged and gradually replaced by oil palm plantations, which now occupy more than 40 per cent of the total land area in the catchment. Concurrently, small-scale farming systems have also changed. Formerly dominant Iban shifting cultivation practices are increasingly being replaced by cash crop production on permanent fields and impacted by off-farm activities involving many ethnic groups. It is argued that land cover changes are continuous and complex processes involving a large number of variables which can be analysed for different time periods at various scales.

    Notes: This paper adopts ideas from critical geopolitics to explore how Western human rights discourses have been enabled and used to construct Myanmar by Australian governments and nongovernmental aid organisations. Three distinct human rights scripts are observed that position donors as either punishers, saviours or partners. It is argued that the scripts are enabled by factors external to the actual abuses themselves, including the traditional scripting of Burma within Australian geopolitical memories; the feminine Burmese identity and the networks embodied by the opposition leader, Daw Suu Kyi; and Australia's shifting geopolitical interests in the Southeast Asia region. The case study is used to argue for a broader geography of human rights that recognises their spatiality. Such geographies can improve and broaden human rights politics and debates beyond state-based campaigning, ensuring that campaigns are in the interests of the most marginalised, rather than the most vocal.

    Notes: It has been posited that small-scale industry – businesses with less than 30 workers – provides a large share of employment and income in Ghana. This paper examines the proposition that while such enterprises in the informal sector are said to act as a sponge to soak up surplus labour in marginal activities, they are unprofitable. Using data from a survey in 1998 of 175 micro and small-scale enterprises in the Central Region of Ghana, the paper also confirms problematic aspects of employment in this sector including the lack of formal contracts, irregular pay, low remuneration, non-existent social protection and only marginal employment growth. The implication is that the small-scale industry sector is not economically sustainable in its present form in Ghana.

    Notes: Advocacy of the advantages accruing to business clusters in developing economies has followed that in developed economies. With doubts emerging about the evidence for these in developed economies, it is therefore appropriate to review the experience in the developing world. A model of cluster emergence and growth has guided accounts of developing country clusters. Drawing on experience from Indonesia, doubts are raised about that model's prediction of the emergence of successful joint action in maintaining cluster advantages. Any advantages from clustering are insufficient to face development challenges arising from the globalisation of economic activity. The significance of business clusters in low income economies needs to be reviewed in the light of actual experience and the reappraisal emerging in developed or high income countries.

    Notes: The EPIC (Erosion Productivity Impact Calculator) crop model, developed by scientists of the United States Department of Agriculture (USDA), has been successfully applied to the study of erosion, water pollution, crop growth and production in the US but is yet to be introduced for serious research purposes in other countries or regions. This paper reports on the applicability of the EPIC 8120 crop model for the assessment of the potential impacts of climate variability and climate change on crop productivity in sub-Saharan West Africa, using Nigeria as the case study. Among the crops whose productivity has been successfully simulated with this model are five of West Africa's staple food crops: maize, millet, sorghum (guinea corn), rice and cassava. Thus, using the model, the sensitivities of maize, sorghum and millet to seasonal rainfall were demonstrated with coefficients of correlation significant at over 98 per cent confidence limits. The validation tests were based on a comparison of the observed and the model-generated yields of rice and maize. The main problems of validation relate to the multiplicity of crop varieties with contrasting performances under similar field conditions. There are also the difficulties in representing micro-environments in the model. Thus, some gaps appear between the observed and the simulated yields, arising from data or model deficiencies, or both. Based on the results of the sensitivity and validation tests, the EPIC crop model could be satisfactorily employed in assessing the impacts of and adaptations to climate variability and climate change. Its use for the estimation of production and the assessment of vulnerabilities need to be pursued with further field surveys and field experimentation.

    Notes: Data collected on rainfall events covering one wet season in Ilorin, Kwara State, Nigeria, were found to be highly correlated with the rate of erosion on all the representative land surface types found in the city – namely bare land, fallow land, short grass and tall tree cover. Of all the rainfall components, rainfall total was best at predicting the rate of soil erosion on all these surfaces. In addition, the study found that a significant constituent of all surfaces, silt which fell through seiver grades 1.18 mm did not cohere, therefore increasing the rate of erosion. However, when grass cover (regenerating with the rains) was at least 95 per cent, erosion was well controlled. Therefore, grass coverage before the onset of the wet season is recommended as an efficient and simple measure to manage erosion in this typical developing urban centre in Nigeria.

    Notes: Rural-urban classification constitutes an important framework for the collection and compilation of population data in many countries. While “urban” is often specifically defined, “rural” is treated simply as a residual category. The criteria defining urban also differ from country to country. This paper argues that these rural and urban statistical categories are also highly significant for local governance, increasingly so in recent years given the emphasis on local governance and its restructuring. In India, constitutional amendments have given constitutional status to local bodies in the federal structure of the country. Local bodies are thus now expected to draw up their own plans and initiate development works, which requires them to generate their own resources and lessen their dependence on central government funding. It is thus necessary to reorganize urban space into viable spatial units in terms of their revenue base. While rural-urban classification is the task of the Census of India, state governments are responsible for granting municipal status to urban centres. This paper examines the criteria and limitations of the rural-urban classification followed by the Census, its congruence with the dynamics of state-accorded municipal/non-municipal status and some implications for municipal governance in India.

    Notes: This paper explores the activities and problems of indigenous women's organisations in Ghana. The survey data that form the basis of the analysis were collected in 1998 from a sample of 66 women's organisations in the districts of Atebubu and Techiman of Brong Ahafo Region in Ghana. The results suggest that despite the debilitating impacts of patriarchy, poverty and illiteracy, the women in both districts have managed to use their grassroots organisations to improve the socioeconomic circumstances of their members in particular and their communities in general. Given the dearth of theoretical and empirical analyses on grassroots mobilisation of women in sub-Saharan Africa, the results presented here are intended to stimulate further research and discussion on the subject.

    Notes: Using a general statistical model, this study attempts to characterise the trend of deforestation in the northeast region (Isan) of Thailand between 1975 and 1991, a period when the kingdom had sustained high rates of economic growth and steady increases in population. Using data obtained directly from government bodies on the 17 provinces comprising this heavily deforested region, the study examines the correlations between forest area and a set of six variables: population density, agricultural area, real per capita income, accumulated irrigated area, agricultural credit levels, and distance from Bangkok, the national political and economic centre. It also considers the effect of the two logging bans instituted in 1979 and since 1989. The study found a negative correlation between forest area and population density in particular, followed in ranking by agricultural credit, per capita income, the logging bans and distance from Bangkok. Viewed together with more recent data showing that rates of deforestation in the kingdom as a whole have slowed and appear to be stabilising, these results also suggest the beginnings in the 1990s of a forest transition – from an industrial to a post-industrial stage in forest utilisation – in Thailand.

    Notes: Applied psychologists have long been interested in the relationship between applicant personality and employment interview ratings. Analysis of data from two studies, one using a situational interview and one using a behavioral interview, suggests that the correlations of structured interview ratings with self-report measures of personality factors are generally rather low. Further, a small meta-analysis integrates these two studies and the limited previous literature to arrive at a similar conclusion – there is relatively little relationship between structured interviews and self-reported personality factors.

    Notes: An international foundation for the promotion of basic research in biomedicine, the Boehringer Ingelheim Fonds (B.I.F.), reports that fellowships are awarded to post-graduate researchers according to the following main criteria: (1) scientific quality as demonstrated by the applicant's achievements to date, (2) the originality of the proposed research project, and (3) the scientific standing of the laboratory where the research will be conducted. Using the Boolean probit statistical technique, this study examines the multiple conjunctural causation that a fellowship will be awarded only if all three of these criteria are assessed positively by the B.I.F. peer review committee. In agreement with the prescriptive principles of the foundation the results suggest that the B.I.F. approves applications only if all of the three criteria are rated positively.

    Notes: The French system of continuing vocational training was set up in 1970 by an intersectoral collective agreement that was then followed by legislation. More than 30 years on, the economic and social context has changed and the system now faces new challenges. In 2001 and again in 2003 the social partners embarked upon new negotiations in a bid to redesign the system. Analysis of the negotiation process and of its outcomes  reveals  both  the  persistence  of  societal  characteristics  and  the  emergence  of significant innovations, particularly in the individualisation of training. This dual trend is explained by the characteristics of the actors involved as well as by the weakness of the state.

    Notes: Although recruitment researchers often discuss influences on decisions to apply for jobs, few studies assess actual application behavior. This study of individuals who expressed an interest in a firefighter job revealed that applying was related to recruiting source informativeness and organizational familiarity. Source informativeness was not related to self-selection out of the process after application or performance on the selection process, but was related to perceptions of familiarity and to applicant demographics. Implications for recruitment are discussed.

    Notes: Standardized tests are commonly used to select individuals in both pre-employment and educational settings. Nevertheless, research has yet to sufficiently explore the relationship between test anxiety (TA) and test performance in selection contexts. The goal of this study was to assess the dimensionality of TA, the relations between TA and test performance, and the impact of gender on TA within the realm of personnel selection. Test anxiety was found to be bi-dimensional and negatively related to performance on a cognitively based selection instrument. Gender moderated the relation between TA and test performance, with stronger associations for males. This finding is consistent with the sex-linked anxiety coping theory that is advanced here. Implications of this study and avenues for future research are discussed.

    Notes: Recruiters infer personal traits from job applicants' resumés and use these inferences in evaluating job applicants' employability. No research to date, however, has determined if resumé reviewers' inferences of applicants' personality drawn solely from resumé biographical data are valid. In the present study, resumé reviewers (N=52) examined one of two applicant resumés and then described the applicant's personality based on the Big 5 taxonomy. Validities of reviewers' inferences concerning applicant personality were assessed by correlating resumé reviewers' judgments with applicants' self-reported Big 5 personality scores. Results suggested that valid personality inferences are possible based solely on resumé evaluation. We also found evidence suggesting that attending a brief training session may enhance reviewers' accuracy when inferring applicants' personality from resumé information.

    Notes: In response to Schmidt, Le, and Ilies' (2003) call for further assessment of transient error variance in measures of key individual difference variables, the current study examines transient error estimates for eight speeded cognitive ability scales and a composite scale. One hundred twenty-three participants were tested on two occasions with a 1-week interval between administrations. Estimates of transient error ranged from a low of .01 to a high of .18, with an average value of .09. The coefficient of equivalence, based on the split-half method, resulted in overestimates of reliability by as little as 1.90% to as much as 25.96%, with an average 12.96%.

    Notes: This article reports on the union recognition dispute that took place at the McDonald's food-processing plant in Moscow. It examines this dispute in the context of McDonald's employment practices worldwide, the interventions made by international and local unions, and Russian government bodies. Despite these interventions it became impossible to either organise the workforce or establish a collective agreement. The case illustrates the difficulties facing both local unions and global union federations when confronted by intransigent multinational companies, especially in low-skilled sectors in transitional economies.

    Notes: This paper is concerned with exploring the barriers to cross-border trade union cooperation among graphical workers in Europe. Highlighting the increasing globalisation of graphical multinationals, the paper illustrates that graphical unions cannot restrict their activities to developments within national boundaries. As such it highlights some of the obstacles that unions face in attempting to instigate transnational cooperation strategies. These obstacles include: the role of employers, the prevailing practices of the unions, employee reticence, the role of the law and resource constraints. It concludes by stressing the continuing need to strengthen cross-border trade union networks with the ability to impose effective sanctions against the unbridled growth of multinational power.

    Notes: This paper identifies the constraints and opportunities facing trade unions in post-socialist societies through a comparison of the Russian and Chinese cases. The paper charts the changing role of trade unions in the integration of state-socialist economies into global capitalism and argues that this presents the trade unions with a dilemma: whether to reconstitute their traditional role through collaboration with management and the state apparatus, or whether to develop their ability to defend the rights and interests of workers. While the former offers the line of least resistance, there are progressive elements pushing for change in both trade union movements.

    Notes: A situational judgment test (SJT) and a Big 5 personality test were administered to 203 participants under instructions to respond honestly and to fake good using a within-subjects design. Participants indicated both the best and worst response (i.e., Knowledge) and the most likely and least likely response (i.e., Behavioral Tendency) to each situation. Faking effect size for the SJT Behavioral Tendency response format was (d=.34) when participants responded first under honest instructions and (d=.15) when they responded first under faking instructions. Those for the Big 5 dimensions ranged from d=.26 to d=1.0. For the Knowledge response format results were inconsistent. Honest condition Knowledge SJT scores were more highly correlated with cognitive ability (r=.56) than were Behavioral Tendency SJT scores (r=.38). Implications for researchers and practitioners are discussed.

    Notes: In this study, we investigated the moderating role of interdependence (a personality trait that measures the extent to which individuals desire working in a group-based, cooperative setting) on the relationship between competitiveness and one-year objective sales performance. On the basis of data from 133 sales representatives, results indicated that: (a) competitiveness was related to objective sales performance; and (b) interdependence moderated the relationship between competitiveness and objective sales performance such that competitiveness predicted objective sales performance more strongly for those who scored low in interdependence versus those who scored high in interdependence. Implications for sales selection and reward systems and directions for future research are considered.

    Notes: Although there has been empirical attention paid to the criterion-related validity of predictor composites, there has been much less attention paid to the standardized ethnic group differences associated with these composites. One important area of inquiry in predictor composite research is the influence of adding predictors to a test of general mental ability. The limited empirical literature on this practice is mixed, but the prevailing expectation is that there is likely to be higher validity and less adverse impact. Unfortunately, much of the previous work is limited by the presence of inaccurate validity and standardized ethnic group difference values. In this analysis we formed meta-analytic matrices to more accurately estimate the validity and standardized ethnic group differences of several composites that combine a measure of cognitive ability with measures of conscientiousness, a structured interview, or biodata. While results were somewhat complex, we found that adding alternative predictors does not result in a situation in which validity automatically goes up and adverse impact potential automatically goes down. In fact, the reductions in adverse impact (if any) from adding “non-cognitive” predictors were more modest than much of the literature suggests.

    Notes: Type IV secretion systems are employed by a wide variety of Gram-negative microorganisms for the translocation of macromolecules across the cell envelope. The translocated substrates (proteins, protein–DNA complexes and DNA) are as diverse as the organisms on the donor and recipient side of the translocation process. Over the course of evolution, these macromolecular transporters were adapted to many different purposes, but their basic mechanism was conserved. They impact human life in various ways, as there are driving forces of horizontal gene transfer, which spreads biodegradative capabilities of environmental bacteria as well as antibiotic resistance of pathogens in hospitals. Also, they translocate toxins and other effectors, which have an effect on host cell metabolism and are essential for the virulence of bacterial pathogens. We here present recent developments of research on the mechanism of type IV secretion focusing on the energetization of transport and assembly processes, formation of the translocation channel and of surface-exposed pili, which initiate host cell interactions.

    Notes: Mitochondrial dysfunction has been shown to elicit broad effects on nuclear gene expression. We show here that transcription dependent on the prototypical acidic activator Gal4 is responsive to mitochondrial dysfunction. In cells with no mitochondrial DNA, Gal4-dependent gene expression is elevated. A minimal Gal4 activator containing the DNA binding and activation domain is sufficient for this response. Transcription dependent on a fusion of Gal4 to a heterologous DNA binding domain is similarly elevated in a mitochondrial mutant. Analysis of different Gal4-dependent promoters and gel mobility shift assays suggest that the effect of mitochondrial dysfunction on Gal4 activity is related to increased DNA binding to the cognate Gal4 element. Given that fermentation is the only means to obtain energy in respiratory deficient cells, it is possible that higher Gal4 activity in cells with dysfunctional mitochondria works to promote more efficient fermentation of galactose.

    Notes: In an enterohemorrhagic Escherichia coli (EHEC) O157:H7 outbreak caused by salted salmon roe that occurred in Japan, 1998, a food isolate (F2) was NaCl-resistant and a patient isolate (P5) was sensitive to NaCl. We show here that hydrogen peroxide, like NaCl, induced a significant loss of culturability in P5. The BacLight assay suggested that the EHEC O157:H7 entered a viable but nonculturable (VNC) state. We used the passage through mice in an attempt to model this transition in phenotype. Mouse-passaged isogenic variants of F2 became NaCl- and oxidation-sensitive, entered the nonculturable state in response to either of these stresses, and could be resuscitated by sodium pyruvate. Since the expression of RpoS in response to these stresses correlated with the isolates' culturabilities, we concluded that in vivo passage negatively modulated RpoS expression, and the subsequent stress exposure induced the VNC state in the EHEC O157:H7 isolates.

    Notes: In the present study a procedure combining a cell extraction method and Fluorescence In Situ Hybridization (FISH) for molecular monitoring and quantification of bacteria in soil and aquifer samples is presented. FISH was applied to bacterial cells extracted from the matrix by density gradient centrifugation. This separation method was applied to soil and aquifer samples and produced high cell recovery of 76.5%± 4.4 and 78.0%± 3.2, respectively. FISH, performed on the harvested cells, permitted a perfect visualization and quantification of bacteria. This approach is therefore promising for in situ detection of indigenous bacterial communities in complex samples.

    Notes: A screening for h ydrogen up take (hup) genes in Rhizobium leguminosarum bv. viciae isolates from different locations within Spain identified no Hup+ strains, confirming the scarcity of the Hup trait in R. leguminosarum. However, five new Hup+ strains were isolated from Ni-rich soils from Italy and Germany. The hup gene variability was studied in these strains and in six available strains isolated from North America. Sequence analysis of three regions within the hup cluster showed an unusually high conservation among strains, with only 0.5–0.6% polymorphic sites, suggesting that R. leguminosarum acquired hup genes de novo in a very recent event.

    Notes: Recent molecular approaches for the study of microbial communities such as PCR-cloning have enabled the detection and identification of as-yet-unculturable taxa. Cloning and sequencing of multiple samples is extremely laborious and expensive to perform thoroughly due to the large diversity involved. For this purpose, techniques such as denaturing gradient gel electrophoresis (DGGE) may be better suited. There is increasing evidence suggesting that DGGE of complex polymicrobial communities may be limited by co-migration of different sequences. In this study, we attempt to address this limitation by excising individual bands and running them through a shorter denaturant gradient, a process we have termed “denaturing gradient gel electrophoresis gel expansion” (DGGEGE).

    Notes: As an initial step to understand the mobile nature of class II mercury resistance transposon TnMERI1, the effect of the recA gene on translocation of mini-TnMERI1 was evaluated. A higher transposition frequency in the LE392 strain (2.4 ± 1.2 × 10−5) than in the recA-deficient DH1 strain (1.2 ± 0.8 × 10−6) indicated participation of the recA gene in mini-TnMERI1 transposition. Introduction of the recA gene into the DH1 strain complemented the transposition frequency at the same level as in LE392 and confirmed participation of the recA gene in transposition. However, treatment of cells by stress agents, including irradiation of up to 3000 J m−2 UV doses, did not alter the transposition frequency and suggested independence of RecA from the SOS stress response. Further analysis of transconjugants indicated participation of RecA in the resolution of the cointegrate structure of the transposon. These results suggested that RecA is a constitutive cellular factor that increases translocation of mini-TnMERI1 and may participate in dissemination of TnMERI1-like transposons.

    Notes: The chromosome of Bacillus subtilis codes for seven extracytoplasmic function sigma factors the activity of which is modulated normally by a cognate anti-sigma factor. While inducing factors and genes for four of them (σM, σW, σX, and σY) have been identified, those of the remaining three sigma factors including σV remain elusive. The objective of the present study was the unequivocal identification of its anti-sigma factor and of genes controlled by σV. In many cases reported so far the gene coding for the anti-sigma factor is located immediately downstream of the gene coding for the sigma factor, and both form a bicistronic operon. We could show by two different experimental approaches that this is also the case for sigV and rsiV. Under conditions of overproduction of σV, 13 genes could be identified being induced several-fold by the DNA macroarray technique. Induction of three of them was confirmed by Northern blots, and the potential promoter of sigV was identified by primer extension. This led to the deduction of a consensus sequence recognized by σV.

    Notes: Noroviruses are positive strand RNA viruses that have received increased attention in recent years because their role as etiologic agents in acute gastroenteritis outbreaks is now clearly established. Much has been learned about the epidemiology of these viruses and the extent of genetic diversity among circulating strains. In contrast, progress on understanding the basic mechanisms of virus replication has been far slower due to the inability to cultivate virus in the laboratory. Despite this limitation, significant progress has been made in defining some basic functions of the norovirus proteins, and the structures of two have been solved to near atomic resolution. This minireview summarizes these recent advances in understanding the structure and function of the norovirus proteins and provides speculation about what roles they may play in the virus replication cycle.

    Notes: The contribution of target gene mutations and active efflux to varying levels of quinolone resistance in Irish Campylobacter isolates was studied. The Thr-86-Ile modification of GyrA did not correlate with the level of quinolone resistance. The efflux pump inhibitor Phe-Arg-β-Naphthylamide (PAβN) had no effect on the MICs to ciprofloxacin. In contrast, a PAβN sensitive efflux system contributed to the low-level nalixidic acid resistance phenotype. The lack of effect of PAβN in high-level resistant nalidixic isolates may be attributable to mutations identified in the CmeB efflux pump of these isolates. PAβN may have limited diagnostic value in the assessment of the contribution of efflux pump activity to ciprofloxacin resistance in Campylobacter.

    Notes: We have constructed a dam mutant of Yersinia pestis GB. In BALB/c mice inoculated subcutaneously, the median lethal dose of the mutant was at least 2000-fold higher than the wild type. Mice inoculated with sub-lethal doses of the mutant were protected against a subsequent challenge with virulent Y. pestis. The effect of dam inactivation on gene expression was examined using a DNA microarray, which revealed increased expression of a number of genes associated with the SOS response. These results confirm the key role of Dam in the regulation of virulence, and its potential role as a target for the generation of attenuated strains of pathogenic bacteria.

    Notes: Physiologically significant levels of intracellular coenzyme A were identified in Pyrococcus furiosus, Thermococcus litoralis, and Sulfolobus solfataricus, suggesting a role for CoA as an important low molecular mass thiol in the thermophilic Archaea. In P. furiosus, cells grown in the presence of sulfur showed significantly higher levels of oxidized CoA compared with those grown in the absence of S0. T. litoralis showed strikingly similar CoA levels, although with low disulfide levels in both the presence and absence of S0. S. solfataricus showed similarly high levels of CoA thiol, with correspondingly low levels of the CoA disulfide. These results are consistent with the identification of a coenzyme A disulfide reductase (CoADR) in P. furiosus and horikoshii as well as the presence of CoADR homologues in the genomes of S. solfataricus and T. kodakaraensis.

    Notes: An inactive (R)-3-hydroxyacyl-acyl carrier protein:coenzyme A transacylase (PhaGPm) was cloned from a newly isolated Proteobacteria Pseudomonas mendocina LZ. It is the first characterized native inactive PhaG protein. Sequence analysis indicated that there were only two sites where the amino acid sequence differed between this inactive protein and the functional PhaGPp from P. putida. The differences were located at position 78 and in the region 109–113 in the amino acid sequence. Mutagenesis was carried out to investigate these two sites. A recombinant strain harboring a S78C PhaGPp mutant accumulated polyhydroxyalkanoates (PHA) at 11.9% of the cellular dry weight, as compared to the 21.6% PHA produced by the recombinant harboring the wild-type PhaGPp. On the other hand, the changes in the amino acid region 109–113 of PhaGPp to its corresponding region of PhaGPm resulted in negligible PHA accumulation. This demonstrated that region 109–113 in PhaG is relatively important for transacylase activity, while position 78 just plays a supporting role for the enzyme. Furthermore, 3-D structural models of PhaGPp and PhaGPm developed by computational prediction revealed that the variation in amino acids at 109–113 leads to the destruction of the PhaG catalytic center, resulting in the loss of enzyme activity.

In the present study, we used two-dimensional gel electrophoresis, mass-spectrometry and microarray analyses to identify the listerial genes that depend on DegU for expression. We found that the two L. monocytogenes operons encoding flagella-specific genes and the monocistronically transcribed flaA gene are positively regulated by DegU at 24 °C, but are not expressed at 37 °C.

    Notes: Blastocrithidia culicis and Crithidia deanei are trypanosomatid protozoa of insects that normally contain intracellular symbiotic bacteria. The protozoa can be rid of their endosymbionts by antibiotics, producing a cured cell line. Here, we analyzed the glycoconjugate profiles of endosymbiont-harboring and cured strains of B. culicis and C. deanei by Western blotting and flow cytometry analyses using lectins that recognize specifically sialic acid and mannose-like residues. The absence of the endosymbiont increased the intensity of the lectins binding on both trypanosomatids. In addition, wild and cured strain-specific glycoconjugate bands were identified. The role of the surface saccharide residues on the interaction with explanted guts from Aedes aegypti gut was assessed. The aposymbiotic strains of B. culicis and C. deanei presented interaction rates 3.3- and 2.3-fold lower with the insect gut, respectively, when compared with the endosymbiont-bearing strains. The interaction rate of sialidase-treated cells of the wild and cured strains of B. culicis and C. deanei was reduced in at least 90% in relation to the control. The interaction of B. culicis (wild strain) with explanted guts was inhibited in the presence of mucin (56%), fetuin (62%), sialyllactose (64%) and α-methyl-d-mannoside (80%), while in C. deanei (wild strain) the inhibition was 53%, 56%, 79% and 34%, respectively. Collectively, our results suggest a possible involvement of sialomolecules and mannose-rich glycoconjugates in the interaction between insect trypanosomatids and the invertebrate host.

    Notes: Legionella pneumophila is a pathogenic bacterium found in freshwater environments that is responsible for pneumonia. People become infected through inhalation of contaminated droplets from water devices, such as cooling towers and showers. It is important to find new treatments that decrease the development of Legionella. We found a Staphylococcus warneri strain that inhibits Legionella growth. This activity is due to a molecule secreted by S. warneri. This molecule displayed a high heat-stability and its activity was lost after protease treatments, suggesting that it might be a bacteriocin. Its purification led us to conclude that this anti-Legionella molecule is an highly hydrophobic peptide. It has an original and very specific spectrum of activity, directed only toward the Legionella genus. This is the first description of an antibacterial peptide active against Legionella.

    Notes: The antibiotic susceptibility of wild Listeria monocytogenes strains and their corresponding nisin resistant variants was assessed. The resistant strains were more sensitive to most of the tested antibiotics than their wild-type counterparts. A slight increase in MIC was observed for a few antibiotics including the membrane disturbing polymixin B. Cross-resistance was detected with two synthetic antimicrobial peptides. A lower C15/C17 ratio in the membrane fatty acid composition of the nisin resistant strains was found, and one strain pair showed a significant difference in surface hydrophobicity. As judged by these results, no clear correlation could be established between resistance to nisin and to worldwide-used antibiotics.

An E. coli–L. hongkongensis inducible expression shuttle vector was further constructed by ligating a 2105-bp fragment that contains the tetracycline repressor and tetracycline-inducible promoter region of pALC2084 to the 8897-bp fragment of pPW380, deletion of the green fluorescent protein gene, and insertion of a multiple cloning site. This inducible expression system was able to express two commonly used reporter genes, the green fluorescent protein gene and the glutathione S-transferase gene, efficiently in E. coli and L. hongkongensis.

    Notes: A moderately halotolerant, Gram-positive, aerobic, motile, spore-forming bacterium, designated as strain YAS1, was isolated from an olive-brine fermentation rich in aromatic compounds, after enrichment on tyrosol. Strain YAS1 grew between 25 and 45 °C and optimally at 37 °C. It grew in the presence of 0–15% (v/w) NaCl, with an optimum of 3–6% (v/w) NaCl. The DNA G + C content was found to be 49.9 mol%. Phylogenetic analysis of the 16S rRNA gene revealed that this isolate was a member of the genus Bacillus. The newly isolated strain YAS1 represents the first moderately halotolerant bacterium transforming tyrosol to p-hydroxyphenylacetic acid (PHPA) in the presence of yeast extract.

    Notes: Sixteen isolates of Bacillus thuringiensis, derived from various soil samples collected in Australia, are highly toxic to larvae of the sheep blowfly (Lucilia cuprina). The toxin gene from one of the strains (CAA890) was cloned by genome walking, and sequencing of the cloned fragments revealed a new cry gene, encoding a protein of 1134 amino acid residues, with a theoretical molecular mass of 139,209 Da. Based on the amino acid sequence comparison with known Cry δ-endotoxins, the gene was designated cry47Aa. Homology modelling based on known crystal structures of the Cry toxins reveals the differences to be located in the loops of domain II in the putative toxin-receptor binding surfaces between Cry47Aa and the dipteran active Cry2Aa. We also showed that the cry47Aa gene is present in the other isolates that are highly toxic to the sheep blowfly.

    Notes: Condensation (C) domains in the nonribosomal peptide synthetases are capable of catalyzing peptide bond formation between two consecutively bound various amino acids. C-domains coincide in frequency with the number of peptide bonds in the product peptide. In this study, a phylogenetic approach was used to investigate structural diversity of bacterial C-domains. Phylogenetic trees show that the C-domains are clustered into three functional groups according to the types of substrate donor molecules. They are l-peptidyl donors, d-peptidyl donors, and N-acyl donors. The fact that C-domain structure is not subject to optical configuration of amino acid acceptor molecules supports an idea that the conversion from l to d-form of incorporating amino acid acceptor occurs during or after peptide bond formation. l-peptidyl donors and d-peptidyl donors are suggested to separate before separating the lineage of Gram-positive and Gram-negative bacteria in the evolution process.

    Notes: HypR has recently been described as the first transcriptional regulator involved in the oxidative stress response and in the intracellular survival of Enterococcus faecalis within macrophages. In order to characterize the HypR regulon, real-time quantitative RT-PCR experiments were performed. The expression of four genes involved in the oxidative stress response encoding catalase, glutathione reductase, and the two subunits of alkyl hydroperoxide reductase were down regulated in the hypR background under H2O2 condition. These findings show that HypR acts as a transcriptional activator, especially during oxidative stress. In addition, DNAse I footprinting assays allowed us to identify the HypR-protected DNA regions corresponding to the “HypR box” in the hypR promoter. Moreover, the effect of the hypR mutation on the virulence of E. faecalis was evaluated in comparison with the wild-type JH2–2 strain using a mouse peritonitis model. Our results revealed that HypR appears to be an important virulence factor in E. faecalis.

    Notes: Burkholderia pseudomallei and B. mallei are the causative agents of melioidosis and glanders, respectively. As iron regulation of gene expression is common in bacteria, in the present studies, we have used microarray analysis to examine the effects of growth in different iron concentrations on the regulation of gene expression in B. pseudomallei and B. mallei. Gene expression profiles for these two bacterial species were similar under high and low iron growth conditions irrespective of growth phase. Growth in low iron led to reduced expression of genes encoding most respiratory metabolic systems and proteins of putative function, such as NADH-dehydrogenases, cytochrome oxidases, and ATP-synthases. In contrast, genes encoding siderophore-mediated iron transport, heme-hemin receptors, and a variety of metabolic enzymes for alternative metabolism were induced under low iron conditions. The overall gene expression profiles suggest that B. pseudomallei and B. mallei are able to adapt to the iron-restricted conditions in the host environment by up-regulating an iron-acquisition system and by using alternative metabolic pathways for energy production. The observations relative to the induction of specific metabolic enzymes during bacterial growth under low iron conditions warrants further experimentation.

    Notes: In the biosynthesis of type B trichothecenes, four oxygenation steps remain to have genes functionally assigned to them. On the basis of the complete genome sequence of Fusarium graminearum, expression patterns of all oxygenase genes were investigated in Fusarium asiaticum (F. graminearum lineage 6). As a result, we identified five cytochrome P450 monooxygenase (CYP) genes that are specifically expressed under trichothecene-producing conditions and are unique to the toxin-producing strains. The entire coding regions of four of these genes were identified in F. asiaticum. When expressed in Saccharomyces cerevisiae, none of the oxygenases were able to transform trichodiene-11-one to expected products. However, one of the oxygenases catalyzed the 2β-hydroxylation rather than the expected 2α-hydroxylation. Targeted disruption of the five CYP genes did not alter the trichothecene profiles of F. asiaticum. The results are discussed in relation to the presence of as-yet-unidentified oxygenation genes that are necessary for the biosynthesis of trichothecenes.

    Notes: The ArcA/ArcB two-component signal transduction system of Escherichia coli regulates gene expression in response to the redox conditions of growth. In this study, uvrA gene expression was repressed when ArcA was induced in E. coli. Transcription of uvrA increased in ΔarcA and ΔarcB strains more than in the wild-type strain, whose trend was remarkable under the anaerobic condition. In the wild-type strain grown in the presence of DTT (10 mM), the uvrA gene expression was also repressed. Furthermore, the results of in vitro transcription and DNase I footprinting experiments indicated that ArcA specifically bound to the ArcA box [(A/T)GTTAATTA(A/T)] in the uvrA promoter and represses its transcription. These results suggest that the ArcA/ArcB two-component system works to negatively regulate uvrA gene expression.

    Notes: Lactobacillus salivarius subsp. salivarius UCC118 is a probiotic bacterium that was originally isolated from human intestinal tissues and was subsequently shown in a pilot study to alleviate symptoms associated with mild-moderate Crohn's disease. Strain UCC118 can adhere to animal and human intestinal tissue, and to both healthy and inflamed ulcerative colitis mucosa, irrespective of location in the gut. In this study, an enzymatic technique has been combined with proteomic analysis to correlate bacterial growth phase with the presence of factors present in the cell wall of the bacterium. Using PAGE electrophoresis, it was determined that progression from lag to log to stationary growth phases in vitro correlated with increasing prominence of an 84 kD protein associated with in vitro adherence ability. Isolated proteins from the 84 kD band region were further separated by two-dimensional electrophoresis, resolving this band into 20 individual protein spots at differing isoelectric points. The protein moieties were excised, trypsin digested and subjected to tandem mass spectrometry. The observed proteins are analogous to those reported to be associated with the Listeria monocytogenes cell-wall proteome, and include DnaK, Ef-Ts and pyruvate kinase. These data suggest that at least some of the beneficial attributes of probiotic lactobacilli, and in particular this strain, may be due to nonpathogenic mimicry of pathogens and potentially be mediated through a form of attenuated virulence.

    Notes: Sclerotinia sclerotiorum is unusual among necrotrophic pathogens in its requirement for senescent tissues to establish an infection and to complete the life cycle. A model for the infection process has emerged whereby the pathogenic phase is bounded by saprophytic phases; the distinction being that the dead tissues in the latter are generated by the actions of the pathogen. Initial colonization of dead tissue provides nutrients for pathogen establishment and resources to infect healthy plant tissue. The early pathogenicity stage involves production of oxalic acid and the expression of cell wall degrading enzymes, such as specific isoforms of polygalacturonase (SSPG1) and protease (ASPS), at the expanding edge of the lesion. Such activities release small molecules (oligo-galacturonides and peptides) that serve to induce the expression of a second wave of degradative enzymes that collectively bring about the total dissolution of the plant tissue. Oxalic acid and other metabolites and enzymes suppress host defences during the pathogenic phase, while other components initiate host cell death responses leading to the formation of necrotic tissue. The pathogenic phase is followed by a second saprophytic phase, the transition to which is effected by declining cAMP levels as glucose becomes available and further hydrolytic enzyme synthesis is repressed. Low cAMP levels and an acidic environment generated by the secretion of oxalic acid promote sclerotial development and completion of the life cycle. This review brings together histological, biochemical and molecular information gathered over the past several decades to develop this tri-phasic model for infection. In several instances, studies with Botrytis species are drawn upon for supplemental and supportive evidence for this model. In this process, we attempt to outline how the interplay between glucose levels, cAMP and ambient pH serves to coordinate the transition between these phases and dictate the biochemical and developmental events that define them.

    Notes: The cytolethal distending toxin B (CdtB) of the mouse pathogen Helicobacter hepaticus has cation binding and DNA catalysis residues in common with members of the mammalian deoxyribonuclease I (DNase I) family. The purpose of the present study was to characterize CdtB nuclease. To establish optimal digestion conditions and to evaluate co-factor requirements, a novel and sensitive fluorometric assay that quantitatively determines double stranded DNA digestion was developed. Although the Ca2+- and Mg2+-dependence and neutral properties of CdtB were similar to DNase I, hydrolysis of DNA by CdtB was approximately 100-fold less active than DNase I and was considerably more resistant to inhibition by ZnCl2 and G-actin.

    Notes: Nutrition influenced growth, sporulation and virulence of the insect pathogenic fungus, Metarhizium anisopliae. Virulent conidia were produced on susceptible insect hosts, 1% yeast extract, 2% peptone, osmotic stress medium (OSM) and CN 10:1 medium. Several strain independent markers were identified that could be used to predict the virulence of M. anisopliae conidia. Virulent conidia typically had high levels of spore bound Pr1, an important cuticle degrading protease, and high germination rates. We also show for the first time that virulent conidia have an endogenous CN ratio below 5.2:1. Real Time PCR revealed that virulent conidia from insects contained significantly higher levels of transcripts of pr1 A and other pathogenicity-related genes than inoculum from artificial media. Of the artificial media studied, 1% yeast extract medium yielded the most virulent conidia, these had higher levels of transcripts of these pathogenicity-related genes than the least virulent conidia from the high conidia yielding CN 35:1 medium (= SDA), however, the levels were significantly lower than those in insect-derived conidia. Our study shows for the first time that the passaged inoculum is virulent irrespective of the original culture medium or insect host. Virulent conidia were consistently produced on OSM even though growth and sporulation were poor. We postulate that starvation conditions, whether in vivo or in vitro, results in de-repression of Pr1 and that elevated levels of this enzyme enhance fungal virulence.

    Notes: In ageing, carbon-depleted cultures of Aspergillus nidulans strain FGSC 26 progressing apoptotic-type cell death was detected, characterised by increasing numbers of Annexin V and TUNEL stained cells after protoplastation. DAPI staining of autolysing mycelia revealed numerous nuclei with elongated, stick-like morphology, which was not observed in surviving hyphal fragments representing a cell population adapted to carbon starvation. Apoptotic cell death was also progressing in aging cultures of the non-autolysing loss-of-function fluG and ΔbrlA mutants, indicating that apoptotic cell death and autolysis were regulated independently. In accordance with this, sphingosine derivatives added to A. nidulans cultures increased cell death rates without influencing autolytic biomass losses and hydrolase production.

    Notes: PMR1, the Ca2+/Mn2+ ATPase of the secretory pathway in Saccharomyces cerevisiae was the first member of the secretory pathway Ca2+ ATPases (SPCA) to be characterized. In the past few years, pmr1Δ yeast have received more attention due to the recognition that the human homologue of this protein, hSPCA1 is defective in chronic benign pemphigus or Hailey–Hailey disease (HHD). Recent publications have described pmr1Δ S. cerevisiae as a useful model organism for studying the molecular pathology of HHD. Some observations indicated that the high Ca2+ sensitive phenotype of PMR1 defective yeast strains may be the most relevant in this respect. Here we show that the total cellular calcium response of a pmr1Δ S. cerevisiae upon extracellular Ca2+ challenge is decreased compared to the wild type strain similarly as observed in keratinocytes. Additionally, the novel magnesium sensitivity of PMR1 defective yeast is revealed, which appears to be a result of competition for uptake between Ca2+ and Mg2+ at the plasma membrane level. Our findings indicate that extracellular Ca2+ and Mg2+ competitively influence the intracellular Ca2+ homeostasis of S. cerevisiae. These observations may further our understanding of HHD.

    Notes: Biofilm formation is an important step in the etiology of periodontal diseases. In this study, in vitro biofilm formation by Treponema denticola and Porphyromonas gingivalis 381 displayed synergistic effects. Confocal microscopy demonstrated that P. gingivalis attaches to the substratum first as a primary colonizer followed by coaggregation with T. denticola to form a mixed biofilm. The T. denticola flagella mutant as well as the cytoplasmic filament mutant were shown to be essential for biofilm formation as well as coaggregation with P. gingivalis. The major fimbriae and Arg-gingipain B of P. gingivalis also play important roles in biofilm formation with T. denticola.
