    Description: Stable isotopic values on planktonic foraminifera in a suite of cores from basins across the SE Baffin Shelf are used to extract a record of meltwater events during Termination I deglaciation. Resolution and Hatton basins lie on the SE Baffin Shelf at water depths 〉 500 m, seaward of major conduits for ice drainage from the eastern sector of the Laurentide Ice Sheet (LIS). Accelerator mass spectrometry 14C dates are used to constrain our chronology of events in ten cores. In Resolution Basin, three cores have 14C AMS dates on foraminifera of 〉 20 ka at their bases; whereas Hatton Basin cores terminate in sediments 〈 13 kyr. Sedimentation rates varied between 0.1 to 4.5 m/ka. Stable oxygen and carbon isotopic ratios were obtained on 146 samples of the planktonic foraminifera Neogloboquadrina pachyderma (Ehrenberg) sinistral, from seven of the ten cores. No evidence was found to indicate that test morphology or size affected ∂18O. Between 7 and 13.5 ka the surface water on the shelf was on average 1 ‰ lower than the open ocean signal. Significant temporal variations were found in both ∂18O and ∂13C. Evidence for significant low ∂18O events occurred between 13 and 8 ka. The ∂13C record from the planktonic foraminifera suggests a threefold division of events between 13 and 7 ka, with positive values between 10.8 and 13.0 ka, negative values between 9 and 10.8 ka, and positive values from 7 to 9 ka. The ∂18O data suggest the presence of meltwater on the shelf some 3,000 years prior to the first late glacial dates on terrestrial deglaciation (at circa 10.4 ka). “Hudson Strait must be the real key to the importance of the calving process during deglaciation, because it is potentially the largest marine outlet for the Laurentide Ice Sheet and because it leads into the very center of the ice sheet.....the rates of calving through Hudson Strait during the period of initial ∂18O rise unfortunately are unknown.”

    Description: Insects with complex life-cycles should optimize age and size at maturity during larval development. When inhabiting seasonal environments, organisms have limited reproductive periods and face fundamental decisions: individuals that reach maturity late in season have to either reproduce at a small size or increase their growth rates. Increasing growth rates is costly in insects because of higher juvenile mortality, decreased adult survival or increased susceptibility to parasitism by bacteria and viruses via compromised immune function. Environmental changes such as seasonality can also alter the quantitative genetic architecture. Here, we explore the quantitative genetics of life history and immunity traits under two experimentally induced seasonal environments in the cricket Gryllus bimaculatus. Seasonality affected the life history but not the immune phenotypes. Individuals under decreasing day length developed slower and grew to a bigger size. We found ample additive genetic variance and heritability for components of immunity (haemocyte densities, proPhenoloxidase activity, resistance against Serratia marcescens), and for the life history traits, age and size at maturity. Despite genetic covariance among traits, the structure of G was inconsistent with genetically based trade-off between life history and immune traits (for example, a strong positive genetic correlation between growth rate and haemocyte density was estimated). However, conditional evolvabilities support the idea that genetic covariance structure limits the capacity of individual traits to evolve independently. We found no evidence for G × E interactions arising from the experimentally induced seasonality.

    Description: In the North Atlantic we define H-0 as a Heinrich-like event which occurred during the Younger Dryas chron. On the SE Baffin shelf prior to 11 ka, surface water productivity was reasonably high, as measured by the numbers of diatom and planktic foraminifera per gram, but an abrupt increase in detrital carbonate (DC-0 event) (from approximately 15% up to 50% carbonate by weight) occurred at 11 ± 14C ka and continued to circa 10 ka. These deposits, 2–6 m thick, are dominated by detrital calcite and silt- and clay-sized sediments. During this event (DC-0/H-0), ice extended onto the inner shelf but did not reach the shelf break and probably originated from a center over Labrador-Ungava. As a consequence, the pattern of ice-rafted debris and sediment provenance shown by H-O in the North Atlantic is different from that during H-1 (14.5 ka) or H-2 (20 ka) when the ice sheet extended along the axis of Hudson Strait and may have reached the shelf break; for example, there is no concrete evidence for DC-O is cores on the floor of the Labrador Sea due east of Hudson Strait (HU75-55,-56), but H-O has been noted in cores off Newfoundland and west of Ireland. A coeval carbonate event to DC-0, but this one dominated by dolomite, occurs in HU82-SU5 on the west side of Davis Strait with a source either from northern Baffin Bay or Cumberland Sound. Although other sources for North Atlantic detrital carbonate cannot be totally excluded, our evidence suggests that H-0 represents the expression of glaciological instability of the Laurentide Ice Sheet within the general region of Hudson Strait and probably to the north (Cumberland Sound and northernmost Baffin Bay). There is one younger DC event, dated circa 8.4 ka, present in sediments along the Labrador margin and in Hudson Strait, which represents the final collapse of the ice sheet within Hudson Strait and Hudson Bay.

    Description: During the discovery and description of seven New Zealand methane seep sites, an infaunal assemblage dominated by ampharetid polychaetes was found in association with high seabed methane emission. This ampharetid-bed assemblage had a mean density of 57,000 ± 7800 macrofaunal individuals m−2 and a maximum wet biomass of 274 g m−2, both being among the greatest recorded from deep-sea methane seeps. We investigated these questions: Does the species assemblage present within these ampharetid beds form a distinct seep community on the New Zealand margin? and What type of chemoautotrophic microbes fuel this heterotrophic community? Unlike the other macro-infaunal assemblages, the ampharetid-bed assemblage composition was homogeneous, independent of location. Based on a mixing model of species-specific mass and isotopic composition, combined with published respiration measurements, we estimated that this community consumes 29–90 mmol C m−2 d−1 of methane-fueled biomass; this is 〉 290 times the carbon fixed by anaerobic methane oxidizers in these ampharetid beds. A fatty acid biomarker approach supported the finding that this community, unlike those previously known, consumes primarily aerobic methanotrophic bacteria. Due to the novel microbial fueling and high methane flux rates, New Zealand's ampharetid beds provide a model system to study the influence of metazoan grazing on microbially mediated biogeochemical cycles, including those that involve greenhouse gas emissions

    Description: To investigate diel calcium carbonate (CaCO3) dynamics in permeable coral reef sands, we measured pore-water profiles and fluxes of oxygen (O2), nutrients, pH, calcium (Ca2+), and alkalinity (TA) across the sediment-water interface in sands of different permeability at Heron Reef, Australia. Background flushing rates were high, most likely as a result of infaunal burrow irrigation, but flux chamber stirring enhanced pore-water exchange. Light and pore-water advection fueled high rates of benthic primary production and calcification in sunlit surface sediments. In the light, benthic photosynthesis and calcification induced surface minima in Ca2+ and TA and peaks in pH and O2. Oxygen penetration depth in coarse sands decreased from ∼ 1.2 cm during the day to ∼ 0.6 cm at night. Total oxygen uptake (TOU) in dark chambers was three to fourteen times greater than diffusive uptake and showed a direct effect of pore-water advection. Greater sediment oxygen consumption rates were observed in higher permeability sands. In the dark, TA release was not stimulated by increasing TOU because of a damping effect of pore-water advection on metabolic CaCO3 dissolution efficiency. On a daily basis, CaCO3 undergoes net dissolution in Heron Reef sands. However, pore-water advection can reverse the CaCO3 budget and promote CaCO3 preservation under the most energetic conditions.

    Description: To test the hypothesis that calcium carbonate (rather than opal) carries most organic carbon to the deep sea, total hydrolysable amino acid (THAA) analysis was applied to deep-sea (3000 m) sediment trap material from the northeast Atlantic (PAP Site), a variable but intrinsically carbonate-dominated system. THAAs were analyzed in conjunction with total organic carbon, biogenic silica, calcium carbonate, and inferred lithogenic fluxes. The THAA-based degradation state of organic carbon could not be systematically explained by changes in the flux of different mineral phases, which could account for only 16% of the observed variability. In addition amino acid parameters indicative of source organisms indicate that diatom cell walls are an important residual component of organic carbon reaching the deep ocean, a finding supported by comparison with data from previous studies of diverse oceanic environments. Finally, during 2001, very high organic carbon fluxes were associated with elevated lithogenic fluxes and low organic matter degradation relative to surrounding years. In accordance with other recent experimental and observational studies, the data indicate that under specific export scenarios, lithogenic fluxes can act as highly significant mediators of organic carbon transfer to the deep ocean.

    Description: Iron limitation often results in increased cellular silica contents of diatoms, suggesting that diatoms grow thicker and possibly mechanically stronger frustules when limited. We performed stability measurements for six diatom species grown under iron-limitation and iron-sufficient conditions. Frustule strength increased in all species when grown under iron limitation, with this effect being statistically significant for four of them. Valve morphology and silica content of the pennate Fragilariopsis kerguelensis and the centric Coscinodiscus wailesii changed under iron limitation but only valve morphology changes were significant; F. kerguelensis grew thicker costae while C. wailesii had smaller pores, especially in the outer part of the valves. These morphological changes are clearly in agreement with increased mechanical strength. Increased cellular silica concentrations in diatoms grown under iron limitation do result in increased frustule strength, most likely improving their protection against grazers.

    Description: Seiche-induced turbulence and the vertical distribution of dissolved oxygen above and within the sediment were analyzed to evaluate the sediment oxygen uptake rate (JO2), diffusive boundary layer thickness (δDBL), and sediment oxic zone depth (zmax) in situ. High temporal-resolution microprofiles across the sediment-water interface and current velocity data within the bottom boundary layer in a medium-sized mesotrophic lake were obtained during a 12-h field study. We resolved the dynamic forcing of a full 8-h seiche cycle and evaluated JO2 from both sides of the sediment-water interface. Turbulence (characterized by the energy dissipation rate, ε), the vertical distribution of dissolved oxygen across the sediment-water interface (characterized by δDBL and zmax), JO2, and the sediment oxygen consumption rate (RO2) are all strongly correlated in our freshwater system. Seiche-induced turbulence shifted from relatively active (ε = 1.2 × 10-8 W kg-1) to inactive (ε = 7.8 × 10-12 W kg-1). In response to this dynamic forcing, δDBL increased from 1.0 mm to the point of becoming undefined, zmax decreased from 2.2 to 0.3 mm as oxygen was depleted from the sediment, and JO2 decreased from 7.0 to 1.1 mmol m-2 d-1 over a time span of hours. JO2 and oxygen consumption were found to be almost equivalent (within ~ 5% and thus close to steady state), with RO2 adjusting rapidly to changes in JO2. Our results reveal the transient nature of sediment oxygen uptake and the importance of accurately characterizing turbulence when estimating JO2.

    Description: Benthic fluxes of dissolved ferrous iron (Fe2+) and phosphate (TPO4) were quantified by in situ benthic chamber incubations and pore-water profiles along a depth transect (11°S, 80–1000 m) across the Peruvian oxygen minimum zone (OMZ). Bottom-water O2 levels were 〈 2 µmol L-1 down to 500-m water depth, and increased to ~40 µmol L-1 at 1000 m. Fe2+ fluxes were highest on the shallow shelf (maximum 316 mmol m-2 yr-1), moderate (15.4 mmol m-2 yr-1) between 250 m and 600 m, and negligible at deeper stations. In the persistent OMZ core, continuous reduction of Fe oxyhydroxides results in depletion of sedimentary Fe :Al ratios. TPO4 fluxes were high (maximum 292 mmol m-2 yr-1) throughout the shelf and the OMZ core in association with high organic carbon degradation rates. Ratios between organic carbon degradation and TPO4 flux indicate excess release of P over C when compared to Redfield stoichiometry. Most likely, this is caused by preferential P release from organic matter, dissolution of fish debris, and/or P release from microbial mat communities, while Fe oxyhydroxides can only be inferred as a major P source on the shallow shelf. The benthic fluxes presented here are among the highest reported from similar, oxygen-depleted environments and highlight the importance of sediments underlying anoxic water bodies as nutrient sources to the ocean. The shelf is particularly important as the periodic passage of coastal trapped waves and associated bottom-water oxygenation events can be expected to induce a transient biogeochemical environment with highly variable release of Fe2+ and TPO4.

    Description: Cell-based therapies, such as adoptive immunotherapy and stem-cell therapy, have received considerable attention as novel therapeutics in oncological research and clinical practice. The development of effective therapeutic strategies using tumor-targeted cells requires the ability to determine in vivo the location, distribution, and long-term viability of the therapeutic cell populations as well as their biological fate with respect to cell activation and differentiation. In conjunction with various noninvasive imaging modalities, cell-labeling methods, such as exogenous labeling or transfection with a reporter gene, allow visualization of labeled cells in vivo in real time, as well as monitoring and quantifying cell accumulation and function. Such cell-tracking methods also have an important role in basic cancer research, where they serve to elucidate novel biological mechanisms. In this Review, we describe the basic principles of cell-tracking methods, explain various approaches to cell tracking, and highlight recent examples for the application of such methods in animals and humans.
